To address the limitations of low relaxivity and physiological toxicity in commercial gadolinium-based contrast agents for magnetic resonance imaging (MRI), a novel manganese chelate macromolecular system was developed using a flash nano-preparation technique. Herein, the approach applying an instantaneous fluid device incorporated gallic acid, dopamine and Mn2+ to perform in-situ polymerization of dopamine and covalent binding with albumin in a nanoconfined environment. This controllable self-assembly process characterized by its scalability and reproducibility was suitable for industrial-scale production. Under optimized flow rates and material ratios, the synthesized ultrasmall protein-based system, Mn-GA@BSA@DA, exhibited excellent aqueous dispersion with an average size of approximately 18 nm, allowing for long-term lyophilized powder storage. More importantly, the nanosystem demonstrated superior MRI-T 1 relaxivity, significantly surpassing that of clinical Gadopentetate dimeglumine, with a high value around 18.5 mM−1 s−1 and a low r 2/r 1 ratio (< 5 at 3.0 T). Furthermore, this Mn-GA@BSA@DA contrast agent was endowed with tumor-targeting effects and long MRI monitoring window period for the liver, gallbladder and renal tubules. The metal chelation within the nanoagent minimizes Mn2+ release; importantly, the antioxidant components, gallic acid and dopamine, significantly inhibit the Fenton reaction-induced toxicity, enhancing biocompatibility. Therefore, this study presents a simple and scalable production technique for a kind of MRI-T 1 weighted contrast agent with high relaxivity and biocompatibility, offering a promising alternative to commercial Gd chelates.

Emerging nanotheranostic systems have promoted the development of dual-mode imaging techniques (i.e. T1/T2-weighted MRI) to meet the increasing requirements of accurate personalized treatment for cancer. Nevertheless, slight tumor accumulation and poor penetration have limited the efficacy of dual-mode theranostic agents. Furthermore, under the premise of guaranteeing imaging capability, most current research studies hardly focused on optimizing theranostic agents to achieve considerable therapeutic effects. Here, we developed a hyaluronic acid (HA)-stabilized iron oxide nanocluster (Fe2O3@PFDH NC) as an intelligent-degradable theranostic nanoagent for dual-mode MRI-guided chemo-photothermal therapy. The obtained Fe2O3@PFDH NC with high longitudinal and transverse relaxivities offers strong contrast in T1/T2-weighted MR imaging. Meanwhile, according to MR images, the intravenous Fe2O3@PFDH NC could accumulate at the hyaluronidase-rich tumor location gradually. More interestingly, it could break into smaller nanoparticles with quick DOX release for deep penetration, accompanied by highly effective photothermal ablation of the tumor under laser irradiation. In conclusion, the versatile tumor-penetrating Fe2O3@PFDH nanocluster could serve as a T1/T2 dual-mode MRI contrast agent with highly effective combination chemo-photothermal therapy, and would be an ideal theranostic nanocarrier with translational potential for future clinical diagnosis and treatment of cancer.

The impact of nanoparticle size on the effectiveness of magnetic resonance imaging (MRI) using sulfurized manganese nanoparticles (MnS@PAA) stabilized with polyacrylic acid (PAA) as a binder was thoroughly investigated. MnS@PAA nanoparticles of varying sizes were synthesized by altering the ratio of ethylene glycol (EG) to diethylene glycol (DEG) during the synthesis process. These nanoparticles exhibited a uniform size distribution and demonstrated high T1 relaxation rates, along with a notable pH-responsive behavior. As the nanoparticle size increased, the T1 relaxation rate decreased, indicating that size plays a crucial role in their MRI performance. Additionally, research has revealed that the efficiency of tumor uptake by these nanoparticles is size dependent. Specifically, MnS@PAA nanoparticles with a core size of 100 nm (MS100) exhibited greater tumor accumulation and provided enhanced MRI contrast. Once within the acidic environment of a tumor, MS100 decomposes into Mn2+ and H2S. Mn2+ ions promote the generation of hydroxyl radicals, which leads to lipid peroxidation and induces ferroptosis. Concurrently, the release of H2S inhibits catalase activity, resulting in elevated levels of hydrogen peroxide (H2O2), achieving a synergistic effect between chemodynamic therapy (CDT) and gas therapy. This study explores the influence of nanoparticle size on its potential applications as an MRI contrast agent and as a therapeutic agent in cancer treatment.

