The first snail shell-shaped helical polyacetylenes (SHPAs) were synthesized from the copolymerization between l -serine-grafted propargylamine (LSA) and propargylamine (PA). The novel structure was obtained at the monomer input proportion of 50%. The monomer (LSA) and copolymers (poly(LSA m - co -PA n )) were characterized by 1 H nuclear magnetic resonance, Fourier transform infrared spectroscopy, ultraviolet visible spectroscopy, circular dichroism and transmission electron microscopy. The infrared emissivity of poly(LSA 50 - co -PA 50 ) investigated at 8–14 μm was 0.451, which was much lower than the other copolymers. This was because the poly(LSA 50 - co -PA 50 ) could form the obvious intramolecular interaction and self-assembled to a snail-shell shape by intermolecular hydrogen bonding in MeOH solution. Meanwhile, the conjugation performance of the main chain was enhanced. Consequently, the abundant hydrogen bonds formed through N–H and C=O bonding and the strong conjugate properties both could decrease the unsaturation degree, as well as change the heat conduction mode in the molecules, thereby reducing the infrared emissivity. This strategy provided a new direction to achieve the controllable infrared stealth.

Natural biofilms developing on mineral substrates at the soil-water interface in freshwater ecosystems are important sinks of per- and polyfluoroalkyl substances (PFAS). However, the role of mineral-biofilm interactions in regulating their trophic transfer remains poorly understood. This study investigates how Fe 2 O 3 is associated with changes in the distribution of PFAS within the biofilm matrix in a carbon chain length-dependent manner, which may further affect PFAS trophic transfer to snails. Fe 2 O 3 preferentially localizes within the tightly bound extracellular polymeric substances (TB-EPS) layer, coincides with stimulated EPS production and altering PFAS partitioning within biofilms, and correlates with the redistribution of long-chain PFAS (e.g., PFNA) from loosely bound to tightly bound EPS fractions. In addition, Fe 2 O 3 alleviates PFNA-induced toxicity and reshapes microbial community composition, increasing the relative abundance of Cyanobacteria and improving biofilm nutritional quality, which in turn may enhance snail grazing. Consequently, Fe 2 O 3 is associated with a significant increase in the trophic transfer factor of PFNA from biofilms to snails (approximately 35%), while showing negligible effects on short-chain PFAS. Overall, these results suggest that Fe 2 O 3 may enhance the trophic transfer of long-chain PFAS through combined effects on contaminant partitioning and biofilm quality, highlighting the important role of mineral-associated biofilms in PFAS bioaccumulation and providing new insights for ecological risk assessment in freshwater ecosystems.

Benzo[a]pyrene (BaP) and its metabolic end product benzo(a)pyren-7,8-dihydrodiol-9,10-epoxide (BPDE), are known toxic environmental pollutants. This study aimed to analyze whether sub-chronic BPDE exposure initiated pulmonary fibrosis and the potential mechanisms. In this work, male C57BL6/J mice were exposed to BPDE by dynamic inhalation exposure for 8 weeks. Our results indicated that sub-chronic BPDE exposure evoked pulmonary fibrosis and epithelial-mesenchymal transition (EMT) in mice. Both in vivo and in vitro , BPDE exposure promoted nuclear translocation of Snail. Further experiments indicated that nuclear factor erythroid 2-related factor 2 (Nrf2) and p62 were upregulated in BPDE-exposed alveolar epithelial cells. Moreover, Nrf2 siRNA transfection evidently attenuated BPDE-induced p62 upregulation. Besides, p62 shRNA inhibited BPDE-incurred Snail nuclear translocation and EMT. Mechanically, BPDE facilitated physical interaction between p62 and Snail in the nucleus, then repressed Snail protein degradation by p62-dependent autophagy-lysosome pathway, and finally upregulated transcriptional activity of Snail. Additionally, aryl hydrocarbon receptor (AhR) was activated in BPDE-treated alveolar epithelial cells. Dual-luciferase assay indicated activating AhR could bind to Nrf2 gene promoter. Moreover, pretreatment with CH223191 or α-naphthoflavone (α-NF), AhR antagonists, inhibited BPDE-activated Nrf2-p62 signaling, and alleviated BPDE-induced EMT and pulmonary fibrosis in mice. Taken together, AhR-mediated Nrf2-p62 signaling contributes to BaP-induced EMT and pulmonary fibrosis.

