This study introduces a novel modification method for micron-sized zero-valent iron, utilizing Shewanella oneidensis MR-1 to simultaneously colonize its surface, disrupt the passivation layer, and alter the crystal structure of the core. This process yields a composite that retains its reducing activity even in high concentrations of pollutants (500 ppm Cr(VI)) and under neutral to slightly acidic conditions. Our findings indicate that the modified system induces the Kirkendall effect in zero-valent iron, leading to the formation of amorphous zero-valent iron with a non-spherical core structure, in contrast to conventional direct mixing approaches. The modified system achieves complete reduction of 100 ppm Cr(VI) to Cr(III) within 8 h. Notably, within the first 4 h of reaction, the reduction rate is approximately three times higher than that of the conventional direct mixing system. XPS, XRD, SEM, EEM and TEM analyses reveal that the surface passivation layer of the biochemically modified zero-valent iron is disrupted, leading to changes in its crystal structure, the formation of hollow spherical structures, and the development of a biological protective film on its surface. Interestingly, electrochemical and ETSA analyses indicate that the electron transfer rate of the biochemically modified composite is significantly enhanced. This study provides fundamental insights into the removal of Cr(VI) from groundwater using biochemically modified micron-sized zero-valent iron (mZVI).

In materials science, “green” synthesis has gotten a lot of interest as a reliable, long-lasting, and ecofriendly way to make a variety of materials/nanomaterials, including metal/metal oxide nanomaterials . To accommodate various biological materials, green synthesis of metallic nanoparticles has been used (e.g., bacteria, fungi, algae, and plant extracts). In this work, Shewanella oneidensis MR-1 was used to biosynthesize palladium nanoparticles (bio Pd) under aerobic conditions for the Cr(VI) bio-reduction. The size and distribution of bio-Pd are controlled by adjusting the ratio of microbial biomass and palladium precursors. The high cell: Pd ratio has the smallest average particle size of 6.33 ± 1.69 nm. And it has the lowest electrocatalytic potential (−0.132 V) for the oxidation of formic acid , which is 0.158 V lower than commercial Pd/C (5%). Our results revealed that the small size and uniformly distributed extracellular bio-Pd could achieve completely catalytic reduction of 200 mg/L Cr(VI) solution within 10 min, while the commercial Pd/C (5%) need at least 45 min. The bio-Pd materials maintain a high reduction during five cycles. Microorganisms play an important role in the whole process, which can fully disperse palladium nanoparticles, completely reduce Cr(VI), and effectively adsorb Cr(III). This work expands our understanding and provides a reference for the design and development of efficient and green bio-Pd catalysts for environmental pollution control under simple and mild conditions.

Shewanella baltica is the specific spoilage microorganism of Pseudosciaena crocea during low-temperature storage. Exploring the correlation between cold adaptability and spoilage potential may provide a new perspective for prolonging shelf life of aquatic products. In the present study, we investigated the synthesis pathway of unsaturated fatty acid (UFA) responsible for regulating cold adaptability in Shewanella baltica and its effect on spoilage potential. FabR and FadR, as key regulators of membrane unsaturated fatty acids synthesis pathway, were identified in S. baltica . FabR was significantly down-regulated at 4 °C compared to at 30 °C, yet FadR displayed the opposite results. By overexpressing fabR and fadR genes at 4 °C, we found that FabR and FadR had negative and positive effects on UFA content and membrane fluidity as well as spoilage potential, respectively. These data indicated that FabR and FadR functioned collectively to increase the membrane fluidity for better cold adaptability at low temperature, resulting in the maintenance of spoilage potential of S. baltica .

How metal-reducing bacteria transfer electrons during dissimilatory energy generation under electron acceptor-limited conditions is poorly understood. Here, we incubated the iron and manganese-reducing bacterium Shewanella oneidensis MR-1 without electron acceptors. Removal of soluble extracellular organic compounds (EOCs) dramatically retarded transfer of electrons to an experimental electron acceptor, Cr(VI), by MR-1. However, the return of either high MW (>3000 Da) or low MW (<3000 Da) soluble EOCs produced by MR-1 to washed cells restored Cr(VI) reduction though Cr(VI) reduction was fastest when both size fractions were added together. Spectral and electrochemical characterization of EOCs indicated the presence of flavins and c-type cytochromes (c-Cyts). A model of the kinetics of individual elementary reactions between cells, flavins, released c-Cyts, and Cr(VI), including the direct reduction of flavins, released c-Cyts, and Cr(VI) by cells and the indirect reduction of Cr(VI) by reduced forms of flavins and released c-Cyts, was developed. Model results suggest that released c-Cyts could act as electron mediators to accelerate electron transfer from cells to Cr(VI), and the relative contribution of this pathway was higher than that mediated by flavins. Hence, extracellular c-Cyts produced by MR-1 likely play a role in extracellular electron transfer under electron acceptor-limited conditions. These findings provide new insights into extracellular electron shuttling and the metabolic strategy of metal-reducing bacteria under electron acceptor-limited conditions.

