Carbohydrate antigen 19-9 (CA19-9) is a key biomarker for early diagnosis of various malignancies, including lung cancer. Herein, a ZnPc/Ti 3 C 2 heterojunction photosensitive material was synthesized via a novel one-pot strategy and utilized for the first time to construct a photoelectrochemical sandwich immunosensor. Taking advantage of the reducing property of Ti 3 C 2 , gold nanoparticles (AuNPs) were in-situ generated on ZnPc/Ti 3 C 2 to form ZnPc/Ti 3 C 2 /AuNPs nanocomposite, which was then immobilized onto a fluorine-doped tin oxide (FTO) electrode. The AuNPs provided a large surface area and biocompatible interface for efficient loading of capture antibody (Ab1). Upon target CA19-9 recognition, the polydopamine-labeled detection antibody (PDA-Ab2) was introduced, triggering a dual photocurrent quenching mechanism through which PDA acted as an electron trap, capturing photoelectrons from ZnPc/Ti 3 C 2 /AuNPs, while the bulky immunocomplex competed with the electrode for light absorption, reducing irradiation of the photosensitive material. This synergistic quenching enabled highly sensitive CA19-9 detection with a linear range of 0.001–200 U/mL and a detection limit of 0.53 mU/mL. The proposed sensor was successfully applied to CA19-9 determination in serum samples with a relative error within 10% compared with hospital reference values, demonstrating its potential for clinical diagnostics.

Background How to ingeniously design multi-effect photosensitizers (PSs), including multimodal imaging and multi-channel therapy, is of great significance for highly spatiotemporal controllable precise phototherapy of malignant tumors. Methods Herein, a novel multifunctional zinc(II) phthalocyanine-based planar micromolecule amphiphile (ZnPc 1) was successfully designed and synthesized, in which N atom with photoinduced electron transfer effect was introduced to enhance the near-infrared absorbance and nonradiative heat generation. After simple self-assembling into nanoparticles (NPs), ZnPc 1 NPs would exhibit enhanced multimodal imaging properties including fluorescence (FL) imaging (FLI) /photoacoustic (PA) imaging (PAI) /infrared (IR) thermal imaging, which was further used to guide the combined photodynamic therapy (PDT) and photothermal therapy (PTT).Results It was that under the self-guidance of the multimodal imaging, ZnPc 1 NPs could precisely pinpoint the tumor from the vertical and horizontal boundaries achieving highly efficient and accurate treatment of cancer. Conclusion Accordingly, the integration of FL/PA/IR multimodal imaging and PDT/PTT synergistic therapy pathway into one ZnPc 1 could provide a blueprint for the next generation of phototherapy, which offered a new paradigm for the integration of diagnosis and treatment in tumor and a promising prospect for precise cancer therapy.

The impact of encapsulating gemcitabine (GEM) into nanoparticles on its delivery remains underexplored, with the potential benefits of targeted drug delivery and stimuli-responsive release yet to be fully clarified. Herein, we designed a near-infrared (NIR) light-responsive polymeric nanoparticle, ZnPc@P(PEG-CMA-TKGEM), which integrates reactive oxygen species (ROS) generation and cell imaging capabilities. The self-assembled polymeric micelles exhibit a hydrodynamic size of ~134 nm. Under NIR irradiation, the cumulative drug release rate reaches 51% within 48 h, which is three times higher than that of the non-irradiated control group. In cytotoxicity assays, the cell viability of the NIR-irradiated drug-loaded group is approximately 17%, while that of the NIR-irradiated blank group (without drug loading) remains above 80%. These results confirm that the nanocarriers successfully deliver GEM to target cells and achieve controlled drug release via NIR stimulation.

Because of the good fluorescence of zinc phthalocyanine-phenolic resin (ZnPc-PFR) photosensitizer and large specific surface area of mesoporous silica nanoparticles (MSNs), a highly efficient nano-drug carrier system, denoted as ZnPc-PFR@MSN, was constructed for photothermal therapy (PTT) and pH-sensitive drug delivery. The facile hydrothermal reaction was used to synthesize ZnPc-PFR nanoparticles in one-step. After loading the as-synthesized ZnPc-PFR nanoparticles into MSNs, and a good high drug-loading rate (143.7 mg g −1 ) to the anticancer drug of Adriamycin (DOX) could be obtained. Thus, a novel nanosphere with the merits of good fluorescence, high drug-loading rate (143.7 mg g −1 ), better sustained-release properties, and photothermal properties (reached 43.23°C within 260 s) was prepared. The as-synthesized multifunctional composites make it a good candidate in fluorescence imaging, PTT, and drug delivery.