Trans-endothelial migration (TEM) is a critical rate-limiting barrier in tumor metastasis, yet its mechanistic basis remains poorly defined. Here we show that α-ketoglutarate (α-KG) and its metabolic enzyme glutamate dehydrogenase 1 (GLUD1) constitute a pivotal regulatory axis governing this process. Mechanistically, GLUD1 translocates to the nucleus, where α-KG binds to its non-catalytic site (K147) to induce a conformational change that stabilizes the GLUD1-SNAIL interaction, promoting assembly of a nuclear ternary complex. Notably, this complex is preferentially enriched in tumors, and specific blockade of K147 disrupts this assembly and abrogates metastatic effects. Our findings reveal a non-canonical role for GLUD1 and α-KG in transcriptional regulation and propose a therapeutic paradigm that selectively interferes with metabolite-dependent complex formation rather than enzymatic activity, offering a precise strategy to target TEM-driven metastatic cells.

A SDIE system characterized by a biomimetic snail-shell structure is fabricated • The highest water evaporation of 2.21 kg m −2 h −1 was obtained from CNT-Ag@PA12 • Antibacterial and salt-resistant design ensures 20-day stable operation without decay • Collected water showed potential for irrigation and household water use. Solar-driven interfacial evaporation can help reduce the problem of global freshwater shortages. However, it remains challenging to integrate devices that simultaneously achieve high-efficiency light absorption, rapid water transport capability, excellent evaporation effect, and strong salt tolerance. This work develops a bio-inspired snail-shell-shaped evaporation device by optimizing the design with the aid of simulations and experiments. A shell-shaped structure with capillary channels that can rapidly convey water to sustain an ultra-thin-water layer stability. In addition, we applied silver-nanoparticle-decorated carbon nanotubes (CNT Ag) composites that contain carbon nanotubes and silver nanoparticles on the top for photo-thermal functions. Multi-physics simulation determined the main geometric parameters required by this structure; experimentally validated the optimized design of this structure. The CNT-Ag film exhibited a broad-band solar absorption coefficient of up to 99.1% and efficiently localized heat at the evaporation boundary. As a result of combining the strengths of both structures and materials, the evaporator achieved an evaporation rate of 2.21 kg m −2 h −1 under one-sun irradiation. Furthermore, the directional salt-formation ability brought about by the structure and the antibacterial and anti-biocontamination capabilities caused by silver ions enable the evaporator to exhibit long-term stability and effective seawater desalination capabilities. In summary, this work presents a practical approach to building durable and effective solar desalination using a combination of biologically inspired structure design, composite photothermal materials, and regulation of heat and mass transfer. Multispecies biofilm exhibited high resistance to nanotoxicity by secreting extracellular polymeric substances (EPS) and undergoing alterations in the community composition. Scarce information was available to assess how these changes could further influence the transfer of nanoparticles (NPs) through the biofilm-based food chain. Photic biofilm was exposed to two distinct NPs (ZnO and TiO 2 ) and subsequently grazed by snails. Exposure to different NPs led to variations in biomass, chlorophyll content, EPS productivity, alpha diversity, and community composition of the photic biofilm. The presence of ZnO NPs facilitated the growth of phylum Cyanobacteria while TiO 2 promoted EPS productivity of photic biofilm. EPS were capable of embedding NPs (TiO 2 and ZnO) within its matrix, thereby mitigating their aggregation within the biofilm matrix. These alterations were subsequently confirmed to have an impact on the trophic transfer factors (TTF) of NPs through the constructed biofilm-snail food chain. The TTF of ZnO was lower than that of TiO 2 in feeding scenario 1 (only fed on TiO 2 or ZnO biofilm) but higher than that of TiO 2 in feeding scenario 2 (fed on TiO 2 and ZnO biofilm simultaneously), which was attributed to the shifts in the algae composition and a smaller size of ZnO NPs in EPS. This study demonstrated that the response of photic biofilm to NPs further affected the TTFs of NPs through the food chain.

Diabetic foot ulcers (DFUs) as a nonhealing wound remain a clinical challenge, and the development of pro-healing and cost-effective drugs is in urgent need. Herein, we reported a novel galactosylated glycosaminoglycan (GAG) from the snail Helix lucorum , as an effective pro-healing compound. The snail GAG is composed of a heparan sulfate-like main chain and galactose side chains at C-3 of GlcNAc residue. Its main chain has a repeating disaccharide structure of → 4)-α-D-GlcNAc-(1 → 4)-α-L-IdoA 2S (1 →. This is the first example of glycosaminoglycan with galactose branches from mollusks. Pharmacological experiments showed that the H. lucorum GAG significantly promoted skin wound healing in both healthy and diabetic mice by accelerating granulation tissue regeneration, angiogenesis, and collagen deposition. The distinctive galactosylated substitution may play an important role on its pro-healing activity. Our discovery enriches the diversity of non-anticoagulant heparan sulfate-like glycosaminoglycans, and provides a potential candidate of pro-healing drug for treating diabetic wound.

