Degradation and Transformation Mechanisms of Zanthoxylum Alkylamides Exposed to UVB Light

Rui Wang,Chaolong Rao,Qiuyan Liu,Xiong Liu

Zanthoxylumoleoresin, a concentrated extract derived fromZanthoxylum bungeanum, is rich in non-volatile, intensely flavorful substances and amide compounds, such as hydroxy-α-sanshool, hydroxy-β-sanshool, and hydroxy-ε-sanshool. The production process ofZanthoxylumoleoresin remains unstandardized, and there is still a lack of research on the precise classification and quantification of its key chemical constituents, as well as the stability of these compounds when produced using different extraction methods. This study utilized preparative liquid chromatography to extract and purify amide compounds fromZanthoxylumoleoresin, successfully isolating three sanshools: hydroxy-α-sanshool, hydroxy-β-sanshool, and hydroxy-ε-sanshool. The stability of three these sanshools under UVB irradiation in different solvents was explored in UVB-simulated sunlight conditions to investigate the degradation or transformation mechanism ofZanthoxylumalkylamides under UVB irradiation. The findings indicate a rapid decrease in the hydroxy-α-sanshool content under UVB ultraviolet light, aligning with the second-order kinetics. This study revealed alterations in the contents of hydroxy-α-sanshool, hydroxy-β-sanshool, and hydroxy-ε-sanshool and the formation of a new compound following exposure to UVB light. This new compound, along with the three sanshools, possesses a uniformm/z264 and shares similar chemical structures. Further analysis also uncovered that these compounds are capable of undergoing isomerization reactions under UVB irradiation. This demonstrates that UVB irradiation of certain intensities can modify the concentrations and chemical structures of theseZanthoxylumalkylamides. These insights offer crucial guidance for future studies on the processing and preservation ofZanthoxylumalkylamides and their derivatives. Keywords:Zanthoxylumalkylamides;UVB light;degradation;transformation;mechanism

Alpha-Ketoglutarate Attenuates UVB-Induced Skin Photoaging by Restoring Mitochondrial Redox Homeostasis

Wenrui Zhang,Yijia Zhang,Xinyuan Wang,Yujuan Chen,Yixuan Li,Yanan Sun

Chronic ultraviolet B (UVB) radiation drives cutaneous photoaging—clinically manifesting as erythema, edema, scaling, deep wrinkling, loss of elasticity, and barrier disruption—through mitochondrial reactive oxygen species (mtROS) overproduction and quality-control failure. Here we identify α-ketoglutarate (AKG; also known as 2-oxoglutarate), a TCA-cycle intermediate and essential co-substrate for α-ketoglutarate-dependent dioxygenases (α-KGDDs), as a metabolic corrector of mitochondrial redox homeostasis in UVB-induced photoaging. In a 10-week chronic UVB SKH1 hairless mouse model, microneedle-assisted transdermal delivery of AKG dose-dependently attenuated macroscopic erythema, scaling, and erosive lesions, restored skin barrier function and dermal elasticity, preserved epidermal–dermal architecture, and protected collagen and elastic fiber integrity, with efficacy comparable to all-trans retinoic acid. Mechanistically, AKG reactivated α-KGDD/prolyl hydroxylase (PHD) catalytic function and promoted proteasomal clearance of aberrantly stabilized HIF-1α under normoxia; this was accompanied by restored AMPK Thr172 phosphorylation downstream of constitutive LKB1 and recovery of PGC-1α-driven mitochondrial biogenesis. AKG preferentially attenuated mitochondrial superoxide over total cellular ROS through a co-substrate-mediated mechanism distinct from direct radical scavenging, and its protective effects were largely abrogated by DMOG (an α-KGDD inhibitor) or compound C (an AMPK inhibitor). These findings position AKG, delivered via microneedle-assisted topical application, as a candidate metabolite-based intervention targeting the α-KGDD/HIF-1α/AMPK axis for photoaging.

