The effects of Rhodiola extract (RE) on longevity and stress resistance in Caenorhabditis elegans (C. elegans), and the underlying molecular mechanisms were explored in the present study. Results showed that the lifespan of C. elegans was remarkably prolonged by 37.1% after treated with high-dose RE (480 μg mL−1). Intervention with RE alleviated aging-related declines in the C. elegans model, and enhanced the stress resistance against heat shock, ultraviolet radiation and paraquat. Moreover, RE reduced the levels of reactive oxygen species (ROS) and malondialdehyde (MDA) and increased the activities of superoxide dismutase (SOD) and catalase (CAT). RE also upregulated the gene expression of sod-3, gst-4, daf-16, skn-1 in C. elegans, downregulated the gene expression of daf-2 and age-1, and accelerated the translocation of DAF-16 and SKN-1 into the nucleus. Furthermore, the daf-16(mu86) and skn-1(zu169) mutants reversed the extension of lifespan triggered by RE, indicating that these genes were involved in RE-regulated longevity. These results demonstrated that RE could enhance lifespan extension, healthspan and stress resistance of C. elegans via insulin/IGF signaling and SKN-1 pathways. Therefore, the present findings suggested Rhodiola as a potential candidate to ameliorate the symptoms of aging.

Aim: Nanobiotechnology has grown rapidly in recent years and constitutes an integral part of modern disease diagnosis and treatment. The goal of this study was to evaluate the protective effect of synthesized selenium nanoparticles (SeNPs) against ionizing radiation (IR)-induced hematopoietic system injury. Methods: An aqueous extract of Rhodiola (RHO) was used for the synthesis of SeNPs (RHO-SeNPs). The resulting nanoparticles were characterized by ultraviolet–visible spectrophotometry and transmission electron microscopy, followed by biological assessment (antioxidant and cytotoxicity). Experiments with BALB/c mice and mouse hematopoietic stem cells (HSCs) exposed to 60Co γ-rays were used to evaluate the therapeutic efficiency of RHO-SeNPs. Immunohistochemistry, ELISA, JC-1 staining, CCK-8, TUNEL, flow cytometry, and Western blot were employed to investigate the underlying mechanisms. Results: RHO-SeNPs showed excellent antioxidant performance and had no toxicity in the major organs of BALB/c mice when administered at ≤20 mg/kg. RHO-SeNPs dose-dependently improved hematopoietic function and survival rate of IR-exposed BALB/c mice, in addition to suppressing mitochondrial oxidative stress, inflammatory responses, and bone marrow cell apoptosis. In vitro studies revealed that RHO-SeNPs limited Kelch-like ECH-associated protein 1 (KEAP1) expression and induced nuclear translocation of nuclear factor erythroid 2-related factor 2 (NRF2), thereby attenuating oxidative stress and HSC apoptosis. Both in vivo and in vitro experiments demonstrated that the disruption of NRF2 nuclear translocation partly abolished the radioprotection of RHO-SeNPs. Conclusion: These findings identify the mechanisms underlying the radioprotection of RHO-SeNPs and their potential role as a medical countermeasure against IR exposure.

Ethnopharmacological relevance We previous study verified that decadienyl-L-carnitine (C10:2) biosynthesis has therapeutic-target capacity for RCE inhibiting pulmonary vascular remodeling to modulate experimental pulmonary hypertension(PH). However, the profound molecular mechanism remains incompletely elucidated. Aim of study This study aim to investigate whether C10:2 biosynthesis regulates pulmonary vascular remodeling by activating cell pyroptosis. Materials and methods Rats and pulmonary artery smooth musle cells (PASMCs) model with PH were successfully induced with monocrataline (MCT) and platelet-derived growth factor-BB (PDGF-BB) in present study. Following RCE treatment, cell targeted metabolomics assay combining biological information analysis and molecular biological methods were used to investigate the modulatory function of C10:2 on pulmonary vascular remodeling by activating cell pyroptosis and accordingly pharmacological mechanism of RCE against cell pyroptosis. Results We found that C10:2 activated NLRP3/Caspase-1/GSDMD signaling pathway to promote PASMCs pyroptosis, and decreased the level of azelaic acid in PASMCs. RCE can significantly upregulate miR-149-5p to targeting bind Cpt1a mRNA, thus, to decrease CPT1A expression, by which low level of C10:2 was maintained to further promote the biosynthesis of anti-inflammatory azelaic acid in vivo, along with this functional molecule is also one main compound of RCE. Our data further demonstrated that the biosynthesis of azelaic acid directly regulates NLRP3/Caspase1/GSDMD axis to inhibit pyroptosis by targeting NLRP3, we therefore argue that azelaic acid is a key functional compound of RCE for treating PH by inhibiting cell pyroptosis. Conclusion Collectively, our work characterized that azelaic acid is a key functional compound in RCE to express pharmacological efficacy against PH, which was observed to inhibit cell pyroptosis to ameliorate pulmonary vascular remodeling in rats through the regulation of decadienyl-L-carnitine/NLRP3/GSDMD axis. Such work will provide essentially molecular basis for developing anti-PH strategy and associated drug discovery.

