Renal-targeted tFNA-TPP nanoagonist treats acute kidney injury by amplifying mitophagy

Jinguo Xu,Yujun Li,Jiafeng Xu,Ruobing He,Fengchi Jiang,Jiaxue Lu,Tao Xu,Jie Wang,Chengxin Zhang

Acute kidney injury (AKI) is a critical clinical syndrome closely associated with mitochondrial dysfunction. Its etiology encompasses both intrinsic renal factors and extrarenal factors, such as cardiac complications following heart surgery. AKI remains a formidable therapeutic challenge due to the absence of strategies capable of precisely targeting and reversing mitochondrial dysfunction, a key pathological driver. To address this, we developed a renal-targeted mitochondrial autophagy inducer, tFNA-TPP, by covalently linking tetrahedral framework nucleic acid (tFNA) with triphenylphosphine (TPP) using click chemistry. This nanocomposite demonstrates a 91.6% efficiency in targeting the kidneys and functions as an integrated system for ROS scavenging and mitophagy activation. The tFNA component promotes renal accumulation and neutralizes reactive oxygen species, while the TPP moiety ensures precise delivery to damaged mitochondria, enhancing organelle-specific autophagy. In AKI models induced by both cisplatin and ischemia-reperfusion, tFNA-TPP effectively enhanced mitophagic flux, as evidenced by a 4.2-fold increase in the LC3-II/LC3-I ratio and a reduction in p62 expression, facilitating the clearance of impaired mitochondria and the restoration of renal function. By concurrently addressing oxidative stress and mitochondrial integrity, our study establishes a versatile platform for organelle-level therapy, offering a transformative approach for treating AKI and other conditions driven by mitochondrial dysregulation.

Study on the relationship between crosslinking degree and properties of TPP crosslinked chitosan nanoparticles

Cunli Pan,Junqing Qian,Changyan Zhao,Haiyang Yang,Xiaohua Zhao,Hui Guo

In order to reveal the relationship between the degree of crosslinking and the properties of chitosan nanoparticles, a potassium polyvinyl sulfate (PVSK) titration method was established for the determination of free amino group in chitosan, which showed that there was a window effect in the crosslinking degree and particle size of three kinds of molecular weight chitosan cross-linked by different content of tripolyphosphate (TPP), and the nanoparticles with moderate crosslinking degree were smaller. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) showed the moderate degree of cross-linking nanoparticles had strong antibacterial properties. IR analysis showed that the interaction between chitosan nanoparticles with different crosslinking degree and TPP were different. X-ray diffraction analysis showed that the cross-linked chitosan nanoparticles were amorphous. Above all, the crosslinking degree of TPP cross-linked chitosan nanoparticles was related to the particle size and antibacterial properties.

Preparation and biological activity studies of resveratrol loaded ionically cross-linked chitosan-TPP nanoparticles

Jie Wu,Yaping Wang,Hao Yang,Xiangyu Liu,Zhong Lu

Nanoparticles with size range of 10–500   nm can be efficiently delivered into cancer cells by the Enhanced Permeability and Retention (EPR) effect. Here, we prepared resveratrol (Res) loaded chitosan (CS) nanoparticles with the size of 172–217   nm by an ionic cross-linking method, with sodium tripolyphosphate (TPP) as the cross-linking agent, to improve the stability, solubility and tumors targeting of the natural anti-cancer drug Res. The prepared Res loaded CS-TPP nanoparticles presented long-term storage stability and UV light stability. The cumulative drug release from nanoparticles in mimetic tumor tissue condition (pH 6.5) was higher than that in physiological condition (pH 7.4). Further, Res-loaded CS-TPP nanoparticles maintained the antioxidant activity of Res even after UV light irradiation. Cell viability study shows that the as prepared drug loaded nanoparticles had similar antiproliferative activity on hepatocellular carcinoma cells SMMC 7721 and lower cytotoxicity on normal hepatocyte cells L02 compared with free Res. Fluorescence microscopy observation revealed that the nanoparticles were efficiently taken in by SMMC 7721 cells. This work indicates the potential use of drug loaded CS-TPP nanoparticles for the efficient delivery of bioactive Res for chemotherapy.

