Fe3O4@GO magnetic nanocomposites protect mesenchymal stem cells and promote osteogenic differentiation of rat bone marrow mesenchymal stem cells

He Zhang,Sirong Li,Yufeng Liu,Yijun Yu,Shichao Lin,Quan Wang,Leiying Miao,Hui Wei,Weibin Sun

Fe3O4 nanoparticles (Fe3O4 NPs) are typical magnetic materials for bone tissue regeneration. However, the accompanying oxidative stress during the reaction process of Fe3O4 NPs and H2O2 in bone remodeling and disease may hinder their application. In order to reduce this side effect, we selected graphene oxide (GO) to modify Fe3O4 NPs. We showed that Fe3O4@GO magnetic nanocomposites (Fe3O4@GO MNCs) eliminated 30% of H2O2 in 3 h, and reduced the amount of ˙OH, the intermediate product of the Fenton reaction. The cellular study demonstrated that Fe3O4@GO MNCs reduced the cell damage caused by reactive oxygen species (ROS) and improved the activity of mesenchymal stem cells (MSCs). Moreover, when the magnetic field and bone morphogenetic protein-2 (BMP2) delivered by Fe3O4@GO MNCs worked together, osteogenic differentiation of MSCs in vitro was well promoted.

Stem cell Janus patch for periodontal regeneration

Na Yan,Bin Hu,Junchao Xu,Rong Cai,Zhenhai Liu,Dapeng Fu,Beibei Huo,Zhenhua Liu,Yuliang Zhao,Chunying Chen,Weihua Xu

Periodontitis places a substantial burden on healthcare worldwide. Due to the complexity of the periodontium, regenerative periodontal therapy in healing periodontal tissue damage caused by periodontitis has long been difficult using traditional tissue engineering techniques. The current clinical treatment, guided tissue regeneration (GTR), has shown limitations in reconstructing an osteogenic microenvironment . Here, we designed a stem cell-containing patch (stem cell “Janus” patch; SCJP) to acclimatize stem cells in the osteogenic and odontogenic microenvironment for periodontal regeneration. The SCJP promoted periodontal regeneration within its functional layer, while its barrier layer blocked gingival epithelial cells (GECs) invasion. Specifically, embedded dental pulp stem cells (DPSCs) in the functional layer facilitated the regeneration of dentin, cementum and alveolar bone, all of which orchestrated complete periodontal structure regeneration. We characterized the biophysical properties of the SCJP barrier layer, which has a compact structure and appreciable resistance to mechanical stress, suggesting the patch may be effective in preventing epithelium invasion. Successful ectopic mineralization and further osteogenic and odontogenic differentiation in mouse, rat and swine periodontal defect models further validated the patch’s potential in accelerating periodontal tissue regeneration. Our study describes a safe and effective stem cell-base strategy for a therapeutic that not only realizes periodontal repair but also protects the restoring periodontium from epithelial cell invasion.

Bioglass enhanced wound healing ability of urine-derived stem cells through promoting paracrine effects between stem cells and recipient cells

Yunlong Zhang,Xin Niu,Xin Dong,Yang Wang,Haiyan Li

In cell therapy, tissue regeneration ability of stem cells relies on the paracrine effects between stem cells and recipient cells. Our recent studies demonstrated that, in tissue engineering, bioactive silicates could stimulate paracrine effects between stem cells and recipient cells, which enhanced tissue generation. Therefore, we proposed that, in cell therapy, it may be an effective method to improve tissue regeneration ability of stem cells through activating the paracrine effects between stem cells and recipient cells with bioactive silicates. As urine-derived stem cells (USCs) have been injected for wound healing and bioglass (BG) have shown bioactivity for various types of stem cells, in this study, we activated USCs with effective BG ionic products. Then the conditioned medium of BG-activated USCs were used to culture endothelial cells and fibroblasts as well as co-cultures of endothelial cells and fibroblasts. Results showed that growth factor expression in BG-activated USCs was upregulated. In addition, paracrine effects between USCs and recipient cells in wound healing were stimulated, which resulted in enhanced capillary-like network formation of endothelial cells and matrix protein production as well as myofibroblast differentiation of fibroblasts. Finally, the BG-activated USCs were applied on full-thickness excisional wounds. Results confirmed that BG-activated USCs had better wound healing ability through improving angiogenesis and collagen deposition in wound sites as compared with USCs without any treatment. Taken together, BG can be used to promote wound healing ability of USCs by enhancing paracrine effects between USCs and recipient cells.

