TMT Quantitative Proteomics Reveals the Molecular Mechanism Behind Meat Quality Changes in Nile Tilapia Exposed to Environmental Concentrations of Microcystin-LR

Yichao Li,Huarong Xiao,Jun Xie,Liping Liu,Fajun Jiang,Jingqiu Liao,Ermeng Yu

This study investigated the effects of chronic MC-LR exposure (0 μg/L [Control], 1 μg/L [M1], 3 μg/L [M3], 10 μg/L [M10], and 30 μg/L [M30]) on the muscle nutrient composition, meat quality, and muscle proteomic profile of Nile tilapia (Oreochromis niloticus). In the high-dose group (M30), MC-LR exposure compromised the muscle antioxidant status of Nile tilapia, resulting in reduced meat quality, as evidenced by decreased pH value and water-holding capacity, elevated lipid/protein oxidation, and altered texture parameters (shear force and fragmentation index). Proteomic analysis further revealed a downregulation of proteins associated with ribosomes, suggesting an impairment of muscle protein synthesis in the M30 group. Moreover, despite chronic exposure, only low levels of MC-LR accumulated in the muscle tissue, indicating a negligible health risk to consumers. Collectively, these findings offered valuable insights into the impact of environmental MC-LR contamination on fish muscle quality and nutritional value.

TMT-based quantitative proteomics and non-targeted metabolomics reveal the antibacterial mechanism of rhein against Bacillus cereus and its potential application as a food preservative

Yanmei Li,Zixi Chen,Zigan Xu,Jinmei Wu,Dexin Yang,Yeye Yu,Ting Xie,Yong Dai,Shaodong Luan,Hanchao Gao

The study aims to investigate the antibacterial effect and mechanism of rhein against Bacillus cereus using Tandem Mass Tag (TMT) quantitative proteomics combined with non-targeted metabolomics. The results showed that rhein exhibited strong antimicrobial effect against B. cereus , with a minimum inhibitory concentration (MIC) and a minimum bactericidal concentration (MBC) of 1.2 μg/mL and 2.4 μg/mL, respectively. Additionally, rhein treatment increased membrane permeability, induced reactive oxygen species generation, and disrupted the cell morphology of B. cereus cells. Notably, rhein could effectively inhibit the proliferation of B. cereus in cooked rice and skimmed milk during the storage at 4 °C. Proteomics and metabolomics analyses identified 288 differentially expressed proteins (DEPs) and 402 differential metabolites (DMs) in rhein-treated B . cereus . These molecules were involved in multiple biological pathways related to primary metabolism, carbon metabolism, amino acid biosynthesis and energy production, as well as the synthesis of secondary metabolites and stress response-related signaling pathways. Bioinformatics analysis further revealed that rhein disrupted key cellular processes, including amino acid metabolism, nucleotide metabolism, glycolysis/gluconeogenesis and flagellar structure. These disruptions significantly impaired bacterial metabolic functions and motility, ultimately inhibiting bacterial growth and leading to cell death. This research provides valuable understanding into the mechanism of rhein's action against B . cereus and lays theoretical foundations for its potential use as an antibacterial agent in the food industry.

Buyang huanwu decoction improves synaptic plasticity of ischemic stroke by regulating the cAMP/PKA/CREB pathway

Jingyuan Mo,Weiguo Liao,Jinyan Du,Xiaoling Huang,Yaxin Li,Anyu Su,Lanying Zhong,Mingyu Gong,Pengcheng Wang,Zai Liu,Huizhen Kuang,Lisheng Wang