Construction of TPP@MCA hybrid for enhancing fire safety and mechanical properties of epoxy resin

Zhiyi Pei,Yanpeng Chu,Tingting Chen,Yuan Yu,Yajie Bu

Enhancing the fire safety and mechanical properties of widely used epoxy resin (EP) composites is essential for safeguarding critical infrastructure and protecting public safety. By recrystallising triphenylphosphine (TPP) on the melamine cyanurate (MCA) surface, a TPP-decorated MCA hybrid (TPP@MCA) was constructed via a safe room-temperature recrystallization method and applied to prepare a series of EP/TPP@MCA composites with different compositions. EP/10TPP@MCA demonstrated a high limiting oxygen index (LOI) of 38.2% and fast self-extinction in vertical combustion rating (UL-94), an inherent property that forms the basis for intrinsic fire safety. The peak heat release rate and total heat release of EP/10TPP@MCA only 579.6 kW/m 2 and 48.1 MJ/m 2 , which was reduced by 44.3% and 44.2%, respectively, compared with those of pure EP. Char residue analysis indicated that the dense phosphate-containing structure and the nitrogen-rich carbon layer acted as synergistic, dual flame-retardant mechanisms in the condensed phase. Specifically, they served as both a flame retardant and a heat insulator to protect the EP matrix. A notable increase in the flexural strength of EP/5TPP@MCA with enhancements observed by 17.0%. Thus, the TPP@MCA hybrid offers an intrinsic safety solution by integrating multi-mode flame retardant mechanisms directly into the EP matrix, effectively suppressing the original fire hazards.

A TPP-Fabricated Fiber Tip Fabry-Perot Hydrogen Sensor with a Scissor-Like Clamped Beam

Xing Liu,Xinyue Yu,Wanru Tang,Ruogu Wang,Dexin Ba,Yongkang Dong

Fabry-Perot interferometers (FPIs) fabricated on fiber tips by two-photon polymerization (TPP) have shown promising sensitivity for hydrogen sensing. However, conventional TPP-fabricated FPI hydrogen sensors typically employ a single-ended cantilever beam, whose one-sided fixation provides limited structural stability, making it inherently susceptible to environmental vibrations and gas-flow disturbances that may cause false alarms during practical hydrogen detection. To solve the problem, a double-ended clamped beam FPI is proposed for hydrogen sensing. To address the sensitivity degradation caused by the increased stiffness of the double-ended configuration, we systematically investigate the influence of scissor-like structural parameters including parallelogram hollow dimensions and row number-on the mechanical response of the clamped beam through finite element analysis, and propose a scissor-like clamped beam configuration featuring a single-row parallelogram hollows beam. The proposed sensor exhibits a high hydrogen sensitivity of 1.2 nm/%, a fast response time of 15.7 s, and strong robustness against flow-rate fluctuations, with a flow-rate dependence of only 0.84 pm/SCCM, representing a 3.25-fold reduction compared with a conventional cantilever beam structure. This work provides a compact all-optical platform for safe hydrogen leak detection and monitoring in harsh environments.

Extracellular vesicle-encapsulated GDF15 alleviates doxorubicin-induced cardiotoxicity via the SUB1/TERT/TPP signalling axis

Gu Qingqing,Chen Qianwen,Deng Ye,Deng Li,Huang Jianya,Wang Jingyi,Ji Yuan,Wang Ruxing,Wang Qingjie,Sun Ling

Doxorubicin (DOX) is a highly effective chemotherapeutic agent, but its clinical application is limited by cardiotoxicity. Mitochondrial dysfunction and apoptosis are key mechanisms underlying DOX-induced cardiomyopathy (DIC), while the role of telomerase reverse transcriptase (TERT) and its specific regulatory mechanisms remain unclear. An in vitro DOX-induced H9C2 cardiomyocyte injury model was established. TERT was overexpressed via adenoviral transfection or inhibited using BIBR1532. An in vivo mouse model of DOX-induced acute myocardial injury was constructed, with TERT expression regulated via intramyocardial adenoviral injection. Cardiac function was evaluated by echocardiography, and myocardial pathology was observed via staining. Additionally, exosome-encapsulated growth differentiation factor 15 (EXO-GDF15) was extracted and identified. Its regulatory mechanism on TERT was explored using DNA pull-down and luciferase reporter gene assays. Metabolomics was used to analyze the effect of TERT on mitochondrial metabolites in cardiomyocytes. TERT overexpression reduced DOX-induced apoptosis, increased mitochondrial membrane potential, and promoted autophagy in cardiomyocyte. In mice, TERT overexpression improved cardiac function and alleviated myocardial fibrosis, inflammation, and cardiomyocyte hypertrophy. Conversely, TERT inhibition exacerbated these injuries. EXO-GDF15 could be endocytosed by cardiomyocytes and alleviated DOX-induced myocardial injury by downregulating SUB1 to promote TERT transcription. Luciferase reporter assays showed that SUB1 binds to the BS4 site of the TERT promoter. Metabolomics showed that TERT increased the mitochondrial metabolite thiamine pyrophosphate (TPP) in cardiomyocytes, which alleviated myocardial injury by participating in the tricarboxylic acid cycle. EXO-GDF15 alleviates doxorubicin-induced cardiotoxicity via the SUB1/TERT/TPP signaling axis, thereby providing advanced insights for future DIC treatment.

