Response of Brassica chinensis L to the stress of combined pollution of microplastics and cypermethrin

Li Hua,Meiting Wang,Xinlong Wei

Microplastics (MPs) can coexist with pesticides in soil for a long period, forming combined pollution. Brassica chinensis L was used as the test plant and the differences in the effects of traditional microplastic polyethylene (PE MPs) and biodegradable microplastic polylactic acid (PLA MPs) alone and combination with cypermethrin pesticide on plants were studied. The results showed that the inhibitory effect of the PLA MPs on Brassica chinensis L was greater than PE MPs and that the inhibitory effect of microplastics in combination with cypermethrin pesticides was greater than that of microplastics alone. Brassica chinensis L avoided oxidative damage by increasing the stress level at a low PE MP concentration (2%) and the antioxidant defense capability gradually weakened as the concentration increased. As the concentration of PLA MPs increased, the oxidative damage of plant roots was further aggravated. The biomass and quality of Brassica chinensis L showed the pattern of ‘low promotion and high inhibition’ whether PE MPs were treated alone or combined with cypermethrin pesticides. While PLA MPs treatment groups showed a pattern of ‘stronger inhibition with increasing microplastic concentration’.

Toxic effect and mechanism of β-cypermethrin and its chiral isomers on HTR-8/SVneo cells

Chaojin Si,Hongjun Yang,Xiaoyan Wang,Qiaoxin Wang,Min Feng,Huayue Li,Yuqi Feng,Jiayuan Zhao,Ying Liao

Beta-cypermethrin (β-CYP) consists of four chiral isomers, acting as an environmental estrogen and causing reproductive toxicity, neurotoxicity, and dysfunctions in multiple organ systems. This study investigated the toxic effects of β-CYP, its isomers, metabolite 3-phenoxybenzoic acid (3-PBA), and 17β-estradiol (E2) on HTR-8/SVneo cells. We focused on the toxic mechanisms of β-CYP and its specific isomers. Our results showed that β-CYP and its isomers inhibit HTR-8/SVneo cell proliferation similarly to E2, with 100 μM 1S-trans-αR displaying significant toxicity after 48 h. Notably, 1S-trans-αR, 1R-trans-αS, and β-CYP were more potent in inducing apoptosis and cell cycle arrest than 1R-cis-αS and 1S-cis-αR at 48 h. AO/EB staining and flow cytometry indicated dose-dependent apoptosis in HTR-8/SVneo cells, particularly at 100 μM 1R-trans-αS. Scratch assays revealed that β-CYP and its isomers variably reduced cell migration. Receptor inhibition assays demonstrated that post-ICI 182780 treatment, which inhibits estrogen receptor α (ERα) or estrogen receptor β (ERβ), β-CYP, its isomers, and E2 reduced HTR-8/SVneo cell viability, whereas milrinone, a phosphodiesterase 3 A (PDE3A) inhibitor, increased viability. Molecular docking studies indicated a higher affinity of β-CYP, its isomers, and E2 for PDE3A than for ERα or ERβ. Consequently, β-CYP, its isomers, and E2 consistently led to decreased cell viability. Transcriptomics and RT-qPCR analyses showed differential expression in treated cells: up-regulation of Il24 and Ptgs2 , and down-regulation of Myo7a and Pdgfrb , suggesting the PI3K-AKT signaling pathway as a potential route for toxicity. This study aims to provide a comprehensive evaluation of the cytotoxicity of chiral pesticides and their mechanisms.

Effect of cypermethrin on thyroid follicular epithelial cell injury via the ROS-NF-κB-NLRP3 pathway

Leina Jia,Xiaoning Meng,Yu Ma,Jinyu Luo,Xiaoqi Luo,Huifang Yang,Jian Zhou

Cypermethrin (CYP, IUPAC name: [cyano-(3-phenoxyphenyl)methyl] 3-(2,2-dichloroethenyl)-2,2-dimethylcyclopropane-1-carboxylate), a type II pyrethroid insecticide, is suggested to have potential adverse effects on human endocrine, immune, and neurotoxic systems. Objectives: In this study, we aimed to investigate whether CYP causes oxidative stress and pyroptosis in thyroid follicle epithelial cells through the activation of the ROS-NFκB-NLRP3 pathway, thereby causing inflammatory responses. Methods: Nthy-ori 3-1 cells were used as an in vitro model and exposed to CYP at different doses (0, 100, 200, 400, 800, and 1600 μmol/L) for 24   h. Results: CYP treatment enhanced oxidative damage in Nthy-ori 3-1 cells, characterized by cellular crumpling, indistinct borders, increased cellular gaps, appearance of intercellular grease droplets, and increased pyroptosis. Subsequent treatment with the caspase-1 inhibitor VX-765 enhanced cell viability in the VX-765+CYP group, improving cell morphology, reducing pyroptotic vesicles, suppressing oxidative stress, and downregulating Interleukin-18(IL-18) (an inflammatory factor) level. Conclusion: The results demonstrate that CYP induces pyroptosis in Nthy-ori 3-1 cells through activation of the reactive oxygen species (ROS)-NF-κB-NLRP3 pathway.

