The exact mechanism by which berberine alleviates depression remains unclear. In this study, we explored the relationship between the antidepressant effect of berberine and the microbiota–brain–gut axis. The levels of IL-1β, TNF-α, IL-6, corticosterone, serotonin, and brain-derived neurotrophic factor (BDNF) were quantified using enzyme-linked immunosorbent assays (ELISA) and Western blotting. Variations in the composition of the gut microbiota were examined using 16S rRNA gene sequencing. Berberine significantly mitigated depressive behaviors in mice with CRS, as manifested by increased total distance traveled and central zone duration in the open-field examination, increased time and number of entries into the outstretched arms during the elevated and maze tests, and an increase in the exercise time during the tail suspension and forced swimming tests. Histopathological analysis indicated that berberine ameliorated CRS-induced hippocampal and colonic damage in mice. Additionally, berberine substantially restrained the generation of proinflammatory cytokines and corticosterone in mice with CRS, while increasing the levels of BDNF and serotonin. Importantly, berberine significantly ameliorated CRS-induced depression-like behaviors ( p < 0.01) and restored gut microbial diversity and short-chain fatty acid (SCFA) levels (fold-change: acetate 1.8-fold, butyrate 2.2-fold; p < 0.05). Furthermore, berberine restored the CRS-induced alterations in SCFA production. Our results indicate that berberine may exert antidepressant effects via a pleiotropic mechanism that modulates the microbiome–brain–gut axis.

This study aimed to optimize the structure and efficacy of xylooligosaccharides (XOSs) from corn cobs in constipated mice. Structural analysis revealed that XOSs from corn cobs were composed of β-Xyl-(1 →4)-[β-Xyl-(1→4)]n-α/β-Xyl (n = 0–5) without any other substituents. XOS administration significantly reduced the defecation time, increased the gastrointestinal transit rate, restored the gastrointestinal neurotransmitter imbalance, protected against oxidative stress, and reversed constipation-induced colonic inflammation. Fecal metabolite and microbiota analysis showed that XOS supplementation significantly increased short chain fatty acid (SCFA) levels and improved the gut microbial environment. These findings highlighted the potential of XOSs from corn cobs as an active ingredient for functional foods or as a therapeutic agent in constipation therapy.

Hypercholesteremia or high cholesterol is one of the important factors leading to atherosclerosis and other cardiovascular diseases. The application of probiotics with cholesterol-lowering characteristics has become increasingly popular over the past decade due to their contribution to human health. This study aimed to evaluate the probiotic effects of Lactobacillus fermentum ZJUIDS06 and Lactobacillus plantarum ZY08 on hyperlipidemic golden hamsters. A hyperlipidemic model was established through a high cholesterol diet in golden hamsters, after which lyophilized Lactobacillus fermentum ZJUIDS06 and Lactobacillus plantarum ZY08 were orally administered individually for 8 weeks. The physiological characteristics of golden hamsters and short chain fatty acid (SCFA) in the colon were assessed by automatic Biochemical Analyzer and gas choromatograph, respectively. A MiSeq sequencing-based analysis of the bacterial 16S rRNA gene (V3–V4 region) in the cecum content was performed to analyze the cecum microbiota. Correlations between sets of these variables were also investigated using the R package “corrplot.” Results showed that neither Lactobacillus fermentum ZJUIDS06 nor Lactobacillus plantarum ZY08 inhibited body weight increase. However, supplementation with Lactobacillus fermentum ZJUIDS06 for 8 weeks increased colon SCFA levels (P < 0.05), decreased serum low-density lipoprotein, total cholesterol, and triglycerides levels, and also induced changes in the cecum microbiota of hyperlipidemic golden hamsters. Remarkably, oral administration of Lactobacillus fermentum ZJUIDS06 increased the relative abundance of Parabacteroides in the cecum, which served as a biomarker for colon SCFA production and improvement of serum cholesterol levels. In a word, Lactobacillus fermentum ZJUIDS06 improved hyperlipidemia in golden hamsters, which correlated with an increase in SCFA levels and relative abundance of Parabacteroides, indicating its potential importance in functional foods that can help lower cholesterol.

