The purified fraction 3 of polysaccharides from Fuzhuan brick tea (FBTPS-3) could attenuate the colitis and modulate the gut microbiota. However, the relationship between anti-inflammatory effect of FBTPS-3 and the gut microbiota is still unknown. Thus, the anaerobic fermentation in vitro was used to investigate the potential mechanism. FBTPS-3 could be utilized and degraded by gut microbiota from inflammatory bowel disease (IBD) subjects. Furthermore, FBTPS-3 could modulate the composition and structure of IBD gut microbiota toward to that of healthy group. FBTPS-3 showed a superior modulated effect on IBD gut microbiota by increasing Bacteroides and decreasing Escherichia/Shigella . Furthermore, the fermentation solution rather than FBTPS-3 itself showed anti-inflammatory effects on lipopolysaccharide-treated RAW264.7 macrophages, which might be due to the metabolites such as short-chain fatty acids (SCFAs). Thus, FBTPS-3 can be expected as novel prebiotics for treatment of IBD via modulating gut microbiota, and promoting the production of SCFAs.

Conventional magnesium supplementation relies on passive
diffusion
in the small intestine, resulting in poor bioavailability. To address
this, we developed an oxidized starch-chitosan-Mg 2+ composite
gel that directs more Mg 2+ to the colon, where resistant
polysaccharides are fermented to SCFAs that may actively enhance absorption.
Mg 2+ incorporation increased the gel’s resistant
component to 52.42 ± 0.63%. Increasing Mg 2+ from 0.5%
to 2% reduced cumulative upper gastrointestinal release from 70.61
± 0.79% to 43.62 ± 1.01% ( p < 0.05),
with higher concentrations showing no further improvement. In Mg 2+ -deficient mice, the gel dose-dependently restored serum
Mg 2+ to healthy levels. Comparison at equal Mg 2+ doses revealed that the polysaccharide carrier independently contributed
to enhanced absorption. Mechanistically, resistant polysaccharides
were associated with upregulation of SCFAs receptors (FFAR2/FFAR3/HCAR2)
and Mg 2+ transporters (TRPM6/TRPM7/CNNM4), with immunofluorescence
indicating TRPM7 upregulation. This work provides proof-of-concept
evidence for a paradigm shift from passive supplementation to active
regulation via SCFAs-associated transporter modulation.

Background: Marine algal oligosaccharide (MAO) molecular structural characteristics are closely related to their bioactivity. However, an understanding of the specific relationships between MAO molecular structure (including polymerization degree [DP] and sulfate groups) and bioactivity is lacking. Methods: In this work, laboratory-prepared DP4 and DP8 MAOs, including agar- (AOs) and κ-, ι-, and λ-carrageenan-oligosaccharides (COs), were examined for their anti-inflammatory activity in RAW264.7 macrophages and therapeutic effect on colitis in mice; additionally, the potential mechanisms involved were explored. Results: The results showed that CO (ι- and λ-COs) anti-inflammatory activity was higher than that of AOs and was positively correlated with sulfate group content. MAOs with different structures all alleviated colitis symptoms; those with higher sulfate group content and DP showed excellent therapeutic efficacy. MAOs modulated gut microbiota and promoted the enrichment of microbiota-derived short-chain fatty acid production (butyrate), which potentially induced PPARγ signaling and contributed to the impacts of MAOs on mucosal homeostasis restoration. Conclusions: Therefore, MAOs with higher sulfate content and DP showed improved therapeutic efficacy compared with those with lower contents, which involved higher SCFA production.

The intestinal fermentability of pectic polysaccharides is largely determined by its molecular size. In this study, fermentation properties of enzymatic-modified apple pectin (AP) and homogalacturonans (HG) with high, medium and low molecular weight (Mw) were evaluated by in vitro fermentation model , and their structural changes were also investigated. Results showed that Mw, monosaccharide contents and molecular linearity of the AP hydrolysates were reduced after microbial degradation . On the other hand, culture media supplemented with low-Mw AP (60,300 g/mol) and low-Mw HG (861 g/mol) exhibited lower pH (5.1 and 5.7, respectively) and produced higher total short-chain fatty acid contents (SCFA, 230.40 mmol/L and 187.19 mmol/L, respectively). However, reduced trends in abundance of the pectinolytic microorganisms Faecalibacterium and Eubacterium were showed as Mw of the HG decreased, whereas growth of the SCFA-producer genera Bifidobaacterium , Megasphaera and Allisonella were improved. This work confirmed that low-Mw pectin and homogalacturonan generated more beneficial metabolites, developing structure-microbiota-gut health relationship.