It has been challenging to integrate various medical imaging modalities into an ultra-small nanoparticle with good biocompatibility to build highly efficient multimodal imaging nanoprobes. A new manganese and dysprosium codoped carbon quantum dots (Mn,Dy-CQDs) with a mean diameter of 1.77 nm was synthesized for fluorescence imaging, T1/T2-weighted magnetic resonance imaging (T1/T2-weighted MRI), and x-ray computed tomography (CT) imaging using a simple one-step hydrothermal approach. The obtained Mn,Dy-CQDs showed good water solubility, long-term stability, strong stable fluorescence property (fluorescence quantum yield of 31.62%), and excellent biocompatibility. The cell imaging verified that the Mn,Dy-CQDs have high efficiency of fluorescence imaging. The Mn,Dy-CQDs, on the other hand, had a superior x-ray absorption performance (47.344 HU l g−1), a higher longitudinal relaxivity (r1 = 7.47 mM−1 s−1), a higher transverse relaxivity (r2 = 42.686 mM−1 s−1). In vitro T1/T2-weighted MRI and CT imaging showed that Mn,Dy-CQDs can produce a strong contrast enhancement impact. To summarise, the Mn,Dy-CQDs may be used as a T1/T2-weighted MRI/CT/fluorescent quadri-modal imaging nanoprobe, indicating that they have a lot of uses in biomedical multimode imaging and clinics.

Nowadays, about 30% of magnetic resonance imaging (MRI) exams need contrast agents (CAs) to improve the sensitivity and quality of the images for accurate diagnosis. Here, a multifunctional nano-agent with ring-like vortex-domain iron oxide as core and gadolinium oxide as shell (vortex nanoring Fe 3 O 4 @Gd 2 O 3 , abbreviated as VNFG) was firstly designed and prepared for highly enhanced T 1 -T 2 dual-modality magnetic resonance imaging (MRI)-guided magnetic thermal cancer therapy. After thorough characterization, the core-shell structure of VNFG was confirmed. Moreover, the excellent heat generation property (SAR=984.26 W/g) of the proposed VNFG under alternating magnetic fields was firmly demonstrated. Furthermore, both in vitro and in vivo studies have revealed a good preliminary indication of VNFG’s biological compatibility, dual-modality enhancing feature and antitumor efficacy. This work demonstrates that the proposed VNFG can be a high-performance tumor diagnosis and theranostic treatment agent and may have great potential for clinical application in the future.

Theranostic nanomedicine designs integrating diagnostic and therapeutic paradigms have been achieving considerable success at clinics. However, the toxicity of nanocomposites in these designs remains a major obstacle to the success of personalized medicine. Herein, highly biocompatible iron and hydroxyapatite materials were selected to design a Fe 3 O 4 -HAp nano-theranostic system with controlled dimensions using simple solvothermal synthesis . The biocompatibility of Fe 3 O 4 -HAp magnetic hydroxyapatite nanocomposites (MHNCs) was further enhanced by Food and Drug Administration- (FDA-) approved triblock copolymers , namely, Pluronic® F-127. Indeed, polymer modification improved the drug encapsulation capacity of HAp, and 70% anti-TROP 2 was loaded onto the surface of MHNCs for intracellular targeting of breast cancer. The Fe 3 O 4 component of MHNCs possessed a high T 1 -MR imaging enhancement feature with an excellent r 1 value, demonstrating these MHNCs as capable substitutes for Gd-based toxic contrast agents. Moreover, indocyanine green (ICG-) coupled-MHNCs revealed strong absorption in the near-infrared region and produced enough temperature (75 °C), even at low (50 ug/mL) concentrations, to ablate cancerous cells upon 808 laser irradiation to induce cell toxicity and apoptosis. The promising diagnostic and photothermal therapeutic performance of these designed MHNCs make them an ideal nano-theranostic agent.