Rothia mucilaginosa membrane vesicles stabilize labile iron to alleviate UVB-induced ferroptosis

Siyuan Fan,Xinyue Cai,Weiqi Wang,Meiling Wu,Xiaoyao Huang,Feng Ding,Hao Guo,Geng Dou,Haotian Luo,Shuai Shao,Jing Guo,Zhenlai Zhu,Peisheng Liu,Linfeng Cheng,Siying Liu,Zihan Li,Kun Xuan,Shiyu Liu

Ultraviolet irradiation, particularly ultraviolet B (UVB), damages keratinocytes, potentially causing actinic cheilitis. Commensal bacteria help maintain barrier function and protect the host. However, it is unclear if commensal bacteria can protect the host from UVB irradiation. Here, we demonstrate that Rothia mucilaginosa ( R. mucilaginosa )-derived membrane vesicles (RMVs) contain ferrochelatase, which stabilizes labile iron in host cells to alleviate UVB-induced ferroptosis. We demonstrate that R. mucilaginosa abundance on lip vermilion inversely correlates with actinic cheilitis severity in patients. Mechanistically, we find that UVB induces R. mucilaginosa to release RMVs, which are internalized by host cell lysosomes. The ferrochelatase contained within these RMVs catalyzes conversion of Fe 2+ and porphyrin into heme, thereby alleviating UVB-induced iron overload and ferroptosis. Topical application of RMVs relieves actinic cheilitis in patients (ChiCTR, no. ChiCTR2500100015). Collectively, we reveal an iron stabilization mechanism through which commensal bacteria protect the host against UVB and expand our understanding of the relationship between commensal bacteria and hosts.

Ultraviolet B (UVB) Photosensitivities of Tea Catechins and the Relevant Chemical Conversions

Meng Shi,Ying Nie,Xin-Qiang Zheng,Jian-Liang Lu,Yue-Rong Liang,Jian-Hui Ye

Ultraviolet B (UVB) photosensitivities of eight catechins were screened. In both water and ethanol, epicatechin (EC, 575 μM) and catechin (C, 575 μM) exhibited low photostabilities under 6 h UVB radiation with the generation of yellow photoproducts, while other catechins (epigallocatechin gallate, epigallocatechin, epicatechin gallate, gallocatechingallate, gallocatechin, catechin gallate) were relatively UVB-insensitive. Photoisomerization and photolysis were two important UVB-induced reactions to EC whereas photolysis was the dominant reaction for C. The influencing factors of time (2–10 h), solvent (water, ethanol) and substrate concentration (71.875–1150 μM) on UVB-induced chemical conversions of EC and C were investigated, and eight photoproducts were identified through ultra performance liquid chromatography-diode array detection-tandem mass spectrometry (UPLC-DAD-MS/MS) and1H nuclear magnetic resonance (1H-NMR analysis). Photolysis reaction involved two pathways, including radical reaction and photo-induced electron transfer reaction. The 2,2-diphenylpicrylhydrazyl (DPPH) scavenging abilities of eight catechins did not change upon 6 h UVB irradiation. EC and C are photosensitive catechins among eight catechins causing deep color. Keywords:photostability;catechins;photoisomerization;photolysis;photoproducts

The Effect of α-Arbutin on UVB-Induced Damage and Its Underlying Mechanism

Peng Shu,Yuan Wang,Lanyue Zhang

Ultraviolet radiation can heighten tyrosinase activity, stimulate melanocyte production, impede the metabolism of numerous melanocytes, and result in the accumulation of plaques on the skin surface. α-Arbutin, a bioactive substance extracted from the arbutin plant, has been widely used for skin whitening. In this study, the whitening effect of α-arbutin by inhibiting tyrosinase activity and alleviating the photoaging effect induced by UVB are investigated. The results indicate that α-arbutin can inhibit skin inflammation, and its effectiveness is positively correlated with concentration. Moreover, α-arbutin can reduce the skin epidermal thickness, decrease the number of inflammatory cells, and down-regulate the expression levels of IL-1β, IL-6 and TNF-α, which are inflammatory factors. It also promotes the expression of COL-1 collagen, thus playing an important role in anti-inflammatory action. Network pharmacology, metabolomics and transcriptomics further confirm that α-arbutin is related to the L-tyrosine metabolic pathway and may interfere with various signaling pathways related to melanin and other photoaging by regulating metabolic changes. Therefore, α-arbutin has a potential inhibitory effect on UVB-induced photoaging and possesses a whitening effect as a cosmetic compound.