Development of membrane-targeting TPP+-chloramphenicol conjugates to combat methicillin-resistant staphylococcus aureus (MRSA) infections

Tao Li,Xiaoli He,Wenlan Tao,Ruixue Zhang,Qiaolin He,Hongzhi Gong,Ye Liu,Dong Luo,Maojie Zhang,Cheng Zou,Shao-Lin Zhang,Yun He

Infections caused by drug-resistant bacteria have become a new challenge in infection treatment, gravely endangering public health. Chloramphenicol (CL) is a well-known antibiotic which has lost its efficacy due to bacterial resistance . To address this issue, herein we report the design, synthesis and biological evaluations of novel triphenylphosphonium chloramphenicol conjugates (TPP + -CL). Study results indicated that compounds 39 and 42 possessed remarkable antibacterial effects against clinically isolated methicillin-resistant Staphylococcus aureus (MRSA) with MIC values ranging from 1 to 2 μg/mL, while CL was inactive to the tested MRSA strains. In addition, these conjugates exhibited rapid bactericidal properties and low toxicity, and did not readily induced bacterial resistance , obviously outperforming the parent drug CL. In a mouse model infected with a clinically isolated MRSA strain, compound 39  at a dose of 20 mg/kg exhibited a comparable or even better in vivo anti-MRSA efficacy than the golden standard drug vancomycin , while no toxicity was observed.

Antitumor and toxicity study of mitochondria-targeted triptolide derivatives using triphenylphosphine (TPP+) as a carrier

Huina Song,Wenlan Xing,Xiaojia Shi,Tao Zhang,Hongxiang Lou,Peihong Fan

Based on the higher mitochondrial membrane potential (Δψ m ) of tumor cells than normal cells, a mitochondria-targeting strategy using delocalized lipophilic cations as carriers is a promising way to improve the antitumor effect of small molecules and to reduce toxicity. Triptolide (TP) has a strong antitumor effect but is limited in the clinic due to high systemic toxicity. Mitochondria-targeted TP derivatives were designed and synthesized using triphenylphosphine cations as carriers. The optimal derivative not only maintained the antitumor activity of TP but also showed a tumor cell selectivity trend. Moreover, the optimal derivative increased the release of lactate dehydrogenase and the production of ROS , decreased Δψ m , and arrested HepG2 cells in G0/G1 phase. In a zebrafish HepG2 xenograft tumor model, the inhibitory effect of the optimal derivative was comparable to that of TP, while it had no obvious toxic effect on multiple indicators in zebrafish at the test concentrations. This work provided some evidence to support the mitochondria-targeting strategy.

Photocontrollable release and enhancement of photodynamic therapy based on host–guest supramolecular amphiphiles

Lei Xu,Wenyan Zhang,Haibo Cai,Feng Liu,Yong Wang,Yun Gao,Weian Zhang

A light-controlled porphyrinic photosensitizer release system was developed based on host–guest TPP–Azo/PEG–β-CD supramolecular amphiphiles, which could significantly enhance the efficiency of photodynamic therapy. TPP–Azo was prepared via esterification between TPP and Azo derivatives, and further used to construct supramolecular amphiphiles with PEG–β-CD. TEM images showed that TPP–Azo/PEG–β-CD amphiphiles can self-assemble into spherical micelles, and then disaggregate under irradiation with 365 nm UV light. The phototoxicity and bio-distribution of TPP–Azo/PEG–β-CD micelles against the MCF-7 cell line were also evaluated by MTT assay, flow cytometry and confocal scanning laser microscopy, respectively. According to MTT assay, the IC50 of the spherical micelle was 31.2 μg mL−1, but it became 20.6 μg mL−1 upon pre-irradiation with 365 nm UV light. This shows for the first time that UV light can well stimulate the release of porphyrin photosensitizers and improve the photodynamic therapy efficiency based on the system of host–guest supramolecular amphiphiles.