Ethnopharmacological relevance Ischemic stroke is an acute central nervous system disease that poses a threat to human health. It induces a series of severe pathological mechanisms, ultimately leading to neuronal cell death in the brain due to local ischemia and hypoxia. Buyang Huanwu decoction (BYHWD), as a representative formula for treating ischemic stroke, has shown good therapeutic effects in stroke patients. Aim of the study This study aimed to explore the mechanism of BYHWD in promoting neural remodeling after ischemic stroke from the perspective of neuronal synaptic plasticity, based on the cAMP/PKA/CREB signaling pathway. Materials and methods A modified suture technique was employed to establish a rat model of MCAO. The rats were divided into sham, model, and BYHWD (20 g/kg) groups. After the corresponding intervention, rat brains from each group were collected. TMT quantitative proteomics technology was employed for the research. Following proteomics studies, we investigated the mechanism of BYHWD in the intervention of ischemic stroke through animal experiments and cell experiments. The experimental animals were divided into sham, model, and BYHWD (5 g/kg, 10 g/kg, and 20 g/kg) groups. Infarct volume and severity of brain injury were measured by TTC staining. HE staining was utilized to evaluate alterations in tissue morphology. The Golgi staining was used to observe changes in cell body, dendrites, and dendritic spines. Transmission electron microscopy was used to observe the ultrastructure of synapses in the cortex and hippocampus. TUNEL staining was conducted to identify apoptotic neurons. Meanwhile, a stable and reliable (OGD/R) SH-SY5Y cell model was established. The effect of BYHWD-containing serum on SH-SY5Y cell viability was measured by CCK-8 kit. The apoptosis situation of SH-SY5Y cells was determined by Annexin V-FITC/PI. Immunofluorescence was employed to measure the fluorescence intensity of synaptic-related factors Syt1, Psd95, and Syn1. Synaptic plasticity pathways were assessed by using RT-qPCR and Western blot to determine the expression levels of cAMP, Psd95, Prkacb, Creb1/p-Creb1, BDNF, Shank2, Syn1, Syt1, Bcl-2, Bcl-2/Bax mRNA and proteins. Results After treatment with BYHWD, notable alterations were detected in the signaling pathways linked to synaptic plasticity and the cAMP signaling pathway-related targets among the intervention targets. This trend of change was also reflected in other bioinformatics analyses, indicating the important role of synaptic plasticity changes before and after modeling and drug intervention. The results of vivo and vitro experiments showed that BYHWD improved local pathological changes, and reduced cerebral infarct volume, and neurological function scores in MCAO rats. It increased dendritic spine density, improved synaptic structural plasticity, and had a certain neuroprotective effect. BYHWD increased the postsynaptic membrane thickness, synaptic interface curvature, and synaptic quantity. 10% BYHWD-containing serum was determined as the optimal concentration for treatment. 10% BYHWD-containing serum significantly reduced the overall apoptotic rate of (OGD/R) SH-SY5Y cells. Immunofluorescence experiments demonstrated that 10% BYHWD-containing serum could improve synaptic plasticity and increase the relative expression levels of synaptic-related proteins Syt1, Psd95, and Syn1. BYHWD and decoction-containing serum upregulated the mRNA and protein expression levels in (OGD/R) SH-SY5Y cells and MCAO rats, suggesting its ability to improve damaged neuronal synaptic plasticity and enhance transmission efficiency, which might be achieved through the regulation of the cAMP/PKA/CREB pathway. Conclusions This study may provide a basis for clinical medication by elucidating the underlying experimental evidence for the promotion of neural plasticity after ischemic stroke by BYHWD.

Effect of additives on stabilization and inhibition of mercury re-emission in simulated desulphurization slurry

Wu H.,Sun J.,Zhou C.,Yang H.

The introduction of additives into desulfurization slurry may inhibit the mercury re-emission from slurry and improve the stabilization of mercury in slurry. In this paper, the effect of common additives, such as sodium sulfide (Na 2 S), 2,4,6-trimercaptotriazine trisodium (TMT-18), sodium dithiocarbamate (DTCR-2) and Fenton reagent, on mercury distribution in gas–liquid–solid phase was investigated under typical operating conditions. Meanwhile, the effect of different additive dosages on the inhibition performance of mercury re-emission was studied. Furthermore, the inhibition mechanisms of different additives were also discussed. The experimental results showed that Na 2 S, TMT-18, DTCR-2 and Fenton reagent could inhibit the mercury re-emission from desulfurization slurry and some mercury that might reemit into the gas phase before would be restrained in the solid phase. For additives such as Na 2 S, TMT-18 and DTCR-2, with the increase in additive dosages, the inhibition performances of mercury re-emission were improved, but the enhancing effects tended to level off. More mercury could be restrained in the solid phase by forming HgS, Hg-TMT and Hg-DTCR. However, for additives such as Fenton reagent, the inhibition performance of mercury re-emission was first increased and then decreased with the proportion of Fe 2+ and H 2 O 2 in Fenton reagent increasing. There existed an optimal proportion. The oxygenated free radical generated by Fenton reagent with Fe 2+ as catalysis was the main reason for the inhibition of mercury re-emission, and more mercury would be oxidized and then restrained in solid phase due to the formation of HgSO 4 and Fe(OH) 3 .

Proteomic profiling of eIF3a conditional knockout mice

Zhuo Wei,Chen Juan,Jiang Shilong,Zheng Juyan,Huang Hanxue,Xie Pan,Li Wei,Lei Mengrong,Yin Jiye,Gao Ying,Liu Zhaoqian

