Myco–macromolecular symbiosis: Chitosan–folate ZnO nanoplatforms from Trametes versicolor for dual-functional oncological and antibacterial therapy

Mojtaba Ranjbar,Masumeh Ahmadpour,Mahmoud Kiani,Mostafa Govahi

The increasing prevalence of antibiotic-resistant bacteria and cancer necessitates the development of novel, targeted therapeutic strategies. This study aimed to develop a multifunctional nanoplatform combining antibacterial and anticancer properties through green synthesis and strategic surface functionalization. Zinc oxide nanoparticles (ZnO NPs) were biosynthesized using Trametes versicolor extract as a reducing and capping agent, then surface-functionalized with chitosan (Cs) for enhanced biocompatibility and conjugated with folic acid (FA) for targeted delivery. The NPs were characterized using multiple analytical techniques and evaluated for antibacterial activity against Gram-positive ( Bacillus subtilis , Staphylococcus aureus ) and Gram-negative ( Escherichia coli ) bacteria. Cytotoxicity was assessed in cancer cells (MDA-MB-231) and normal cells (MCF-10 A), followed by gene expression analysis of key oncogenic and apoptotic markers. Characterization confirmed the formation of crystalline, core-shell structures with successful ligand attachment. Cs-ZnO NPs demonstrated significantly enhanced antibacterial activity against all tested bacterial strains compared to bare ZnO NPs. FA-Cs-ZnO NPs exhibited selective cytotoxicity toward cancer cells while maintaining biocompatibility with normal cells. Gene expression analysis revealed down-regulation of cancer stemness genes ( CD44 , SOX2 ) and proliferation markers ( mTOR , β-catenin ), coupled with up-regulation of the apoptotic marker Caspase 3 . This green-synthesized, dual-functional nanoplatform demonstrates promising potential for combined antibacterial and targeted anticancer therapy, warranting further in vivo evaluation for clinical translation.

Phoxim sublethal effect induces vitellogenin mediated reproductive enhancement and alters microbial symbiosis across generations in Hylyphantes graminicola

Fengjie Liu,Peijie Cheng,Lei Li,Wei Li,Chengjie Tu,Jing Shan,Wenjing Xiao,Jie Liu,Yu Peng,Yang Zhu

BACKGROUND Phoxim, a widely used organophosphate insecticide, poses potential risks to non-target natural enemies. Hylyphantes graminicola is a dominant predatory spider in agroecosystems, yet the sublethal effects and transgenerational impacts remain poorly characterized. This study aimed to systematically evaluate the physiological, molecular, and microbial changes in H. graminicola induced by low lethal concentration of phoxim exposure across two successive generations. RESULTS Laboratory bioassays determined the LC 30 of phoxim to be 9.442 mg/L. Exposure at this concentration significantly reduced female longevity but increased fecundity in both F 0 and F 1 generations, suggesting a potential hormetic effect. Transcriptomic analysis revealed that reproduction-related genes were significantly upregulated in the F 0 generation, whereas detoxification genes were markedly expressed in the F 1 generation. Functional validation through RNAi confirmed that vitellogenin ( Vg ) and cytochrome P450 ( CYP2J1 ) are crucial in reproduction and detoxification, respectively. Furthermore, acetylcholinesterase ( AChE ) was also found to be involved in regulatory phoxim exposure. Moreover, microbiome profiling demonstrated substantial shifts across generations, including decreased Wolbachia and increased Candidatus_Cardinium abundance, which may be related to the observed increase in fecundity. The results showed that a low lethal concentration of phoxim exposure can trigger complex physiological and microbial changes across generations. CONCLUSION These findings underscore the necessity of optimizing insecticide application intervals within Integrated Pest Management (IPM) frameworks to preserve biological control provided by beneficial arthropods. © 2026 Society of Chemical Industry.