Functional analysis of the regulation of beta-cypermethrin resistance by Deg-CYP-3 in Dermanyssus gallinae

Jing Liu,Zhengjie Wang,Shuo Yin,Zhonghao Wang,Meng Wu,Jianhua Qin,Junlong Liu,Chuanwen Wang

Dermanyssus gallinae is a significant haematophagous ectoparasite of laying hens. Despite the worldwide application of beta-cypermethrin for mite control, resistance to this compound has been developed by D. gallinae . P450s, as key detoxification enzymes involved in the metabolism of exogenous and endogenous chemical substances, have not been functionally characterized in D. gallinae . Here, the role of the P450s gene (Deg-CYP-3) of D. gallinae in acaricide resistance was studied. Firstly, P450s activities of the five beta-cypermethrin resistant strains (RR) against p-nitroanisole were significantly higher than that of the susceptible strain (RS), suggesting enhanced enzymatic activities might contribute to mite beta-cypermethrin resistance. Both mRNA expression level and DNA copy number of Deg-CYP-3 were significantly higher in RR than in RS, except for DK RR strain. Moreover, recombinant Deg-CYP-3 (rCYP-3) expressed in Escherichia coli displayed high enzymatic activity against p-nitroanisole, while Qulaton-100, acetone, isoacetone, avermectin, and ivermectin were found to inhibit this activity. Immunohistochemical analysis revealed the presence of Deg-CYP-3 protein in the digestive tract and Malpighian tubules. Finally, silencing of Deg-CYP-3 via RNAi resulted in decreased enzyme activity and increased susceptibility of PG RR strain to beta-cypermethrin, confirming that Deg-CYP-3 is crucial for beta-cypermethrin detoxification. RNAi also resulted in enhanced mortality, as well as reduced oviposition and fecundity in the PG RR strain. Collectively, these findings indicate that resistance of D. gallinae to beta-cypermethrin is associated with elevated P450s protein activity and increased Deg-CYP-3 expression levels. The results provide insights into the metabolic resistance mechanisms of D. gallinae and offer scientific guidance for the management and control of mite.

Maternal exposure to beta-Cypermethrin disrupts placental development by dysfunction of trophoblast cells from oxidative stress

Xiaoli Tang,Yanqing Geng,Rufei Gao,Zhuxiu Chen,Xinyi Mu,Yan Zhang,Xin Yin,Yidan Ma,Xuemei Chen,Fangfang Li,Junlin He

As a broad-spectrum and efficient insecticide, beta-Cypermethrin (β-CYP) poses a health risk to pregnancy. It matters the mechanisms of maternal exposure to β-CYP for impacting reproductive health. The placenta, a transient organ pivotal for maternal-fetal communication during pregnancy, plays a crucial role in embryonic development. The effect of β-CYP exposure on the placenta and its underlying molecular mechanisms remain obscure. The objective of this study was to investigate the effect of β-CYP exposure on placental development and the function of trophoblast, as well as the underlying mechanisms through CD-1 mouse model (1, 10, 20   mg/kg.bw) and in vitro HTR-8/SVneo cell model (12.5, 25, 50, 100   μM). We found slower weight gain and reduced uterine wet weight in pregnant mice with maternal exposure to β-CYP during pregnancy, as well as adverse pregnancy outcomes such as uterine bleeding and embryo resorption. The abnormal placental development in response to β-CYP was noticed, including imbalanced placental structure and disrupted labyrinthine vascular development. Trophoblasts, pivotal in placental development and vascular remodeling, displayed abnormal differentiation under β-CYP exposure. This aberration was characterized by thickened trophoblast layers in the labyrinthine zone, accompanied by mitochondrial and endoplasmic reticulum swelling within trophoblasts. Futher researches on human chorionic trophoblast cell lines revealed that β-CYP exposure induced apoptosis in HTR-8/SVneo cells. This induction resulted in a notable decrease in migration and invasion abilities, coupled with oxidative stress and the inhibition of the Notch signaling pathway. N-acetylcysteine (an antioxidant) partially restored the impaired Notch signaling pathway in HTR-8/SVneo cells, and mitigated cellular functional damage attributed to β-CYP exposure. Collectively, exposure to β-CYP induced oxidative stress and then led to inhibition of the Notch signaling pathway and dysfunction of trophoblast cells, ultimately resulted in abnormal placenta and pregnancy. These findings indicate Reactive Oxygen Species as potential intervention targets to mitigate β-CYP toxicity. The comprehensive elucidation contributes to our understanding of β-CYP biosafety and offers an experimental basis for preventing and managing its reproductive toxicity.

