Despite the importance of exogenous electron shuttles (ESs) in extracellular electron transfer (EET), a lack of understanding of the key properties of ESs is a concern given their different influences on EET processes. Here, the ES-mediated EET capacity of Shewanella putrefaciens 200 (SP200) was evaluated by examining the electricity generated in a microbial fuel cell. The results indicated that all the ESs substantially accelerated the current generation compared to only SP200. The current and polarization parameters were linearly correlated with both the standard redox potential (EES0) and the electron accepting capacity (EAC) of the ESs. A thermodynamic analysis of the electron transfer from the electron donor to the electrode suggested that the EET from c-type cytochromes (c-Cyts) to ESs is a crucial step causing the differences in EET capacities among various ESs. Based on the derived equations, both EES0 and EAC can quantitatively determine potential losses (ΔE) that reflect the potential loss of the ES-mediated EET. In situ spectral kinetic analysis of ES reduction by c-Cyts in a living SP200 suspension was first investigated with the EES, Ec-Cyt, and ΔE values being calculated. This study can provide a comprehensive understanding of the role of ESs in EET.

Lignin, as the abundant carbon polymer, is essential for carbon cycle and biorefinery. Microorganisms interact to form communities for lignin biodegradation, yet it is a challenge to understand such complex interactions. Here, we develop a coastal lignin-degrading bacterial consortium (LD), through “top-down” enrichment. Sequencing and physiological analyses reveal that LD is dominated by the lignin degrader Pluralibacter gergoviae (>98%), with additional rare non-degraders. Interestingly, LD, cultured in lignin-MB medium, significantly enhances cell growth and lignin degradation as compared to P. gergoviae alone, implying a role of additional outliers. Using genome-scale metabolic models, metabolic profiling and culture experiments, modeling of inter-species interactions between P. gergoviae , Vibrio alginolyticus , Aeromonas hydrophila and Shewanella putrefaciens , unravels cross-feeding of amino acids, organic acids and alcohols between the degrader and non-degraders. Furthermore, the sub-population ratio is essential to enforce the synergy. Our study highlights the unrecognized role of outliers in lignin degradation.

Summary The study aimed to assess the in vitro antibacterial effect of thymol (Thy) in conjunction with ultrasound (US) on Shewanella putrefaciens ( S. putrefaciens ). The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of Thy against S. putrefaciens were 0.50 and 1.00 mg mL −1 , respectively. US was found to augment the antibacterial efficacy of Thy, resulting in a significant reduction of 2.36 log CFU mL −1 in the total colony count of S. putrefaciens , ensuring that the bacterial population maintained a consistently low growth rate. Quantitative detection of crystal violet revealed that the combined treatment exhibited a pronounced inhibitory effect on biofilm formation, with a clearance rate of mature biofilms reaching 91.60%. Alkaline phosphatase (AKPase), lactate dehydrogenase (LDHase), UV absorbing substance permeability, and other indicators were employed to measure changes in the bacterial cell membrane and internal environment. Scanning electron microscopy (SEM) was further used to observe the bacteria morphology before and after treated by US and Thy, a notable collapse of bacterial cells was observed, accompanied by ruptured outer membranes, damaged cell walls, and extensive leakage of cellular contents. This study presents a novel approach for controlling bacterial growth in food preservation methods.

Background In order to detect the antimicrobial mechanism of combined treatment of cinnamon oil and gamma irradiation (GI), the membrane fatty acids and proteins characteristics of Shewanella putrefaciens ( S. putrefaciens ) treated with cinnamon oil and GI, and the distribution of cinnamon oil in S. putrefaciens were observed in this study. Results The membrane lipid profile of S. putrefaciens was notably damaged by treatments of cinnamon oil and the combination of cinnamon oil and GI, with significantly fatty acids decrease in C14:0, C16:0, C16:1, C17:1, C18:1 ( p < 0.05). The SDS-PAGE result showed that GI did not have obvious effect on membrane proteins (MP), but GI combined with cinnamon oil changed the MP subunits. Cinnamaldehyde, the main component of cinnamon oil, can not transport into S. putrefaciens obviously. It was transformed into cinnamyl alcohol in the nutrient broth with the action of S. putrefaciens . This indicated that the antimicrobial action of cinnamon oil mainly happened on the membrane of S. putrefaciens.
Conclusion Cinnamon oil could act on the membrane of S. putrefaciens with the damage of fatty acids and proteins, and GI would increase the destructive capability of cinnamon oil on the membrane fatty acids and proteins of S. putrefaciens .