Aim: To develop precise targeting and versatile Fe3O4@SiO2-P123/PTX-ZnPc nanoparticles (FSP-PTX-ZnPc NPs) to reverse paclitaxel (PTX)-induced multidrug resistance in breast cancer. Materials & methods: PTX and zinc (II) phthalocyanine (ZnPc) co-loaded FSP-PTX-ZnPc NPs were designed. The resulting multifunctional NPs were evaluated systematically in vitro and in vivo, and the mechanism of drug-resistance reversal was investigated. Results: The NPs enhanced drug uptake in MCF-7/PDR cells by increasing drug solubility and impairing P-glycoprotein efflux. Additionally, magnetic targeting and enhanced permeation and retention (EPR) effect enhanced drug accumulation in tumor, facilitating the chemotherapeutic and photodynamic therapy effects. Moreover, FSP-PTX-ZnPc NPs could penetrate the blood–brain barrier, a desirable trait for brain disease therapy. Conclusion: The multifunctional FSP-PTX-ZnPc NPs are an effective tool for overcoming drug resistance in breast cancer.

Clinical pancreatic ductal adenocarcinoma (PDAC) treatment is severely limited by lack of effective KRAS suppression strategies. To address this dilemma, a reactive oxygen species (ROS)-responsive and PDAC-targeted nanodrug named Z/B-PLS was constructed to confront KRAS through dual-blockade of its downstream PI3K/AKT/mTOR and RAF/MEK/ERK for enhanced PDAC treatment. Specifically, photosensitizer zinc phthalocyanine (ZnPc) and PI3K/mTOR inhibitor BEZ235 (BEZ) were co-loaded into PLS which was constructed by click chemistry conjugating MEK inhibitor selumetinib (SEL) to low molecular weight heparin with ROS-responsive oxalate bond. The BEZ and SEL blocked PI3K/AKT/mTOR and RAF/MEK/ERK respectively to remodel glycolysis and non-canonical glutamine metabolism. ZnPc mediated photodynamic therapy (PDT) could enhance drug release through ROS generation, further facilitating KRAS downstream dual-blockade to create treatment-promoting drug delivery-therapeutic positive feedback. Benefiting from this broad metabolic modulation cascade, the metabolic symbiosis between normoxic and hypoxic tumor cells was also cut off simultaneously and effective tumor vascular normalization effects could be achieved. As a result, PDT was dramatically promoted through glycolysis-non-canonical glutamine dual-metabolism regulation, achieving complete elimination of tumors in vivo . Above all, this study achieved effective multidimensional metabolic modulation based on integrated smart nanodrug delivery, helping overcome the therapeutic challenges posed by KRAS mutations of PDAC.

Both chemodynamic therapy and photodynamic therapy, based on the production of reactive oxygen (ROS), have excellent potential in cancer therapy. However, the abnormal redox homeostasis in tumor cells, especially the overexpressed glutathione (GSH) could scavenge ROS and reduce the anti-tumor efficiency. Therefore, it is essential to develop a simple and effective tumor-specific drug delivery system for modulating the tumor microenvironment (TME) and achieving synergistic therapy at the tumor site. In this study, self-assembled nanoparticles (named CDZP NPs) were developed by copper ion (Cu2+), Doxorubicin (Dox), Zinc phthalocyanine (ZnPc) and a trace amount of poly (2-(di-methylamino) ethylmethacrylate)-poly [(R)-3-hydroxybutyrate]-poly (2-(dimethylamino) ethylmethacrylate (PDMAEMA-PHB-PDMAEMA) through chelation, π-π stacking and hydrophobic interaction. This triple factors-responsive (pH, laser and GSH) nanoparticles demonstrated unique advantages through the synergistic effect. Highly controllable drug release ensured its effectiveness at the tumor site, Dox-induced chemotherapy and ZnPc-mediated fluorescence (FL) imaging exhibited the distribution of nanoparticles. Meanwhile, Cu2+-mediated GSH-consumption not only reduced the intracellular ROS elimination but also produced Cu+ to catalyze hydrogen peroxide (H2O2) and generated hydroxyl radical (∙OH), thereby enhancing chemodynamic and photodynamic therapy. Herein, this study provided a green and relatively simple method for preparing multifunctional nanoparticles that can effectively modulate the TME and improve synergetic cancer therapy.