Investigation of Ca2Al2SiO7: Pr3+/Gd3+ UVB upconverting phosphor and its optical thermometric properties

Shanshan Zhao,Saima Kanwal,Weiwen Su,Hui Lin,Zhaoxia Han,Songlin Zhuang,Dechao Yu,Dawei Zhang

On basis of fluorescence intensity ratio (FIR) over certain narrow coupled levels of rare-earth ions, up-conversion of RE-doped materials perform excellent temperature-sensing abilities for thermometry applications. A series of Pr 3+ /Gd 3+ co-doped Ca 2 Al 2 SiO 7 phosphors (Ca 2 Al 2 SiO 7 :Pr 3+ /Gd 3+ ) were prepared by a conventional high-temperature solid-state reaction. Under excitation of blue ∼445 nm, the Ca 2 Al 2 SiO 7 :Pr 3+ /Gd 3+ phosphor yields intense photoluminescence (PL) bands around 489, 528, 537, 580, 604, 613, and 646 nm due to electronic transition of 3 P 0 → 3 H 4 , 3 P 0 → 3 H 5 , 3 P 1 → 3 H 5 , 3 P 1 → 3 H 6 , 1 D 2 → 3 H 4 , 3 P 0 → 3 H 6 , and 3 P 0 → 3 F 2 of Pr 3+ , respectively. Their relative intensities are sensitively determined by Pr 3+ contents, which, from 0.1 % to 5 %, tune the emission color from pink to warm white under excitation of 445 nm. It is of great interest that intense irradiation using a blue ∼445 laser does make efficient ultraviolet B (UVB) up-conversion at about 297, 306, and 313 nm, just stemming from the 6 P 3/2  →  8 S 7/2 , 6 P 5/2  →  8 S 7/2 , and 6 P 7/2  →  8 S 7/2 of Gd 3+ , respectively. A typical two-photon absorption process was proved to account for the UVB up-conversion, i.e. successive absorption of two blue ∼445 nm photons populating Pr 3+ 4f5d excited state, which further efficiently transfer its energy to the 6 I J / 6 P J states of Gd 3+ neighbor for UVB emission. Moreover, the thermally coupled levels (TCLs) of Gd 3+ : 6 P 3/2 - 6 P 7/2 , and Gd 3+ : 6 P 5/2 - 6 P 7/2 were thoroughly investigated for temperature sensing in 298–598 K. Maximum relative sensitivity of the 6 P 3/2 - 6 P 7/2 TCLs, i.e. S r (I 306 /I 313 ), is proved to be 0.96 % K −1 at 298 K, and that of the 6 P 5/2 - 6 P 7/2 TCLs, i.e. S r (I 297 /I 313 ), is 1 % K −1 at 373 K. The relevant thermal stabilities of the Ca 2 Al 2 SiO 7 :Pr 3+ /Gd 3+ thermometric candidate was well validated with 10 cycles of heating and cooling processes in 298–598 K. Such continuous explorations of Pr 3+ /Gd 3+ co-doping system will significantly promote the development of robust UVB up-converting phosphors as well as their practical applications in the field of high-resolution optical thermometry.