Identification and biochemical characterization of a carboxylesterase gene associated with β-cypermethrin resistance in Dermanyssus gallinae

Xuedi Zhang,Yue Zhang,Kai Xu,Jianhua Qin,Dehe Wang,Lijun Xu,Chuanwen Wang

ABSTRACT Dermanyssus gallinae is a major hematophagous ectoparasite in layer hens. Although the acaricide β-cypermethrin has been used to control mites worldwide, D. gallinae has developed resistance to this compound. Carboxylesterases ( CarEs ) are important detoxification enzymes that confer resistance to β-cypermethrin in arthropods. However, CarEs associated with β-cypermethrin resistance in D. gallinae have not yet been functionally characterized. Here, we isolated a CarE gene ( Deg-CarE ) from D. gallinae and assayed its activity. The results revealed significantly higher expression of Deg-CarE in the β-cypermethrin-resistant strain ( RS ) than in the susceptible strain ( SS ) toward α-naphthyl acetate ( α-NA ) and β-naphthyl acetate ( β-NA ). These findings suggest that enhanced esterase activities might have contributed to β-cypermethrin resistance in D. gallinae . Quantitative real-time PCR analysis revealed that Deg-CarE expression levels were significantly higher in adults than in other life stages. Although Deg-CarE was upregulated in the RS, significant differences in gene copy numbers were not observed. Additionally, Deg-CarE expression was significantly induced by β-cypermethrin in both the SS and RS. Moreover, silencing Deg-CarE via RNA interference decreased the enzyme activity and increased the susceptibility of the RS to β-cypermethrin, confirming that Deg-CarE is crucial for β-cypermethrin detoxification. Finally, recombinant Deg-CarE ( rDeg-CarE ) expressed in Escherichia coli displayed high enzymatic activity toward α/β-NA. However, metabolic analysis indicated that rDeg-CarE did not directly metabolize β-cypermethrin. The collective findings indicate that D. gallinae resistance to β-cypermethrin is associated with elevated CarEs protein activity and increased Deg-CarE expression levels. These findings provide insights into the metabolic resistance of D. gallinae and offer scientific guidance for the management and control of D. gallinae .

A molecularly imprinted fluorescence nanosensor based on upconversion metal–organic frameworks for alpha-cypermethrin specific recognition

Hu Xuelian,Cao Yichuan,Tian Yanyan,Qi Ying,Fang Guozhen,Wang Shuo

A sensitive molecularly imprinted fluorescent nanosensor based on zeolitic imidazolate frameworks-8 (ZIF-8) and upconversion nanoparticles (UCNPs) was developed for the determination of trace alpha-cypermethrin (α-CPM) for the first time. The sensor was synthesized by a layer-by-layer self-assembly strategy. UCNPs with a maximum emission wavelength of 544.5 nm under 980 nm excitation were firstly prepared as the luminous core. Then, ZIF-8 with the large specific surface and porosity was introduced, which not only improved the mass transfer and adsorption capacity of the sensor but also increased the fluorescence intensity of UCNPs as a protective layer. Finally, molecularly imprinted polymers (UCNPs@ZIF-8@MIPs) were fabricated in mixed solutions containing UCNPs@ZIF-8 (support material), α-CPM (template), acrylamide (functional monomer), and divinylbenzene (cross-linker). Under the optimal condition, the fluorescence intensity of UCNPs@ZIF-8@MIP was linearly quenched with increasing concentration of α-CPM in the range 0.10–12 mg L −1 with a detection limit of 0.03 mg L −1 (S/ N  = 3). The developed UCNPs@ZIF-8@MIP probe was used to detect α-CPM in real samples; the satisfactory results obtained were consistent with those obtained by GC-MS. Graphical abstract

Distribution, metabolism and metabolic disturbances of alpha-cypermethrin in embryo development, chick growth and adult hens

