Fluorescent gold nanoclusters (Au NCs) have attracted considerable interest in biological application. Here, we reported a novel Au NCs with blue fluorescence, conjugated with glycine (Gly) and rotenone (R). The structure and properties of Au–BSA–Gly–R conjugates were characterized by transmission electron microscopy (TEM), UV–visible, fluorescence spectroscopy, Fourier transform infrared spectroscopy (FTIR) and Zeta potential measurement. The results of fluorescent microscopic imaging revealed that the Au NCs with different ligands were capable of being selectively uptaken by Spodoptera frugiperda (Sf9) cells. Furthermore, the gold content of Au–BSA–Gly–R in Sf9 cells was 2.2 and 2.6 times higher than that of Au–BSA and Au–BSA–R, respectively. It illustrated that nanoparticles after conjugated with Gly could gain specificity to intracellular transportation. This study indicated that the Au NCs after mediated by Gly not only had great potential for applications in insect cell imaging, but also could play an important role of amino acid for agrochemicals delivery in agricultural fields.

BACKGROUND Spodoptera frugiperda and Mythimna loreyi are sympatric pests of corn and other gramineous crops. In addition, the two species share three primary pheromone components Z 9-14:Ac, Z 7-12:Ac and Z 11-16:Ac, resulting in capture of males from both species by commercially available lures, which poses a great challenge for species-specific population monitoring. RESULTS To increase the specificity of the pheromone blend used in lures, we conducted comparative investigations between the two species, with respect of female calling rhythms, components in female pheromone glands, male electrophysiological and sexual excitement responses to pheromone gland components. Based on differences between the two species obtained from these investigations, we further designed and tested a variety of blend formulations for the species-specificity in the field trapping assays, with the reported ternary blend for S. frugiperda ( Z 9-14:Ac/ Z 7-12:Ac/ Z 11-16:Ac = 88/1.0/11) as a positive control. The results showed that the ratio of component Z 7-12:Ac is the major determinant of the male trapping specificity of the blends in lures. Compared to the positive control, increasing the ratio of Z 7-12:Ac to 10 in the ternary blend greatly improved the specificity and captures for M. loreyi males, while reducing its ratio to 0.4 significantly improved the specificity and retained the capture ability for S. frugiperda males. CONCLUSION The optimal blend was determined to be Z 9-14:Ac/ Z 7-12:Ac/ Z 11-16:Ac at a ratio of 88/0.4/11 for S. frugiperda , and 88/10/11 for M. loreyi . These findings offer highly species-specific sex pheromone formulations for population monitoring and male trapping of the two sympatric pests. © 2025 Society of Chemical Industry.

The effectiveness, tolerance, and safety of pesticides must be established before their scientific or rational. This study evaluates the field control efficacy of broflanilide, tetraniliprole, and chlorantraniliprole in combating Spodoptera frugiperda in maize crops, as well as the resistance of S. frugiperda to these three diamide pesticides after exposure. By assessing field control efficiency, toxicity, effects on development and reproduction, and detoxification enzyme activity of these diamide pesticides on S. frugiperda, highlights broflanilide's significant insecticidal potential. A highly sensitive and efficient method using QuEChERS/HPLC MS/MS was developed to simultaneously detect residues of these three pesticides on maize. Initial concentrations of broflanilide, tetraniliprole, and chlorantraniliprole ranged from 2.13 to 4.02 mg/kg, with their respective half-lives varying between 1.23 and 1.51 days. Following foliar application, by the time of harvest, the terminal residue concentrations of these pesticides were all under 0.01 mg/kg. Chronic dietary intake risk assessments and cumulative chronic dietary exposure for three pesticides indicated that the general population's terminal residue concentration was within acceptable limits. Not only does this research provide valuable insights into field control efficiency, insecticidal effects, resistance, residues, and risk assessment results of broflanilide, tetraniliprole, and chlorantraniliprole on maize, but additionally, it also paves the way for setting suitable Maximum Residue Limits (MRLs) values based on pre-harvest interval values, rational dosage, and application frequency.

