Surface biofunctional bFGF-loaded electrospun suture accelerates incisional wound healing

Yi Li,Hangqi Luo,Yue Li,Peipei Huang,Jie Xu,Jian Zhang,Peihan Cai,Huacheng He,Jiang Wu,Xiaokun Li

Surgical suture plays an important role in increasing wound repair. However, the healing efficacy of sutures currently is still not satisfactory due to their failure in regulating healing biology. Therefore, developing functional sutures are highly required. Herein, we fabricated a new degradable suture to control the release of basic fibroblast growth factor (bFGF) for incisional wound healing. The suture ( [email protected] ) was simply made by anchoring bFGF onto the surface of the electrospun polycaprolactone (PCL) suture through the assistance of dopamine with a bFGF loading efficiency around 80 %. The introduction of dopamine to the suture also increased the mechanical strength of suture from 17 kPa to 27 kPa. The in vitro data evidenced that the suture could effectively prolong bFGF release up to 10 days and enhance cell adhesion and proliferation. Further in vivo results demonstrated that the suture could increase wound healing and induce a complete wound closure within 13 days. Moreover, the suture was found to accelerate skin mechanical restoration with a recovery of maximum strength to 87.1 % of the healthy skin. The suture was also revealed to help the skin wound healing by promoting granulation tissue formation, collagen deposition, reepithelization, and basement membrane reconstruction.

Tough and self-adhesive zwitterionic hydrogels with mechano-responsive release of bFGF for tympanic membrane repair

Shengjia Chen,Xiangshu Guo,Yanyu Yang,Junjie Deng,Ting Xu,Zhechen Yuan,Hao Xue,Longxing Niu,Rong Wang,Yi Shen

The tympanic membrane (TM) is constantly in a state of vibrating. However, there is currently a lack of drug-delivery scaffolds suitable for the dynamic environment of TM perforation. In this study, a mechano-responsive tough hydrogel was developed. It consists of basic fibroblast growth factor (bFGF)-loaded sodium alginate (SA) microspheres, polysulfobetaine methacrylate (polySBMA), and gelatin methacrylate (GelMA). This hydrogel was designed to serve as a TM scaffold to promote perforation healing under dynamic conditions. bFGF was encapsulated in SA microspheres, which were then incorporated into polySBMA-GelMA hydrogels through photo-initiated free radical polymerization. The mechanical properties, tissue adhesiveness, swelling properties, and degradation of the hydrogels were evaluated before and after microsphere incorporation. It was observed that incorporating bFGF-loaded SA microspheres did not significantly impact the adhesion and degradation mechanisms of the hydrogel. The compressive strength and tensile strength of the microsphere-incorporated hydrogel were up to 6.6 MPa and 64.1 kPa, respectively, suitable for a TM scaffold. The release behavior of bFGF from the hydrogel could be controlled by vibration stimulation without significantly affecting the hydrogel's mechanical properties, indicating a mechano-responsive nature of the hydrogel. The in vitro cytotoxicity assay demonstrated that the hydrogels showed no cytotoxic effects. Moreover, cell culture assays demonstrated that vibration stimulation could enhance the release of bFGF, significantly promoting cell proliferation and migration. The results demonstrate the significant potential of the mechano-responsive hydrogel as a scaffold for repairing TM perforations.

Electrospun collagen/hyaluronic acid dressing co-loaded with EGF and bFGF for enhanced diabetic wound healing

Liang Wu,Wei Wang,Linxinyi Chen,Duohui Li,Yijun Ke,Yanghui Xu,Bo Zhu,Fangqi Hu,Lishang Dai,Xiwu Shi