Bioinspired GO nanofluids achieve near-zero wear under 2.46 GPa. • Hydrodynamic lubrication and tribofilms enable durable performance. • Strategy offers enhanced near-zero wear for engineering steel. Near-zero wear behavior of engineering steel is instrumental to achieving unprecedented performance in longevity, reliability, and precision for advanced mechanical systems. However, achieving near-zero wear is typically accomplished under mild conditions and with specific friction pairs, which makes it difficult to meet the strategic requirements for high loads and general applicability. Herein, inspired by the advanced lubrication of snail mucus, biomimetic graphene-based nanofluids (GO NFs) were synthesized as lubricants by grafting graphene oxide (GO) with organosilane (KH560) and polyethyleneimine (PEI) via covalent linkage. Importantly, the obtained nanofluids demonstrate significant near-zero wear performance on steel substrate under a high contact pressure of 2.46 GPa, achieving an ultralow wear rate of 1.32 × 10 −10 mm 3 ·N −1 ·m −1 and corresponding friction coefficient reduced to 0.068. Further analysis of the worn surfaces and lubrication regimes calculations reveals that the synergistic combination of hydrodynamic lubrication, replenishable adsorption films, and protective tribochemical films collectively contributed to the effective friction reduction and near-zero wear on the steel substrate. This work offers new insights into the lubrication mechanisms of GO NFs and provides guidance for designing high-performance lubricants with enhancing near-zero wear characteristics of engineering steel. Flexible supercapacitors (SCs) have attracted considerable attention for wearable electronics, and developing high-performance electrolytes is critical for their practical application. While hydrogels have been widely investigated as solid electrolytes, studies on double-network (DN) hydrogel electrolytes specifically addressing the electrode–electrolyte interface stability under mechanical deformation remain relatively scarce. A major obstacle is maintaining a stable electrode–electrolyte interface under large mechanical deformation. Drawing inspiration from the mucus of a snail, which effectively adheres to various surfaces in challenging conditions, we present a self-healing xanthan gum/hydrophobically associated polyacrylamide/NaCl (XG/HPAAm/NaCl) hydrogel polymer electrolyte (HPE) that facilitates the creation of flexible SCs with improved mechanical and electrochemical properties. The optimized 2 wt% XG/HPAAm/0.4 M NaCl DN HPE exhibits a high ionic conductivity of 4.0 S/m, a tensile strength of 0.43 MPa, and an elongation at break of 11.7 mm/mm, along with a high adhesive energy of 254.7 J/m 2 . The tough HPE was coated with a mixed adhesive of 502 cyanoacrylate glue and triethyl citrate (TEC) to create a surface coating resembling “mucus”, onto which activated carbon (AC)-modified carbon cloth (CC) electrodes (CC/AC) were affixed on both sides to construct the flexible SCs. Investigations into the HPE’s characteristics and the SCs’ electrochemical performance at various bending angles reveal that the “mucus-coating” HPE exhibits strong electrode adhesion and significantly improved electrochemical performance. The assembled flexible SC delivers a high specific capacitance of 249.3 F/g at 0.30 A/g, retains 73.4% of its initial capacitance after 20,000 cycles, and maintains 86.9% capacitance retention under 180° bending, outperforming SCs assembled with original HPEs in both performance and stability. This approach provides a versatile method for improving the interfacial properties between electrodes and HPEs, paving the way for innovative applications in robust, self-healing, and flexible devices.