Cyclic diguanylate (c-di-GMP) is a second messenger involved in the regulation of various physiological processes in bacteria. However, its function in spoilage bacteria has not yet been addressed. Here, we studied the function of a tandem GGDEF-EAL domain protein, Sbal_3235, in the spoilage bacterium Shewanella baltica OS155. The deletion of sbal_3235 significantly reduced the c-di-GMP level, biofilm formation, and exopolysaccharide, trimethylamine (TMA), and putrescine production; sbal_3235 deletion also downregulated the expression of the torS and speF genes and affected membrane fatty acid composition. Site-directed mutagenesis in conserved GGDEF and EAL motifs abolished diguanylate cyclase (DGC) and phosphodiesterase (PDE) activity, respectively. These data indicate that Sbal_3235 is an essential contributor to the c-di-GMP pool with bifunctional DGC and PDE activity, which is involved in the biofilm formation and spoilage activity of S. baltica OS155. Our findings expand the biochemical role of c-di-GMP and uncover its link to spoilage activities, providing novel targets for food quality and safety controlling.

An exoelectrogens, Shewanella oneidensis MR-1 ( S. oneidensis MR-1), was supplied to a microbial fuel cell (MFC) to enhance the degradation of a recalcitrant organic compound, o -xylene. The experimental results revealed that, with the addition of the S. oneidensis MR-1, the o -xylene removal efficiency increased by 35–76% compared with the original MFC. The presence of the S. oneidensis MR-1 not only improved the activity of the biofilm in the bioanode but also developed the connections between the bacteria by nanowires . Therefore, the maximum power density increased from 52.1 to 92.5 mW/m 3 after the addition of the S. oneidensis MR-1. The microbial community analysis showed that adding the S. oneidensis MR-1 increased the biodiversity in bioanode. The dominant exoelectrogens shifted from Zoogloea sp., Delftia sp., Achromobacter sp., Acinetobacter sp., Chryseobacterium sp., and Stenotrophomonas sp. to Zoogloea sp., Delftia sp., Shewanella sp., Achromobacter sp., Hydrogenophaga sp., Sedimentibacter sp. and Chryseobacterium sp.. Furthermore, the cyclic voltammetry analysis showed that the outer membrane bound protein complex of OmcA–MtrCAB was involved as direct electron transfer pathway in the S. oneidensis MR-1 containing bioanode. We believed that this work is promising to provide optional strategy for efficient VOCs degradation by adjusting the microbial community in the bioanode.

The synergetic integration of living microorganisms and nonliving matrices into a new artificial micro-ecological system is a crucial challenge in bio-manufacturing especially towards the development of sustainable energy technology. Here we develop a way of creating a Shewanella oneidensis based conductive micro-niche to boost hydrogen production by integrating graphene, polydopamine and alginate. We reveal that both the respiratory metabolism induces localized hypoxic conditions inside the micro-niche and the wired-up extracellular electron reverses transfer pathway from the outer environment to the periplasmic hydrogenases, contributing to the enhanced hydrogen production rate which is about 12.7 folds enhancement compared with that of the free Shewanella oneidensis in solution. Significantly, the whole assembly process shows good biocompatibility, ensuring that the Shewanella oneidensis inside the micro-niche could maintain hydrogen production for 30 days. Overall, it is anticipated the hybrid integration of microorganisms and abiotic material will provide an effective strategy to modulate green biomanufacturer as well as enhance biological hydrogen production.

Some nano-biochars (nano-BCs) as electron mediators could enter into cells to directly promote intracellular electron transfer and cell activities. However, little information was available on the effect of nano-BCs on SMX degradation. In this study, nano-BCs were prepared using sludge-derived humic acid (SHA) and their effects on SMX degradation by Shewanella oneidensis MR-1 were investigated. Results showed that nano-BCs (Carbon dots, CDs, <10 nm) synthesized using SHA performed a better accelerating effect than that of the nano-BCs with a larger size (10–100 nm), which could be attributed to the better electron transfer abilities of CDs. The degradation rate of 10 mg/L SMX in the presence of 100 mg/L CDs was significantly increased by 84.6% compared to that without CDs. Further analysis showed that CDs could not only be combined with extracellular Fe(III) to accelerate its reduction, but also participate in the reduction of 4-aminobenzenesulphonic acid as an intermediate metabolite of SMX via coupling with extracellular Fe(III) reduction. Meanwhile, CDs could enter cells to directly participate in intracellular electron transfer, resulting in 32.2% and 25.2% increases of electron transfer system activity and ATP level, respectively. Moreover, the activities of SMX-degrading enzymes located in periplasm and cytoplasm were increased by around 2.2-fold in the presence of CDs. These results provide an insight into the accelerating effect of nano-BCs with the size of <10 nm on SMX degradation and an approach for SHA utilization.