Summary The antibacterial activity and mechanism of allicin (All) and antioxidant of bamboo leaves (AOB) against Shewanella putrefaciens was studied. The minimum inhibitory concentration (MIC) and growth curve were used to evaluate the antibacterial activity of combining All and AOB against S. putrefaciens . The inactivation mechanism was reflected by the changes in Na + /K + -ATPase activity, confocal laser scanning microscopy (CLSM), and the leakage of nucleic acid and protein. Scanning electron microscopy (SEM) was used to observe the morphology of bacterial cells and crystal violet biofilm reflected the ability to inhibit the formation of biofilm. Results showed that after being treated by All and AOB, S. putrefaciens was effectively inactivated, and the combined treatment had the strongest inactivation effect. The cell wall and cell membrane were destroyed by All and AOB, resulting in the release of cell contents. The results of Na + /K + -ATPase activity demonstrated that the membrane permeability could be changed, and the cell structural proteins could be destroyed by All and AOB. SEM observation further proved the effects of All and AOB on bacterial morphology destructing. Therefore, the combination of All and AOB can be used as an efficient method for the preservation of aquatic products.

This study aimed to enhance U(VI) removal from water by constructing a ternary system of Shewanella putrefaciens ( S. putrefaciens ), chlorite, and humic acid (HA). The ternary system achieved significantly higher U(VI) immobilization than single or binary systems, owing to synergistic effects of adsorption enrichment, electron shuttling, and bioreduction. Under optimal conditions, the U(VI) removal efficiency reached 91.6%. Importantly, pre-forming the ternary composite before U(VI) addition avoided false removal caused by HA-U(VI) complexation, ensuring accurate assessment of reductive immobilization. This work provides a reliable strategy for uranium-contaminated water remediation. Graphical abstract

Effluents loaded with various synthetic dyes are considered as a huge burden to the surrounding ecosystems. Sudan dyes are relatively difficult to decolorize due to its water-insolubility. In the present study, the strain Shewanella putrefaciens CN32 was firstly applied to decolorize Sudan dyes under the anaerobic condition , and the physicochemical parameters on the decolorization were optimized. The results demonstrated that the suitable decolorization condition was temperature 26 °C, initial pH 7.0–8.0 and NaCl concentrations 0–20 g/L. Electron competitive acceptors including nitrite, nitrate, dimethyl sulphoxide and oxygen could cause the significant inhibition to the decolorization of Sudan dyes. Biosurfactant rhamnolipid played a positive role in enhancing the decolorization of Sudan I. The co-culture of S. putrefaciens CN32 and Bacillus circulans BWL1061 is reported for the first time to accelerate the decolorization through improving the synergistic effect of enzymatic degradation and biological reductive effect. The highest decolorization of 90.23% to Sudan I was achieved within 108 h, suggesting that co-culture technique has a good potential in the treatment of dyeing wastewater. Furthermore, the microbial toxicity tests indicated that the toxicity of Sudan I to Escherichia coli BL21 and Bacillus subtilis 168 was obviously decreased after the decolorization.

ABSTRACT Plasma-activated water (PAW) is a new sanitizer that has received considerable attention for application in food industries. This research aimed to evaluate the effect of PAW on the inactivation of Shewanella putrefaciens and quality attributes of Yellow River carp ( Cyprinus carpio ) fillets. The carp fillet samples were immersed in sterile deionized water (SDW) or SDW activated by plasma discharge for 120 s (PAW120) for 1.5, 3.0, 4.5, or 6.0 min. After being treated by PAW120 for 6 min, the population of S. putrefaciens on carp fillets was significantly ( P < 0.05) decreased by 1.03 log CFU/g. Compared with SDW-treated samples, the L* value of PAW120–treated carp fillets was increased and the a* value was decreased after PAW120 treatment, whereas there was no significant ( P > 0.05) difference in the b* value. Compared with SDW, PAW120 caused no significant ( P > 0.05) changes in sensory properties and texture attributes of carp fillets including hardness, springiness, gumminess, and cohesiveness. However, 6-min PAW120 treatment caused a significant increase in the lipid oxidation level and a decrease in the pH value of the carp fillets. This work provides a basis for the potential application of PAW in the preservation of aquatic products.