Background Although the presence of intermediate snails is a necessary condition for local schistosomiasis transmission to occur, using them as surveillance targets in areas approaching elimination is challenging because the patchy and dynamic quality of snail host habitats makes collecting and testing snails labor-intensive. Meanwhile, geospatial analyses that rely on remotely sensed data are becoming popular tools for identifying environmental conditions that contribute to pathogen emergence and persistence. Methods In this study, we assessed whether open-source environmental data can be used to predict the presence of human Schistosoma japonicum infections among households with a similar or improved degree of accuracy compared to prediction models developed using data from comprehensive snail surveys. To do this, we used infection data collected from rural communities in Southwestern China in 2016 to develop and compare the predictive performance of two Random Forest machine learning models: one built using snail survey data, and one using open-source environmental data. Results The environmental data models outperformed the snail data models in predicting household S. japonicum infection with an estimated accuracy and Cohen’s kappa value of 0.89 and 0.49, respectively, in the environmental model, compared to an accuracy and kappa of 0.86 and 0.37 for the snail model. The Normalized Difference in Water Index (an indicator of surface water presence) within half to one kilometer of the home and the distance from the home to the nearest road were among the top performing predictors in our final model. Homes were more likely to have infected residents if they were further from roads, or nearer to waterways. Conclusion Our results suggest that in low-transmission environments, leveraging open-source environmental data can yield more accurate identification of pockets of human infection than using snail surveys. Furthermore, the variable importance measures from our models point to aspects of the local environment that may indicate increased risk of schistosomiasis. For example, households were more likely to have infected residents if they were further from roads or were surrounded by more surface water, highlighting areas to target in future surveillance and control efforts.

Copper oxide nanoparticles (CuO-NPs) may pose high ecological risks to aquatic ecosystems. While sediments are the final destinations for CuO-NPs, little is known about the potential ecotoxicity of sediment-associated CuO-NPs on freshwater deposit-feeding macroinvertebrates. The gastropod Bellamya aeruginosa was chosen as an ecotoxicological test species. Adult snails were exposed to Cu (180 μg/g dry weight (DW)) added to sediments in the form of CuO-NPs, CuO microparticles (CuO-MPs, size control), and CuSO 4 (solubility control) for 7, 14, and 28 days, Cu burdens in different tissues and biomarkers of oxidative stress were determined to understand Cu accumulation differences among tissues, potential mechanisms of Cu uptake, time-effect relationships, particle size effects, and the relative contribution of toxicity from CuO-NPs and its soluble Cu ions. There was no difference in Cu ion concentrations in porewaters between the CuO-NPs and CuO-MPs treatments. In addition, relatively low Cu ion concentrations in porewater might indicate their remarkably low solubility. The hepatopancreas and gonad of B. aeruginosa are the primary target tissues for Cu accumulation. Cu accumulation in the hepatopancreas and gonad from CuSO 4 treatments was consistently higher than that from the CuO-NPs and CuO-MPs treatments. After long-term exposure, Cu accumulation was higher from CuO-NPs than from CuO-MPs, especially, the Cu accumulation rate from CuO-NPs was greater than that from CuSO 4 . Short-term exposure to the three Cu forms caused oxidative stress to the hepatopancreas. CuO-MPs did not cause oxidative damage. Long-term exposure to CuO-NPs and CuSO 4 resulted in oxidative damage. Overall, prolonged exposure to CuO-NPs will increase the ecotoxicity risk to B. aeruginosa . Although there was no difference in Cu accumulation between the CuO-NPs and CuO-MPs treatments after 14 days of exposure, pronounced oxidative damage was caused by exposure to CuO-NPs but not to CuO-MPs, implying that toxicity of CuO-NPs could be attributed to specific nanoparticulate effects.

Methionine sulfoxide reductase B1 (MsrB1) can catalyze both free and protein-bound R-methionine sulfoxides (R-MetO) to methionine (Met). It has been reported that MsrB1 plays an important role in the development of HCC and human bone osteosarcoma. However, little is known about the functions of MsrB1 in human colorectal cancer (CRC). Herein, we detected MsrB1 expression level in CRC tissue and cell lines, and investigated the effect of MsrB1 knockdown on CRC phenotypes and possible mechanisms involved in. The results showed that MsrB1 was highly expressed in both CRC tissues and cell lines, and that cell proliferation, migration and invasion were significantly inhibited, but apoptosis was increased after MsrB1 knockdown in colorectal cancer HCT116 and RKO cell lines, compared to control siRNA group. In addition, E-cadherin protein level was increased, vimentin and Snail protein were greatly decreased after knockdown of MsrB1 in cells. Furthermore, pGSK-3β (Ser9) and β-catenin protein levels were reduced, the promoter activity of TCF/LEF construction was inhibited after MsrB1 knockdown in cells, suggesting that GSK-3β/β-catenin signaling axis was involved in the tumorigenesis of CRC. In conclusion, the oncogenic role and related mechanisms of MsrB1 in CRC discovered in our work determined the potential role of MsrB1 as a biomarker and may provide a new target for clinical therapy of CRC.