T 1 –T 2 multi-modality submicron particles were widely used in clinic imaging, biochemical analysis and biosensor. Here, we prepared a new magnetic submicron particle through the spatial-gap of cellulose nanocrystal (CNC) dispersion as a template. The obtained submicron particles were ellipsoid and had a better magnetic response and relaxation effects. By varying the Fe(acac) 3 to Gd(acac) 3 ratio, it was possible to tune the surface morphology, crystal structure and T 1 –T 2 relaxation effect. More importantly, the pre-drying and burning process had influence on the polycrystalline growth process of Fe(acac) 3 and Gd(acac) 3 mixture, and the slow removal process for the CNC template could endow the compounds with different relaxation effects. By using the spatial-gap of the cellulose nanocrystal dispersion as a template makes it possible to fabricate T 1 –T 2 multi-modality crystalline submicron particles directly and is promising for preparing other CNC-templating based functional materials.

Nanostructure-based materials for multimodal imaging have attracted great attentions. Here we report a new formulation integrated gadolinium (Gd) and magnetite (Fe 3 O 4 ) nanoparticles as magnetic resonance (MR) imaging contrast agent at both T 1 mode and T 2 mode. Polyetherimide coated iron oxide (Fe 3 O 4 @PEI) nanoparticles were first synthesized and then conjugated to diethylenetriamine pentaacetic acid (DTPA), which further chelated with Gd 3+ ions (Fe 3 O 4 @PEI@DTPA-Gd). Both phantom and MR studies showed Fe 3 O 4 @PEI@DTPA-Gd nanocomposites could be applied in two modes ( r 1 = 4.7 mM −1 s −1 , r 2 = 131.82 mM −1 s −1 ). MTT and fluorescent staining results also suggested that the nanocomposites had good biocompatibility. Besides, Fe 3 O 4 @PEI@DTPA-Gd nanocomposites exhibited high entrap efficiency when the mass ratio of nanocomposites to DNA was up to 30:1. The in vitro gene transfection ability was also evidenced. Therefore, Fe 3 O 4 @PEI@DTPA-Gd nanocomposites can be employed as a potential candidate for MRI guided clinical diagnosis and gene delivery guided therapy.

Despite the progress in the design and synthesis of theranostic agents, limitations on efficiency and safety offer significant room for improvement in these agents. Inspired by the natural binding ability of polydopamine nanospheres (PDAs) with iron ion , a simple and versatile synthesis strategy is developed to prepare biodegradable coordination polymer (CP) encapsulated PDAs nanocomplex ( [email protected] x , x = 3, 6, 9). We found that the [email protected] 3 can serve as a T 1 /T 2 dual mode contrast agent (DMCA) for magnetic resonance imaging (MRI), which possesses high longitudinal (r 1 = 7.524 mM −1 s −1 ) and transverse (r 2 = 45.92 mM −1 s −1 ) relaxivities. In this system, benefitting from the high photothermal conversion efficiency derived from PDAs, DOX loaded [email protected] 3 nanocomplex is able to not only destroy the tumor directly by heat, but also stimulate the chemotherapy by enabling NIR-responsive on demand delivery of DOX. To the best of our knowledge, this is the first example exploring the potential of [email protected] x nanocomplex for T 1 /T 2 dual mode MRI-guided chemo-photothermal synergistic therapy. This work extends the currently available theranostic agents, and opens up new avenues to rationally design the high-performance T 1 /T 2 DMCA.

