Low Strength Wastewater Treatment Using a Combined Biological Aerated Filter/Anammox Process

Wanying Xie,Ji Li,Tao Song,Yong Li,Zhenlin Wang,Xiaolei Zhang

To achieve the in situ capacity expansion of the post-denitrification biological aerated filter (BAF-DN), the integration of BAF with the anammox process (BAF/AX) was proposed. With the objective of maximizing retaining ammonia nitrogen, the operational optimization of BAF was achieved by two distinct strategies. The treatment performance of BAF demonstrated that the removal efficiencies of chemical oxygen demand (COD) and ammonia nitrogen (NH4+-N) was 66.3~67.3% and 4~12%, respectively, under conditions of low aeration intensity (0.4 m3·m−2·h−1) or a shortened empty bed residence time (EBRT) of 30 min. ResidualNH4+-Nin the BAF effluent served as the ammonia substrate for the subsequent anammox process, which was successfully launched by using ceramic particles and sponges as carriers. Notably, the sponge carrier facilitated a shorter start-up period of 41 to 44 days. Furthermore, the sponge-based anammox reactor exhibited a superiorNH4+-Nremoval capacity (≥85.7%), under operations of a shorter EBRT of 40 min, low influentNH4+-Nconcentrations (≤30 mg/L), and COD levels of ≤67 mg/L. In addition, a comprehensive evaluation of the BAF/AX process was conducted, which considered performance, cost-effectiveness, and engineering feasibility. The performance results illustrated that the effluent quality met the standard well (with a COD level of ≤ 50 mg/L, and a TN of ≤3.1~10.5 mg/L). Following a comparison against the low aeration intensity operation, it was recommended to operate BAF at a low EBRT within the BAF/AX process. Consequently, the treated volume was double the volume of the standalone BAF-DN, synchronously achieving low costs (0.413 yuan/m3).

Quantification of enhanced nitrogen removal pathways of pyrite interaction with anammox sludge system

Fan Feng,Caiyan Qu,Jia Tang,Xing Wu,Xi Tang,Fubing Yao,Liyuan Chai,Ruiyang Xiao,Chong-Jian Tang

Fe and S could improve the anammox performance by coupling with the N cycle. However, the coexistence of Fe and S complicates the metabolic process, troubling the quantification of nitrogen removal pathways. Herein, the nitrogen removal pathways were quantificationally analyzed by 15 N stable isotope tracing and elaborate batch tests, and the enhancing mechanism was illuminated by physiological and metatranscriptome analyses in pyrite (FeS 2 )-amended anammox system. We found that anammox, denitrification and Feammox (NH 4 + oxidation coupled with Fe(III) reduction) contributed to 80.22 %, 19.53 % and 0.25 % nitrogen removal in this process. With pyrite addition, Fe, S release and the extracellular polymeric substances secretion were responsible for the enhancement of nitrite and nitrate removal via chemical, autotrophic and heterotrophic denitrification, improving nitrogen removal by 14 % and 19 %. The network analyses further confirmed that the multiple nitrogen removal pathways (i.e., anammox, Feammox and denitrification) contribute to pyrite-induced nitrogen removal enhancement in the anammox system. This work enables the in-depth understanding, future design and optimization of the pyrite-amended anammox system, providing a fundamental basis for developing Fe- and S-based anammox enhancement technologies .

Free nitrous acid prediction in ANAMMOX process using hybrid deep neural network model

Junlang Li,Jilan Dong,Zhenguo Chen,Xiaoyong Li,Xiaohui Yi,Guoqiang Niu,Jiaan He,Simin Lu,Yuxiang Ke,Mingzhi Huang

Free nitrous acid (FNA) is a critical metric for stabilization of ANAMMOX but can not be directly and immediately measured by sensors or chemical measurement method, which hinders the effective management and operation for ANAMMOX. This study focuses on FNA prediction using hybrid model based on temporal convolutional network (TCN) combined with attention mechanism (AM) optimized by multiobjective tree-structured parzen estimator (MOTPE), called MOTPE-TCNA. A case study in an ANAMMOX reactor is carried out. Results show that nitrogen removal rate (NRR) is highly correlated with FNA concentration, indicating that it can forecast the operational status by predicting FNA. Then, MOTPE successfully optimizes the hyperparameters of TCN, helping TCN achieve a high prediction accuracy, and AM furtherly improves model accuracy. MOTPE-TCNA obtains the highest prediction accuracy, whose R 2 value gets 0.992, increasing 1.71–11.80% compared to other models. As a deep neural network model, MOTPE-TCNA has more advantages than traditional machine learning methods in FNA prediction, which is beneficial to maintain the stable operation and easy control for ANAMMOX process.