For the first time, a mixoploid plants and callus cultures of Rhodiola rosea L. were established. Flow cytometry analysis revealed that seed treatment with 1000 µM colchicine for 24 hours induced mixoploidy in 30% of the samples, while the remaining 70% remained diploid. The combination of 5 µM thidiazuron and 5 µM α-naphthaleneacetic acid resulted in a 100% callus induction rate from cotyledonary leaves and supported callus growth up to 50 g/L dry weight. The phenylpropanoids identified in the rhizomes of in vitro plants (control, 2n) were rosavin, rosin, unidentified phenylpropanoids (PP1, PP2, and PP3), and cinnamyl alcohol. In the mixoploid calluses, the same phenylpropanoids were detected, with the exception of PP1. In the diploid calluses, no phenylpropanoids were detectable within the sensitivity limits of the analytical method used. ANOVA confirmed the significant influence of both UV exposure and cinnamyl alcohol concentration on phenylpropanoid accumulation in mixoploid calluses, with levels reaching up to 484 mg/100 g dry weight. Rosin was identified as the major phenylpropanoid in mixoploid calluses under standard conditions. However, after 72 hours of UV irradiation and cinnamyl alcohol supplementation, PP3 became the dominant compound. This study demonstrates that combining polyploidization, biotransformation, and elicitation is a promising alternative strategy for enhancing metabolite production in Rhodiola rosea cell cultures. Moreover, the targeted application of these stress factors can serve as a tool for ‘metabolic engineering’ in its callus culture. This approach may facilitate the sequential scale-up of metabolite biosynthesis in bioreactors to meet industrial demands.

Objective: An integrated approach combining network pharmacology and experimental verification was used to elucidate the effects and mechanisms of the Scutellaria baicalensis (Huangqin [HQ]) and Rhodiola rosea (Hongjingtian [HJT]) herbal pair in treating ischemic stroke (IS). Materials and Methods: A compound–disease network for the HQ-HJT herbal pair and IS was constructed for the network pharmacology analysis. In addition, the efficacy and targets of the HQ-HJT herbal pair were verified using a classical in vitro IS model and molecular docking methods. Results: A total of 82 overlapping targets were identified between the HQ-HJT herbal pair and IS. Kaempferol was the most active compound in the compound–disease network. The protein-protein interaction network analysis showed that Interleukin-6, tumor necrosis factor-α (TNF-α), and HIF-1α were core targets. Kyoto Encyclopedia of Genes and Genomes analysis identified that the HQ-HJT herbal pair exerts therapeutic effects on IS through the TNF signaling and other related pathways. Our experimental analyses confirmed that the key components (kaempferol, salidroside, and baicalein) had neuroprotective effects in the oxygen-glucose deprivation/reoxygenation injury model. Notably, kaempferol suppressed inflammatory responses and showed strong binding affinity to six key targets within the HIF-1α/NLRP3/Caspase-1 signaling axis. Conclusions: HQ-HJT may exert an anti-IS effect by inhibiting the inflammatory cascade. Kaempferol may serve as a key bioactive compound in the HQ-HJT pair.