Ethnopharmacological relevance Dingxin Recipe Ⅲ (DXR Ⅲ) is a traditional Chinese medicine compound used for hyperlipidemia treatment in clinical practice. However, its curative effects and pharmacological mechanisms in hyperlipidemia have not been clarified to date. Aim of the study Studies have demonstrated that gut barrier was strongly implicated in lipid deposition. Based on gut barrier and lipid metabolism, this study examined the effects and molecular mechanisms of DXR Ⅲ in hyperlipidemia. Materials and methods The bioactive compounds of DXR Ⅲ were detected by ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry, and its effects were evaluated in high-fat diet-fed rats. Specifically, the serum levels of lipids and hepatic enzymes were measured using the appropriate kits; colon and liver sections were obtained for histological analyses; gut microbiota and metabolites were analyzed by 16S rDNA sequencing and liquid chromatography-MS/MS; and the expression of genes and proteins was determined by real-time quantitative polymerase chain reaction and western blotting and immunohistochemistry, respectively. The pharmacological mechanisms of DXR Ⅲ were further explored by fecal microbiota transplantation and short-chain fatty acid (SCFAs)-based interventions. Results DXR Ⅲ treatment significantly downregulated serum lipid levels, mitigated hepatocyte steatosis and improved lipid metabolism. Moreover, DXR Ⅲ improved the gut barrier, specifically by improving the physical barrier in the colon, causing part composition changes in the gut microbiota, and increasing the serum SCFAs level. DXR Ⅲ also upregulated the expression of colon GPR43/GPR109A. Fecal microbiota transplantation from rats treated with DXR Ⅲ downregulated part hyperlipidemia-related phenotypes, while the SCFAs intervention significantly improved most of the hyperlipidemia-related phenotypes and upregulated the expression of GPR43. Moreover, both DXR Ⅲ and SCFAs upregulated the expression of colon ABCA1. Conclusion DXR Ⅲ protects against hyperlipidemia by improving the gut barrier, particularly the SCFAs/GPR43 pathway.

Iota-carrageenan oligosaccharides (iCOs), derived from marine red algae, are traditionally used as antithrombotic and anti-inflammatory agents in folk medicinal practice. Despite the prevailing emphasis on these aspects in their applications, the potential of iCOs as a prebiotic agent for gut health and its subsequent impact on intestinal disorders such as colitis remains largely unexplored. A DSS-induced colitis model was employed in C57BL/6 male mice to analyze the gut microbiota via 16S rRNA sequencing. Fecal microbiota transplantation (FMT) was used to assess the therapeutic effects of iCOs on colitis. RNA sequencing (RNA-Seq) identified pathways and genes affected by iCOs. ELISA measured inflammatory cytokines, while western blot and RT-qPCR evaluated protein and gene expressions, respectively. The iCOs increased beneficial bacteria, such as Lactobacillus , Bifidobacterium , and Akkermansia . They enhanced short-chain fatty acid production and upregulated GPR41, GPR43, and GPR109A mRNA, influencing cytokine secretion. The iCOs reduced mRNA of SPHK1 , BDKRB1 , LCN2 , and so on, potentially through PI3K-Akt pathway inhibition, and promoted tight junction protein expression. Our findings highlight the novel therapeutic potential of iCOs in colitis, indicating a multifaceted approach to treatment that includes gut microbiota modulation, intestinal barrier restoration, and the suppression of inflammatory responses. The present study aimed to explore the possible mechanisms underlying Dendrobium officinale leaf polysaccharides of different molecular weight to alleviate glycolipid metabolic abnormalities, organ dysfunction and gut microbiota dysbiosis of T2D mice. An ultrafiltration membrane was employed to separate two fractions from Dendrobium officinale leaf polysaccharide named LDOP-A and LDOP-B. Here, we present data supporting that oral administration of LDOP-A and LDOP-B ameliorated hyperglycemia, inhibited insulin resistance, reduced lipid concentration, improved β-cell function. LDOP-A with lower molecular weight exhibited improved effect on diabetes than LDOP-B, concurrent with increased levels of colonic short-chain fatty acids (SCFAs) i.e., butyrate, decreased ratio of Firmicutes to Bacteroidetes phyla, and increased abundance of the gut beneficial bacteria i.e., Lactobacillus , Bifidobacterium and Akkermansia . These results suggest that LDOP-A possesses a stronger effect in ameliorating T2D than LDOP-B which may be related to the distinct improved SCFAs levels produced by the change of intestinal flora microstructure.