Anammox granules show strong resistance to engineered silver nanoparticles during long-term exposure

Zheng-Zhe Zhang,Ya-Fei Cheng,Lian-Zeng-Ji Xu,Yu-Hui Bai,Ren-Cun Jin

The increasing use of engineered Ag nanoparticles (NPs) in consumer products raises great concerns about their environmental impacts, but their potential impact on anaerobic ammonium oxidation (anammox) bacteria in wastewater treatment remains unclear. In this study, the response of anammox consortia inhabiting in granules to long-term exposure of AgNPs was investigated. The gradually increased stress of AgNPs (1, 10, 50 mg L −1 ) showed no adverse effects on the nitrogen removal performance and on the relative abundance of Ca. Kuenenia inhabiting in granules. Notably, in response to the escalating burden of AgNPs, anammox consortia tended to secrete more protein but less polysaccharide, and meanwhile increase the relative abundances of Ag + efflux protein SilA and regulatory protein SilR. Overall, these results suggested that the strong resistance of anammox granules to AgNPs might be mainly attributed to the defense role of extracellular polymeric substances and gene-based regulation of ion transport.

Linear anionic surfactant (SDBS) destabilized anammox process through sludge disaggregation and metabolic inhibition

Zheng-Zhe Zhang,Yu Zhang,Ya-Fei Cheng,Ren-Cun Jin

The increase of emerging contaminants, such as surfactants, is one of the major challenges to biological wastewater treatment. However, the potential impact of linear alkylbenzene sulphonates (LAS), a major class of anionic surfactants, on anammox process is unclear. The long-term effects of sodium dodecyl benzene sulfonate (SDBS, as a model LAS) on reactor performance, microbial community and sludge properties were investigated in this study. The presence of 5 mg L −1 SDBS promoted the release of extracellular microbial products from anammox granules and the wash-out of anammox population via effluent. Despite sludge disaggregation, the reactor performance was robust to the exposure of 5 mg L −1 SDBS due to functional redundancy. With the further increase of SDBS to 10 mg L −1 , the metabolic activity of anammox biomass and the transcription and post-translation of hydrazine dehydrogenase were significantly decreased. The potential mechanism might be associated with the damage on cell membrane that induced the leakage of intracellular matrix. These results highlight the need to consider the potential risk of LAS to operation of anammox process in biological wastewater treatment plant.

Alleviating the nitrite stress on anaerobic ammonium oxidation by pyrolytic biochar

Jiajia Xu,Chao Li,Nanwen Zhu,Yanwen Shen,Haiping Yuan

The nitrite (NO 2 − ) inhibition in anaerobic ammonium oxidation (anammox) process is widely reported. Here, the effects of three pyrolytic biochars (CS300, CS550 and CS800) were investigated to alleviate NO 2 − stress on anammox process under exposure of varied NO 2 − -N concentrations (70, 200, 400 and 600 mg L −1 ). No nitrite inhibition was observed at 70 mg N L −1 . However, the total nitrogen removal efficiency (TNREs) decreased with NO 2 − -N concentration increased, while the biochar-amended groups achieved higher TNREs than the control (CK). At 200 mg N L −1 , the TNREs were 60.2%, 99.0%, 98.5% and 86.6% for CK, CS300, CS550 and CS800, respectively. At 400 mg N L −1 , the TNREs were 23.3%, 56.0%, 37.1% and 29.7% for CK, CS300, CS550 and CS800, respectively. At 600 mg N L −1 in which severe inhibition was observed, the TNREs were increased by 231% ( p  = 0.002), 149% ( p  = 0.014), and 51.0% ( p  = 0.166) for CS300, CS550 and CS800, respectively, as compared to CK, with the corresponding specific anammox activity increased by 3.1-, 2,0- and 1.1-folds, respectively. CS300 enriched the relative abundance of Candidatus Kuenenia and increased the gene copies of functional genes ( hzs A, hdh , nir S and nir K). Besides, CS300 effectively alleviated the suppression of three membrane-associated enzyme complexes for anammox electron transport chain, indicating the possible contribution of redox-active moieties of CS300 to energy conversion metabolism for mitigating the NO 2 − -N inhibition. This study provided an effective strategy for alleviating NO 2 − -N stress by applying an environmentally compatible material (biochar) on anammox process.