Preparation of Multifunctional Drug-loaded Microcapsules with Core-shell Three-dimensional Network Structure and their Application in Colon Targeted Release

Qiujie Fang,Na Chen,Shan Hu,Sheng Hu,Jianfeng Ye,Xiaozhou Ye,Yun Wang

Some special oral drugs may cause adverse reactions and degrade in the stomach; therefore, it is necessary to develop a new generation of multifunctional drug delivery carries to deal with such issues. Herein, by polyelectrolyte composite method using KGM and CS as raw material, CKGM/CS/TPP/5-ASA microcapsules are prepared using 5-ASA as model drug, and then coated with an acid-resistant polyacrylic acid polymer layer. The size of CKGM/CS/TPP/5-ASA microcapsules is relatively uniform, with average particle size 1.78 ± 0.02 mm. The inner part of microcapsules has a core-shell three-dimensional network structure, and the 5-ASA embedded in the network is in its crystal form. The microcapsules exhibited good encapsulate properties, with 495.5 µg mg −1 drug loading amount and 82.3% encapsulation efficiency under optimized conditions. The in vitro release properties of microcapsules are also studied, and results showed that the drug is released little in Simulated Gastric Fluid solution (pH 1.2) but fully released in Simulated Colonic Fluid or Phosphate Buffered Solution (pH 7.0), and the release rate is higher in SCF solution, indicating that β-mannanase could facilitate release. All these results indicated that CKGM/CS/TPP microcapsules have good pH-controlled release, enzymatic release, and sustained release properties, hence they are promising multifunctional colon-targeted release carriers.

The Apoptosis of Liver Cancer Cells Promoted by Curcumin/TPP-CZL Nanomicelles With Mitochondrial Targeting Function

Li Wanyu,Chen Yanan,He Kun,Cao Tianshou,Song Daibo,Yang Huiling,Li Li,Lin Jiantao

The mitochondrion is one of the most important cellular organelles, and many drugs work by acting on mitochondria. Curcumin (Cur)-induced apoptosis of HepG2 in liver cancer cells is closely related to the function of inhibiting mitochondria. However, the mitochondrion-targeting curcumin delivery system was rarely been reported. It is important to develop a high-efficiency mitochondrion-targeting curcumin vector that can deliver curcumin into mitochondria directly. Here, a special mitochondrion-targeting delivery system based on triphenylphosphine bromide (TPP)-chitosan-g-poly-(N-3-carbobenzyloxy-l-lysine) (CZL) with TPP functional on the surface is designed to perform highly efficient mitochondria-targeting delivery for effective liver cancer cell killing in vitro. The TEM images showed that the nanomicelles were spherical; the results of fluorescence test showed that TPP-CZL nanomicelles could promote the cellular uptake of drugs and finally targeted to the mitochondria. The results of cell survival rate and Hoechst staining showed that curcumin/TPP-CZL nanomicelles could promote the apoptosis of liver cancer cells. Curcumin/TPP-CZL nanomicelles could significantly reduce the mitochondrial membrane potential, increase the expression of pro apoptotic protein Bcl-2, and reduce the expression of antiapoptotic Bax protein, and these results were significantly better than curcumin/CZL nanomicelles and curcumin. It is a potential drug delivery system with high efficiency to target mitochondria of liver cancer cells.

Singlet oxygen generation proportion from triplet state of porphyrin in water

Lu Li,Feng Qin,Yongda Wang,Zhiguo Zhang

The proportion of singlet oxygen ( R ) produced from the triplet state of porphyrin in aqueous solutions was investigated by substituting porphyrin molecules with the metalloporphyrin Platinum(II) meso -tetraphenylporphyrin (Pt-TPP). The feasibility of using platinum porphyrin as a substitute for porphyrin to study the photophysical properties in photodynamic therapy (PDT) was confirmed. The model of first-order decay in a water-methanol mixed solution encompasses potential deactivation processes of the excited triplet states and was built, which was confirmed by monitoring the change in the total transition rate of the T 1 state ( A 0 ) value at different water contents under anoxic environments, obtained through measuring the phosphorescence lifetime ( τ p ) of Pt-TPP. Combined with A 0 and the quenching rate constant of T 1 by O 2 ( k q ), the formula for R in relation to changes in oxygen can be deduced in pure water.