Magnetic resonance contrast agent employs the use of gadolinium chelates, which has many limitations for clinical use including a low contrast effect, a short diagnostic window, and a brief blood circulation time. On this basis, we designed a gadolinium-labeled dendrimer nanocluster (GdDN) loaded graphene oxide nanosheet (GO-GdDN) to boost T1 contrast ability for imaging in vivo and improve blood circulation time. GO-GdDN presented an ultrahigh r1 relaxivity up to 19.07 mM–1 s–1 in a 9.4 T MR scanner, and a bright contrast image in vitro experiment. In addition, GO-GdDN could be internalized by HepG2 cells and presented strong cell contrast enhancement and reduction of the T1 value in HepG2 cells. In vivo, the retention time of GO-GdDN was significantly improved, so that the accumulation of GO-GdDN in liver was enhanced. Moreover, compared to Gd-DTPA, systemic delivery of GO-GdDN dramatically enhanced the signal-to-noise ratio of liver images, which was helpful for accurate imaging of liver and detection of liver lesions in vivo. Thus, this study demonstrates the utility of a powerful diagnosis tool for liver tumor or lesions.

Clinically, magnetic resonance imaging (MRI) often uses contrast agents (CAs) to improve image contrast, but single-signal MRI CAs are often susceptible to calcification, hemorrhage, and magnetic sensitivity. Herein, iron acetylacetone and gadolinium acetylacetone were used as raw materials to synthesize a T1–T2dual-mode imaging gadolinium-doped iron oxide (GdIO) nanocluster. Moreover, to endow the nanoclusters with targeting properties and achieve antitumor effects, the cyclic Arg-Gly-Asp (cRGD) peptide and docetaxel (DTX) were attached to the nanocluster surface, and the efficacy of the decorated nanoclusters against pancreatic cancer was evaluated. The final synthesized material cRGD-GdIO-DTX actively targeted αvβ3on the surface of Panc-1 pancreatic cancer cells. Compared with conventional passive targeting, the enrichment of cRGD-GdIO-DTX in tumor tissues improved, and the diagnostic accuracy was significantly enhanced. Moreover, the acidic tumor microenvironment triggered the release of DTX from cRGD-GdIO-DTX, thus achieving tumor treatment. The inhibition of the proliferation of SW1990 and Panc-1 pancreatic cancer cells by cRGD-GdIO-DTX was much stronger than that by the untargeted GdIO-DTX and free DTX in vitro. In addition, in a human pancreatic cancer xenograft model, cRGD-GdIO-DTX considerably slowed tumor development and demonstrated excellent magnetic resonance enhancement. Our results suggest that cRGD-GdIO-DTX has potential applications for the precise diagnosis and efficient treatment of pancreatic cancer.Keywords:T1–T2;DTX;MRI;drug delivery;pancreatic cancer

Carbon quantum dots (CQDs) with good biosafety can be doped with metals to construct multimodal imaging probes with a wide range of applications in fluorescence and molecular imaging. However, developing CQDs with both long-wavelength emission and a variety of imaging modes remains a challenge. In this article, novel bismuth- and gadolinium-codoped carbon quantum dots (Bi,Gd-CQDs) were developed using a hydrothermal method for green and red fluorescence imaging, X-ray computed tomography (CT) imaging, and T1-weighted magnetic resonance imaging (T1-weighted MRI). In addition to exceptional water solubility, strong steady fluorescence characteristics, and great biocompatibility, the obtained Bi,Gd-CQDs have a high X-ray attenuation coefficient and a short magnetic resonance T1 relaxation time. Additionally, cell imaging involved red fluorescence imaging with deep penetration. Therefore, the prepared Bi,Gd-CQDs with green and red dual-emissions can be used as nanoprobes for fluorescence, CT, and T1-weighted MR imaging, expanding their application prospects in the field of biomedicine.
