A new electrochemical sensor for Taxifolin based on RGO-Co3S4@MoS2 modified electrode

Di Huang,Lingxi Yang,Xing lin Li,Lina Zou,Baoxian Ye

A novel electrochemical sensor was designed and constructed for high-sensitive detection of Taxifolin based on Co 3 S 4 @MoS 2 loaded on reduced graphene oxide (RGO-Co 3 S 4 @MoS 2 ) as modified materials. It’s the first time for Co 3 S 4 @MoS 2 was exploited to constructed electrochemical sensor. Owing to the Co 3 S 4 @MoS 2 providing a huge surface area to enrich more Taxifolin and the excellent electric conductivity of RGO, Taxifolin was detected sensitively with a linear range from 5.0 × 10 −9 to 1.0 × 10 −6  mol L − 1 and limit of detection down to 1.67 × 10 −9  mol L − 1 (S/N = 3). In addition, the sensor also demonstrated strong anti-interference properties in the presence of some metal ions and organic compounds. The proposed method was successfully applied for determination of Taxifolin in traditional Chinese medicine in fructus polygoni orientalis with satisfactory results.

In vitro and in vivo inhibitory activity of taxifolin on three digestive enzymes

Hang Su,Yi-Ting Ruan,Ying Li,Ji-Guang Chen,Zhong-Ping Yin,Qing-Feng Zhang

The inhibitory activity of taxifolin on three digestive enzymes were investigated in both vitro and vivo. Taxifolin exhibited inhibitory effect on α-glucosidase, α-amylase and pancreatic lipase with IC50 values of 0.038, 0.647 and 0.993 mg/mL, respectively. Inhibitory kinetics indicated that taxifolin was more like a competitive inhibitor of α-glucosidase and α-amylase, while it was a non-competitive inhibitor of pancreatic lipase. The binding of taxifolin caused the quenching of intrinsic fluorescence intensity of enzymes, and the binding constant (lgK a ) and the number of binding site (n) were further calculated through fluorescence titration. The values of lgK a were in the range of 4.93‐6.65, and the values of n were all close to 1. Molecular docking indicated that taxifolin could interact with α-glucosidase and α-amylase through many kinds of secondary interaction, such as hydrogen bond, π-π stack, etc. In vivo study revealed that pre-administration with taxifolin can significantly improve the postprandial hyperglycemia in rat. Furthermore, its can also decrease triglyceride absorption through the inhibition of pancreatic lipase.

Interaction with taxifolin reduces the digestibility of corn starch in vitro and in vivo

Li Mang-Mang,Peng Rui-Yan,Wang Wen-Jun,Xu Hai-Xia,Yin Zhong-Ping,Chen Ji-Guang,Zhang Qing-Feng

Reducing the hydrolysis of starch in the gastrointestinal tract can improve postprandial hyperglycemia, which is beneficial for diabetes. The blends of corn starch with taxifolin (CS-Tax) were prepared with different taxifolin proportion. Iodine binding test showed that taxifolin can occupy the helix of starch, and the affinity is stronger than that of iodine. FTIR and XRD characterization revealed secondary interaction between taxifolin and starch in the blends, and the crystallization of taxifolin was suppressed. Taxifolin decreased the gelatinization temperature and enthalpy for starch. Scanning electron microscopy showed that the morphology of the blends was the same as starch granules. In vitro digestion experiment showed that taxifolin could slow down starch hydrolysis. Compared with native starch, the content of rapidly digestible starch in CS-10%Tax decreased from 56.35 to 49.20%, while the content of resistant starch increased from 17.62 to 29.61%. The comparison in vivo showed that after intragastric administration of CS-10%Tax in rats, the increase of blood glucose slowed down and the maximum decreased from 7.1 to 6.2 mM. Hence, taxifolin is prospective in preparing low glycemic index starchy foods and deserves further study.

Simultaneous Determination of Three Flavonoids in Rat Plasma of Masson Pinus Needles (Pinus massoniana L.) Extracts by UHPLC-MS/MS and Application to Pharmacokinetics Studies after Oral and Transdermal Administration

Xia Chenjie,Zhu Hongliu,Hu Changhui,Liu Jie,Hua Haibing,Yang Jianqiu,Chen Zhipeng,Gao Yihong