Chronic non-healing wounds are a major complication of diabetes and require advanced therapeutic strategies. Although epidermal growth factor (EGF) and basic fibroblast growth factor (bFGF) are known to promote tissue regeneration, their poor stability and short half-life have severely restricted clinical translation. To overcome these challenges, a collagen/hyaluronic acid dressing loaded with EGF and bFGF (CH-EGF/bFGF) was prepared via electrospinning. CH-EGF/bFGF exhibited a fibrous network-like entanglement and possessed good water retention and mechanical properties. In vitro experiments, it was observed that CH-EGF/bFGF not only had good biosafety but also promoted the adhesion, proliferation, and migration of HaCaT cells through activation of the ERK1/2 and p38 MAPK signaling pathways. Furthermore, CH-EGF/bFGF effectively accelerated the wound-healing process in db/db diabetic mice, and markedly enhanced the expression of vascular endothelial growth factor, Ki67, CD31, alpha-smooth muscle actin, collagen I, and collagen III at the wound site. This study achieves dual growth factor stabilization and controlled release through an electrospun biomimetic matrix that simultaneously mimics the extracellular matrix and maintains a moist wound-healing milieu. In conclusion, CH-EGF/bFGF demonstrated promising potential for promoting diabetic wound healing, offering a novel and viable therapeutic approach for the clinical management of diabetic wounds.

Bioactive ECM Mimic Hyaluronic Acid Dressing via Sustained Releasing of bFGF for Enhancing Skin Wound Healing

Anqi Chen,Wen Huang,Liang Wu,Ying An,Tengxiao Xuan,Huacheng He,Mengqi Ye,Lamei Qi,Jiang Wu

Successful dermal wound regeneration requires the coordination of repair cells and cellular signals with the extracellular matrix (ECM), which serves as an indispensable mechanical and biological supporter for cell functions and communications with varied cytokines during healing processes. Here, we developed an injectable bioactive wound dressing, methacrylated hyaluronic acid (Me-HA)-based hydrogel loading with basic fibroblast growth factor (bFGF), endowing the dressing with the pleiotropic bioactivity to mimic natural ECM. This bFGF@Me-HA dressing was applied to a mouse with full-thickness excisional wounds to investigate its positive roles in wound repair owing to the complementary functions of HA with sustained release of bioactive bFGF. Compared with the single Me-HA and bFGF group, bFGF@Me-HA hydrogel dressings significantly enhanced wound healing with accelerated re-epithelialization, granulation formation, collagen, deposition and skin appendage regeneration. Further investigations showed significantly promoted cell proliferation and vascularization in the bFGF@Me-HA group, which was mediated by the upregulation of transforming growth factor-β (TGF-β) and vascular endothelial growth factor (VEGF) expressions. In conclusion, this bFGF@Me-HA hydrogel realized the optimization of simple ECM mimic dressing via introducing the bioactive effector, bFGF, and has the potential to be widely used as an effective bioactive ECM-based wound dressing in future wound care.

Core-shell chitosan microsphere with antimicrobial and vascularized functions for promoting skin wound healing

Hao Li,Feng Wang

It is imperative to develop a wound dressing that can effectively treat bacterial infection in the wound location and speed up wound healing. In this study, chitosan (CS) microspheres with a core–shell structure were prepared using high-voltage electrostatic drop technology. The antimicrobial polypeptide (PonG1) and bFGF, were encapsulated in the core of the CS microsphere. The preparation of CS microspheres was studied by changing the parameters. The surface morphology of CS microspheres and the release quantities of PonG1 and bFGF were observed and detected. In vivo experiments were conducted to study the neovascularization and anti-inflammatory abilities of bFGF and PonG1. The repair ability of CS microspheres was evaluated using a rat model of full-thickness skin wound. Both bFGF and PonG1 exhibited slow release from the CS microspheres within 14 days. In vivo experiments showed that CS microspheres containing PonG1 (3 mg/ml) and bFGF (300 μg/ml) had the best neovascularization and anti-inflammatory effects. CS microspheres containing both bFGF and PonG1 had the strongest ability to repair skin wound. CS microspheres with antimicrobial and vascularized functions can be used as dressings for the repair of hard-to-heal wounds.

