Antibacterial Adhesion of Poly(methyl methacrylate) Modified by Borneol Acrylate

Xueli Sun,Zhiyong Qian,Lingqiong Luo,Qipeng Yuan,Ximin Guo,Lei Tao,Yen Wei,Xing Wang

Poly(methyl methacrylate) (PMMA) is a widely used biomaterial. But there is still a challenge facing its unwanted bacterial adhesion because the subsequent biofilm formation usually leads to failure of related implants. Herein, we present a borneol-modified PMMA based on a facile and effective stereochemical strategy, generating antibacterial copolymer named as P(MMA-co-BA). It was synthesized by free radical polymerization and studied with different ratio between methyl methacrylate (MMA) and borneol acrylate (BA) monomers. NMR, GPC, and EA, etc., were used to confirm their chemical features. Their films were challenged with Escherichia coli (Gram-negative) and Bacillus subtilis (Gram-positive), showing a BA content dependent antibacterial performance. The minimum effective dose should be 10%. Then in vivo subcutaneous implantations in mice demonstrated their biocompatibilities through routine histotomy and HE staining. Therefore, P(MMA-co-BA)s not only exhibited their unique antibacterial character but also suggested a potential for the safe usage of borneol-modified PMMA frame and devices for further implantation.

Folate and Borneol Modified Bifunctional Nanoparticles for Enhanced Oral Absorption

Yifan Yang,Yunzhi Yin,Jun Zhang,Tiantian Zuo,Xiao Liang,Jing Li,Qi Shen

Oral delivery is considered the preferred route of administration due to its convenience and favorable compliance. Here, docetaxel (DTX) loaded polylactic-co-glycolic acid (PLGA) nanoparticles, coated with polyethyleneimine–folic acid (PEI-FA) and polyethyleneimine–borneol (PEI-BO), were designed to enhance oral absorption (FA/BO-PLGA-NPs). The FA/BO-PLGA-NPs were spherical and smooth with an average size of (137.0 ± 2.1) nm. Encapsulation efficiency (EE%) and drug loading (DL%) were (80.3 ± 1.8)% and (2.3 ± 0.3)%, respectively. In vitro release studies showed that approximately 62.1% of DTX was released from FA/BO-PLGA-NPs in media at pH 7.4. The reverted gut sac method showed that the absorption of FA/BO-PLGA-NPs in the intestines was approximately 6.0 times that of DTX. Moreover, cellular uptake suggested that the obtained FA/BO-PLGA-NPs could be efficiently internalized into Caco-2 cells via FA-mediated active targeting and BO-mediated P-glycoprotein (P-gp) inhibition. Pharmacokinetics study demonstrated that after oral administration of DTX at a dose of 10 mg/kg in FA/BO-PLGA-NPs, the bioavailability of FA/BO-PLGA-NPs was enhanced by approximately 6.8-fold compared with that of DTX suspension. FA/BO-PLGA-NPs caused no obvious irritation to the intestines. Overall, the FA/BO-PLGA-NP formulation remarkably improved the oral bioavailability of DTX and exhibited a promising perspective in oral drug delivery.Keywords:docetaxel;nanoparticles;dual-functional;folic acid;borneol;oral absorption

All-Natural Injectable Antibacterial Hydrogel Enabled by Chitosan and Borneol

Zhijie Zhao,Xiao Fan,Xinyu Li,Yuwei Qiu,Yunfeng Yi,Yuping Wei,Yong Wang

Hydrogels with intrinsic antimicrobial capabilities based on natural strategies have been studied as a hot topic in biomedicine. Nevertheless, it is highly challenging to thoroughly develop a bacteriostatic natural hydrogel. Borneol as a traditional Chinese medicine possesses a unique broad-spectrum antibacterial activity under a membrane-breaking mechanism. In this study, a range of fully natural antibacterial hydrogels are designed and synthesized via the Schiff base cross-linking of carboxymethyl chitosan and dialdehyde dextran grafted natural borneol. The borneol with three configurations is hydrophilically modified onto dextran to boost its antibacterial activity. Also, the synergism of hydrophilic-modified borneol groups and positively charged ammonium ions of carboxymethyl chitosan make the hydrogels totally constrict the E. coli and S. aureus growth during 24 h. Furthermore, the hydrogels exhibit good in vitro cytocompatibility through cytotoxicity, protein adhesion, and hemolytic tests. In view of the injectability, the hydrogels can be delivered to the target site through a minimally invasive route. In short, this work offers a potential tactic to develop antibacterial hydrogels for the treatment of topical wound infections.

