Purification and Structural Characterization of Polysaccharides from Polygonum multiflorum Thunb. and Their Immunostimulatory Activity in RAW264.7 Cells

Yan Gou,Donglin Gu,Jing Fan,Jianbo Yang,Ke Zan,Jingjing Liu,Hongyu Jin,Ying Wang,Feng Wei,Shuangcheng Ma

Polygonum multiflorumThunb. (PM) and derived products are broadly utilized in Chinese traditional medicine. According to our previous research, PM mostly contains polysaccharides, which display a wide range of biological activities. Two water-soluble polysaccharides (PMPs-1 and PMPs-2) were obtained from PM by DEAE-Cellulose and Sephadex G-100 column chromatography. Colorimetry, HPGPC-MALLS-RID, HPLC-PDA, methylation, FT-IR, NMR, and SEM were used to characterize these polysaccharides. PMPs-1 and PMPs-2 had average molecular weights of 255.5 and 55.7 kDa, respectively. PMPs-1 consisted of Man, Glc, Gal, and Ara at 0.9:78.6:1.0:1.6 and was a glucan with → 4)-Glcp-(1 → as a backbone. Meanwhile, PMPs-2, an acidic polysaccharide, comprised Rha, GalA, Glc, Gal, and Ara at 3.2:20.3:2.7:1.0:8.3. PMPs-1 and PMPs-2 significantly improved the proliferation of RAW 264.7 cells and induced NO, TNF-α, and IL-6 release. This study reveals that these two polysaccharides can be explored as novel immunomodulators and provide a basis for further development of PM in food and pharmaceutical industries.

The chemical structure and biological activities of a novel polysaccharide obtained from Fructus Mori and its zinc derivative

Ping-Ping Wang,Qiang Huang,Chun Chen,Li-Jun You,Rui Hai Liu,Zhi-Gang Luo,Mou-Ming Zhao,Xiong Fu

A novel polysaccharide (MFP4P) was isolated from mulberry fruits ( Morus alba L . ) pulp using a combination of DEAE-Sepharose fast-flow and Sephadex G-100 chromatographic columns. Structural characterization revealed that MFP4P was composed of arabinose, rhamnose, galactose, glucose and galacturonic acid with a molecular weight of 198.2 kDa. The linkage types of MFP4P were proved to be (1 → 6)-linked α-D-Glc, (1 → 2)-linked α-L-Rha, (1 → 3)-linked α-D-Gal, (1 → 3)-linked β-L-Rha and (1 → )-linked α-L-Ara. The MFP4P showed good antioxidant activity with an oxygen radical absorbance capacity value of 121.8 ± 16.5 μM trolox equivalent/g. In addition, the MFP4P promoted pancreatic cell proliferation, increased insulin secretion, and enhanced glucose consumption. In the non-toxic range, the polysaccharide and zinc complexes (MFP4P-Zn) exhibited higher antioxidant and hypoglycemic activities than MFP4P at the same concentration. In vivo hypoglycemic study, MFP4P and MFP4P-Zn both showed decreased blood glucose level and glucose tolerance compared with the model control group.

A peptide-Fe(II) complex from Grifola frondosa protein hydrolysates and its immunomodulatory activity

Biao Yuan,Cong Zhao,Chen Cheng,De-chun Huang,Shu-jie Cheng,Chong-jiang Cao,Gui-tang Chen

Grifola frondosa protein was hydrolysed using Alcalase, and the resulting hydrolysates (CGFP) were filtered using 5 and 1 kDa nominal cut-off ultrafiltration membranes and separated using Sephadex gel chromatography . Two main fractions, named GFP-1 and GFP-2 were obtained. CGFP-Fe and GFP-Fe were synthesized using ferrous chelation. Results showed that the GFP-2 fraction with molecular weight of 963 Da had the highest Fe(II) chelating activity (2680 ± 40 μg/g). UV spectrum showed that the Fe(II) shifted the maximum absorption to shorter wavelengths. The peptides measured using FTIR showed that the GFP-Fe peptide and the un-chelated peptide had different C–O and C= O absorption peaks. The effects of CGFP, CGFP-Fe, GFP2-2 and GFP-Fe on splenocytes proliferation and cytokines secretion were studied in vitro . Both the GFP-2 and GFP-Fe had good immune-enhancing activity. Furthermore, GFP-Fe showed an ability to promote the proliferation of splenocytes and peritoneal macrophages. The iron-chelating peptide of Grifola frondosa still maintained its immune enhancing activity after in vitro gastrointestinal digestion. The results suggested that GFP-Fe might be beneficial as a new iron supplement and as an immunological enhancement .