Xueke Liu,Chang Liu,Peng Wang,Yiran Liang,Jing Zhan,Zhiqiang Zhou,Donghui Liu

Alpha-cypermethrin (Alpha-CP), an important pyrethroid pesticide , has been widely used for pest control in agriculture and parasite control in livestock farms. Thus, alpha-CP is easily exposed to wild birds and poultry, which may pose a potential risk to birds. Alpha-CP and its metabolites have been detected in many environmental samples, including poultry and wild birds. We studied the distribution and metabolism of alpha-CP and its metabolites in embryo development and newborn chick. The results showed that metabolites were the main residual forms of alpha-CP in different stages of life and might increase the exposure risk of bird and its offspring . Metabolomics investigation of newborn chick exhibited that the metabolic profiles of chicks were disturbed, especially lipid metabolism . The concentrations of cis-DCCA and trans-DCCA were high in the first and second weeks of chick growth, indicating that chicks have limited ability to further metabolize and excrete cis-DCCA and trans-DCCA during the early stages of chicks. Toxicokinetics of alpha-CP in adult hens showed that alpha-CP was rapidly metabolized to acid metabolites, which could be further metabolized and excreted. The results about metabolism of alpha-CP in different stages of chicken indicate that the ability of the embryo and early chick to metabolize alpha-CP and its metabolites was the weakest. Therefore, it is of important significance to focus on evaluating the ecological risk of cypermethrin on birds at different stages of life cycle.

Dietary cypermethrin exposure reshapes the rumen microbiota and enriches antibiotic resistance genes: Metagenomic evidence of co-selection

Xiaoli Zhang,Shihao Han,Anlin Zhao,Bingni Wei,Xuan Chang,Siwei Song,Yongju Zhao,Zhongquan Zhao,Xiangfei Zhang,Juncai Chen

Pesticide residues in crop-derived feedstocks represent a pervasive environmental stressor in agro-ecosystems, yet their role in driving the non-antibiotic co-selection of antimicrobial resistance (AMR) within the ruminant gut reservoir remains poorly understood. This study investigated the physiological trade-offs and indirect mechanisms of resistome expansion in a ruminant model exposed to environmentally relevant levels of cypermethrin. Integrated metagenomic and phenotypic assays revealed that cypermethrin exposure did not impair growth performance, but significantly increased daily feed intake and shifted fermentation profiles toward acetate. This metabolic compensation was supported by a reshaped core microbiome, characterized by increased abundance of fibrolytic consortia (e.g., Fibrobacter , Ruminococcus ), enrichment of carbohydrate-active enzymes (GH3, GH5, GH13, and GH43), and upregulation of glycolysis and acetate-producing pathways. However, this metabolic adaptation came at a severe physiological cost, evidenced by systemic oxidative injury and hepatic dysfunction in the host. Crucially, cypermethrin acted as a potent non-antibiotic selective agent that expanded the ruminal resistome and mobilome, specifically, enriching efflux pumps (e.g., oqxA , MexB ) confirmed target alteration genes (e.g., vanE ). Consequently, dietary cypermethrin exposure forces microbial metabolic compensation at the expense of host hepatic health, while turning the ruminant gut into an overlooked repository for AMR. These findings highlight the critical ecological risks of pesticide-induced resistance co-selection, threatenting the One Health framework. Future research should incorporate multi-dose gradients, evaluate long-term exposure effects using sequential temporal sampling, and utilize non-invasive baseline monitoring across diverse ruminant species to fully elucidate these ecological risks.

Characterization of Four Carboxylesterases Involved in Detoxification of β-Cypermethrin, λ-Cyhalothrin, and Malathion in Helicoverpa armigera

Yongmei Chang,Weihuan Xu,Shan Wang,Mengyao Zhu,Yanan Ru,Zhiheng Xu,Guangyou Chen,Yongqiang Li

Helicoverpa armigera Hübner (Lepidoptera: Noctuidae) is a globally devastating pest and has evolved with varying levels of resistance to a broader spectrum of insecticides. CarEs are major enzymes involved in insecticide detoxification and metabolic resistance. Four CarE genes were identified and cloned from H. armigera, and the expression pattern analyses indicated that they were predominantly expressed in the detoxifying tissues and larval feeding periods. The insecticide inductive assays further suggested that these CarE genes with expressions were significantly upregulated after beta-cypermethrin, lambda-cyhalothrin, malathion, and chlorpyrifos exposures. The purified recombinant proteins of both CarE016B and CarE016C by bacterial expression exhibited significantly higher catalytic efficiency toward α-naphthyl acetate and also showed relatively stronger binding with both β-cypermethrin and λ-cyhalothrin compared to the CarE006C and CarE015A. In vitro metabolism assay with HPLC suggests that CarE016B and CarE016C have the ability to metabolize β-cypermethrin, λ-cyhalothrin, and malathion with varying hydrolase activities. GC-MS/MS analysis identified 3-phenoxybenzaldehyde (3-PBAld) as the metabolite of both β-cypermethrin and λ-cyhalothrin. Homology modeling and molecular docking analyses further indicate that these three types of insecticides could be anchored into the active pocket of both CarEs. Collectively, these results demonstrate that both CarE016B and CarE016C play a critical role in the detoxification of pyrethroid and organophosphate insecticides in H. armigera. This study provides the foundations for a comprehensive understanding of the role of the CarEs family in insecticide detoxification and resistance in H. armigera.