Chronic low-grade inflammation (CLI) is a common pathological basis for many major diseases, whose systemic regulatory mechanism remains unclear. Based on Pu-erh tea’s potential to improve gut inflammation and dysfunction in the “gut-liver/brain” axis, this study systematically explored the multi-target regulatory mechanisms of Pu-erh tea through establishing a dextran sodium sulfate-induced CLI model. The results demonstrated that CLI triggered an inflammatory cascade in the colon, liver, and brain, accompanied by liver metabolism disorders (downregulation of FXR/SREBP-1c) and gut-brain barrier damage (downregulation of ZO-1/Occludin). Pu-erh tea significantly reversed these pathological changes by reshaping the gut microbiota (increased levels of norank_f__Muribaculaceae, Lactobacillus, Akkermansia , and Dubosiella ) and promoting the biosynthesis of short-chain fatty acids (SCFAs, butyric acid and isobutyric acid), which stemmed from improved mitochondrial dysfunction. The “gut microbes-SCFAs-mitochondria” axis may hold promise as a potential strategy for phytochemicals to improve body inflammation.

Polyoxometalates (POMs), a versatile and environmentally-friendly inorganic material, have been extensively studied and applied in chemical catalytic oxidation and biological nutrients removal processes. However, little is known about effects of POMs pretreatment on anaerobic sludge fermentation. This study thereby filled such knowledge gap and provided insights into the underlying mechanisms. Results demonstrated the maximal short-chain fatty acids (SCFAs) production increased by 6.18 times with POMs rising from 0 to 0.05 g/g TSS. Mechanistic investigations revealed that the oxidation stress of POMs as well as reactive oxygen species (ROS) activated by POMs were responsible for the disintegration of waste activated sludge (WAS). More importantly, POMs pretreatment improved the biodegradability of organics released, providing more biodegradable substrates for SCFAs generation. Furthermore, the inhibition of POMs to SCFAs producers was less severe than that to SCFAs consumers, leading to SCFAs accumulation. Microbial community analysis exhibited that increased the population of hydrolysis (i.e., Longilinea ) and SCFAs generation microbes (i.e., Acinetobacter and Fusibacter ). Further evaluation showed that the POMs-based technology is economically and environmentally attractive for the pretreatment of WAS. Finally, a “closed-loop” concept of the reutilization of renewable POMs may provide an important implication of WAS management in the future.

Short-chain fatty acids (SCFAs) play key roles in maintaining health and treating disease. Quantification of important fecal SCFAs is necessary to facilitate the clarification of their biological roles. However, the existing quantifying methods mainly depend on complicated precolumn derivatization, and/or are unable to determine formic acid, a SCFA commonly associated with toxicity. In this study, a direct gas chromatography-mass spectrometry (GC-MS) method for simultaneous quantification of ten SCFAs including formic acid in rat feces was developed. The approach was optimized in terms of chromatographic and spectrometric conditions as well as sample preparation. DB-FFAP capillary column with temperature programming was used to get baseline separation and symmetrical peak shape of SCFAs without precolumn derivatization in a relatively short running time (8 min). Multiple reaction monitoring (MRM) scan mode was employed to enhance the sensitivity and selectivity of SCFAs. Acidification with 50% HCl and immediate extraction with diethyl ether were utilized to achieve sample preparation of ten SCFAs from feces. Furthermore, the developed method was validated with wide linear range, high sensitivity and precision, low matrix effect and acceptable accuracy. The established method was successfully applied to compare the contents of fecal SCFAs between normal and immunosuppressed animal models.