Symbiosis culture of probiotic Escherichia coli Nissle 1917 and Lactobacillus rhamnosus GG using lactate utilization protein YkgG

Yu-Chi Tseng,Chengfeng Xue,I-Son Ng

The abundance and biodiversity of intestinal bacteria play a critical role in immune system. Escherichia coli Nissle 1917 (EcN), a tractable probiotic cell has emerged as a ‘smart microbe’ that can provide therapeutic modalities at the sites of disease. Additionally, Lactobacillus rhamnosus GG (LGG) represents one of the key probiotics in clinical studies with the ability to enhance intestinal immunity in symbiosis with other cells. To fully exploit the advantages of EcN and LGG, we optimized the commensal culture medium for LGG and EcN, succeeded by the establishment of a novel fluorescence-based colony quantification. From the metabolite analysis, more lactate was consumed when LGG is co-cultured with EcN; thus, YkgG was further explored as it was foreseen to be the enzyme responsible for lactate conversion to pyruvate. The YkgG enzyme from EcN has V max of 64.5 mM min -1 and catalytic ability k cat /K m of 2055 mM -1 min -1 , that is 6-fold compared to E. coli MG1655. Co-culture strategy of EcN with LGG in the mimic gastrointestinal tract increased the CFU by 50 % and 40 % within 5 mins and 10 mins, respectively. This practical, feasible, and promising symbiosis system aims to accelerate the application of probiotic in healthy food industries.

Symbiosis of 1 T and 2H phases in the basal plane of defective MoS2 nanoflowers for efficient hydrodesulfurization

Zhe Bai,Luyao Wang,Hongwei Cao,Xiangwen Zhang,Guozhu Li

Hydrodesulfurization (HDS) is an effective way to reduce sulfur content in oil for the enforcement of more stringent environmental regulations. MoS 2 has been widely used as an effective component for catalyzing HDS, and needs to be extensively studied in depth. Herein, MoS 2 nanoflowers with the symbiosis of 1T-2H phases in the basal planes and coexistence of S and Mo vacancies are prepared via the assembly of many nanosheets by a one-pot solvothermal method. The morphology, crystal phase structure and defects of these 1T-2H MoS 2 samples were well characterized. The 1T/2H ratio and the state of vacancies were effectively tuned by changing the solvothermal conditions. Active and stable 1T-2H mixed MoS 2 nanoflowers with high content of 1T phase and moderate S and Mo vacancies were obtained by the regulation of oxalic acid in the mixture of ethanol and water. In the HDS of dibenzothiophene at 300 °C and 4 MPa, the optimal catalyst of MoS 2 -water–ethanol-OA exhibited a substrate conversion of 89.3% in 10 h. Moreover, the activity of MoS 2 -water–ethanol-OA was well retained without any decrease in the recycling experiment.

Symbiosis between the components of a soft composite material responding to osmotic shock: The case of three-liquid systems

Wei Chen,Paul Clegg,Tao Li

Hypothesis For conventional high internal phase emulsions (HIPEs) with an external osmotic pressure greater than Laplace pressure, once the osmotic balance is broken, the swelling or shrinking of the aqueous phase can easily trigger phase separation. Mixing two immiscible dispersed phases in a double HIPE can evolve differently following an osmotic shock, which is expected to create a synergistic effect that can frustrate the phase separation of the system. Experiments Osmotic responses of double HIPEs were studied at the surface of a NaCl solution at a range of molarities . Fluorescence confocal microscopy studies were carried out to track the responses on microscopic scales. Measurements on surface tensions revealed the interfacial behaviors of the used surfactant . Findings A synergistic effect is achieved by a symbiotic process between the dispersed oils, where one type of droplets become more stable and pack around the other ones to halt their coalescence. The essential drive comes from the adsorption/desorption of surfactant molecules at oil–water interfaces. By directly adjusting the osmotic pressure difference, transitions between osmotic down-shock and osmotic up-shock can also be realized. This symbiosis greatly expands the potential technological applications of multiple-liquid systems, and can be used to design novel multi-functional composite materials.

Synchronous intervention of TCA cycle and lactic acid metabolism through self-enhanced cuproptosis and acidosis for tumor metabolic symbiosis destruction