Co-transplantation of bone marrow mesenchymal stem cells and monocytes in the brain stem to repair the facial nerve axotomy

Li Wu,Dan Han,Jie Jiang,Xiaojie Xie,Xunran Zhao,Tengfei Ke,Wen Zhao,Liu Liu,Wei Zhao

After the facial nerve axotomy (FNA), the distal end of the axon would gradually decay and disappear. Accumulated evidence shows that transplantation of bone marrow mesenchymal stem cells (BMSCs) reveals potential in the treatment of nervous system diseases or injuries. This study is aimed at investigating the therapeutic effects of co-transplantation of BMSCs and monocytes in FNA. We found that co-culture significantly elevated the CD4+/CD8+ ratio and CD4+ CD25+ T cell proportion compared with monocytes transplantation, and enhanced the differentiation of BMSCs into neurons. After the cell transplantation, the lowest apoptosis in the facial nerve nucleus was found in the co-transplantation group 2 (BMSCs:monocytes= 1:30). Moreover, the lowest expression levels of pro-inflammatory cytokines and the highest expression levels of anti-inflammatory cytokines were observed in the co-transplantation group 2 (BMSCs: monocytes= 1:30). The highest expression levels of protein in the JAK/STAT6 pathway and the SDF-1/CXCR4 axis were found in the co-transplantation group 2. BMSC/monocyte co-transplantation significantly improves the microenvironment in the facial nerve nucleus in FNA rats; therefore these findings suggest that it could promote the anti-/pro-inflammatory balance shift towards the anti-inflammatory microenvironment, alleviating survival conditions for BMSCs, regulating BMSC the chemotaxis homing, differentiation, and the section of BMSCs, and finally reducing the neuronal apoptosis. These findings might provide essential evidence for the in-hospital treatment of FNA with co-transplantation of BMSCs and monocytes.

Conversion of glucose to levulinic acid and upgradation to γ-valerolactone on Ru/TiO2 catalysts

Yubo Liu,Guoqiang Ding,Guoping Zhao,Haohao She,Yulei Zhu,Yong Yang

Combining glucose dehydration and the subsequent hydrogenation in one pot is a preferable approach for process development as such a method allows in situ generation of the reactive intermediate to undergo further reaction without extra energy-intensive separation. Herein, phosphotungstic acid and various types of titania (anatase, rutile, P25) supported Ru-based catalysts were considered as the dehydration and hydrogenation catalysts, respectively. Modulating the different reactant media (N2, H2), various products were obtained with GBL–H2O as the solvent. A considerable yield (42%) of levulinic acid (LA) and γ-valerolactone (GVL) (40%) were obtained in nitrogen and subsequent hydrogen. Ru/TiO2 (rutile) was the favorable hydrogenation catalyst among the three types of Ru/TiO2. Meanwhile, a certain amount of sorbitol (36%) was obtained in pure hydrogen. The hydrogenation of glucose is more likely to occur than the glucose dehydration. The physicochemical properties of the catalysts were characterized by XRD, BET, TPR, STEM and in situ CO/FT-IR, and the results show that well-dispersed Ru particles are located on the rutile crystallites, which facilitated the hydrogenation of LA. A strong metal support interaction (SMSI) was responsible for the various microstructure properties and the different hydrogenation reactivity. This work allows a better understanding of the reaction paths of glucose conversion.

Induced pluripotent stem cell-derived mesenchymal stem cells (iMSCs) inhibit M1 macrophage polarization and reduce alveolar bone loss associated with periodontitis

Liang Chen,Yuanqing Liu,Chenhao Yu,Pei Cao,Yiming Ma,Yiran Geng,Yu Cai,Yong Zhang,Jia Liu,Yang Li,Qingxian Luan