A MIF-p38-GSDMD inflammatory loop in keratinocytes underlies UVB-induced cutaneous lupus

Guo Chipeng,Luo Siweier,Luo Jigang,Lu Siyao,You Xiaomei,Cao Junlin,Du Yufei,Lv Haoran,Liang Hanzhi,Wang Le,Wang Liangchun,Liu Tao,Zhou Yiming

Ultraviolet B (UVB) is a well-recognized trigger of cutaneous lupus erythematosus (CLE), yet its molecular basis remains largely undefined. Here, using single-cell transcriptomics and a lupus-prone mouse model, we identify keratinocyte-derived macrophage migration inhibitory factor (MIF) as a key amplifier of cutaneous inflammation through a self-sustaining feedback loop. Single-cell RNA sequencing reveals elevated MIF expression specifically within pathogenic, interferon-high keratinocyte subclusters associated with CLE, which is further validated across major CLE subtypes in clinical skin samples. In vitro, UVB irradiation dose-dependently induces the release of MIF from keratinocytes, which in turn promotes inflammatory signaling and matrix remodeling in both keratinocytes and fibroblasts. Mechanistically, we demonstrate that UVB irradiation activates the ribotoxic stress response (RSR), leading to the p38-C/EBPβ-mediated transcriptional upregulation of NLRP3 and GSDMD cleavage in keratinocytes. The ensuing GSDMD-dependent pyroptosis facilitates the release of MIF, primarily through GSDMD pores rather than vesicular secretion, which in turn amplifies the p38-C/EBPβ signaling pathway. Therapeutic disruption of this loop either by gene silencing via AAVs or pharmacological inhibition via microneedles, markedly attenuates epidermal hyperplasia and cytokine imbalance in lupus-prone mice. These findings uncover a previously unrecognized MIF-p38-GSDMD inflammatory loop contributes to the UVB-induced cutaneous lupus, offering both mechanistic insights and translational opportunities for CLE.

Leucine-tryptophan dipeptide organo-bridged mesoporous silica for naringin encapsulation and strengthening UVB protection

Hengchang He,Chenqiu Dai,Ji Zhu,Kaiyin Shi,Yapeng Lu,Peng Wang,Jianqiang Wang

An organo-bridged mesoporous silica with the framework of biomolecule Leucine-Tryptophan dipeptide (Leu-Trp-OBMS) is first constructed through soft-templating sol-gel method. • The dipeptide framework facilitated high naringenin loading through synergistic π-π stacking and hydrogen bonding, conforming to pseudo-second-order kinetics and the Freundlich isotherm. • The optimized 15%-Leu-Trp-OBMS-NAR composite with high SPF values demonstrated excellent UVB shielding performance. • The composite exhibited remarkable retention stability, with less than 3.4% of the loaded naringenin being released in simulated sweat within a 24 h period. The utilization of natural plant polyphenols, such as naringin, as sunscreen agents is hindered by their intrinsic instability. In this research, we devised a novel ultraviolet protection system through the encapsulation of naringin with high capacity (about up to 94.55 mg/g) within a dipeptide organo-bridged mesoporous silica (OBMS), which achieved outstanding UVB protection. A leucine-tryptophan dipeptide-based bridged organosilane precursor (Leu-Trp-BOSP) was synthesized and co-condensed with tetraethyl orthosilicate (TEOS) (5%-20% molar percentage of the precursor) under alkaline conditions, employing cetyltrimethylammonium bromide (CTAB) as a template. The resultant materials (x-Leu-Trp-OBMS) were characterized by XRD, FTIR, SEM, TEM, TG analysis and nitrogen adsorption-desorption analysis, which verified a well-defined mesoporous structure with type IV isotherms. The adsorption of naringin adhered to the pseudo-second-order kinetic model and the Freundlich isotherm, suggesting a multilayer chemisorption process predominantly dominated by cooperative π-π stacking and hydrogen bonding interactions between the tryptophan indole rings, dipeptide amide groups and the aromatic flavonoid core. The optimal formulation (15%-Leu-Trp-OBMS-NAR) attained a sun protection factor (SPF) of 4.45. In simulated sweat release tests, less than 3.4% of the loaded naringin was released over 24 h, demonstrating outstanding retention stability. These results indicate that dipeptide OBMS can serve as an effective scaffold for stabilizing naringin, presenting a promising platform for long-lasting sunscreen formulations. ROS-Responsive Hydrogel Delivering Bio-Nanoselenium for Targeted Therapy of UVB-Induced Skin Photodamage