Purification, structural characterization and immunomodulatory activities of a polysaccharide from the fruiting body of Morchella sextelata

Meng-Ting Kuang,Jing-Yue Xu,Jin-Yu Li,Liu Yang,Bo Hou,Qi Zhao,Jiang-Miao Hu

A novel polysaccharide (MSP-1) was isolated from the fruiting body of Morchella sextelata and purified using DEAE-52 and Sephadex G-75. The molecular weight of MSP-1 was 1.17 × 10 4  Da, as detected by HPLC analysis. The monosaccharide composition of MSP-1 was mannose and glucose at a ratio of 1.00: 1.25. Methylation and NMR results revealed that the backbone of MSP-1 was composed of →4)- β -D-Man p -(1→, →4)- β -D-Glc p -(1→, →4)- α -D-Glc p -(1→, and →4, 6)- α -D-Glc p -(1→. SEM images of MSP-1 presented a dense network structure with porous characterizations. The immunomodulatory activities of MSP-1 were evaluated using RAW264.7 cells, and the results showed that MSP-1 promoted proliferative and phagocytic activity and increased the production of NO, TNF- α and IL-6. These results indicated that MSP-1 exhibited significant immunomodulatory activities.

Protective effects of a polysaccharide from Boletus aereus on S180 tumor-bearing mice and its structural characteristics

Jinling Zheng,Tingting Zhang,Jian Fan,Yongliang Zhuang,Liping Sun

A polysaccharide from the aqueous extract of Boletus aereus fruit (BAP) was isolated. The antitumor activities of BAP and/or cyclophosphamide (CTX) were investigated using the model of S180 tumor-bearing mice. Results indicated that BAP could effectively inhibit the growth of S180 solid tumors and protect the immune organs. Hematoxylin and eosin staining, Annexin V-FITC/PI staining, and mitochondrial membrane potential analysis demonstrated that BAP could induce the apoptosis of S180 tumor cells. In combination with CTX, BAP exhibited a significant synergistic antitumor effect on S180 cells. Furthermore, a novel polysaccharide, namely, BAPF, was purified from BAP by using DEAE Cellulose-52 column and Sephadex G-100 gel column. Structural characterization revealed that BAPF was primarily composed of mannose, glucuronic acid, glucose, galactose, arabinose, and fucose at a proportion of 12.98:1:16.8:16.48:1.08:9.1. Its average molecular weight was 1.79 × 10 6  Da. FTIR and NMR analyses demonstrated that BAPF was a pyranose with α-type and β-type glycosidic residues.

Structural Characterization of Polygonatum Cyrtonema Polysaccharide and Its Immunomodulatory Effects on Macrophages

Ruiding Wen,Lu Luo,Runcheng Zhang,Xudong Zhou,Wei Wang,Limin Gong

A neutralPolygonatumcyrtonemapolysaccharide (NPCP) was isolated and purified fromPolygonatumcyrtonemaby various chromatographic techniques, including DEAE-52 and Sephadex-G100 chromatography. The structure of NPCP was characterized by HPLC, HPGPC, GC-MS, FT-IR, NMR, and SEM. Results showed that NPCP is composed of glucose (55.4%) and galactose (44.6%) with a molecular weight of 3.2 kDa, and the sugar chain of NPCP was →1)-α-D-Glc-(4→1)-β-D-Gal-(3→. In vitro bioactivity experiments demonstrated that NPCP significantly enhanced macrophages proliferation and phagocytosis while inhibiting the M1 polarization induced by LPS as well as the M2 polarization induced by IL-4 and IL-13 in macrophages. Additionally, NPCP suppressed the secretion of IL-6 and TNF-α in both M1 and M2 cells but promoted the secretion of IL-10. These results suggest that NPCP could serve as an immunomodulatory agent with potential applications in anti-inflammatory therapy.

Purification and characterization of a novel angiotensin I-converting enzyme-inhibitory peptide derived from Alaska pollack skins

Guilan Yang,Song Qin,Wenjun Li

Hydrolysates containing angiotensin I-converting enzyme (ACE)-inhibitory peptide were prepared from protein of Alaska pollack skins using alcalase and trypsin. The protein hydrolysate was separated by ultrafiltration, Sephadex G-25 gel filtration chromatography and reversed phase high-performance liquid chromatography (HPLC), from which a novel purified peptide was obtained. Both random coil structure and β-sheet in the purified peptide were revealed in Fourier transform infrared spectrum. The amino sequence of the purified peptide was identified as GPLGVP, VLYPVK, VFLENVLR, and FEEF by HPLC-Q-TOF-MS (HPLC-quadrupole time-of-flight mass spectrometry). The peptide GPLGVP whose molecular weight was 538.31 Da showed the highest ACE inhibitory activity (IC 50 = 105.8 µM). The purified peptide featured a noncompetitive inhibition kinetic mechanism was shown in the Lineweaver–Burk plots and was susceptible to enzymes as indicated in the studies on stability of gastrointestinal proteases. Moreover, the peptide GPLGVP can combine ACE catalytic pocket through hydrogen bonds and other forces with high binding power as disclosed in molecular docking simulation, which provides the inhibitory effect of GPLGVP on ACE.
