The increasing amount of e-waste plastics needs to be disposed of properly, and removing the brominated flame retardants contained in them can effectively reduce their negative impact on the environment. In the present work, TBBPA-bis-(2,3-dibromopropyl ether) (TBBPA-DBP), a novel brominated flame retardant, was extracted by ultrasonic-assisted solvothermal extraction process. Response Surface Methodology (RSM) achieved by machine learning (support vector regression, SVR) was employed to estimate the optimum extraction conditions (extraction time, extraction temperature, liquid to solid ratio) in methanol or ethanol solvent. The predicted optimum conditions of TBBPA-DBP were 96 min, 131 mL g −1 , 65 °C, in MeOH, and 120 min, 152 mL g −1 , 67 °C in EtOH. And the validity of predicted conditions was verified.

With the widespread application of the brominated flame retardant tetrabromobisphenol A (TBBPA), it poses a threat to human health, especially intestinal health. Quercetin (Que) is a flavonol compound with anti-inflammatory and antioxidant properties. However, whether Que can prevent TBBPA-induced intestinal toxicity remains unknown. Therefore, in this study, a TBBPA (75 μM) exposure model and a Que (0.1 μM) treatment model were established using mouse colon epithelial cells (MCEC). The p38 pathway, pyroptosis, and intestinal barrier function-related indicators were analyzed using an oxidative stress reagent kit, immunofluorescence staining, western blotting, and qRT-PCR. The results showed that TBBPA exposure dose-dependently reduced cell viability, impaired the antioxidant function of cells, promoted ROS accumulation, activated the p38 signalling pathway, induced pyroptosis, and decreased the expression levels of ZO-1, Occludin, and Claudin-1. Notably, Que treatment significantly restored the antioxidant enzyme activity in MCEC cells, reduced the ROS level, inhibited the p38 axis, alleviated MCEC cell pyroptosis, and recovered the intestinal barrier function. Further studies found that using LX-3 (a p38 activator) treatment disrupted the therapeutic effect of Que. In summary, Que can exert a protective effect by inhibiting the ROS-mediated p38 pathway, thereby alleviating the damage of TBBPA to MCEC cells.

On the basis of H2O2-mediated growth of gold nanoparticle (AuNPs), a novel plasmonic enzyme-linked immunosorbent assay (pELISA) was developed with a polyclonal antibody for the ultrasensitive simultaneous naked-eye detection of tetrabromobisphenol A bis(2-hydroxyetyl) ether (TBBPA DHEE) and tetrabromobisphenol A mono(hydroxyethyl) ether (TBBPA MHEE), one of the major derivatives and byproducts of tetrabromobisphenol A (TBBPA), respectively. In this modified indirect competitive pELISA, glucose oxidase (GOx) played an important role leading to the growth of AuNPs through a reaction between GOx and glucose to produce hydrogen peroxide (H2O2). In addition, further signal amplification was achieved via a large number of GOx molecules, which were immobilized on silica nanoparticles carrying poly brushes (SiO2@PAA) to increase the enzyme load, and the whole complex was conjugated on the second antibody. Under the optimized conditions, 10–3 μg/L TBBPA DHEE can be distinguished via the observation of a colored solution, and the limit of detection (LOD) of the method using a microplate reader reaches 3.3 × 10–4 μg/L. In contrast, the sensitivity of the method was 3 orders of magnitude higher than that using conventional colorimetric ELISA with the same antibody. Furthermore, the proposed approach showed good repeatability and reliability after a recovery test fortified with a variety of targets was performed (recoveries, 78.00–102.79%; coefficient of variation (CV), 4.38–9.87%). To our knowledge, this is the first case in which pELISA was applied for the detection of small molecules via the production of H2O2 from GOx and glucose. The method will be widely used for the investigation of TBBPA DHEE and TBBPA MHEE in real environments.