A density gradient of basic fibroblast growth factor guides directional migration of vascular smooth muscle cells

Jindan Wu,Zhengwei Mao,Lulu Han,Yizhi Zhao,Jiabin Xi,Changyou Gao

The migration of vascular smooth muscle cells (VSMCs) is an important process in many physiological events. It is of paramount importance to control the migration rate and direction of VSMCs by biomaterials. In this paper, a density gradient of basic fibroblast growth factor (bFGF) was fabricated using an injection method and the bio-conjugation between heparin and bFGF. The density of bFGF gradually increased with a slope of 17   ng/cm 2 /mm. Adhesion and migration of VSMCs were studied on the bFGF gradient. The VSMCs exhibited preferential orientation and an enhanced directional migration behavior on the gradient surface. Up to 70% cells migrated towards the region with a higher density of bFGF on the gradient. However, the bFGF gradient had no effect on the cell migration rate.

Collagen-Inspired 3D Printing Electrospinning Biomimetic Patch for Abdominal Wall Defect Regeneration

Tao Yinghua,Luo Peiyu,Jing Fengya,Liu Tao,Tan Xin,Lyu Zhiyang,VeerleBernaerts Katrien,Zhang Tianzhu,Jia Ruipeng

Repairing abdominal wall defects presents significant challenges, due to the high infection risk, poor biocompatibility, and insufficient mechanical strength associated with synthetic materials. To overcome these limitations, we developed a bioinspired multifunctional 3DPF patch by integrating 3D printing and electrospinning technologies. The core material of the patch is 4arm-PLGA-GPO (4A-GPO), synthesized by conjugating the Gly-Pro-Hyp (GPO) peptide sequence with 4arm-PLGA(4A), which significantly enhances bioactivity and mechanical properties. Additionally, the patch encapsulates basic fibroblast growth factor (bFGF) to stimulate cell proliferation and migration, while an antibacterial layer composes of emodin (EMO) and tobramycin to prevent infection. In vivo studies demonstrate the 3DPF patch effectively accelerates tissue repair by reducing fibrosis and adhesions, promoting angiogenesis and collagen deposition, and modulating the immune response. Transcriptomic analysis reveals that the patch downregulates IL-17 mediated inflammatory pathways while upregulating cell adhesion molecule-related pathways, synergistically facilitating microenvironment reconstruction. Furthermore, molecular docking studies suggest the patch interacts with key molecules such as VEGF and COL3, enhancing angiogenesis and matrix remodeling. In summary, this biomimetic patch, composed of bioactive materials with well-defined chemical compositions, integrates mechanical support, immune modulation, and antibacterial protection. by offering a comprehensive solution for abdominal wall repair, it holds significant potential for clinical translation in complex tissue engineering applications.

Evaluation of a Photopolymerized Gelatin Hydrogel Network with bFGF-Loaded Alginate Microspheres for Tympanic Membrane Perforation Repair in Rats

Hao Xue,Shengjia Chen,Zhechen Yuan,Yi Hu,Juntao Huang,Yi Shen

Tympanic membrane perforation (TMP) often leads to hearing loss and requires effective repair strategies. However, existing surgical options are invasive and lack ideal biomaterials for scaffold-based healing. Herein, we present a custom-engineered mechano-acoustic responsive hydrogel incorporating bFGF-loaded sodium alginate microspheres, designed for controlled drug release and tissue regeneration under dual stimulation: vibrational simulation and low-frequency acoustic waves (generated by radio devices) — representing its first transition from in vitro characterization to in vivo regenerative application. This polymer-based scaffold exhibits robust adhesion, biocompatibility, and mechanoresponsive release behavior. A rat acute TMP model was employed to evaluate the hydrogel's therapeutic efficacy. Compared with control and blank hydrogel groups, the bFGF-loaded mechanoresponsive hydrogel (SGM) noticeably accelerated tympanic membrane closure, reduced local inflammation, and enhanced early auditory recovery, as confirmed by otoscopic inspection, ABR tests, histological staining, and TEM imaging. Our findings demonstrate that the SGM hydrogel effectively promotes functional tissue regeneration and early hearing restoration in vivo. This work highlights the potential of polymer-based, stimuli-responsive biomaterials in advancing minimally invasive strategies for TMP treatment and offers valuable insights for future tissue engineering applications in otolaryngology.