Synthesis of borneol via isomerization of isoborneol catalyzed by Co/TiO2

Yuansen Xiao,Huimin Li,Zheng Wang,Zhihan Liu,Chenyang Li,Bin Wu,Longfei Yan,Huidong Zheng,Fuweng Zhang

Borneol synthesis faces challenges from co-generated impurities (isoborneol/camphor) and competing dehydration/dehydrogenation during isomerization. Here, Co/TiO 2 is demonstrated as a selective catalyst for isoborneol-to-borneol isomerization, effectively suppressing structural degradation. Controlled acidity and cobalt dispersion are identified as critical factors: TiO 2 's moderate acidity promotes isomerization, while strongly acidic supports (e.g., Mordenite) favor dehydration to camphene. Reduced cobalt particle size enhances active-site accessibility, lowering activation energy and boosting borneol yield. A one-pot two-step process achieves >62 % borneol content with <0.5 % camphor, meeting ChPC purity standards. This strategy bypasses traditional purification bottlenecks, offering a scalable route for high-purity borneol synthesis. The mechanistic insights into acid strength and metal dispersion provide a framework for rational catalyst design in industrial isomerization applications.

Electrospun borneol/PVP antibacterial coating for postoperative management of keratoprosthesis implantation

Yan Luo,Tengxiang Wang,Ke Yang,Taixiang Liu,Xue Bai

Keratoprosthesis (K-Pro) implantation and postoperative recovery often lead to severe ocular infections called keratoprosthesis-associated infections (KPAI). Antibiotics are commonly used; however, prolonged antibiotic use can lead to bacterial resistance and disrupt microbial balance in the eye. Borneol provides possibilities for developing antibiotic-independent antimicrobial K-Pro. Therefore, the development of an antibiotic-independent self-antimicrobial K-Pro is crucial for postoperative management. Herein, electrospinning technology was employed to fabricate a soluble borneol-based antibacterial coating on K-Pro surfaces for postoperative management of keratoprosthesis implantation. The modified K-Pro exhibited a distinctive nanofibrous morphology (100% borneol release ratio within 125 min), surface antibacterial properties (Escherichia coli: 2%, Staphylococcus aureus: 6%), biocompatibility, and negligible cytotoxicity towards epithelial cells. This study aimed to explore the potential application of camphor-based electrospun coatings in the field of K-Pro infection resistance, thereby providing novel strategies and material foundations for the effective postoperative management of KPAI.

Improving glioblastoma therapeutic outcomes via doxorubicin-loaded nanomicelles modified with borneol

Lingwei Meng,Xiaoyang Chu,Haoyue Xing,Xuan Liu,Xin Xin,Liqing Chen,Mingji Jin,Youyan Guan,Wei Huang,Zhonggao Gao

Glioblastoma is a grade IV malignant glioma with high recurrence and metastasis and faces a therapeutic obstacle that the blood-brain barrier (BBB) severely hinders the brain entry and efficacy of therapeutic drugs. Previous studies suggest that borneol (BO) has been used to enhance interested drugs to penetrate the BBB. In this study, a borneol-modified nanomicelle delivery system was established to facilitate the brain entry of doxorubicin for glioblastoma therapy. Herein, we firstly conjugated borneol molecules with DSPE-PEG 2000 -COOH to synthesize a novel carrier DSPE-PEG 2000 -BO and also characterized its structure. Doxorubicin-loaded nanomicelles (DOX BO-PMs) were prepared using DSPE-PEG 2000 -BO via electrostatic interaction and the physicochemical properties were investigated. The average particle size and zeta potential of DOX BO-PMs were respectively (14.95 ± 0.17)nm and (−1.27 ± 0.06)mV, and the drug encapsulation efficiency and loading capacity in DOX BO-PMs were (95.69 ± 0.49)% and (14.62 ± 0.39)%, respectively. The drug release of the DOX BO-PMs exhibited a both time- and pH-dependent pattern. The results demonstrated that DOX BO-PMs significantly enhanced the transport efficiency of DOX across the BBB and also exhibited a quick accumulation in the brain tissues. The in vitro anti-proliferation assay results suggested that DOX BO-PMs exerted a strong inhibitory effect on proliferation of glioblastoma cells. Importantly, in vivo antitumor results demonstrated that DOX BO-PMs significantly inhibited the tumor growth and metastasis of glioblastoma. In conclusion, DOX BO-PMs can improve the glioblastoma therapeutic outcomes and become a promising nanodrug candidate for the application of doxorubicin in the field of glioblastoma therapy.