Pinus massoniana needles, a traditional herb, were applied to prevent hair loss in China. Studies available mainly focused on pine needle flavonoids with various biological activities. However, there has been no pharmacokinetics study of the flavonoids from Pinus needles extract. A selective and sensitive ultra-high performance liquid chromatography–tandem mass spectrometry (UHPLC-MS/MS) method was developed to simultaneously quantify taxifolin, quercetin and catechin in rat plasma. To separate the three constituents, an Agilent Extend-C18 column (2.1 mm × 100 mm, 1.8 μm) was used with a mobile phrase of (A) 0.1% formic acid and (B) acetonitrile. The analytes were measured by multiple reaction monitoring in the negative ionization mode. There was good linearity in the established UHPLC-MS/MS method, with a coefficient of determination (r2) of >0.99. The accuracy, intra-day and inter-day precision and recovery were all satisfactory and these 3 compounds were stable under the tested conditions. The validated method in this study was successfully applied to pharmacokinetic study in healthy rats after oral and transdermal administration of Pinus needles extract. The results could provide further research foundation for pine needle extract as external preparations.

Non-covalent interactions between whey protein isolate and taxifolin and their potential application in yogurt

Chenligen Bao,Mi Yan,Fangyu Li,Liqing Jiang,Tiehua Zhang

This study investigated the interaction mechanism of non-covalent complexes between whey protein isolate (WPI) and taxifolin (Tax) and subsequently applied the complexes into yogurt to evaluate their effects on the quality and functionality of yogurt during storage. The results demonstrated that the WPI and Tax can form granular aggregates with average particle sizes between 184.57 and 287.03 nm. As Tax concentration increases, WPI exhibits enhanced UV–visible absorption alongside intensified static fluorescence quenching, primarily driven by conventional hydrogen bonding and π-alkyl interactions between the two compounds. And the incorporation of the WPI + Tax complex into yogurt significantly improves its water-holding capacity and reduces syneresis, with the WPI + 40 μM group showing the optimal effect. The complexes also enhanced yogurt's antioxidant activity and inhibitory effects against α-glucosidase and α-amylase, which reached the highest levels on day 14 of storage, while maintaining good sensory acceptability.

Taxifolin-loaded cellulose/l-arginine-chitosan hydrogel promoting bone defect repair through osteogenesis and angiogenesis

Jiali Yang,Yuanyuan Han,Lifeng Zhang,Qiteng Ding,Shuwen Sun,Shuai Zhang,Yue Wang,Ning Wang,Hewei Wei,Taojing Yu,Wencong Liu,Chuanbo Ding

Bone defects have always been a difficult problem in clinical practice. Taxifolin (TAX) is beneficial to bone regeneration. In order to obtain more attractive biomaterials, we extracted cellulose (LC) from larch sawdust and prepared a TAX-loaded hydrogel (DCT) together with L-arginine chitosan (CA). The hydrogel had a uniform porous structure and good mechanical properties. After drug loading, the adhesion, antibacterial, and antioxidant properties of the hydrogel were improved. In addition, DCT has good biocompatibility. In vitro cell experiment results showed that DCT can promote cell proliferation and migration, and DCT has a certain ability to promote the osteogenic activity and enhance the expression of osteoblastic factors (OCN and Osx) and vascular endothelial growth factor (VEGF) in MC3T3-E1 cells. In addition, in vivo studies showed that the DCT treatment could promote bone regeneration by increasing the levels of osteogenic markers (OPN, OCN, and Osx) and VEGF. Transcriptome analysis showed that DCT intervention affected the hematopoiesis function in bone tissue. Meanwhile, KEGG analysis showed that DCT promoted bone regeneration mainly by activating the PI3K/AKT signaling pathway, which was verified by a western blot. Therefore, DCT is a good material for bone repair.

Taxifolin Inhibits WSSV Infection and Transmission by Increasing the Innate Immune Response in Litopenaeus vannamei

Xu Zhang,Li-Peng Shan,Qi Zhao,Lei Liu,Xu OuYang,Yang Hu,Chen-Jie Fei,Jiong Chen

An outbreak of white spot syndrome virus (WSSV) can hit shrimp culture with a devastating blow, and there are no suitable measures to prevent infection with the virus. In this study, the activity of active molecules from Chinese herbs against WSSV was evaluated and screened. Taxifolin had the highest rate (84%) of inhibition of the WSSV infection. The viral infectivity and genome copy number were reduced by 41% when WSSV virion was pretreated with taxifolin prior to shrimp infection. A continuous exchange of taxifolin significantly reduced the mortality of shrimp infected with WSSV. Due to the WSSV virion infectivity being affected by taxifolin, the horizontal transmission of the virus was blocked with an inhibition rate of up to 30%, which would further reduce the cost of a viral outbreak. Additionally, the viral genome copy number was also reduced by up to 63% in shrimp preincubated in taxifolin for 8 h. There may be a connection to the enhancement of innate immunity in shrimp that resulted in a 15% reduction in mortality for taxifolin-fed shrimp after the WSSV challenge. After dietary supplementation with taxifolin, the resistance of larvae to WSSV was improved, indicating that taxifolin may be a potential immunostimulant for shrimp to prevent WSD. Therefore, the results indicate that taxifolin has application potential for blocking a WSSV outbreak and reducing the loss of shrimp culture. Keywords:taxifolin;WSSV;antiviral;Chinese herbs;Litopenaeus vannamei