Ethnopharmacological relevance Rhodiola crenulata (Hook.f. & Thomson) H.Ohba has a long history of clinical application for the prevention and treatment of acute mountain sickness (AMS) in traditional Chinese medicine . However, gaps in knowledge still exist in understanding the underlying mechanisms of Rhodiola crenulata against AMS. Aims To address this problem, a comprehensive method was established by combining UHPLC-Q-TOF-MS/MS analysis and network pharmacology . Materials and methods The ingredients of Rhodiola crenulata were comprehensively analyzed using UHPLC-Q-TOF-MS/MS method. On this basis, a network pharmacology method incorporated target prediction, protein-protein interaction network, gene enrichment analysis and components-targets-pathways network was performed. Finally, the possible mechanisms were verified through molecular docking, in vitro and in vivo experiments. Results A total of 106 constituents of Rhodiola crenulata were charactered via UHPLC-Q-TOF-MS/MS. The 98 potentially active compounds out of 106 were screened and corresponded to 53 anti-AMS targets. Gene enrichment analysis revealed that hypoxia and inflammation related genes may be the central factors for Rhodiola crenulata to modulate AMS. Molecular docking revealed that TNF, VEGFA and HIF-1α had high affinities to Rhodiola crenulata compounds. Subsequently, Rhodiola crenulata extract was indicated to inhibit the protein expression level of TNF in hypoxia induced H9c2 cells. Lastly, Rhodiola crenulata extract was further verified to ameliorate heart injury and decreased the heart levels of TNF, VEGFA and HIF-1α in acute hypoxia-induced rats. Conclusions This study used UHPLC-Q-TOF-MS/MS analysis and a network pharmacology to provide an important reference for revealing the potential mechanism of Rhodiola crenulata in the prevention and treatment of AMS.

Salidroside from Rhodiola rosea L. has hypoglycemic effects, and has anti-osteoporosis effect in diabetic rats. Whether it has beneficial effects on diabetic nephropathy (DN) has not been fully reported. Here, a DN rat model was established by streptozotocin (STZ). Salidroside (10, and 20 mg kg −1 d −1 ) were orally giving for eight weeks. The purity of salidroside was 99%. The results showed that eight weeks salidroside supplementation dose-dependently decreased blood glucose, serum insulin, serum creatinine, blood urea nitrogen (BUN), as well as urinary albumin of DN rats, repaired the pathological changes of kidney tissue, the renal sclerosis and fibrosis in the kidney of DN rats were improved, and the activities of oxidative defense system associated enzyme such as superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPX) were also improved. Serum inflammatory cytokines (including tumor necrosis factor-α, monocyte chemoattractant protein-1, interleukin-1β, and interleukin-6) were decreased significantly in DN rats after eight weeks salidroside supplementation. Immunohistochemical results showed that fibrosis related proteins such as transforming growth factor-β1 (TGF-β1), phospho-Smad2 (p-Smad2), and phospho-Smad3 (p-Smad3) expression levels were decreased significantly in the kidney of salidroside treated group. Our results indicate that daily salidroside consumption could alleviate DN in rats, the main mechanisms are anti-inflammatory, anti-oxidative stress, and inhibition of the TGF-β1/Smad2/3 pathway. This study suggested that salidroside could be used as a nutrient against DN, as well as diabetes.

Rhodiola species are edible medicinal plants, which have been traditionally used in both Asia and Europe as an adaptogen, a tonic, an anti-depressant and anti-inflammatory supplement. However, whether it presents a therapeutic effect on colitis or not remains unknown. The aim of this study is to investigate the protective effect of a Rhodiola crenulata extract (RCE) on mice with DSS-induced colitis. RCE significantly alleviated the pathological abnormalities in colitic mice, including the correspondingly increased colon length, ameliorated colonic injury and reduced pro-inflammatory factors. The protective effect was similar to that of the positive control, 5-aminosalicylic acid. The DSS-induced epithelial apoptosis and maintained intestinal barrier function were attenuated by RCE through the upregulation of the level of tight junction proteins such as ZO-1 and occludin. Notably, RCE prevented gut dysbiosis in colitic mice by restoring the microbial richness and diversity, and decreasing the abundance of Proteobacteria phylum and opportunistic pathogenic Parasutterella and Staphylococcus, as well as increasing the abundance of beneficial microbes in Lactobacillus and Bifidobacterium, which were closely correlated with its protective effect against colitis. Meanwhile, chemical characterization of RCE was performed by UPLC-HR-MS to explain its material basis. A total of 63 compounds were identified, while the content of two bioactive ingredients (salidroside, 1.81%; rosavin, 0.034%) was determined.
