Extraction, purification, structural characterization, and anti-skin aging activity of a homogeneous polysaccharide from Durvillaea antarctica

Wu Jiahui,Yuan Yonglei,Qu Liping

As an edible algae with rich nutrients and high medicinal value, Durvillaea antarctica has certain potential in anti-aging, but there is currently limited research on the separation, purification and activity of its anti-aging active ingredients. We successfully extracted and purified a new β-(1,3)-D-glucan with a special branched structure from Durvillaea antarctica using subcritical water extraction combined with ultrafiltration membrane (SWE-UFM). The polysaccharide, named DAG-1, consists of glucose (92.70%) and has a weight-average molecular weight of 10149 Da. The GC–MS and 1D/2D NMR confirmed that DAG-1 was composed of 3)-β-D-Glcp-(1 → , → 3,6)-β-D-Glcp-(1 → , β-D-Glcp-(1 → and → 6)-α-D-Glcp-(1 → , and β-glycosidic linkages between these sugar units. Animal experiments showed that DAG-1 has significant anti-skin aging effects by inhibiting the reduction of superoxide dismutase (SOD) activity, accumulation of malonic dialdehyde (MDA), and decline in hydroxyproline (HYP) content in collagen fibers of D-gal-induced mouse skin. To further elucidate the mechanism of DAG-1, we found that DAG-1 can alleviate senescence in human dermal fibroblasts (HDFa), inhibit degranulation of human cutaneous mast cells (HCMC), and modulate inflammatory factors (TNF-α, IL-6) production. Additionally, DAG-1 may bind to c-KIT receptors and downregulate the expression of KITLG/KIT, regulating the growth and activation of mast cells, and thus alleviate skin aging. These findings suggest that DAG-1 is a potential anti-aging ingredient for functional food, cosmetic, and medicine development.

The regulatory effects of fucoidan and laminarin on functional dyspepsia mice induced by loperamide

Tianxu Liu,Mengting Zhang,Ismail Muhammad Asif,Yonglin Wu,Bin Li,Ling Wang

Gastrointestinal dysmotility is a common cause of functional dyspepsia. As two kinds of polysaccharides derived from brown algae, fucoidan and laminarin possess many physiological properties; however, their relative abilities in regulating gastrointestinal motility have not been illustrated yet. In this study, we aimed to investigate the regulatory effect of fucoidan and laminarin on functional dyspepsia mice induced by loperamide. Mice with gastrointestinal dysmotility were treated with fucoidan (100 and 200 mg per kg bw) and laminarin (50 and 100 mg per kg bw). As a result, fucoidan and laminarin reversed the dysfunction mainly through regulating gastrointestinal hormones (motilin and ghrelin), the cholinergic pathway, the total bile acid level, c-kit protein expression, and gastric smooth muscle contraction-related gene expression (ANO1 and RYR3). Moreover, fucoidan and laminarin intervention modulated the gut microbiota profile including the altered richness of Muribaculaceae, Lachnospiraceae, and Streptococcus. The results indicated that fucoidan and laminarin may restore the rhythm of the migrating motor complex and regulate gut microecology. In conclusion, we provided evidence to support that fucoidan and laminarin might have potential abilities to regulate gastrointestinal motility.

Characteristics of nobiletin-induced effects on jejunal contractility

Yong-Jian Xiong,Da-Peng Chen,Bo-Chao Lv,Fang-Fei Liu,Li Wang,Yuan Lin

Nobiletin , a citrus polymethoxylated flavone , exhibits multiple biological properties including anti-inflammatory, anti-carcinogenic, and anti-insulin resistance effects. The present study found that nobiletin exerted significant stimulatory effects on the contractility of isolated rat jejunal segments in all 6 different low contractile states, and meanwhile significant inhibitory effects in all 6 different high contractile states, showing characteristics of bidirectional regulation (BR). Nobiletin-exerted BR on jejunal contractility was abolished in the presence of c-kit receptor tyrosine kinase inhibitor imatinib or Ca 2   + channel blocker verapamil . In the presence of neuroxin tetrodotoxin , nobiletin only exerted stimulatory effects on jejunal contractility in both low and high contractile states. Hemicholinium-3 and atropine partially blocked nobiletin-exerted stimulatory effects on jejunal contractility in low-Ca 2   + -induced low contractile state. Phentolamine or propranolol or l -NG-nitro-arginine significantly blocked nobiletin-exerted inhibitory effects on jejunal contractility in high-Ca 2   + -induced high contractile state respectively. The effects of nobiletin on myosin light chain kinase (MLCK) mRNA expression, MLCK protein content, and myosin light chain phosphorylation extent were also bidirectional. In summary, nobiletin-exerted BR depends on the contractile states of rat jejunal segments. Nobiletin-exerted BR requires the enteric nervous system, interstitial cell of Cajal, Ca 2   + , and myosin phosphorylation-related mechanisms.