Preparation of nanocomposite membranes loaded with taxifolin liposome and its mechanism of wound healing in diabetic mice

Qiteng Ding,Chuanbo Ding,Xinglong Liu,Yinan Zheng,Yingchun Zhao,Shuai Zhang,Shuwen Sun,Zanwen Peng,Wencong Liu

In this study, a new wound dressing was developed to speed up the healing process of diabetic wounds. First of all, taxifolin liposome (TL) was manufactured in this study. Then, taxifolin (TAX) and TL were mixed with polyvinyl alcohol (PVA) and chitosan (CS) by electrostatic spinning to prepare nanocomposite membranes. Finally, the mechanism of nanocomposite membranes to accelerate diabetic wound healing was investigated. The diameter of TL-loaded polyvinyl alcohol/chitosan nanocomposite membranes (PVA/CS/TL) was 429.43 ± 78.07 nm. The results of in vitro experiments demonstrated that the PVA/CS/TL had better water absorption, water vapor transmission rate (WVTR), porosity, hydrophilicity, mechanical properties, slow-release, antioxidant capacity, and antibacterial properties. The results of in vivo experiments demonstrated that the wound healing rate of mice treated with PVA/CS/TL for eighteen days was 98.39 ± 0.34 %. Histopathological staining, immunohistochemical staining, and western blot experiments also demonstrated that PVA/CS/TL could promote wound healing in diabetic mice by inhibiting the activation of inhibitor kappa B alpha (IκBα)/nuclear factor-kappa B (NF-κB) signaling pathway and related pro-inflammatory factors to increase the expression of CD31 and VEGF in skin tissues. These results suggested that PVA/CS/TL could be a potential candidate for wound dressing to promote chronic skin wound healing.

Self-assembly of MoS2 nanosheet on functionalized pomelo peel derived carbon and its electrochemical sensor behavior toward taxifolin

Xia Zhang,Qiumin Cao,Zhuohuan Guo,Meiyue Zhang,Meng Zhou,Zhenzhen Zhai,Yuandong Xu

MoS 2 nanosheet arrays grown on functional pomelo peel derived carbon (PPC) is constructed via in situ self-assembly method. The obtained MoS 2 /PPC composite is further used to fabricate electrochemical sensor for taxifolin detection. The current response of taxifolin on MoS 2 /PPC is significantly enhanced due to the synergistic effect between MoS 2 and PPC. It is found that the charge transfer impedance and the Ohmic resistance corresponding to MoS 2 /PPC are the lowest, indicating fast electron transfer rate and good conductivity. Further investigation reveals that the redox peak current is linearly related to the concentration of taxifolin in the range of 7 × 10 −8 –1 × 10 −5 mol·L −1 and the limit of detection is calculated to be 2.3 × 10 −8 mol·L −1 (S/N = 3). In addition, no obvious interfering phenomenonhas been observed in the detection process of taxifolin, demonstrating that MoS 2 /PPC presents good selectivity toward taxifolin determination.

Effect of the C2C3 double bond and sodium taurodeoxycholate addition on the quercetin-hemoglobin and taxifolin-hemoglobin interactions

Yu Guo,Hanlu Jiang,Xinru Zhang,Mengyao Guo,Hongying Fang,Qian Zhang,Jie Liu

The structure of polyphenolic compounds plays a pivotal role in determining their functional efficacy. Moreover, the presence of bile salt surfactants can significantly impact their metabolism. Hence, this study employed multi-spectroscopic methods to explore the binding mechanism of Quercetin (Que) and Taxifolin (Tax, dihydroquercetin) with bovine hemoglobin (HB), influenced by the addition of sodium taurodeoxycholate (NaTDC). The results indicate that, through the static quenching mechanism, nearly a single Que. or Tax molecule can spontaneously form a complex on each HB chain. This process is driven by Van der Waals forces, hydrophobic interaction, and hydrogen bonds. During the interaction, the secondary structure of HB chains undergoes changes with an enhanced hydrophobicity. The hydrogenation of C2 C3 double bond of flavonols increases the structural flexibility and helps to close to the active sites of HB, resulting in the stronger binding affinity of Tax on HB chains than Que. Additionally, the presence of this double bond exerts a notable influence on the physicochemical properties of polyphenols, such as solubility, antioxidant activity, and antibacterial activity. Specifically, the solubility of pure Tax in water, and its bacteriostatic effect against Klebsiella Pneumoniae (KP) and Methicillin-resistant Staphylococcus Aureus (MRSA), are superior to that of Que. The C2 C3 double bond of Que. make it exhibiting better antioxidant ability than Tax. After interacting with HB, the antioxidant capacity are enhanced, and the bacteriostatic effects are either weakened or remain essentially unchanged compared to those of pure Que./Tax. The addition of NaTDC can enhance the low solubility of Que./Tax and regulate their interaction strength with HB, thereby further influencing their bioavailability. For the Tax/Que-HB mixed system, the presence of NaTDC strengthens their antioxidant capacity, but does not affect the antimicrobial activity basically. These findings hold significant practical implications for enhancing the therapeutic efficacy and bioavailability of natural products.