Simple SummaryAcrylamide (AA), a prevalent foodborne contaminant formed during high-temperature food processing, poses significant health risks; however, effective and practical dietary countermeasures remain scarce. Our study provides compelling evidence that a prebiotic, xylo-oligosaccharides (XOS) derived from corncobs, effectively mitigates AA-induced gut–liver toxicity in mice, characterized by alleviation of colonic and hepatic inflammation, amelioration of intestinal barrier dysfunction, and reduction in hepatic oxidative stress. These protective effects were closely associated with the remodeling of gut microbiota, characterized by a marked enrichment of short-chain fatty acid (SCFA)-producing genera and a concomitant reduction in pro-inflammatory taxa, resulting in significantly elevated SCFA levels. These findings indicate that corncob-derived XOS represents a potential prebiotic candidate for counteracting AA-induced toxicity and support further investigation into its value for future applications in the food safety field.Acrylamide (AA) is a common foodborne toxicant formed during the thermal processing of starch-rich foods and poses significant risks to the gut–liver axis. Although corncob-derived xylo-oligosaccharides (XOS) have emerged as promising prebiotics with multiple bioactivities, their protective effects against foodborne toxicants have not been explored. This study investigated the ameliorative effects of dietary corncob-derived XOS on AA-induced gut–liver toxicity and the underlying mechanisms in mice. Female C57BL/6J mice were gavaged with AA and concurrently fed diets with or without 7.0% XOS for 7 weeks. The results showed that XOS effectively alleviated AA-induced gut–liver toxicity, as evidenced by attenuated intestinal and hepatic inflammation (decreased pro-inflammatory cytokines IL-6 in colon, TNF-α, and IL1-β in liver; increased anti-inflammatory IL-10 in colon and liver), restored intestinal barrier integrity (upregulatedZo-1,Occludin, andClaudin-1mRNA, along with increased goblet cell numbers and mucus secretion), and ameliorated hepatic oxidative stress (upregulatedNrf2/Nqo1/Ho1mRNA expression, restored CAT activity, and reduced H2O2accumulation). 16S sequencing revealed that XOS supplementation significantly increased the Shannon index in AA-exposed mice compared with the AA group, suggesting enhanced cecal microbial diversity, although AA exposure did not significantly reduce α-diversity. Further analysis showed that XOS reshaped the community composition, particularly by reducing pro-inflammatory genera (Dubosiella,Desulfovibrio,Faecalibaculum, andFaecalimonas) and enriching short chain fatty acid (SCFA)-producing bacteria (Bifidobacterium,norank_f__Muribaculaceae, andAkkermansia). Furthermore, XOS significantly elevated SCFA levels in AA-treated mice, including acetic, propionic, butyric, valeric, and isovaleric acids. Collectively, these findings demonstrate that dietary XOS ameliorates AA-induced gut–liver toxicity in mice when co-administered with AA. This protective effect was associated with modulation of the gut microbiota and SCFA metabolism, providing a basis for future studies exploring the potential of XOS against foodborne contaminant toxicity.

Ficus pumila L. has been reported to alleviate pulmonary inflammation, its impact on chronic obstructive pulmonary disease (COPD) pathobiology-specifically via modulation of the gut-lung signaling pathway-has yet to be mechanistically defined. This study investigated how Ficus pumila L. polysaccharides (FP-P) and aqueous extracts (FP-E) remodel the gut microbiome-SCFA network and restore microbial metabolic function in a cigarette smoke–induced COPD mouse model. Microbiota composition was profiled by high-resolution 16S rRNA amplicon sequence variant (ASV) analysis, with concomitant quantification of caecal SCFA using targeted gas chromatography–mass spectrometry (GC–MS) and inference of metagenome function by PICRUSt2. Results demonstrated that FP-P and FP-E alleviated pulmonary pathology, reduced inflammatory cytokine secretion, and significantly restored gut microbiota α-diversity in COPD mice. At the family level, FP-P selectively expanded SCFA-producing Clostridiaceae , and Staphylococcaceae , whereas it contracted pro-inflammatory Helicobacteraceae and Campylobacteraceae . Caecal total SCFA concentration increased by 41.90 %, driven primarily by elevations in butyrate (+23.41 %) and propionate (+45.45 %), without significant changes in acetate. PICRUSt2-inferred metagenomes showed up-regulation of butanoate biosynthesis (PWY-5677), metabolism of cofactors and amino acid (P162-PWY and NAD-BIOSYNTHESIS-II), and carbohydrate degradation (P341-PWY), all of which underpin SCFA production. These functional shifts were accompanied by increased abundance of microbial genes encoding ribosomal proteins and ATP-binding cassette transporters, indicating barrier reinforcement. Collectively, FP-P and FP-E mitigate CS-induced COPD pathology through a gut microbiota-SCFA-lung signaling signaling pathway, highlighting the gut-to-lung communication within the broader gut-lung axis. These findings establish a mechanistic link between microbial metabolism and pulmonary inflammation while acknowledging that the reverse lung-to-gut feedback remains to be elucidated. Future studies will investigate this bidirectional crosstalk and the receptor-mediated signaling of SCFAs in lung tissue.