Reactivation and Nitrogen Removal Performance of Idle Anammox Sludge Enhanced by Rape Straw Biochar

Qiang Chen,Yi Ding,Zhicheng Xu,Haibin Zhou,Ruoyu Zhang,Jiao Chen,Yixin Lu,Wenlai Xu

Low reactivation efficiency of idle anaerobic ammonia oxidation (anammox) sludge hinders its reapplication. To address this issue, rape straw biochar (RSB) was added in the reactivation process of idle anammox sludge, and its effects on the nitrogen transformation and sludge characteristics were investigated, and the mechanism of RSB to enhance the reactivation performance was explored. Results indicated that adding 5 g/L RSB for 35 days successfully reactivated anammox sludge that had been idle for 270 days. The reactivation time was reduced by 34% compared to the control without RSB. During the stable operation period, the average TN removal efficiency reached 90.6%, and the sludge exhibited higher activity. After completion of reactivation, the specific surface area, total pore volume, and average pore diameter of RSB decreased by 59.4%, 66.9%, and 55.2%, respectively, compared with that before reactivation, and the carbon–oxygen functional groups also changed. RSB not only provided a habitat for the enriched growth of nitrogen transforming functional flora but also possessed the potential to supply sufficient electron donors and acceptors for the nitrogen transforming process, which promoted the synergistic removal of nitrate by denitrification, resulting in an effective enhancement of reactivation efficiency and nitrogen removal performance. The addition of RSB provides a novel strategy to enhance the reactivation efficiency of idle anammox sludge, which is of positive significance in promoting its efficient reuse and stable operation.

Regulation mechanism and resistant strain based bioaugmentation of anammox consortia in response to triclocarban

Jia-Hui Yang,Wei-Lin Guo,Zhe-Qi Lian,Qi Su,Xin-Yi Zhao,Wen-Li Guo,Nian-Si Fan,Ren-Cun Jin

Triclocarban was emerging pollutants of increasing concern in wastewater treatment process, posing a high risk to ecological and human health. Anaerobic ammonium oxidation (anammox) process was a potential emerging and efficient biological wastewater treatment technology, which converted NH 4 + and NO 2 − into N 2 under anaerobic condition. After the long-term monitoring and analysis of anammox consortia responses to different concentrations of triclocarban, the bioaugmentation strategy based on resistant bacteria inoculation was proposed in this study. Nitrogen removal performance and anammox sludge activity were barely affected by 50 μg L −1 triclocarban, but significantly inhibited by 200 μg L −1 triclocarban. Meanwhile, the extracellular polymeric substances and reactive oxygen species production significantly increased, and cell damage of anammox consortia were also observed. Batch assays revealed the obvious adsorption of triclocarban onto anammox sludge, while molecular docking simulation illustrated the potential binding area of triclocarban and protein molecules. The density functional theory calculations indicated that triclocarban was probably degraded via hydrolysis or dechlorination. The richness and diversity of the anammox consortia decreased under triclocarban exposure. In particular, the relative abundance of Candidatus Kuenenia gradually decreased from 13.1 % to 3.9 %, showing a concentration-dependent manner. The abundance of nitrogen removal-related functional genes also exhibited a downward trend. In addition, three triclocarban-resistant strains were isolated from various aquatic environments, identified as Stenotrophomonas acidaminiphila , Achromobacter mucicolens and Pseudomonas mosselii . They could maintain the high ATP level, NADH content and proportion of viable bacteria under triclocarban stress. Their triclocarban removal efficiency reached 87.5 % in batch experiments. This resistant bacteria-based bioaugmentation is expected to achieve the quick startup and stabilization of anammox system to the stress of non-antibiotic antimicrobial agents, accelerating the widespread application of anammox process in such wastewater treatment.

An overlooked nanofluids effect from Fe3O4 nanoparticles enhances mass transfer in anammox granular sludge

Dongdong Xu,Aqiang Ding,Yang Yu,Ping Zheng,Meng Zhang,Zhetai Hu

Magnetite (Fe 3 O 4 ) particles have been widely reported to enhance the anammox's activity in anammox granular sludge (AnGS), yet the underlying mechanisms remain unclear. This study demonstrates that both Fe 3 O 4 microparticles (MPs) and nanoparticles (NPs) at a dosage of 200 mg Fe 3 O 4 /L significantly increased the specific anammox activity (SAA) of AnGS. Additionally, the transcriptional activities of the hzs and hdh genes involved in the anammox process, as well as the heme c content in AnGS, were also notably enhanced. Notably, Fe 3 O 4 NPs were more effective than MPs in boosting anammox activity within AnGS. Mechanistically, Fe 3 O 4 MPs released free iron, which anammox bacteria utilized to promote the synthesis of key enzymes, thereby enhancing their activity. Compared to MPs, Fe 3 O 4 NPs not only elevated the synthesis of these key enzymes to a higher level but also induced a nanofluids effect on the surface of AnGS, improving substrate permeability and accessibility to intragranular anammox bacteria. Moreover, the nanofluids effect was identified as the primary mechanism through which Fe 3 O 4 NPs enhanced anammox activity within AnGS. These findings provide new insights into the effects of nanoparticles on granular sludge systems, extending beyond AnGS.