Eukaryotic translation initiation factor 3 subunit A (eIF3a) is the largest subunit of the eukaryotic translation initiation factor 3 (eIF3). eIF3a plays an integral role in protein biosynthesis, hence impacting the onset, development, and treatment of tumors. The proteins regulated by eIF3a are still being explored in vivo. In this study, a Cre-loxP system was used to generate eIF3a conditional knockout mice. Tandem mass tag (TMT) labeling with LC-MS/MS analysis was used to identify differentially expressed proteins (DEPs) in fat, lungs, skin, and spleen tissue of the eIF3a knockout mice and controls. Bioinformatics analysis was then used to explore the functions and molecular signaling pathways of these protein landscapes. It was observed that eIF3a is essential for life sustenance. Abnormal tissue pathology was found in the lungs, fat, skin, spleen, and thymus. In total, 588, 210, 324, and 944 DEPs were quantified in the lungs, fat, skin, and spleen, respectively, of the eIF3a knockout mice as compared to the control. The quantified differentially expressed proteins were tissue-specific, except for eight proteins shared by the four tissues. A broad range of functions for eIF3a, including cellular signaling pathway, immune response, metabolism, defense response, phagocytes, and DNA replication, has been revealed using bioinformatics analysis. Herein, several pathways related to oxidative stress in the Kyoto Encyclopedia of Genes and Genomes (KEGG) database, including nitrogen metabolism, peroxisome, cytochrome P450 drug metabolism, pyruvate metabolism, PPAR signaling pathway, phospholipase D signaling pathway, B-cell receptor signaling pathway, ferroptosis, and focal adhesion, have been identified. Collectively, this study shows that eIF3a is an essential gene for sustaining life, and its downstream proteins are involved in diverse novel functions beyond mRNA translational regulation.

Shengli Fu,Mingmei Ding,Qingjian Liang,Yanjian Yang,Meng Chen,Xiufang Wei,Anli Wang,Shaoan Liao,Jianmin Ye

The immune mechanism elicited in pufferfish ( Takifugu obscurus ) against the invasion of Aeromonas hydrophila is still poorly understood. We examined the spleen of pufferfish at the transcriptome and proteome levels by using Illumina-seq and TMT coupled mass spectrometry after 12 h infection by A. hydrophila , respectively. A total of 2,339 genes (1,512 up-regulated and 827 down-regulated) and 537 (237 up-regulated and 300 down-regulated) proteins were identified. GO and KEGG analyses revealed that the responses to stimulus were the main biological processes , intestinal immune network for IgT production and calcium signaling pathway . Fourteen genes (8 up-regulated and 6 down-regulated) and proteins (5 up-regulated and 9 down-regulated) involved immune responses or signal transduction were validated by qRT-PCR and parallel reaction monitoring to confirm the reliability of the transcriptomic and proteomic analyses, respectively. Moreover, qRT-PCR and flow cytometry were used to detect dynamics of the genes in calcium signaling pathway and changes of concentration of cytoplasm Ca 2+ in spleen cells within a 72 h challenge. This study provides the findings regarding immune response, especially intestinal immune network for IgT production pathway and calcium signaling pathway at the molecular, protein and cellular in pufferfish after infection by A. hydrophila . These results would provide a new insight and molecular targets into the response to pathogenic infection in pufferfish.

Sensitive quantification of mevalonate pathway intermediates and prediction of relative novel analogs by chemical derivatization-based LC-MS/MS

Meiyu Gao,Jiarui Sun,Qinwen Xiao,Yuanyuan Zhai,Yuan Tian,Zunjian Zhang,Fengguo Xu,Pei Zhang

The mevalonate (MVA) pathway plays a crucial role in the occurrence and progression of various diseases, such as osteoporosis, breast cancer, and lung cancer, etc. However, determining all the MVA pathway intermediates is still challenging due to their high polarity, low concentration, chelation effect with metal compartments, and poor mass spectrometric response. In this study, we established a liquid chromatography-tandem mass spectrometry (LC-MS/MS) method coupled with N 2 , N 2 , N 4 , N 4 -tetramethyl-6-(4-(piperazin-1-ylsulfonyl) phenyl)-1,3,5-triazine-2,4-diamine (Tmt-PP) labeling for the simultaneous analysis of all MVA intermediates in biospecimens. Chemical derivatization significantly improved the chromatographic retention, peak shape, and detection sensitivity of the analytes. Moreover, we employed a method named mass spectrum calculation to achieve the absolute quantification of the isomers, i.e., isopentenyl pyrophosphate (IPP) and dimethylallyl pyrophosphate (DMAPP). The established method was fully qualified and applied to explore the difference of these metabolites in cisplatin-resistant non-small cell lung cancer (NSCLC) cells. Additionally, several MVA intermediate analogs, including isopentenyl monophosphate or dimethylallyl monophosphate (IMP/DMAMP), geranyl monophosphate (GMP), 5-triphosphomevalonate (MTP), and isopentenyl triphosphate or dimethylallyl triphosphate (ITP/DMATP), were identified for the first time using a knowledge-driven prediction strategy. We further explored the tissue distribution of these novel metabolites. Overall, this work developed a sensitive quantification method for all MVA intermediates, which will enhance our understanding of the role of this pathway in various health and disease conditions. The novel metabolites we discovered warrant further investigations into their biosynthesis and biological functions.