Zhen Guo,Renlu Han,Yun Gao,Jie Fu,Zekai Zhuang,Yafei Hou,Jiancheng Yu,Keqi Tang

Abnormal metabolic symbiosis is one of the most typical features of tumors that distinguishes them from normal tissues. Disrupting tumor metabolic symbiosis has been recognized a promising tumor therapeutic approach. However, a single pathway intervention strategy greatly impedes its treatment efficiency. Herein, α-cyano-4-hydroxycinnamic acid (CHCA) and elesclomol (ES) co-loaded Cu 7 S 4 nanoplatform (named as CCE) is developed to synchronously intervene tricarboxylic acid (TCA) cycle and lactic acid metabolism through self-enhanced cuproptosis and acidosis for tumor metabolic symbiosis destruction. CCE could be triggered by up-regulated glutathione (GSH) in cancer cells to release ES, CHCA, Cu ions, and S 2− ions. On the one hand, ES significantly improves the influx efficiency of Cu ions to mitochondria where cuproptosis is initiated and TCA cycle is attacked. Meanwhile, the H 2 S produced by the binding of S 2− to H + in acidic tumor cells could induce mitochondrial damage and inhibit Cu ions efflux by depriving energy supply. Moreover, the GSH consumption reduces its chelation with Cu ions. Collectively, the increased intracellular Cu ions level significantly boosts cuproptosis mediated proteotoxicity of TCA cycle, thus aggravating TCA cycle injury. On the other hand, CHCA down-regulates the expression of monocarboxylate transporters 4 (MCT4) and inhibits lactic acid transport between tumor cells, causing the tumor cytoplasm acidification. Interestingly, H 2 S accelerates lactate production by promoting the glucose uptake to further potent the lactate metabolic disorder. In vitro and in vivo experiments confirm anti-tumor efficacy and biosafety of CCE. This study further enriches tumor therapeutic strategy.

One-Step Symbiosis of Bimetallic Peroxides Nanoparticles to Induce Ferroptosis/Cuproptosis and Activate cGAS-STING Pathway for Enhanced Tumor Immunotherapy

Bin Liu,Xiaorui Chen,Yanlin Zhu,Hao Chen,Jia Tan,Zhuang Yang,Jing Li,Pan Zheng,Lili Feng,Qingqing Wang,Shili Gai,Lei Zhong,Piaoping Yang,Ziyong Cheng,Jun Lin

To improve the efficiency and application prospects of metal peroxides in tumor therapy, the synthesis of bimetallic peroxides via simple yet effective approaches will be highly significant. In this work, hyaluronic acid modified zinc-copper bimetallic peroxides (ZCPO@HA) nanoparticles are synthesized through a one-step symbiotic method by co-hydrolysis of zinc acetate and copper acetate in weakly alkaline solution, followed by modification with sodium hyaluronate. Upon decomposition in the tumor microenvironment, ZCPO@HA nanoparticles can generate a considerable content of hydroxyl radical (·OH) by Fenton-like reaction between Cu 2+ and self-compensating hydrogen peroxide, while downregulating the expression of glutathione peroxidase 4 to induce ferroptosis. The abundant release of Cu 2+ leads to the aggregation of dihydrolipoamide S-acetyltransferase and the reduction of iron-sulfur cluster proteins, causing cuproptosis. The immunogenic cell death of tumor cells releases abundant damage associated molecular patterns, effectively activating the adaptive immune response. Zn 2+ and ·OH cause mitochondrial damage, leading to the release of a substantial amount of mitochondrial DNA. This subsequently activates the cyclic guanosine monophosphate-adenosine monophosphate synthase-stimulator of interferon genes (cGAS-STING) pathway, enhancing the innate immune response. In conclusion, it synthesizes a new type of bimetallic peroxides by one-step symbiosis for activating anti-tumor immunotherapy combined with immune checkpoint inhibitor.

Iron oxide-mediated enhancement of extracellular electron transfer and symbiosis in consortium of electroactive bacteria and microalgae for wastewater treatment

Pingyin Shu,Li Zhao,Xin Wen,Ziqi Wei,Chaowei Yuan,Huazu Liu,Xiangdong Zhang,Xizi Long,Qiang He,Wei Li

This study explores the role of α-Fe₂O₃ in improving extracellular electron transfer (EET) and symbiotic interactions between electroactive Shewanella oneidensis MR-1, its gene-deficient mutants (Δ mtrC , Δ omcA , and Δ cymA ), and microalgae ( Chlorella vulgaris ). The iron oxide facilitates the efficient transfer of electrons generated by MR-1 to microalgal photosystem via the pathway of CymA-MtrC-OmcA to α-Fe₂O₃. This process enhances the removals of TOC, TN, and NH₄⁺-N in the MR-1 bacterial-algal consortium by 9.99%, 12.32%, and 52.25% respectively via OmcA regulation while boosting phosphorus removal by 16.27% through CymA regulation. The consortium exhibits 26.76% lower CO₂ emission and 62.93% higher biomass productivity. When integrated into microbial fuel cells with Δ cymA mutants, α-Fe₂O₃ elevates open-circuit voltage by 283.33%, confirming its ability to compensate for electron deficiencies caused by CymA defects. α-Fe₂O₃ enhances energy metabolisms (TCA cycle, quinone pool, and photosynthesis) to modulate the key metabolites including starch/sucrose, glycolysis, amino acids, lipids, and quorum sensing. These adaptations strengthen the symbiotic interactions and utilization of MR-1 bacterial-algal consortium for carbon and nutrients. Reactor experiments validate that α-Fe₂O₃ integration with the consortium achieves 93.43% COD removal and 55.99% NH₄⁺-N removal, while reducing N₂O emissions by 61.37%. The results reveal the interplay between OmcA, CymA, and iron oxides in optimizing bacterial-algal consortia and underscore the molecular mechanisms underlying iron oxide-enhanced EET for developing low-carbon, resource-efficient wastewater treatment.