Exploring the roles of dominant bacteria to spoilage is important in inhibiting spoilage. This study investigated related quality, protein degradation, and muscle microstructure in refrigerated large yellow croaker inoculated with Shewanella putrefaciens (S) and Pseudomonas fluorescens (P) as mono- or co-culture (SP). P. fluorescens had the most robust spoilage ability. P. fluorescens and the co-culture degraded major myofibrillar proteins and the links to disrupt muscle fibers, induced significant free water loss in muscle and formed amounts of trichloroacetic acid (TCA)-soluble peptides and total volatile basic nitrogen (TVB-N). S. putrefaciens caused severe mutual separation in myofibers. The competitive interactions between the two spoilage bacteria inhibited spoilage in the co-culture, while the numbers of differentially abundant peptides in the co-culture were enriched. Therefore, this study revealed competitive interactions of the co-culture of S. putrefaciens and P. fluorescens , which offered a thorough understanding of bacterial spoilage behavior in large yellow croaker.

Aquatic products are highly susceptible to microbial contamination and spoilage during storage, while traditional thermal sterilization often compromises their nutritional and sensory quality. In this study, a non-thermal dual-photosensitizer photodynamic treatment system composed of curcumin and octyl gallate was developed and activated under 420 nm light. The synergistic action of the two photosensitizers significantly enhanced the generation of reactive oxygen species (ROS), resulting in a 3.30 log reduction in Shewanella putrefaciens counts via membrane disruption, intracellular leakage, and genomic DNA degradation. Furthermore, the curcumin/octyl gallate-photodynamic treatment system was incorporated into a chitosan-based coating and applied to Litopenaeus vannamei . This treatment significantly inhibited increases in total viable counts, TVB-N, TBARS, and pH, thereby effectively extending the refrigerated shelf life. Microbial community analysis revealed marked shifts in bacterial composition, with suppression of dominant spoilage genera such as Psychrobacter, Pseudomonas , and Shewanella , confirming the broad-spectrum antibacterial efficacy of the treatment. Overall, curcumin/octyl gallate-photodynamic treatment represents an innovative, low-dose, and high-efficiency non-thermal sterilization approach for seafood preservation, offering a feasible alternative to traditional methods while maintaining microbial safety and product quality.

Citral exerted its antibacterial activities in a dose-dependent manner in the three groups. • Citral increased intracellular oxidative stress levels of S. putrefaciens and P. fluorescens in mono- and co-culture. • Citral reduced motility and weakened biofilm formation of S. putrefaciens and P. fluorescens in mono- and co-culture. • Citral treatment can prolong the shelf life of large yellow croaker. Shewanella putrefaciens and Pseudomonas fluorescens , as significant specific spoilage organisms in aquatic products, can regulate cell density and a variety of physiological processes through quorum sensing (QS). This research examined the antibacterial mechanism and antivirulence efficacies of citral (CT), as a quorum sensing inhibitor (QSI), against S. putrefaciens and P. fluorescens in mono- and co-culture. Furthermore, the application of CT to large yellow croaker preservation was investigated. Results revealed that CT exerted its antibacterial activities in a dose-dependent manner. CT increased intracellular oxidative stress levels and malondialdehyde (MDA) content, decreased ATP content and peroxidase activity, and promoted the damage of cell membrane, especially in S. putrefaciens group. These two bacteria had a synergistic effect in causing spoilage through biofilm formation and motility. The antivirulence efficacies of CT against the co-culture group were weaker than those of in the mono-culture groups. CT was also found to suppress the expression of key virulence regulators in S. putrefaciens and P. fluorescens virulence, including Autoinducers-2 (AI-2) production in the QS system. Furthermore, CT had the potential capability in the preservation of large yellow croaker. Consequently, these results proved CT could combat bacteria and had antivirulence efficacies against S. putrefaciens and P. fluorescens in mono- and co-culture. Specific spoilage organisms (SSOs) play a crucial role in causing the deterioration of aquatic products. This study investigated the antibacterial activity and mechanism of exogenous autoinducers through biochemical and metabolomics analysis against Shewanella putrefaciens . The potential application of exogenous autoinducers on large yellow croaker preservation was further investigated. C4-HSL, C10-HSL and C14-HSL with the strongest antibacterial activity on S. putrefaciens were selected, from the aspects of growth curve, biofilm formation, and motility ability, which damaged the integrity of cell membrane. Metabolomics analysis demonstrated C10-HSL significantly affected key metabolic pathways including alanine, aspartate and glutamate metabolism, glycine, serine and threonine metabolism, arginine biosynthesis, phenylalanine metabolism, tricarboxylic acid (TCA) cycle, pyrimidine and purine metabolism. Furthermore, C10-HSL exhibited the most effective preservation performance for large yellow croaker. These findings highlighted the antibacterial mechanism of exogenous acylated homoserine lactones (AHLs) and confirmed the potential of exogenous AHLs to prolong the shelf-life of aquatic products.