As a widely used brominated flame retardant, tetrabromobisphenol A (TBBPA) is heavily utilized and has led to major concerns about its environmental and health implications. Although TBBPA has been detected in various environmental and biological matrices globally, its impact on the spleen and the underlying mechanisms remain poorly understood. HMGB1 is a protein released during pyroptosis which can trigger inflammation by activating the TLR4/MyD88/NF-κB pathway, leading to the production of inflammatory cytokines and chemokines. Nonetheless, its impact of HMGB1 on spleen damage caused by TBBPA is still uncertain. This study aims to explore the effect of TBBPA on spleen injury in mice and elucidates the role of HMGB1 in this process. In this study, male ICR mice were treated with TBBPA (50 mg/(kg·day)) for 28 days. Results showed that TBBPA induced splenic injury, characterized by white pulp reduction, red pulp congestion, lymphatic hyperplasia, and inflammatory infiltration. Moreover, vacuolation was observed in the mitochondria of the TBBPA exposed group, with the majority of mitochondrial cristae either disrupted or had vanished. Mechanistically, TBBPA upregulates HMGB1, which activates the TLR4/MyD88/NF-κB pathway and subsequently promotes NLRP3-mediated pyroptosis, ultimately exacerbating the inflammatory response. Our findings indicate that HMGB1 could be an effective target for addressing inflammatory and immune disorders.

This study has focused on the evaluation of the biotoxicity controlling effects during the TBBPA degradation by ozonation, including the acute, chronic and genetic toxicity under different [O3]/[TBBPA] (1:1–11:1), initial solution pH (5.0–9.0) and temperatures (10–40 °C). In addition, the comprehensive biotoxicity of the treated water sample was evaluated by the method of potential ecotoxic effects probe (PEEP). The results showed that TBBPA could be completely degraded with an initial solution pH of 7.0, temperature of 25 °C and an [O3]/[TBBPA] ratio of 9:1. The chronic toxicity of the untreated sample was as high as 41.7 TU, which represented the main toxicity of TBBPA itself. In contrast, the TBBPA showed a much lower acute and genetic toxicity in this study. During the process of TBBPA degradation, the ozonation could effectively control the toxicity of wastewater and showed strong adaptability. When the ratio of [O3]/[TBBPA] was 11:1, the acute and chronic toxicity were reduced to 0.02 TU and 0.76 TU, respectively, with the controlling rates being as high as 96% and 98.2% and meeting the emission standards. The mutagenicity ratio of the water sample was less than 2.0, indicating no genotoxicity risk. The evaluation of the comprehensive biological toxicity showed that ozonation could control the PEEP value below 2.0 in ranges of low [O3]/[TBBPA] ratio (3:1), wide pH (5–9) and temperatures (10–40 °C).Keywords:ozonation;tetrabromobisphenol A;acute toxicity;chronic toxicity;comprehensive biological toxicity

Development of functional porous materials for efficient elimination of environmental pollutants is of great importance in green chemistry and environmental protection. Here we report the design, synthesis, and application of a core–shell magnetic amino-functionalized microporous organic network nanosphere (Fe3O4@MON-NH2) for efficient magnetic adsorption of a typical brominated flame-retardant tetrabromobisphenol A (TBBPA) from aqueous solution. By integration of the hydrophobic networks and hydrogen binding sites within MON-NH2, the synthesized core–shell Fe3O4@MON-NH2 nanospheres gave good adsorption for TBBPA. The adsorption equilibrium of TBBPA (50 mg L–1) on Fe3O4@MON-NH2 was achieved within 1 min, showing ultrafast adsorption kinetics of Fe3O4@MON-NH2 for TBBPA. The adsorption of TBBPA on Fe3O4@MON-NH2 followed the pseudo-second-order kinetics and Langmuir adsorption models, giving a maximum adsorption capacity of 135.9 mg g–1 at 25 °C. The adsorption of TBBPA on Fe3O4@MON-NH2 was a spontaneous and endothermic process controlled by positive entropy. Encouraged by the large adsorption capacity, rapid adsorption kinetics, and the small effect of pH (5–10), dissolved organic matter, and ionic strength on the adsorption, the application of Fe3O4@MON-NH2 for TBBPA removal in real water samples was achieved. The used Fe3O4@MON-NH2 can be easily regenerated and reused at least four times without significant reduction of the adsorption capacity. These results revealed the potential of Fe3O4@MON-NH2 for adsorption and removal of environmental pollutants from aqueous solution.