As one of the major threats to global food security, Spodoptera frugiperda ( S. frugiperda ) is highly gaining consideration due to its severe damage. Matrine is a widely and effectively used botanical insecticide in controlling S.frugiperda but lacks a rapidly available effect. To further improved the insecticidal activity of matrine based on combination principles, this work synthesized five new pyrazole matrine derivatives (PMDs) using Michael addition and investigated insecticidal activity against 2nd instar larvae of S. frugiperda ( in vivo ) and its isolated cell( in vitro ). Our result demonstrated that PMDs show higher pesticidal activity than that matrine in both in vitro and in vivo assays. The most toxic derivatives in vitro and in vivo are PMD-3 and PMD-1, with IC 50 of 2.49 mM and LC 50 of 22.76 mg/L respectively. This research also investigates the anti-proliferation mechanism of PMDs based on isolated cells. PMDs decrease mitochondria membrane potential , arrested cell cycle at the G2/M phase, and upregulated Caspase 3 , Caspase 9 , and Apaf-1 to induce Caspase-dependent apoptosis. For Caspase-independent apoptosis, AIF and Endo G were found to be upregulated. Besides, pro-apoptotic factors like p53, IBM-1, and anti-apoptotic factors like IAP were upregulated. Moreover, we supposed that there was a linkage between lysosomes and PMD-induced apoptosis according to increased apoptosis rate , activated lysosomes, and upregulated Cathepsin B . This research provides new ideas for the synthesis of matrine derivatives and further demonstrated the anti-proliferation mechanism of PMDs.

Damage to the growing ears by Spodoptera frugiperda is a major economic burden, therefore, the development of a novel targeted treatment with safe, environmentally friendly, and effective is necessary to control the damage of S. frugiperda in maize ears. In this study, chitosan/sodium alginate hydrogel rings loaded with chlorantraniliprole (CLAP) were prepared by physical embedding and ionic cross-linking method. The CLAP-loaded hydrogel rings were evaluated for their morphology, loading efficiency, encapsulation efficiency, and release kinetics at different pH levels. The efficacy of the CLAP-loaded hydrogel rings and CLAP suspension were evaluated under field conditions for control of S. frugiperda in maize ears, and the distribution and dissipation of CLAP on maize ears were investigated. The results of the two-year trial showed that the insecticidal effects of the CLAP-loaded hydrogel rings treatment were significantly improved contrasted with the use of CLAP suspension, and increased the transfer efficiency of the pesticide from maize bracts to maize kernels. The terminal residues showed that it is safe to use CLAP suspension and CLAP-loaded hydrogel rings on maize ears. This study provides guidance for the use of pesticides to control pests in maize ears in rainfall-rich areas.

Simple SummaryThe fall armyworm is a notorious lepidopteran pest that consumes many economically important crops. Its recent invasion into China threatens crops in over 19 provinces. This species is susceptible to its homologous nucleopolyhedrovirus (Spodoptera frugiperdamultiple nucleopolyhedrovirus, or SfMNPV). Selection of indigenous isolates that are adaptable in each geographical region is important for developing a virus-based pesticide. In this study, an SfMNPV field isolate was obtained from a natural population of the fall armyworm in Hubei, China. Two genotypes were cloned from the field isolate, and one genotype, SfHub-A, which had similar activity to the field isolate and produced significantly more progeny viruses, was considered to be a suitable strain for the commercial production of SfMNPV. This information will be valuable for developing a virus-based pesticide against fall armyworm in China.AbstractThe fall armyworm,Spodoptera frugiperda, is a new invading pest in China. The baculovirusSpodoptera frugiperdamultiple nucleopolyhedrovirus (SfMNPV) is a pathogenic agent of the fall armyworm and a potential agent for its control in integrated pest management strategies. In this work, we analyze the molecular and biological characteristics of an SfMNPV isolate collected from maize in China (SfMNPV-Hub). Two genotypes were further isolated from SfMNPV-Hub by an in vivo cloning method. The PstI profile of one genotype (SfHub-A) was similar to genotype A of the SfMNPV Colombian isolate, and the other (SfHub-E) was similar to genotype E of the Colombian isolate. The bioactivity of SfHub-A against second-instarS. frugiperdalarvae was not significantly different from that of SfMNPV-Hub, whereas SfHub-E was 2.7–5.5 fold less potent than SfMNPV-Hub. The speed of kill of SfHub-E was quicker than SfMNPV-Hub, while SfHub-A acted slower than SfMNPV-Hub. Occlusion body (OB) production of SfHub-A in anS. frugiperdacadaver was significantly higher than that of SfMNPV-Hub, while SfHub-E yielded far fewer occlusion bodies (OBs) in the host larvae. These results provide basic information for developing a virus-based pesticide against the invading pestS. frugiperda. Keywords:SfMNPV;field isolate;genotype;molecular characterization;biological activity;OB yield

The invasion of Spodoptera frugiperda has imposed a serious impact on global food security. Matrine is a botanical pesticide with a broad spectrum of insecticidal activity which was recommended for controlling Spodoptera frugiperda . In order to discover effective insecticide for Spodoptera frugiperda , two matrine derivatives modified with carbon disulfide and nitrogen-containing groups were systhesized. And their inhibition activities on Sf9 cell were evaluated. The structural configuration of compounds were characterized by IR, HPLC, MS, NMR and XRD, with yields of 52% and 65%, respectively. The IC 50 of the two newly synthesized compounds on Sf9 cell reduced to 0.648 mmol/L and 1.13 mmol/L, respectively, compared with that of matrine (5.330 mmol/L). In addition, microscopic observation of Sf9 cell treated with the compounds showed that the number of adherent cells decreased, the cells shrunk, vacuolated and apoptotic bodies appeared. The two newly synthesized compounds exhibited better inhibitory effect on Sf9 cell than that of the parent matrine, suggesting that the positive effect of the introduction of 1-pyrrolidinecarbodithioate and diethylcarbamodithioate groups to matrine. The morphological observation of Sf9 cell induced by derivatives indicated that apoptosis induction may be a mechanism that inhibits insect cell proliferation and exerts insecticidal effect.