Multi-Target Photoprotection by Taxifolin Against UVB-Induced Keratinocyte Injury Through UVB Filtration, ROS Scavenging and Transcriptomic-Proteomic Reprogramming

Fangfang Chen,Yihan Cai,Jinxiong Wu,Nengzhen Fang,Fei Li,Hongtan Wu,Yu-Pei Chen

Taxifolin, a natural flavonoid, consistently exerts cytoprotective effects against various oxidative stresses. In this study, we systematically evaluated its photoprotective efficacy and underlying mechanisms against ultraviolet B (UVB)-induced injury in human immortalized keratinocytes (HaCaT). Cell viability and apoptosis were assessed by MTT, fluorescence staining, and flow cytometry, while integrative transcriptomic and proteomic analyses were employed to identify core pathways and key mediators. Taxifolin exhibited antioxidant capacity comparable to that of ascorbic acid under identical in vitro radical-scavenging assays. Moreover, it displayed a strong absorption peak at 289 nm that overlaps the UVB spectrum (280–320 nm), enabling it to act as a chemical sunscreen. In UVB-challenged HaCaT cells, taxifolin markedly reduced intracellular reactive oxygen species (ROS) and attenuated JNK/p38 MAPK activation, as evidenced by Western blot, thereby breaking the ROS-MAPK vicious cycle. Multi-omics revealed that taxifolin was associated with attenuation of UVB-imposed G1/S arrest concomitant with restored Cyclin expression, while up-regulating MYC, FOXQ1, HMOX1 and AP-1 components c-Jun/c-Fos and thereby switching on a pro-survival transcriptional program. Consequently, apoptosis was suppressed and survival was significantly improved. Collectively, taxifolin integrated chemical filtration, ROS scavenging and signaling modulation to support a multi-target photoprotective network, which provides mechanistic insight into taxifolin-mediated cytoprotection and identifies candidate molecular nodes for further validation.

Protective effect and mechanism of taxifolin 7-rhamnoside extracted from Hypericum japonicum against LPS/D-GalN-induced acute liver injury

Hengxing Ran,Yan Wen,Yiebi Qin,Kaiwen Kang,Zhengrong Wu,Yifan Li,Yu Wang,Rihui Wu,Dongli Li,Song Ang,Lishe Gan,Jingwei Jin

This study aims to identify the most primary hepatoprotective compound from Hypericum japonicum and elucidate its underlying mechanisms of liver protection. This study screened 15 compounds from Hypericum japonicum against D-GalN-induced AML12 cell injury, identifying TAX ( Compound 8 , taxifolin 7-rhamnoside) as the most hepatoprotective. This study indicates that TAX reduced AST, ALT, and MDA levels, while enhancing SOD and GSH levels in D-GalN-induced AML12 and LPS/D-GalN -induced liver. In addition, TAX attenuated LPS/D-GalN-induced hepatic injury by lowering inflammatory infiltration and pro-inflammatory cytokines (TNF-α, IL-6 and NO) levels. Network pharmacology analysis revealed common targets between TAX and drug-induced liver injury, forming a protein-protein interaction (PPI) network. Functional enrichment (GO/KEGG) and molecular docking indicated strong binding affinity between TAX and AKT1, confirmed by molecular dynamics simulations. Western blot analyses that TAX ameliorated inflammation and oxidative stress through down-regulating the AKT/NF-κB pathways and up-regulating Nrf2/HO-1 pathways in vivo, TAX inhibited apoptosis via up-regulating Bcl-2 and down-regulating Bax in vitro. Collectively, this study demonstrates that TAX ameliorates LPS/D-GalN-induced ALI by attenuating hepatocyte apoptosis, inflammatory responses, and oxidative stress.