The degradation of tetrabromobisphenol A (TBBPA), one of the most widely used brominated flame retardant , was evaluated during peroxymonosulfate (PMS) oxidation. TBBPA degradation was pH-dependent, with peak degradation rate constants occurring at pH 8.0–9.0, which was distinct from some other phenolic compounds . Singlet oxygen and radicals were found to play negligible roles in TBBPA degradation. TBBPA oxidation by PMS mainly proceeded via a direct oxidation pathway and the in-situ formed HOBr was found to greatly accelerate its degradation rates. The values of species-specific second-order rate constants for the reactions of PMS with the TBBPA k HSO5 − +TBBPA , k HSO5 − +TBBPA− and k HSO 5− +TBBPA2− were determined to be (1.11 ± 0.84) × 10 −2 , (8.05 ± 2.31) × 10 −2 , and (1.34 ± 0.25) × 10 −1 M −1 s −1 , respectively, while the reaction rate constants for HOBr/OBr − with TBBPA k HOBr+TBBPA , k HOBr+TBBPA− , k HOBr+TBBPA2− and k OBr−+TBBPA2− were determined to be (9.38 ± 2.10) × 10 3 , (1.59 ± 0.56) × 10 5 , (8.22 ± 0.41) × 10 6 , and (1.81 ± 0.12) × 10 6 M −1 s −1 , respectively. The bromine mass balance analysis showed that bromide ion and HOBr/OBr − occupied 19.5% of total Br and brominated organic compounds accounted for the remaining percentages at pH 7.0. No formation of bromate was observed. Based on the identified products, a reaction pathway was proposed, which included oxidation, β-scission, hydroxylation, and dimerization reaction pathways. The results indicate that unactivated PMS is useful for the remediation of TBBPA contaminated water .

Tetrabromobisphenol A (TBBPA) was one of the most widely used brominated flame retardants. However, it easily contaminates nature and harms the environment and human health during its production and use. Therefore, it is necessary to strictly control the content of TBBPA in electronics. Surface-enhanced Raman spectroscopy has the advantages of being fast and sensitive, but it is difficult to obtain the SERS spectra of TBBPA because the hydrophobic TBBPA molecule is difficult to approach with the hydrophilic surface of common noble metal SERS substrates. In the present work, a hydrophobic Cu-Ag chip was developed for the SERS detection of TBBPA. The integration of the hydrophobic interaction and the Ag-Br bonding promoted the adsorption of TBBPA on the Cu-Ag chip, allowing for SERS detection. It was observed that both the hydrophobicity and bimetallic composition of the Cu-Ag chip played important roles in the SERS detection of TBBPA. Under the optimized conditions, the low limit of detection of the established SERS method for TBBPA was 0.01 mg L−1, within a linear range of 0.1–10 mg L−1. Combined with ultrasonic-assisted extraction, the substrate could be used for the quantitative determination of TBBPA in electronic products. Compared with the HPLC-UV method used as a national standard, the relative error of the SERS method for quantifying the TBBPA content in a mouse cable and shell was ±3% and ±7.7%, respectively. According to the SERS results, the recovery of TBBPA in the spiked mouse shell was 95.6%.Keywords:tetrabromobisphenol A;surface-enhanced Raman scattering;hydrophobic interaction;bi-metal chips;synergistic coupling

A novel potentiometric sensor based on molecularly imprinted polymer (MIP) beads embedded in an asymmetric membrane configuration was developed for the trace monitoring of tetrabromobisphenol A (TBBPA), a widely used brominated flame retardant and persistent environmental pollutant. The sensor was fabricated by incorporating TBBPA-imprinted beads into a poly(vinyl chloride) (PVC) membrane plasticized with dioctyl phthalate (DOP) and drop-cast onto a reduced graphene oxide (rGO)-modified screen-printed electrode. The asymmetric design featured a surface-enriched ion-exchange layer, enabling enhanced accumulation of deprotonated TBBPA at the membrane interface. Under optimized conditions, the sensor exhibited a near-Nernstian slope of −56.6 ± 1.2 mV per decade over a linear range of 5.0 × 10−6 to 10−3 M and a detection limit of 5.5 × 10−7 M. The sensor demonstrated high selectivity toward TBBPA over structurally related phenols and common anions, with the enhanced discrimination attributed to the imprinting effect and nano-scale recognition features. Application to spiked environmental samples including wastewater, sediment, and plastic leachates yielded recoveries between 95.6% and 102.3%, with no significant deviation from high-performance liquid chromatography (HPLC) results (p > 0.05). The proposed MIP-based asymmetric membrane sensor offers a portable, cost-effective, and highly selective platform for on-site environmental monitoring of TBBPA.