The environmental fate and risks of mononitrophenols (mono-NPs), the simplest nitrophenols (NPs) often found in aquatic environments, are profoundly influenced by anaerobic bioreduction and co-existing electron shuttles (ESs), but little is known about the underlying mechanisms. Here, we elucidate the pathways of anaerobic mono-NPs bioreduction by Shewanella oneidensis MR-1 and assess the effect of model ESs on these processes. We found that all three mono-NPs isomers could be readily reduced to their corresponding aminophenols by S. oneidensis MR-1 under anaerobic conditions. CymA, a core component of the Mtr respiratory pathway, performs a dynamic role in these bioreduction, which is highly dependent on the bioreduction kinetics. The exogenous addition of quinones was found to accelerate the mono-NPs bioreduction through interactions with key outer-membrane proteins (e.g., OmcA and MtrC), and all these processes matched well to linear free energy relationships (LFERs). Surprisingly, adding riboflavin did not influence the bioreduction of all three mono-NPs isomers, which may be due to the contribution of OmcA and MtrC to these bioreduction processes and their downregulated expression. This study enhances our understanding of the environmental fate of mono-NPs and their bioconversion processes, providing valuable insights for the bioremediation of nitrophenol-contaminated sites.

The high performance of microbial extracellular electron transfer (EET) is essential for bioremediation and bioenergy generation. Carbon dots (CDs) modified electroactive microorganisms showed an increased electron transfer efficiency. However, the influence of CDs’ concentration the EET has not been illustrated before. Herein, we reported that the addition of CDs increased the current generation by accelerating the EET process. The maximum current output of microbial electrochemical cells with the addition of CDs was increased by more than 5 times compared to pure Shewanella oneidensis MR-1. With CDs addition, the increase of biofilm immobilization and flavins secretion improved the direct and mediated EET process. Electrochemical analyses clarified that the combination of cytochrome c with CDs resulted in a faster electron transfer rate. The overload of CDs would restrict their promotion effect on EET, which may be due to their biotoxicity to bacteria. This work offered the insight of CDs’ concentration on the efficiency of EET, not only promoting the related applications of EET but also inspiring the regulation of microorganism function via nanomaterial.

The microbial hybrid system modified by magnetic nanomaterials can enhance the interfacial electron transfer and energy conversion under the stimulation of a magnetic field. However, the bioelectrocatalytic performance of a hybrid system still needs to be improved, and the mechanism of magnetic field-induced bioelectrocatalytic enhancements is still unclear. In this work, γ-Fe2O3magnetic nanoparticles were coated on aShewanella putrefaciensCN32 cell surface and followed by placing in an electromagnetic field. The results showed that the electromagnetic field can greatly boost the extracellular electron transfer, and the oxidation peak current of CN32@γ-Fe2O3increased to 2.24 times under an electromagnetic field. The enhancement mechanism is mainly due to the fact that the surface modified microorganism provides an elevated contact area for the high microbial catalytic activity of the outer cell membrane’s cytochrome, while the magnetic nanoparticles provide a networked interface between the cytoplasm and the outer membrane for boosting the fast multidimensional electron transport path in the magnetic field. This work sheds fresh scientific light on the rational design of magnetic-field-coupled electroactive microorganisms and the fundamentals of an optimal interfacial structure for a fast electron transfer process toward an efficient bioenergy conversion.

Biogenetic nanocomposites research provides valuable methods for the green synthesis of nanomaterials. As a dissimilatory metal-reducing bacterium, Shewanella oneidensis MR-1 is used to reduce Ag + to Ag nanoparticles (Ag NPs) (diameter about 10 nm) under anaerobic conditions, resulting in the in-situ formation of Ag NPs immobilised on TiO 2 nanotubes (TNTs) (Ag/TNTs nanocomposites). The loading amount of Ag nanocrystals on the TNT surface can be controlled easily through adjusting the AgNO 3 concentration and further influence visible-light absorption efficiency of Ag/TNTs nanocomposites by decreasing the Ag loading amount. Ag/TNTs nanocomposites show superior photocatalytic efficiency under simulated sunlight than single TiO 2 nanomaterials. Moreover, the photocatalytic capacity of Ag/TNTs nanocomposites synergistic by S. oneidensis MR-1 is further enhanced and the degradation ratio of methylene blue reaches 92.3% within 30 min which attributed to a synergistic effect.