Laxative effect and mechanism of Tiantian Capsule on loperamide-induced constipation in rats

Tian Li,Mengmeng Hu,Cuihua Jiang,Dongjian Zhang,Meng Gao,Jianwei Xia,Mengqi Miao,Gaofeng Shi,Hui Li,Jian Zhang,Zhiqi Yin

Ethnopharmacological relevance Tiantian capsule (TTC), as a functional food, which consists of four herb medicines, including Aloe vera Burm.f. (25%), leaf juices, dried; Cucurbita moschata Duch. (25%), fructus, dried; Poria cocos (Schw.) Wolf. (12.5%), sclerotium, dried; Tremella fuciformis Berk. (12.5%), fruiting bodies, dried, and one extract xylooligosaccharides (25%) from Maize Cob by enzymolysis, has been commonly used in China to ameliorate constipation. Aim of the study The aim of the work is to elucidate the potential laxative mechanisms of TTC in loperamide-induced constipated rats. Materials and methods LC-MS/MS was employed for analyzing the TTC extract. The gastrointestinal transit was evaluated by X-ray. The H&E and Alcian-Blue stain were applied to determine the changes of goblet cells and mucus layer, respectively. Meanwhile, levels of neurotransmitters were evaluated by enzyme-linked immunosorbent assay. The protein expressions were also measured by immunohistochemistry and Western blot . Results Our results showed that TTC administration attenuated constipation responses in aspects of fecal pellets number, water content of feces, stomach emptying and gastrointestinal transit. Further investigations revealed that TTC treatment not only induced the recovery of neurotransmitters, such as motilin , substance P , somatostatin , endothelin and vasoactive intestinal peptide , but also up-regulated the expressions of c-kit and stem cell factor (SCF). Additionally, the number of goblet cells and thickness of the mucus layer were elevated, and the guanylate cyclase C-cGMP signal pathway was also up-regulated after TTC treatment. Conclusion Our findings demonstrated that the laxative effect of TTC in constipation rats is probably due to the regulation of bowel movement and intestinal fluid secretion.

Effects of Saccharomyces boulardii on microbiota composition and metabolite levels in the small intestine of constipated mice

Tang Shuai,Li Jia,Li Yi,Du Haitao,Zhu Wenya,Zhang Ru,Wan Jun

Saccharomyces boulardii (S. boulardii) is a fungal probiotic used to treat digestive disorders. However, the mechanism(s) by which S. boulardii affects the small intestine remains unclear. Here, we aimed to explore the effects of S. boulardii on the small intestine and the underlying mechanisms in mice with loperamide-induced constipation. While S. boulardii administration did not fully reverse the alterations in loperamide-induced defecation parameters, it altered the small intestinal floral composition toward a community conducive to alleviate constipation. Moreover, S. boulardii up-regulated the expression of tyrosine-protein kinase Kit (c-Kit), aquaporin 3 (AQP3), interleukin (IL)-10, myosin light chain kinase (MLCK), and phosphorylated myosin light chain 20 (P-MLC20), while concurrently down-regulating the expression levels of inducible nitric oxide synthase (iNOS), p65, and IL-17 A. These alterations indicate a discernible effect of small intestinal water reabsorption, inflammatory factor levels, and smooth muscle contraction. Saccharomyces boulardii also positively regulated small intestinal metabolite levels, such as fructose 6-phosphate, dihomo-alpha-linolenic acid, and 3-(4-hydroxyphenyl) lactate, and participated in metabolic pathways such as arginine biosynthesis, linoleic acid metabolism, and protein digestion and absorption. While not fully reversing defecation changes, Saccharomyces boulardii alters intestinal flora, up-regulates key proteins affecting water reabsorption and inflammation, and positively influences metabolic pathways. Our study provides serves as a basis for further studies on the application of S. boulardii in the treatment of intestinal disorders.