Customized Vascular Repair Microenvironment: Poly(lactic acid)-Gelatin Nanofibrous Scaffold Decorated with bFGF and Ag@Fe3O4 Core–Shell Nanowires

Congyi Yang,Weiwen Yuan,Guoxing Liao,Zhe Tang,Tong Zhu,Yifan Jia,Qianqian Yu,LinGe Wang

Vascular defects caused by trauma or vascular diseases can significantly impact normal blood circulation, resulting in serious health complications. Vascular grafts have evolved as a popular approach for vascular reconstruction with promising outcomes. However, four of the greatest challenges for successful application of small-diameter vascular grafts are (1) postoperative anti-infection, (2) preventing thrombosis formation, (3) utilizing the inflammatory response to the graft to induce tissue regeneration and repair, and (4) noninvasive monitoring of the scaffold and integration. The present study demonstrated a basic fibroblast growth factor (bFGF) and oleic acid dispersed Ag@Fe3O4 core–shell nanowires (OA-Ag@Fe3O4 CSNWs) codecorated poly(lactic acid) (PLA)/gelatin (Gel) multifunctional electrospun vascular grafts (bAPG). The Ag@Fe3O4 CSNWs have sustained Ag+ release and exceptional photothermal capabilities to effectively suppress bacterial infections both in vitro and in vivo, noninvasive magnetic resonance imaging (MRI) modality to monitor the position of the graft, and antiplatelet adhesion properties to promise long-term patency. The gradually released bFGF from the bAPG scaffold promotes the M2 macrophage polarization and enhances the recruitment of macrophages, endothelial cells (ECs) and fibroblast cells. This significant regulation of diverse cell behavior has been proven to be beneficial to vascular repair and regeneration both in vitro and in vivo. Therefore, this study supplies a method to prepare multifunctional vascular-repair materials and is expected to represent a significant guidance and reference to the development of biomaterials for vascular tissue engineering.

A bioactive hydrogel integrating bFGF/VEGFA gene-loaded nanoparticles and platelet-rich plasma for accelerated full-thickness skin wound healing

Yan Duan,Weixin Wang,Yuanyuan Jin,Ming Jiang,Bolin Wang,Jianle Chen,Yun Jiang

Research objectives Full-thickness skin defects present substantial challenges to the healing process, arising from the loss of the dermal layer, inadequate vascular supply, and insufficient growth factor availability. Although previous studies have established that growth factors and platelet-rich plasma (PRP) individually promote tissue repair, the synergistic benefits of their combined application for achieving superior therapeutic outcomes remain unclear. Here, we introduce a novel composite biomaterial: a hydrogel integrating nanoparticles loaded with basic fibroblast growth factor (bFGF) and vascular endothelial growth factor A (VEGFA) genes (bFGF/VEGFA NPs) with PRP (termed bFGF/VEGFA@PRP hydrogel), engineered to enable the synergistic delivery of growth factors and bioactive components. Methods In this study, we hypothesized that the bFGF/VEGFA@PRP hydrogel can promote wound healing. This hypothesis was tested through various experimental perspectives and methods including material characterization, wound healing rate, wound blood flow signal intensity, skin attachment, cell proliferation, apoptosis, collagen deposition, and growth factor levels. Results In a rat cutaneous wound model, this hydrogel accelerated wound closure, enhanced local blood perfusion, and increased the density of skin appendages and collagen fibers. Mechanistic investigations revealed upregulated expression of angiogenic factors bFGF, VEGFA, and platelet and endothelial cell adhesion molecule 1 (CD31), the anti-apoptotic protein BCL2, the cell proliferation marker Ki67, and type III collagen (COL III) in the treated group. Conclusions Collectively, these results demonstrate that the bFGF/VEGFA@PRP hydrogel promotes wound healing through the synergistic enhancement of growth factor expression, angiogenesis, and COL III deposition. The combination of gene therapy with PRP treatment was found to be more effective than either modality alone. This study establishes a promising therapeutic strategy for managing complex full-thickness skin injuries.