This study systematically assessed the regulatory effects of Yunnan Lufeng aromatic vinegar (LFAV) on the intestinal microbiota composition, short-chain fatty acids, and cecal metabolites in mice. 16S rRNA high-throughput sequencing revealed that LFAV intervention significantly altered gut microbiota diversity; the M-L group exhibited 19.98% unique operational taxonomic units, while both Chao1 (496.63 ± 42.14) and Shannon indices (6.68 ± 0.32) increased by 37.46% and 3.25%, respectively, compared to the blank group, indicating enhanced microbiota richness. Species composition analysis demonstrated that the relative abundance of Firmicutes reached 75.4% in the M-L group, a 24.4% increase over the B group, whereas Bacteroidetes abundance decreased to 8.2%. GC-MS analysis detected peak butyric acid levels in the M-L group. Untargeted metabolomics identified 520 metabolites, of which 60 were significant differential metabolites. Cluster heatmap and Z-score analyses demonstrated that LFAV intervention significantly modulated mouse metabolites. KEGG pathway enrichment analysis indicated the upregulation of pathways including neuroactive ligand–receptor interactions and renin secretion. Pearson correlation analysis showed a strong positive correlation (p< 0.01) between Lactobacillus and acetic acid/butyric acid; concurrently, increased Lactobacillus proliferation and elevated butyric acid levels were observed in the M-L and M-M groups. These findings suggest that LFAV intervention promotes the proliferation of beneficial bacteria, which may improve intestinal health. Collectively, LFAV significantly modified gut microbiota structure and metabolites in mice, highlighting its potential as a natural prebiotic or functional food ingredient and providing a scientific basis for developing functional vinegar products.

T-2 toxin is a trichothecene mycotoxin produced by Fusarium species and can cause disease after contaminated food or feed ingestion. This study aimed to investigate the impact of lycopene (Ly) on systemic inflammation, oxidative stress, and dysbiosis in mice exposed to T-2 toxin. A total of 20 male BALB/c mice (6 weeks old, weighing 23.5 ± 0.32 g) randomly divided into four groups ( n = 5): Control (Cont), Ly, T-2, and T-2 + Ly groups. Findings revealed that Ly enhanced the body weight, feedd, and water intake that have been reduced by T-2. Ly increased the villus height (VH) and VH: crypt depth (CD) and decreased CD and villus width (VW) in ileum compared to the T-2 group. In alpha diversity, Staphylococcus and Corynebacterium were the most prevalent in Cont group; Staphylococcus , and Lactobacillus , were the most prevalent in T-2 group; Candidatus-Arthromitus and Staphylococcus were the most prevalent in T-2 + Ly group. Moreover, T-2 mice had significantly ( p < 0.05) greater levels of interleukin-1 beta (IL-1β), interferon gamma (IFN-γ), Interleukin-17 (IL-17), and tumor necrosis factor-alpha (TNF-α) than Cont, while the Ly significantly ( p < 0.05) lowered their concentration. The T-2 mice had significantly ( p < 0.05) higher levels of reactive oxygen species (ROS) and malondialdehyde (MDA), while their catalase (CAT),, glutathione (GSH), adenosine triphosphate (ATP), and superoxide dismutase 1(SOD1) activities were significantly ( p < 0.05) lower than Cont. Ly significantly ( p < 0.05) restored their concentration. Moreover, there was a significant reduction ( p < 0.05) of hexanoic, butyric, acetic, pentanoic, and Heptanoic acids in the T-2 mice compared to Cont. In contrast, treatment with Ly was found to significantly restore their levels compared to T-2 group. We conclude that Ly administration in the context of T-2–induced toxicity may help attenuate systemic oxidative stress and inflammation, possibly through modulation of gut microbiota and fecal short-chain fatty acids (SCFAs).