The influence of oxytetracycline on the degradation and enantioselectivity of the chiral pesticide beta-cypermethrin in soil

Wenqi Jiang,Jing Gao,Zheng Cheng,Wangjing Zhai,Donghui Liu,Zhiqiang Zhou,Peng Wang

Pesticide residues most likely coexist with antibiotics due to the application of animal-based fertilizers in agriculture. In this study, the degradation and enantioselectivity of beta-cypermethrin in soil and chicken manure-amended soil were investigated. The effects of oxytetracycline on the soil microbial community were also estimated. The results showed that the half-life of beta-cypermethrin in the soil was 16.9 days and that the (+)-enantiomer was degraded preferentially in both pairs of enantiomers. The metabolites cis/trans-DCCA(3-(2′,2′-dichlorovinyl)-2,2-dimethylcyclopropane carboxylic acid) and 3-PBA (3-Phenoxybenzoic acid) were detected. The trans-DCCA concentrations ranged from 0.094 to 0.120 mg/kg, which were higher than the concentrations of cis-DCCA (0.091–0.120 mg/kg) and 3-PBA (0.022–0.061 mg/kg). In the presence of oxytetracycline, beta-cypermethrin degradation was inhibited slightly, while the enantioselectivity was not affected. Oxytetracycline increased the enrichment and persistence of the metabolites. Addition of chicken manure decreased the cis-DCCA residue levels in the soil and alleviated the effect of oxytetracycline; however, chicken manure increased the accumulation and persistence of 3-PBA. In addition, oxytetracycline perturbed the structure of the soil microbial community. The abundance of Proteobacteria increased, while the abundances of Firmicutes and Actinobacteria decreased. These changes might affect the biodegradation of beta-cypermethrin and its metabolites. Combined pollution with antibiotics should be considered for its potential impact on pesticide residues.

Monitoring and biochemical characterization of beta-cypermethrin resistance in Spodoptera exigua (Lepidoptera: Noctuidae) in Sichuan Province, China

Xuegui Wang,Xing Xiang,Huiling Yu,Shuhua Liu,Yong Yin,Peng Cui,Yaqiong Wu,Jing Yang,Chunxian Jiang,Qunfang Yang

The beet armyworm Spodoptera exigua , a major pest affecting numerous cultivated crops in China, has developed a serious resistance to many traditional chemical insecticides . The resistance levels of the field-collected populations from different districts in Sichuan Province, China, to nine insecticides were detected with a diet-incorporation bioassay. Compared to the Lab-ZN strain, five (in 2014) and three (in 2016) field populations displayed either high or extremely high levels of resistance to beta-cypermethrin. All the field populations collected in 2014 were susceptible to emamectin benzoate, hexaflumuron, methoxyfenozide, chlorantraniliprole, cyantraniliprole and indoxacarb but exhibited low or moderate levels of resistance to abamectin . The resistances of field populations collected in 2016 were significantly higher than two years earlier, especial for chlorantraniliprole and cyantraniliprole with RRs rising from 173.4- to 582.6-fold and 175.3- to 287.6-fold, respectively, even though the field populations had retained moderate or low levels of resistance to chlorpyrifos and hexaflumuron. The synergism experiment revealed that the resistance of the LS16 population to beta-cypermethrin may be mainly related to cytochrome P450 monooxygenases (P450s), which was responsible for the highest increase ratio of 37.97-fold, for piperonyl butoxide , rather than either carboxylesterase (CarE) or glutathione S-transferase (GST). The cytochrome P450 ethoxycoumarin O -deethylase activity of the LS16 population was also the strongest among the treatments ( P  < 0.05). Non-denaturing polyacrylamide gel electrophoresis (native PAGE) indicated that enhanced E11, E13 and E15-E16 bands in the LS16 population likely contribute to the development of resistance to beta-cypermethrin.