Traditional pesticide spraying methods such as knapsack sprayers are inefficient. Ground application equipment is restricted by the terrain and damages the crops. Pesticides cannot be sprayed over a large area in a short time; thus, when there are concentrated outbreaks of pests with strong migration capacity, such as Spodoptera frugiperda, pests could not be controlled in a small area. Therefore, it is difficult to achieve a satisfactory control effect. Plant protection unmanned aerial vehicles (UAVs) have high efficiency and spray pesticides at low altitude and can suitably prevent and control the outbreak of pests. However, pesticide formulations suitable for UAVs are rare. In this study, we developed ultralow-volume (ULV) sprays suitable for plant protection UAVs. We studied their properties and found that the ULV sprays have good wettability and can be fully spread. The minimum contact angle on corn leaves is up to 19°, and the maximum retention is 2.7 times that of the reference product. They also have superior resistance to rain washout and can effectively extend the duration and utilization of pesticides. When sprayed by plant protection UAVs, the decrease rate of the ULV spray at 3 L/ha to S. frugiperda larvae is 81.31% and the control effect is 83.01%, both higher than those of the reference product 40% chlorantraniliprole–thiamethoxam water-dispersible granules (decrease rate 67.39%, control effect 70.35%), sprayed with a knapsack sprayer.

Simple SummaryInvestigations into the insecticidal activity of novaluron against the fall armyworm (Spodoptera frugiperda) and diamondback moth (Plutella xylostella) provide valuable data on its efficacy against these pests. Utilizing the leaf-dip method to assess toxicity to second-instar larvae and enzyme-linked immunosorbent assays (ELISA) to monitor changes in detoxifying enzymes and ecdysteroid levels, the study highlights distinct detoxification pathways in each pest species. The results indicate that physiological and ecological differences between the fall armyworm and diamondback moth influence their response to novaluron, offering valuable data for its application in integrated pest management. To provide a scientific basis for pest control, this study evaluated the insecticidal activity of novaluron against the fall armyworm (Spodoptera frugiperda) and diamondback moth (Plutella xylostella). The leaf-dip method determined the toxicity of novaluron to second-instar larvae, while corn leaves and cabbage treated with sublethal (LC10) and median lethal concentrations (LC50) of novaluron were used to feed the larvae. Enzyme-linked immunosorbent assays (ELISA) measured the activities of detoxifying enzymes [carboxylesterase (CarE), cytochrome P450 (P450), glutathione S-transferase (GST), and acetylcholinesterase (AChE)] and ecdysteroid (Ecd) levels in the pests after 24 and 48 h of treatment. Results indicated that after 24 h, AChE was involved in diamondback moth metabolism and CarE activity was inhibited. After 48 h, P450 and GST participated in fall armyworm detoxification, whereas P450 and GST were active in diamondback moth detoxification, with other enzyme activities normalizing. Novaluron also altered Ecd levels in both pests. These results demonstrate differing detoxification mechanisms in fall armyworm and diamondback moth, likely due to their unique physiological and ecological traits, and support the potential use of novaluron in pest management strategies.

Simple SummaryThe fall armyworm,Spodoptera frugiperda, is an economically important pest that has caused extensive damage to many crops all around the world over the past decade. Due to the advantages of being bioactive, biodegradable, and ecologically safe, thymol, one of the principal ingredients identified in essential oils, has exhibited its potential in pest control. However, the effects and toxicological mechanisms of thymol onS. frugiperdaare still unclear. In this study, we evaluated the bioactivity of thymol on the larvae ofS. frugiperda. RNA-Seq was performed to explain the preliminary toxicological mechanisms of thymol on larvae by the identification and functional enrichment analyses of differentially expressed genes (DEGs). Overall, our findings indicate that thymol also exhibits the potential for the control ofS. frugiperda. The global migratory pest,Spodoptera frugiperda, has garnered widespread attention due to the serious damage it inflicts on agricultural productivity, particularly in maize. Thymol is a phytochemical that exhibits functional diversification in plant defense, encompassing antibacterial activities and insect pest management. However, the impact of thymol onS. frugiperdais still undetermined. This study examined the growth inhibition and mortality induction inS. frugiperdalarvae after thymol exposure. The detrimental effects of 2.0 and 4.0 mg/g thymol treatments on the growth and development ofS. frugiperdawere also examined. RNA-Seq was used to investigate the probable toxicological mechanism of thymol onS. frugiperda, resulting in the identification of 1754 and 1022 DEGs impacted by 2.0 and 4.0 mg/g thymol treatments, respectively. The DEGs associated with chitin metabolism and cuticle synthesis, hormone biosynthesis, and protein and fat digestion were subjected to additional analysis. Our findings demonstrate the efficacy of thymol in controllingS. frugiperdaand lay the groundwork for understanding the molecular toxicological mechanisms of thymol on larvae.