An immunoregulation PLGA/Chitosan aligned nanofibers with polydopamine coupling basic fibroblast growth factor and ROS scavenging for peripheral nerve regeneration

Xiaokun Chen,Jihai Xu,Feng Qin,Ziyuan Yang,Xueyuan Li,Miao Yu,Ming Li,Yanhua Wang,Wang Xin

The repair and functional recovery of long-segment peripheral nerve injuries are crucial in clinical settings. Nerve conduits are seen as promising alternatives to autologous nerve grafts, but their effectiveness is limited by the controlled delivery of bioactive factors and meeting various functional requirements during different stages of repair. This research developed multifunctional nerve conduits using electrospinning and polydopamine (PDA) coating techniques to integrate bioactive substances. Chitosan-composite PLGA electrospun nerve conduits demonstrated exceptional mechanical properties and biocompatibility. Nanofibers with specific topological structures effectively promoted oriented cell growth. The PDA coating provided ROS scavenging and immune modulation functions. The bFGF growth factor attached to the PDA coating facilitated sustained release, enhancing Schwann cell functionality and stimulating neurite outgrowth. In a rat sciatic nerve defect model with a 10 mm gap, PLGA/CS-PDA-bFGF nerve conduits showed a positive impact on nerve regeneration and functional recovery. Consequently, nerve conduits with multiple functions modified with PDA-coated bioactive molecules are poised to be excellent materials for mending peripheral nerve injuries.

A multifunctional bilayer wound dressing co-loaded with nanosilver and bFGF for enhanced skin regeneration: synergistic antibacterial, hemostatic, and angiogenic effects

Qianqian Wang,Fang Ma,Ying Xian,Xingyan Shi,Shenglan Ma,Rui Zhang,Hualin Zhang

Introduction: Wound closure, infection prevention and accelerated healing are important to consider in the development of multifunctional dressings. In this study, we aimed to design a dressing that can simulate the structure of normal skin, prevent bacterial infection, improve hemostasis and angiogenesis, accelerate cell adhesion and proliferation, and continuously promote skin regeneration. Methods: We created a multifunctional bilayer dressing coloaded with nanosilver and basic fibroblast growth factor (bFGF) via emulsion electrospinning and vacuum freeze-drying methods. The upper hydrophobic layer was composed of a poly(lactic-co-glycolic acid)/wool keratin (PLGA/WK) electrospun membrane containing nanosilver and bFGF, and the lower hydrophilic layer was composed of a chitosan sponge containing bFGF.Results and Discussion: The bilayer dressing allowed the sustained release of nanosilver and bFGF for more than 2 weeks, had good water absorption and water retention rates, had an appropriate water vapor transmission rate, absorbed excess exudate, and maintained a moist wound environment. In vitro, the dressing prevented bacterial penetration, accelerated fibroblast migration, and promoted angiogenesis and coagulation. Furthermore, the bilayer dressing facilitated cell migration and proliferation, promoted vessel formation, shortened the healing time and significantly promoted wound healing in vivo. By day 3, the wound healing rate in the bilayer dressing group (35.28% ± 2.06%) was significantly higher than that in the control group (23.99% ± 4.32%), representing an increase of approximately 47%. Our findings suggest that this multifunctional bilayer dressing coloaded with nanosilver and bFGF is a potential candidate for skin repair and regeneration applications.