Background Periodontitis is a chronic inflammatory disease and macrophages play a pivotal role in the progression of periodontitis. Mesenchymal stem cells (MSCs) have emerged as potential therapeutic agents for the treatment of periodontitis due to their immunomodulatory properties and capacity for tissue regeneration. Compared to conventionally derived MSCs, induced pluripotent stem cell-derived MSCs (iMSCs) offer distinct advantages as promising candidates for MSC-based therapies, owing to their non-invasive acquisition methods and virtually unlimited availability. This study aims to investigate the effects and mechanisms of iMSCs in modulating macrophage polarization and alleviating periodontitis-related alveolar bone loss. Methods iMSCs were generated from iPSCs and characterized for differentiation potential. The effects of iMSCs on macrophage polarization were evaluated using THP-1-derived macrophages under inflammatory conditions (LPS and IFN-γ stimulation). Co-culture assays, cytokine analysis, reactive oxygen species (ROS) detection, transcriptomic analysis, flow cytometry, reverse transcription-quantitative polymerase chain reaction (RT-qPCR), and western blot analysis were performed to elucidate the underlying mechanisms. The therapeutic potential of iMSCs was assessed in a ligature-induced periodontitis mouse model using micro-CT, histological analysis, and immunofluorescence staining. Results iMSCs inhibit M1 macrophage polarization through the suppression of the NF-κB signaling pathway. Additionally, iMSCs reduce the production of pro-inflammatory cytokines (IL-1β, IL-17) and reactive oxygen species (ROS), while enhancing the secretion of anti-inflammatory cytokines (IL-10) and growth factors (VEGF), thereby improving the inflammatory microenvironment. Under inflammatory conditions, iMSCs preserve the osteogenic potential of periodontal ligament stem cells (PDLSCs) and alleviate alveolar bone loss in mice with periodontitis. In vivo, iMSCs reduce the number of M1 macrophages and inhibit the activation of NF-κB in periodontal tissues, supporting their anti-inflammatory and immunomodulatory effects. Conclusion iMSCs demonstrate significant therapeutic potential in periodontitis by modulating macrophage polarization, reducing oxidative stress, and mitigating alveolar bone loss associated with the disease. These findings provide new insights into the mechanisms of iMSCs and their application as cell-based therapies for periodontal diseases. Stem Cell-Recruiting Injectable Microgels for Repairing Osteoarthritis

Yiting Lei,Yuping Wang,Jieliang Shen,Zhengwei Cai,Yongsheng Zeng,Piao Zhao,Junyi Liao,Chengjie Lian,Ning Hu,Xiaoji Luo,Wenguo Cui,Wei Huang

The differentiation potentials and viability of stem cells are often impaired during cell isolation and delivery. Inspired by the phenomenon where islands can recruit seabirds for nesting, “cell island” microgels (MGs), that is, growth factor-loaded methacrylated hyaluronic acid and heparin blend MGs, which can recruit endogenous stem cells and promote chondrogenic differentiation, are constructed using microfluidic technology and photopolymerization processes, followed by non-covalently binding platelet-derived growth factor-BB (PDGF-BB) and transforming growth factor-beta3 (TGF-β3). The loading efficiency of PDGF-BB and TGF-β3 are 96% and 91%, respectively. In vitro and in vivo experiments find that the “cell island” MGs can enhance the migratory capacity of cells and recruit them from their niche via releasing PDGF-BB. Meanwhile, by using hyaluronic acid, the “cell island” MGs provide a suitable microenvironment for cell attachment and spreading. Furthermore, the “cell island” MGs induce chondrogenic differentiation of the recruited cells via releasing TGF-β3 and present a promising therapeutic effect for osteoarthritis. In sum, this developed “cell island” MG might serve as a temporary “nest site” to allow the migration, adhesion, and differentiation of endogenous stem cells, which can be a promising candidate rather than the conventional cell-seeded scaffolds for promoting tissue regeneration.

O-alg-THAM/gel hydrogels functionalized with engineered microspheres based on mesenchymal stem cell secretion recruit endogenous stem cells for cartilage repair

Yucong Li,Linlong Li,Ming Wang,Boguang Yang,Baozhen Huang,Shanshan Bai,Xiaoting Zhang,Nan Hou,Haixing Wang,Zhengmeng Yang,Chong Tang,Ye Li,Wayne Yuk-Wai Lee,Lu Feng,Micky D. Tortorella,Gang Li

Lacking self-repair abilities, injuries to articular cartilage can lead to cartilage degeneration and ultimately result in osteoarthritis. Tissue engineering based on functional bioactive scaffolds are emerging as promising approaches for articular cartilage regeneration and repair. Although the use of cell-laden scaffolds prior to implantation can regenerate and repair cartilage lesions to some extent, these approaches are still restricted by limited cell sources, excessive costs, risks of disease transmission and complex manufacturing practices. Acellular approaches through the recruitment of endogenous cells offer great promise for in situ articular cartilage regeneration. In this study, we propose an endogenous stem cell recruitment strategy for cartilage repair. Based on an injectable, adhesive and self-healable o-alg-THAM/gel hydrogel system as scaffolds and a biophysio-enhanced bioactive microspheres engineered based on hBMSCs secretion during chondrogenic differentiation as bioactive supplement, the as proposed functional material effectively and specifically recruit endogenous stem cells for cartilage repair, providing new insights into in situ articular cartilage regeneration.