Kongdi Zhu,Jiamin Zhao,Tong Li,Shidi Zhang,Chuanming Liu,Zijian Xu,Cong Lv,Guijie Zhang,Yunlu Kang,Jiaqiang Huang

Ultraviolet B (UVB) radiation is a major environmental factor that induces skin photodamage through oxidative stress and inflammation. To address this, we developed a reactive oxygen species (ROS)-responsive hydrogel, SeNPs@HPTA, for the intelligent delivery of biogenic selenium nanoparticles (BioSeNPs) synthesized by Bifidobacterium animalis. The hydrogel was fabricated by cross-linking phenylboronic acid-grafted hyaluronic acid (HA-PBA) with tannic acid (TA), forming dynamic boronic ester bonds that cleave under high ROS conditions to enable on-demand BioSeNPs release. Transcriptomic analysis revealed that B. lactis H15 efficiently reduces selenite (Se(IV)) to elemental SeNPs by upregulating ion transport and NADPH-regenerating pathways. In vitro, SeNPs@HPTA enhanced HaCaT cell viability, scavenged ROS, and promoted cell migration post-UVB irradiation. In vivo, topical application alleviated skin lesions, improved hydration, reduced epidermal hyperplasia, and suppressed collagen degradation. Mechanistically, the hydrogel conferred therapeutic effects by enhancing antioxidant defenses, inhibiting inflammation and attenuating ferroptosis via GPx4/Nrf2 activation. Critically, genetic knockdown of GPx4 abolished its therapeutic effects, establishing a strict dependence on this key selenoenzyme. The formulation exhibited an excellent biosafety profile. This study presents a microbiome-inspired, stimulus-responsive strategy for precise skin photodamage treatment. We anticipate this microbiome-inspired, intelligent delivery system will offer a powerful and precise strategy for treating oxidative stress-related dermatoses.

Azelaic acid alleviates UVB-induced photoaging in keratinocytes by restoring Smad-dependent TGF-β signaling

Wenbing Lai,Lurun Wang,Wei Tang,Yuze Liu

Background Skin photoaging, primarily caused by chronic ultraviolet B (UVB) exposure, is characterized by cellular senescence and extracellular matrix (ECM) degradation. Suppression of transforming growth factor-β (TGF-β)/Smad signaling plays a critical role in UVB-induced collagen loss and impaired skin repair. Azelaic acid (AzA), a clinically established dermatological agent with antioxidant and anti-inflammatory properties, has been reported to maintain skin homeostasis; however, its mechanistic role in protecting against UVB-induced photoaging remains unclear. Objective This study aimed to investigate whether AzA alleviates UVB-induced photoaging in keratinocytes and to determine whether its protective effects are mediated through the restoration of Smad-dependent TGF-β signaling. Methods An in vitro UVB-induced photoaging model was established using HaCaT keratinocytes. Following UVB irradiation, cells were treated with AzA. Cell viability and cytotoxicity were evaluated using CCK-8 and LDH assays, respectively. Cellular senescence was assessed by senescence-associated β-galactosidase staining. ECM-related markers were measured by ELISA. TGF-β/Smad signaling activity was analyzed by Western blotting, quantitative real-time PCR, and immunofluorescence. The selective TGF-β/Smad inhibitor SB431542 was used to verify pathway dependence. A keratinocyte–fibroblast conditioned medium model was also employed to assess the paracrine regulation of fibroblast ECM metabolism. Results AzA significantly attenuated UVB-induced cellular senescence and partially restored cell viability in HaCaT cells. It significantly increased pro-collagen I levels and reduced MMP-1 levels, indicating the restoration of ECM-related protein balance. Mechanistically, AzA reversed UVB-induced suppression of TGF-β/Smad signaling by enhancing Smad2/3 phosphorylation, decreasing Smad7 expression, promoting the nuclear translocation of phosphorylated Smad2/3, and restoring Smad-dependent transcription. Pharmacological inhibition of TGF-β/Smad signaling largely abrogated these protective effects. Conditioned medium from AzA-treated keratinocytes also improved ECM-related responses in fibroblasts in a Smad-dependent manner. Conclusion AzA alleviates UVB-induced photoaging by restoring Smad-dependent TGF-β signaling, preserving ECM homeostasis, and modulating epidermal–dermal crosstalk in vitro. These findings provide mechanistic support for the potential application of AzA in anti-photoaging strategies.