Sewerage sludge-derived biochars (SSDBCs) with high adsorption capacity and excellent recyclability were synthesized to remove tetrabromobisphenol A (TBBPA) in aqueous system. Scanning electron microscopy, elemental mapping via energy dispersive X-ray spectroscopy, X-ray photoelectron spectroscopy, and Fourier transform infrared spectroscopy were used to characterize the morphology, composition, and microstructures. The maximum adsorption capacity of SSDBCs was about 87.02 mg g −1 at 303 K and pH 7.5. The Langmuir isotherm demonstrated that the adsorption was mainly homogeneous and chemical processes. The kinetics of TBBPA removal well fitted the second-order dynamic model. Thermodynamic analysis showed that the adsorption was exothermic. The effect of π−π dispersive force and hydrogen bonding was proven as the main adsorption mechanism. Multiple cycle runs experiment revealed the excellent stability of recycled SSDBCs. This work provided a promising method of sludge resourceful treatment using an efficient, economic, cyclic, and convenient material for typical organic contaminant in the environment.

Tetrabromobisphenol A (TBBPA), a commonly utilized flame retardant, presents potential risks to both environmental and human health, with particular concern regarding its impact on embryonic development. This study employed zebrafish embryos as a model organism to investigate the comprehensive toxicological effects of TBBPA exposure, integrating metabolomics analysis with molecular and biochemical approaches. Embryos exposed to TBBPA concentrations ranging from 0.5 to 1.5 mg/L exhibited significant dose-dependent developmental abnormalities, including pericardial edema, yolk sac enlargement, and body axis curvature. At 96 h, we observed 50 % mortality at 1 mg/L. At 144 h of exposure to 0.1 mg/L TBBPA, automated behavioral analysis revealed significant changes in larval swimming patterns, characterized by reduced total distance moved, shortened active swimming time, impaired acceleration parameters, and abnormal spatial distribution. UHPLC-Q-TOF-MS-based metabolomics analysis revealed substantial perturbations in multiple biochemical pathways, particularly affecting neurotransmitter metabolism, energy homeostasis, and oxidative stress responses. TBBPA exposure significantly disrupted dopamine and serotonin metabolism, evidenced by altered enzyme expression and metabolite levels. Notable changes in oxidative stress markers, including GSH, MDA, and SOD, indicated significant cellular damage, while inflammatory responses showed dysregulation of both pro- and anti-inflammatory cytokines. Energy metabolism was comprehensively affected, with disruptions in glycolysis, TCA cycle, and amino acid metabolism pathways. The study identified key metabolic signatures of TBBPA toxicity and elucidated the interconnected mechanisms underlying its developmental impacts, providing valuable insights for environmental risk assessment and regulatory considerations. These findings emphasize the complex nature of TBBPA toxicity and highlight the need for careful evaluation of its environmental impact, particularly concerning early developmental exposure.

Brominated flame retardants (BFRs) are ubiquitous industrial chemicals. In China, BFRs that are applied in large quantities include decabromodiphenyl ether (BDE-209), tetrabromobisphenol A (TBBPA), and hexabromocyclododecane (HBCD). Although findings are not always unequivocal, mounting evidence in vivo suggests that the BFRs have potential neurotoxicity. The present study aimed to assess and compare the neurotoxic effects of these three BFRs’ exposure. Male mice were orally exposed to BDE-209, TBBPA, or HBCD at 50 and 100 mg/kg bw/day for 28 days. The cognitive behavior, oxidative stress (ROS, MDA, and GSH), apoptosis-related genes (caspase-3, bax, and bcl-2), memory-related proteins (BDNF and PSD-95), and neurotransmitters (AChE and ChAT) were detected comparatively. Results showed that high doses of BDE-209, TBBPA, and HBCD exposure impaired spatial memory of mice, elevated ROS and MDA and reduced GSH levels of hippocampus, upregulated caspase-3 and bax expressions, decreased BDNF and PSD-95 levels, and disordered AChE and ChAT levels. Notably, BDE-209 caused greater adverse effects > HBCD > TBBPA. This study confirms and extends that these three BFRs had similar neurotoxic effects at current concentrations, although they may be more or less toxic.