Well-dispersed iron and nitrogen co-doped hollow carbon microsphere anchoring by g-C3N4 for efficient peroxymonosulfate activation

Likun Chen,Kewen Xing,Qikai Shentu,Yifei Huang,Weiyang Lv,Yuyuan Yao

Developing single-atom Fenton-like catalysts with the maximum utilization of active sites present an attractive potential in environmental remediation. Herein, the single-atom Fe and N co-doped hollow carbon microsphere loaded g-C 3 N 4 catalyst (HFeNC-g-C 3 N 4 ) was prepared by an innovative cascade anchoring strategy using polystyrene as the hard template, iron phthalocyanine, polydopamine and urea as the Fe, N and C precursor, in which the in-situ generated g-C 3 N 4 could not only effectively anchor Fe atom to create the well-dispersed Fe-N x active sites, but also accelerate the electron transfer in peroxymonosulfate (PMS) activation. Taking advantages of such sequential protecting strategy, the as-synthesized HFeNC-g-C 3 N 4 catalyst with single-atom Fe-N x active sites, verified by XRD , XPS and HAADF-STEM, could work as an efficient Fenton-like catalyst for PMS activation, which achieved almost 100% removal of 4-chlorophenol (4-CP) in 5 min with the turnover frequency calculated to be 34.6 times higher than that of the homogeneous Fe 2+ catalyst. The mechanism of O 2 •- dominated radical combined with nonradical 1 O 2 pathway was confirmed by quenching experiments and ESR analysis, which might be interrelated to the improvement of pH adaptability and interference immunity of HFeNC-g-C 3 N 4 /PMS system. Overall, the present findings provided an innovation strategy for the synthesis of excellent single-atom Fe based catalyst in wastewater purification.

Mesenchymal Stem Cell Exosomes and Cancer: Controversies and Prospects

Sirine Yassine,Nada Alaaeddine

Mesenchymal stem cells (MSCs) have displayed a novel therapeutic strategy for a wide range of diseases and conditions. Their secretome and exosome-based paracrine activity are considered as the main processes harboring their diverse therapeutic properties. Several investigations have examined the effects of MSC-derived exosomes on cancer growth, yet, controversial results have always emerged. Although MSC-derived exosomes are able to rigorously enforce the repression of cancer proliferation and progression, it is shown that MSCs exosomal activity displays numerous protumorigenic effects. This discrepancy over the dual effects of MSCs on cancer growth may be mediated by many factors including experimental design, stem cells origins, culture conditions, in addition to cancer-MSCs cross-talks. Despite the controversial effects of MSCs on carcinogenesis, scientists are able to overcome a number of obstacles by modifying MSCs to deliver antioncogenic miRNAs, anticancer drugs, and oncolytic viruses into tumor sites. This review discusses the controversial effects of MSC-derived exosomes on tumorigenesis, investigates the main causes that underlie this discrepancy, summarizes the pattern of engineered-MSCs, and finally highlights how future studies should advance the research in the field of MSCs-based cancer therapies in order to accelerate the transition from preclinical studies to clinical practice.

Yang Lei,Liu Yuxiao,Sun Lingyu,Zhao Cheng,Chen Guopu,Zhao Yuanjin

A novel stem cell delivery core-shell biomass microcapsule was generated by an all-aqueous phase microfluidic electrospray technique. Microcapsule with a certain mechanical strength and porous structure is beneficial for substances exchange between cells and the external environment, together with avoiding cell damage during treatment. Core-shell microcapsule provided a favorable cell growth microenvironment, which mimics a physicochemical microenvironment and protects the cells from the immune attack from body.

Alginate-assisted enrichment and purification of mesenchymal stem cells

Jiao Lin,Xuewen Lu,Puchang Lie,Lingwen Zeng

The umbilical cord contains different populations of stem/progenitor cells such as mesenchymal, hematopoietic, trophoblastic and endothelial cells. However, the isolation methods currently in use are different, and the resulted cells are liable to cross contamination. Thus it is increasingly difficult to compare and contrast the study outcomes, which hinders progress in the field of stem cell applications. In an attempt to address this issue, we have used a magnetism assisted self-assembly method for cell capturing and releasing in combination with antibodies to mesenchymal stem cell antigens, CD105, CD73 and CD90, to identify and localize stem cells within the placenta umbilical cord. This approach is capable of detecting a minimum of 10 000 mesenchymal stem cells, and has also shown excellent specificity to distinguish other types of cells.