The extract ofDendrobium huoshanense, a traditional Chinese medicinal and food homologous plant belonging to the family Orchidaceae, was previously reported to have hypoglycemic and antioxidant effects. In this study, the direct effects of polysaccharide (DHP) and non-polysaccharide (NDHP) components ofD. huoshanense, as well as its water extract (DHWE) were compared with that of metformin (an antidiabetic drug) on the gut microbiota (collected from fecal flora) of rats with streptozotocin-induced type 1 diabetes (T1D) using an in vitro fermentation method. The results showed that DHWE, DHP, and NDHP reduced pH and increased bacterial proliferation and short-chain fatty acid (SCFA) content in fermentation broth. DHWE, DHP, NDHP and metformin promoted the production of acetic and propionic acid, acetic acid, propionic acid and butyric acid, and propionic acid, respectively. DHWE, DHP, and NDHP reduced the abundance ofProteobacteria(subdominant pathogenic bacteria) and increased the abundance ofFirmicutes(dominant beneficial gut bacteria). NDHP also reduced the abundance ofBacteroidetes(beneficial and conditional pathogenic). Metformin increased the abundance ofProteobacteriaand reduced the abundance ofFirmicutesandBacteroidetes. At the genus level, NDHP promoted the proliferation ofMegamonasandMegasphaeraand decreased harmful bacteria (e.g.,Klebsiella), and DHP increased the abundance ofPrevotellaceae(opportunistic and usually harmless). By contrast, metformin increased the abundance of harmful bacteria (e.g.,Citrobacter) and reduced the abundance of beneficial bacteria (e.g.,Oscillospira). Our study indicates that DHWE, DHP, and NDHP are potentially more beneficial than metformin on the gut microbiota of T1D rats in vitro.

Long-term hyperuricemia can induce kidney damage, clinically referred to as hyperuricemic nephropathy (HN), which is characterized by renal fibrosis, inflammation, and oxidative stress. However, currently used uric acid-lowering drugs are not capable of protecting the kidneys from damage. Therefore, uric acid-lowering drugs that can also protect the kidneys are urgently needed. In this study, we first discovered that salinomycin, an antibiotic, can regulate uric acid homeostasis and ameliorate kidney damage in mice with HN. Mechanistically, salinomycin inhibited serum and hepatic xanthine oxidase (XOD) activities and downregulated renal urate transporter 1 (URAT1) expression and transport activity, thus exerting uric acid-lowering effects in mice with HN. Furthermore, we found that salinomycin promoted p-NRF2 Ser40 expression, resulting in increased nuclear translocation of NRF2 and activation of NRF2. More importantly, salinomycin affected the gut microbiota and promoted the generation of short-chain fatty acids (SCFAs) in mice with HN. In conclusion, our results revealed that salinomycin maintains uric acid homeostasis and alleviates kidney injury in mice with HN by multiple mechanisms, suggesting that salinomycin might be a desirable candidate for HN treatment in the clinic.

Pediatric bacterial pneumonia continues to pose a significant global health burden, often resulting in serious complications. Recent studies have emphasized the pivotal role of the gut-lung axis in the progression of respiratory diseases, wherein disruptions in gut microbiota can aggravate pulmonary inflammation. This research investigates the underlying mechanisms by which Ma-Xing-Shi-Gan Decoction (MXSG), a classical formula in traditional Chinese medicine, influences the gut-lung axis in the context of childhood bacterial pneumonia. Clinical data indicated pronounced alterations in intestinal microbial composition and a marked decline in short-chain fatty acid (SCFA) levels among affected pediatric patients—changes that coincided with dysfunctional gut-lung signaling. In a murine model of bacterial pneumonia, MXSG administration restored microbial diversity, elevated SCFA production, and attenuated inflammatory responses by suppressing the HDAC3/NF-κB signaling cascade. Key bioactive constituents of MXSG—namely, quercetin, osajin, nobiletin, and solasodine—were identified as critical regulators, modulating lipid metabolism and promoting SCFA biosynthesis. These results highlight the therapeutic promise of MXSG in reestablishing gut-lung homeostasis and shed light on its mechanistic potential in the treatment of pediatric bacterial pneumonia. Graphical This schematic illustrates how MXSG restores gut microbiota diversity and SCFA levels, suppresses HDAC3/NF-κB signaling, and alleviates lung inflammation in pediatric bacterial pneumonia. Integrated multi-omics and mouse model experiments reveal that MXSG modulates the gut-lung axis and provides a novel mechanism for its clinical efficacy.

Hyperuricemia (HUA) is mainly caused by sustained disturbances in purine metabolism. Although both iridoids and vinegar have been reported to exert anti-hyperuricemic effects, no product combining iridoid-rich ingredients with aromatic vinegar has yet been investigated. In this study, the urate-lowering effects and underlying mechanisms of a combined preparation of iridoids from Gardenia jasminoides Ellis and Zhenjiang aromatic vinegar were evaluated in a rat model of HUA. Both Gardenia jasminoides extract (GET) and its aromatic vinegar formulations (GETV) reduced serum uric acid, creatinine, and urea nitrogen levels, inhibited serum and hepatic xanthine oxidase (XOD) activity, and improved hepatic antioxidant status. GETV showed stronger effects than GET, and the ethanol extract aromatic vinegar group (GEEV) produced the most pronounced overall response. In addition, GET and GETV modulated gut microbiota composition and increased the levels of short-chain fatty acids (SCFAs), especially acetic, propionic, butyric, and valeric acids. These results suggest that GETV exerts multi-target protective effects against HUA and may provide a basis for developing a functional food ingredient for HUA management.