Azadirachtin, a highly effective botanical pesticide, demonstrated notable biological activities against Spodoptera frugiperda , including mortality induction, growth and development inhibition, and antifeedant effects. Neuropeptide F (NPF) has been shown to play a role in various physiological processes in insects. Nonetheless, the functions of Sf- NPF1 in regulating food intake and antifeedant induction by azadirachtin in S. frugiperda larvae are yet to be elucidated. In this study, the antifeedant effects induced by 0.5 and 1.0 μg/g azadirachtin treatments in S. frugiperda larvae were evaluated. The sequence of Sf-NPF1 was identified with two transcript variants, and the proteins exhibit high identity with NPFs found in lepidopteran insects. Spatiotemporal expression analysis indicated that Sf-NPF1 exhibited high expression levels in first instar larvae and male adults, while the lowest expression was observed in non-feeding stages (egg and pupa stages). Besides, Sf-NPF1 also demonstrated notable expression in the midgut tissue of larvae. Azadirachtin treatments inhibited the expression of Sf-NPF1 in the larval midgut. The injection of the Sf- NPF1a mature peptide resulted in increased larval food intake and weight gain during a two-day treatment period. The antifeedant effect induced by 1.0 μg/g azadirachtin was also mitigated by the injection of Sf- NPF1a mature peptide. Our results indicated that Sf- NPF1 may serve as a positive regulator of food intake in S. frugiperda larvae, while azadirachtin could exert an antifeedant effect in larvae via inhibiting the expression of Sf-NPF1 . These findings establish a foundation for comprehending the mechanism behind the antifeedant effect induced by azadirachtin in insect larvae and provide a potential target for developing novel insecticides.

Simple SummaryThe widespread cultivation of genetically engineered crops producing not only toxic proteins from the bacteriumBacillus thringiensis(Bt) but also plant defensive compounds known as allelochemicals, combined with occasional use of insecticides, is the major tactic to manage some economically important pests. Better understanding of the toxicological interactions of the three types of toxins is needed to rationally deploy them to protect crops from pests. The aim of this study is to examine the sequential and simultaneous interactions of the allelochemical flavone, Bt toxin Vip3A, and insecticide emamectin benzoate in the fall armyworm (Spodoptera frugiperda), a worldwide target pest of Bt crops. Bioassays ofS. frugiperdaneonates revealed that all interactions of the three toxins, except for 1-day pre-exposure to a sublethal dose (LC5) of flavone followed by 6-day simultaneous exposure to flavone LC5+ emamectin benzoate LC50, are synergistic or additive. The results suggest that the combined use of the three toxins is basically a great strategy to manageS. frugiperda.AbstractTarget pests of genetically engineered crops producing both defensive allelochemicals andBacillus thuringiensis(Bt) toxins often sequentially or simultaneously uptake allelochemicals, Bt toxins, and/or insecticides. How the three types of toxins interact to kill pests remains underexplored. Here we investigated the interactions of Bt toxin Vip3A, plant allelochemical flavone, and insecticide emamectin benzoate inSpodoptera frugiperda. Simultaneous administration of flavone LC25+ Vip3A LC25, emamectin benzoate LC25+ Vip3A LC25, and flavone LC15+ emamectin benzoate LC15+ Vip3A LC15but not flavone LC25+ emamectin LC25yielded a mortality significantly higher than their expected additive mortality (EAM). One-day pre-exposure to one toxin at LC5followed by six-day exposure to the same toxin at LC5plus another toxin at LC50showed that the mortality of flavone LC5+ Vip3A LC50, emamectin benzoate LC5+ Vip3A LC50, and Vip3A LC5+ emamectin benzoate LC50were significantly higher than their EAM, while that of flavone LC5+ emamectin benzoate LC50was significantly lower than their EAM. No significant difference existed among the mortalities of Vip3A LC5+ flavone LC50, emamectin benzoate LC5+ flavone LC50, and their EAMs. The results suggest that the interactions of the three toxins are largely synergistic (inductive) or additive, depending on their combinations and doses. Keywords:allelochemical;Bt toxin;chemical insecticide;combined toxicity;induced toxicity;synergistic interactions