Ligustrazine-Loaded Borneol Liposome Alleviates Cerebral Ischemia–Reperfusion Injury in Rats

Yu Wen,Zuxian Zhang,Zhongmou Cai,Baoning Liu,Zhehao Wu,Yude Liu

Our team’s pharmacological and clinical trials proved that ligustrazine/borneol spray had a definite effect on ischemic stroke (IS). To solve the shortcomings of ligustrazine/borneol spray, such as low bioavailability, short half-life, and poor compatibility between borneol and ligustrazine, ligustrazine-loaded borneol liposomes (LIP@TMP) were successfully prepared by a thin-film ultrasonication method. The average particle size of LIP@TMP was 282.4 ± 3.6 nm, the drug loading rate was 14.5 ± 0.6%, and the entrapment efficiency was 42.7 ± 1.0%, which had excellent stability and sustained release ability. In addition, live/dead fluorescent staining and the CCK-8 test confirmed that LIP@TMP had good biocompatibility. Moreover, middle cerebral artery occlusion (MCAO) rat model experiments further demonstrated that LIP@TMP could significantly alleviate cerebral ischemia and reperfusion injury by improving neurological scores, reducing cerebral infarct volume, promoting neurogenesis, inhibiting inflammation, and reducing tissue damage. In addition, LIP@TMP enhanced neuronal marker doublecortin (DCX) and neuronal nuclei (NEUN), inhibited inflammatory factors (TNF-α and IL-1β), and reduced apoptosis signal molecules (TUNEL and caspase-3). The findings of this study suggested that the prepared LIP@TMP had tremendous potential for the treatment of cerebral ischemia.

Phosphonium Cationic Borneol Stereochemical Membrane Breaking Arrow: High Antibacterial and Antibiofilm Activity

Xiangbin Sun,Yuanyuan Xie,Xiaobing Ma,Yufeng He,Pengfei Song,Rongmin Wang

Bacterial biofilm contamination poses a substantial threat to public health and safety while also leading to considerable economic losses. In this study, a series of both offensive and defensive poly(quaternary phosphonium salt-borneol) copolymers (P(QPx-BOy)), utilizing stereochemically structured borneol as the membrane breaking arrow and quaternary phosphonium salt cations as the electrostatic driving force, were successfully synthesized. Notably, this work exemplifies a novel integration of natural borneol with quaternary phosphonium cations. Subsequently, the molecular structure, molecular weight distribution, microstructure, and corresponding physicochemical properties of the P(QPx-BOy) were characterized using NMR, FT-IR, GPC, SEM, EDS, CA, and DLS. Leveraging the electrostatic interaction between the phosphonium cation and the stereochemical antibacterial mechanism of borneol, P(QPx-BOy) exhibited remarkable antibacterial efficacy against both E. coli and S. aureus, achieving a 100% antibacterial rate with MIC value as low as 30 μg/mL. Furthermore, crystal violet staining assays revealed that borneol’s unique antibiofilm mechanism exerts a significant inhibitory effect on bacterial biofilm formation. Finally, the biocompatibility and antibacterial potential of P(QPx-BOy) were evaluated through cytotoxicity assays and fabric antibacterial tests. Specifically, the integration of borneol essential oil with hydrophilic quaternary phosphonium salt cations effectively addresses the hydrophobic nature of borneol essential oil while mitigating the cytotoxic effects associated with quaternary phosphonium salts. This synergistic combination leverages the strengths of both components, thereby enhancing their suitability for applications in healthcare and public health sectors.

Borneol-Modified Chitosan Coating with Antibacterial Properties via Layer-by-Layer Strategy

Zixu Xie,Zhiran Zheng,Chen Chen,Guofeng Li,Xing Wang

Chitosan exhibits remarkable broad-spectrum antibacterial activity, especially in acidic environments. However, its poor solubility in water and significantly decreased antibacterial performance after membrane formation greatly limit its extensive application. To address this issue, glycol chitosan-4-pyridylcarboxaldehydeborneol (GCBP) was synthesized by conjugating glycol chitosan (GC) with 4-pyridylcarboxaldehydeborneol ester (BP) through dynamic Schiff base bonds utilizing a layer-by-layer self-assembly strategy. When bacteria come into contact with the surface, the local acidic microenvironment triggers the cleavage of the Schiff base, resulting in the release of bactericidal BP and GC for combined sterilization. In vitro results demonstrated that the antibacterial properties of GCBP were positively related to the modification layers. The excellent antibacterial performance of the GCBP modification demonstrates not only great potential for clinical urinary catheters but also for broad antibacterial applications in the medical field.