Effect and Mechanism of Tricholoma matsutake Extract on UVA and UVB Radiation-Induced Skin Aging

Lu Hu,Zhenhai Huang,Jiyu Weng,Chujie Huang,Lanyue Zhang

Ultraviolet (UV) radiation often causes skin aging, inflammation, cancer and other related skin diseases. In this study, the main components of Tricholoma matsutake extract (TME) were identified using UPLC-Q-TOF-MS, and their anti-photoaging effects were assessed through UV-induced cell and animal models. The key components identified were D-mannitol (27.41%), DL-malic acid (14%), alginate (12.5%), isoleucine (4.82%), and phenylalanine (4.31%), all of which played roles in anti-aging and UV protection. TME (50-100 mg/ml) significantly alleviated UVA/UVB-induced erythema and wrinkles in mice. Pathological staining showed that TME suppressed UV-induced epidermal hyperplasia ( p < 0.05), reduced collagen damage ( p < 0.01), and decreased mast cell infiltration ( p < 0.01), while down-regulating inflammatory markers such as IL-6, IL-1β, and TNF-α. TME also upregulated type I collagen (COL-1). Flow cytometry results demonstrated that high-dose TME inhibited UV-induced apoptosis and reduced reactive oxygen species (ROS) in HaCaT cells ( p < 0.05). Immunofluorescence and scratch migration assays showed that TME promoted PPAR-α expression, reduced inflammation, and supported skin repair ( p < 0.01). Transcriptomic and metabolomic analyses indicated that TME regulated inflammation-related signaling pathways, helping to prevent skin aging. TME is a promising natural product for skin care and treatment of oxidative stress and inflammation-related diseases.

One-Dimensional Ferroelectric CsCu2Br3 Single-Crystal Enables UVB Polarization-Sensitive Photodetector for Encryption Wireless Communications

Kai Tang,Tong Xu,Chengzhou Zhang,Wenjie Li,Yuheng Chen,Caixia Kan,Daning Shi,Xiaosheng Fang,Mingming Jiang

Perovskite ferroelectrics with switchable spontaneous polarization have aroused widespread attention due to their potential applications in nonvolatile memory, optoelectronic detection, and so forth, yet they face challenges like insufficient thermal stability, structural brittleness, lead toxicity concerns, and other aspects. This paper presents the first well-defined synthesis of one-dimensional (1D) lead-free ferroelectric CsCu 2 Br 3 single-crystals (SCs), which possess a wide bandgap (≈4.1 eV), high Curie temperature, in-plane ferroelectric self-polarization, prominent optical anisotropy, and other notable properties. These characteristics enable the samples to sensitively detect weak ultraviolet-B (UVB) signals and precisely discriminate linear polarization states. A prototype polarization-sensitive UVB photodetector based on 1D CsCu 2 Br 3 SCs is successfully fabricated, demonstrating exceptional photoresponse performance with a photoresponsivity of 400 mA W − 1 , a specific detectivity of 1.12 × 10 12 Jones, a rapid response characteristic with rise/fall times of 1.2/1.6 ms, and a high polarization ratio of 4.3 under 320 nm linearly polarized light illumination. By leveraging a dual-parameter optoelectronic signal-polarization encoding technology, the detector realizes encryption communication, with the channel capacity being augmented by 1.45 times. This work establishes CsCu 2 Br 3 SCs as eco-benign perovskite platforms for probing ferroelectricity and multi-physical coupling phenomena, demonstrating their critical role in advancing cross-disciplinary functional material research.