Enhancing biolipid production and self-flocculation of Chlorella vulgaris by extracellular polymeric substances from granular sludge with CO2 addition: Microscopic mechanism of microalgae-bacteria symbiosis

Xiaolei Liu,Bin Ji,Anjie Li

Microalgae-bacteria symbiotic systems were known to have great potential for simultaneous water purification and resource recovery, among them, microalgae-bacteria biofilm/granules have attracted much attention due to its excellent effluent quality and convenient biomass recovery. However, the effect of bacteria with attached-growth mode on microalgae, which has more significance for bioresource utilization, has been historically ignored. Thus, this study attempted to explore the responses of C. vulgaris to extracellular polymeric substances (EPS) extracted from aerobic granular sludge (AGS), for enhancing the understanding of microscopic mechanism of attached microalgae-bacteria symbiosis. Results showed that the performance of C. vulgaris was effectively boosted with AGS-EPS treatment at 12–16 mg TOC/L, highest biomass production (0.32±0.01 g/L), lipid accumulation (44.33±5.69%) and flocculation ability (20.83±0.21%) were achieved. These phenotypes were promoted associated with bioactive microbial metabolites in AGS-EPS (N-acyl-homoserine lactones, humic acid and tryptophan). Furthermore, the addition of CO 2 triggered carbon flow into the storage of lipids in C. vulgaris , and the synergistic effect of AGS-EPS and CO 2 for improving microalgal flocculation ability was disclosed. Transcriptomic analysis further revealed up-regulation of synthesis pathways for fatty acid and triacylglycerol that was triggered by AGS-EPS. And within the context of CO 2 addition, AGS-EPS substantially upregulated the expression of aromatic protein encoding genes, which further enhanced the self-flocculation of C. vulgaris . These findings provide novel insights into the microscopic mechanism of microalgae-bacteria symbiosis, and bring new enlightenment to wastewater valorization and carbon-neutral operation of wastewater treatment plants based on the symbiotic biofilm/biogranules system.

Dual transcriptomic and metabolomic analyses provide novel insights into the role of vitamins A and B metabolism in ectomycorrhizal symbiosis between Pinus yunnanensis and Lactarius deliciosus

Kai Su,Jing Yuan,Ran Wang,Lanlan Huang,Jian Zhan,Xueqiong Zhang,Shanping Wan,Xinhua He,Hans Lambers,Fuqiang Yu,Yanliang Wang

Ectomycorrhizal (EM) fungi play important roles in nutrient cycling and plant community establishment in forest ecosystems. Effects of EM formation on global alterations of the transcriptome and metabolome during plant-fungal interaction and the key metabolites involved in EM development are largely unknown. Here, dual RNA-Seq and untargeted metabolomic analyses were used to reveal stage-specific and core responses of Pinus yunnanensis and Lactarius deliciosus during mycorrhizal colonization. We found that L. deliciosus colonization in P. yunnanensis roots induced different transcriptional changes across three interaction stages, with a small core of genes consistently regulated at all stages. Concentrations of retinol (vitamin A) and retinoic acid increased while that of B group vitamins decreased during EM formation, which was coordinately regulated by these two plant-fungus partners, with L. deliciosus possibly playing a dominant role. Exogenous retinol altered the diameter and mantle thickness of P. yunnanensis - L. deliciosus EM tips and affected host plant growth and phosphorus acquisition . In the absence of L. deliciosus, exogenous retinol increased the root diameter and the number of root tips of P. yunnanensis . Furthermore, the concentration of auxin increased, but that of abscisic acid decreased during EM formation, and the genes involved in plant hormone signal transduction were gradually activated, and auxin and cytokinin signal transduction potentially played a positive role in this EM symbiosis. In conclusion, we propose that the interaction of P. yunnanensis and L. deliciosus alters vitamin metabolism, which may further affect plant hormone biosynthesis and signal transduction, modulating root morphology and EM traits.