Simultaneously ammonium and perchlorate remove via the partial nitrification-anammox coupled sulfur autotrophic system

Guiying Zhang,Jingmei Liu,Yi Han,Liang Xia,Jianbing Zhang,Jianbo Guo,Haibo Li,Yanan Hou,Yuanyuan Song

In this study, partial nitrification-anammox coupled sulfur autotrophic (PNA-SA) system was constructed for removing ammonium and perchlorate from composite wastewater simultaneously. The removal rates of ammonium and perchlorate could reach 94.2% and 93.1%, respectively, at an ammonium concentration of 60 mg N∙L −1 and a perchlorate concentration of 30 mg N∙L −1 . Ammonium was mainly removed by anammox bacteria (AnAOB) in PNA reactor, while perchlorate removal was attributed to perchlorate reducing bacteria (PRB) in SA reactor. Furthermore, combined with Pearson analysis, N-acylhomoserine lactones (AHLs)-mediated quorum sensing regulated the production of TB-EPS, improving biofilm stability and thus ensuring the removal performance of reactors. 16sRNA gene sequencing results indicated that the key functional bacteria in the PNA reactor were Comamonas and Candidatus Kuenenia , and the key functional bacteria in the SA reactor were Sulfurimonas , Thiobacillus , and Defluviimonas . These key functional bacteria ensured PNA-SA system feasibility and stability. Such results indicated that PNA-SA system could be used to treat ammonium and perchlorate composite wastewater, thereby providing a new strategy for removing such composite wastewater.

Formate-facilitated recovery of wastewater anammox granular sludge from oxygen inhibition: Effects and mechanisms

Min Xu,Rui Xiao,Fengqian Xu,Wenbo Chai,Wenlong Liu,Huijie Lu

Oxygen leakage due to poor reactor sealing or accidental aeration can have detrimental impacts on the performance of wastewater anammox processes. Strategies of restoring functionality of anammox consortia are scarce, and providing appropriate chemical reducing reagents may help reduce oxidative damage. This study characterized the responses of anammox granular sludge (AnGS) to oxygen inhibition, and identified formate as an effective promoter capable of facilitating activity recovery, while other organics (acetate, glucose, methanol) were less effective. Upon 12 h of air exposure (DO: 1.2 ± 0.1 mg/L), the total nitrogen removal efficiency of a lab-scale AnGS reactor decreased from 85.15 % to 45.46 %. Compared with natural recovery (without chemical addition), potassium formate at 10 mg COD/L for 3 days significantly increased the total nitrogen removal efficiency, both in the short-term (66.68 % vs 57.09 %, 3 days post inhibition) and long-term (82.34 % vs 72.88 %, 30 days post inhibition). By examining sludge morphology, metagenome and transcriptome, the following potential mechanisms were proposed: 1) rapidly decreasing intracellular ROS; 2) enhancing nitrite turnover; 3) maintaining granular sludge integrity; 4) facilitating interspecies metabolic interactions. The ability of formate to facilitate sludge recovery from oxygen inhibition also implies its potential in mitigating other oxidative stresses in wastewater anammox reactors.

Molecular interference and ecological restructuring: The acute strikes of nitroaromatics on anammox granular sludge

Yan Xia,Shengwen Huang,Wentao Li,Ren-Cun Jin

Nitroaromatic compounds, widely used in the pharmaceutical, dye, and pesticide industries, often coexist with high-strength ammonium in industrial effluents, posing a dual stress on biological nitrogen removal systems. Anaerobic ammonium oxidation (anammox) represents a promising technology due to its high efficiency, low energy consumption, and minimal sludge yield. However, the acute response mechanism of anammox granular sludge (AnGS) to nitroaromatic compounds remains poorly understood. This study systematically investigated the acute effects of nitrobenzene (NB) and its reduction product aniline (AN) on AnGS by integrating nitrogen removal performance, extracellular polymeric substances (EPS) characterization, microbial community evolution, and functional gene prediction. Results demonstrated a concentration-dependent inhibition of anammox activity, with specific anammox activity (SAA) decreasing by 57.91 ± 2.44 % and 25.06 ± 1.38 % at 10 mg L −1 of NB and AN, respectively, and nearly complete suppression at 50 mg L −1 . NB exhibited stronger toxicity than AN, consistent with greater shifts in EPS composition, surface hydrophilicity, and microbial community structure. Molecular docking revealed that NB exhibited a stronger disruptive potential due to its superior affinity and specific interactions with active-site residues. High-throughput sequencing revealed severe inhibition of anammox bacteria like Candidatus_Brocadia , while fermentative genera like Acetoanaerobium were significantly enriched, indicating a functional trade-off under stress. Functional gene analysis further confirmed the downregulation of key anammox genes ( hzs , hdh ). These findings highlight the compound-specific toxicity mechanisms and the resilience of microbial communities through metabolic plasticity, providing new insights into the ecological risks of nitroaromatics in anammox systems.