Biofilm inhibition and eradication activity of citral and borneol against foodborne bacteria

Wen Wang,Tom Coenye,Jianyu Su,Sheng-Xiang Qiu

Biofilms growing on food surfaces, packaging, equipment and processing environments bring huge risks to food safety and result in economic losses. Natural components are receiving increasing interest as “green” antimicrobial agents due to their biocompatibility, broad-spectrum antimicrobial properties, and antibiofilm potential. As a traditional adjuvant in Chinese medicine, borneol has the potential to enhance citral’s antibiofilm efficacy. In this study, the antibiofilm activities of citral, borneol, and the combination citral and borneol (BC) against various foodborne bacteria were evaluated using multiple techniques. Light microscopy, crystal violet (CV) staining, and the 2,3-bis (2-methoxy-4-nitro-5-sulfophenyl)-5-[(phenylaminocarbonyl]-2H-tetrazolium hydroxide (XTT) assay were employed to monitor biofilm formation. Additionally, biofilm eradication was assessed against Staphylococcus aureus and Escherichia coli biofilms grown on polycarbonate membranes. Protein leakage was measured, and field emission scanning electron microscopy (FESEM) was used to examine cell membrane integrity. Results showed that borneol and citral acted synergistically to inhibit biofilms. Borneol, citral, alone or together inhibited biofilm formation in a concentration dependent manner, and they have the ability to inactivate or kill at least 90.7 ± 1.9 % bacteria in preformed biofilms. Both borneol and citral disrupted the membrane integrity of the bacteria, increased permeability, and ultimately led to cell death. These findings suggest that the combination citral and borneol has the potential to be developed as a food preservative or bactericidal agent for controlling foodborne bacterial contamination.

Bioinspired Sulfobetaine Borneol Fluorinated Amphiphilic Polymers for Marine Antifouling and Fouling Release Applications

Zhongqiang Yu,Wen Sun,Lida Wang,Zhengqing Yang,Yine Ren,Yanke Li,Guichang Liu,Qian Zhang

The development of nontoxic antifouling coatings in static marine environments is urgent. Herein, the successful synthesis of sulfobetaine borneol fluorinated polymers (PEASBF) by a free radical polymerization method is reported. The PEASBF coatings exhibit outstanding antifouling activity, which effectively resists the adhesion of Bovine serum albumin (FITC-BSA adhesion rate: 0.5%), Pseudomonas sp. (Biofilm: 1.3 absorbance) and Navicula sp. (Diatom attachment rate: 33%). More importantly, the PEASBF coatings display outstanding fouling release properties, achieving a release rate of 98% for Navicula sp., and the absorbance of the Pseudomonas sp. biofilm is only 0.2 under 10 Pa shear stress. XPS and MD studies showed that the fluorinated/isobornyl groups induce more sulfobetaine groups to migrate toward polymer surfaces for intensify antifouling. Additionally, the chiral stereochemical structure of borneol enhances antifouling and fouling release ability of amphiphilic polymers. Therefore, the PEASBF has the potential for static marine antifouling applications.

Smart l-borneol-loaded hierarchical hollow polymer nanospheres with antipollution and antibacterial capabilities

W. Song,M. Zhang,X. Huang,B. Chen,Y. Ding,Y. Zhang,D.G. Yu,I. Kim

Porous organic polymers having well-defined morphologies and designable functionalities are potentially suitable materials for environmental remediation, e.g., for the removal of pollutants from water. However, single materials generally do not simultaneously possess high adsorption abilities and antibacterial performance, as well as easy processibility. Herein, we reported a processible porous polymer which composed swellable hollow spherical morphology and branched L-borneol antibacterial agents. These features endure a valuable capability of pollutant absorption for represented pollutants such as, organic dye (methylene blue), organic compound (bisphenol A), and the heavy metal (Cu 2+ ). In addition, the L-borneol groups and 3D crosslinked hierarchical structure result in enhancement of antibacterial performance against Escherichia coli and Staphylococcus aureus . Moreover, we demonstrate the preparation of the polymer as a fine powder could anchored on the membranes, processed into the tablet, adhered with the polyurethane foams, and assembled as the dye absorption device. The rationally designed polymers by this method show notable potential as adsorbents for water purification and sewage treatment.