The use of waste activated sludge (WAS) fermentative short-chain fatty acids (SCFAs) as the excellent carbon source of wastewater biological nutrient removal has drawn much attention recently as it can reuse WAS organics and reduce WAS production. This study developed a novel, efficient, and environmentally friendly approach combining atmospheric pressure plasma jet with microbubbles (plasma/MBs) for WAS pretreatment. Compared with the control, plasma/MBs pretreatment enhanced SCFA generation by 92% and acetic acid proportion by 21% with plasma discharge power at 500 W and MBs dosage at 7.9 mg/L dissolved oxygen. The plasma/MBs combination motivated the reaction of various reactive species (such as O 3 , NO x , ONOO − , •OH, e − , 1 O 2 , and •O 2 − ) and enhanced the activity of physical energies (such as light and heat). This synergy promoted sludge cell lysis and biodegradable substance release, significantly boosting acetic acid-enriched SCFA generation from fermentation. Moreover, plasma/MBs pretreatment increased the expression of key genes for SCFA production during fermentation, which also contributed to the production of SCFAs. Besides, plasma/MBs pretreatment favored WAS dewatering, heavy metal removal, and organic pollutant degradation, providing a new approach to advancing WAS resource recovery through innocuous management.

Natural polysaccharides from Astragalus membranaceus have been shown to relieve ulcerative colitis (UC). However, the mechanism and causal relationship between the gut microbiota and Astragalus polysaccharides (APS) treatment of UC are unclear. The results of the present study showed that APS ameliorated colonic injury and the disruption of the gut microbiota and restored intestinal immune homeostasis in mice with DSS-induced colitis. Meanwhile, we found that APS treatment was ineffective in antibiotic-treated colitis mice but was effective when FMT (Fecal microbiota transplantation) was performed on UC mice using APS-treated mice as donors. APS increased the proportion of relevant microbiota that produce SCFAs and both direct administration of APS and administration of APS-adjusted gut microbiota significantly promoted the production of SCFAs in colitis mice. We demonstrated that APS dually inhibited NF-κB activation via the TLR4 and HDAC3 pathways and improved the balance in Th17/Treg cells in UC mice. In conclusion, our study revealed that APS is a promising prebiotic agent for the maintenance of intestinal health and demonstrated that APS may ameliorate colitis in a gut microbiota-dependent manner.

Purpose GLP-1 based therapy represents a new treatment option for inflammatory bowel disease. Ban-Lan-Gen (BLG) granule, a known anti-viral TCM formulation, exhibits potential anti-inflammatory activities in treating various kinds of inflammation. However, its anti-inflammatory effect on colitis and the underlying mechanisms remain unknown. Methods Dextran sulfate sodium (DSS)-induced chronic relapsing colitis in mice was established. The disease activity index, histological sign of damage, and levels of proinflammatory cytokines were performed to assess the protective effects of BLG. Serum GLP-1 level and colonic Gcg, GPR41 and GRP43 expression, the community compositions of gut microbiota, the levels of SCFAs in the feces and GLP-1 release from primary murine colon epithelial cells were performed to characterize the effects of BLG on gut microbiota and gut SCFA derived-GLP-1 production. Results BLG treatment significantly alleviated body weight loss, DAI, colon shortening, colon tissue damage, and pro-inflammatory cytokine levels of TNF-α, IL-1β and IL-6 in the colon tissues. Moreover, BLG treatment could observably restore colonic Gcg, GPR41 and GRP43 expression and serum GLP-1 level of colitic mice, as well as correct the alteration of gut microbiota in colitic mice by increasing the abundances of SCFA-producing bacteria, eg, Akkermansia and Prevotellaceae_UCG-001, and decreasing the abundances of bacteria, eg, Eubacterium_xylanophilum_group, Ruminococcaceae_UCG-014, Intestinimonas, and Oscillibacter. Furthermore, BLG treatment could markedly increase the levels of SCFAs in feces of colitic mice. In parallel, ex vivo assay also showed that and the extract of feces from BLG-treatment mice could greatly stimulate the secretion of GLP-1 from primary murine colon epithelial cells. Conclusion These findings suggest that the anti-colitis effects of BLG are achieved at least partly by regulating gut microbiota and restoring gut SCFA derived-GLP-1 production, and BLG has the potential to be developed as a promising agent for the treatment of chronic relapsing colitis.
