Colorimetric Measurement of Deltamethrin Pesticide Using a Paper Sensor Based on Aggregation of Gold Nanoparticles

Jingyang Zhu,Lifeng Yin,Weiyi Zhang,Meilian Chen,Dongsheng Feng,Yong Zhao,Yongheng Zhu

Deltamethrin (DEL) is one of the most commonly used pyrethroid pesticides that can cause serious harms to the ecological environment and human health. Herein, we have developed a paper-based colorimetric sensor impregnated with gold nanoparticles (AuNPs) for on-site determination of DEL pesticide. AuNPs show obvious color change on paper device with the presence of DEL. Measuring the gray intensity of the AuNPs on the reaction zone of the paper sensor allows accurate quantitative analysis. The detection mechanism of DEL on paper sensor was confirmed by UV-Vis spectrophotometry (UV-Vis), Fourier transform infrared spectroscopy (FT-IR), and transmission electron microscope (TEM). Under optimal conditions, the colorimetric sensor exhibited high sensitivity, rapid detection, and low detection limit within the values stipulated by Chinese detection standards (LOD = 0.584 mg/L). Besides, detecting DEL in vegetable and fruit samples also gave satisfying results, which were much consistent with those obtained by spectrophotometry. Overall, this work provided a user-friendly, cost-effective and visualized detection platform, which could be applied to rapidly detect DEL pesticides in the food safety field. Keywords:colorimetric signal;deltamethrin pesticide;gold nanoparticles;paper sensor;rapid detection

Enhanced Degradation of Deltamethrin in Water through Ferrous Ion Activated Sulfite: Efficiency and Mechanistic Insights

Ying Wan,Fangze Shang,Luming Yin,Hantao Wang,Yang Ping,Jiaqi Ding,Zongping Wang,Pengchao Xie

Deltamethrin’s global use as a potent insecticide against pests is well-established. However, the compound’s diverse levels of toxicity are increasingly under scrutiny, drawing significant attention to treatments of deltamethrin. Transition metal activation of sulfite is a promising technology for micropollutant degradation. In this study, iron-activated sulfite was used for the degradation of deltamethrin. The degradation effects and influencing factors and the underlying mechanism of deltamethrin degradation in the system were investigated. The degradation of deltamethrin was effectively achieved by the Fe (III)/sulfite system. The optimal reaction conditions at laboratory scale were determined to be an initial pH of 4, a Fe (III) concentration of 100 μM, and aHSO3−concentration of 1 mM, where the degradation rate was approximately 69.5%. Dissolved oxygen was identified as an essential factor in the reaction process, with the degradation rate of deltamethrin decreasing by up to 22% under anaerobic conditions. The presence of light facilitated the degradation of deltamethrin within the reaction system, while bicarbonate and natural organic compounds were found to inhibit its degradation. Quenching experiments verified the presence of hydroxyl radicals (HO•) and sulfate radicals (SO4•−) in the reaction system, with HO•being the predominant species. This was further confirmed by EPR experiments. Additionally, density functional theory calculations indicated the propensity for bond breaking between C16 and O21 in deltamethrin molecules, and the degradation pathway was validated through GC-MS analysis of the products formed. Moreover, the Fe (III)/sulfite system demonstrated good degradation performance for deltamethrin in secondary effluent, achieving degradation rates of 46.3%. In particular, the Fe (III)/sulfite system showed minimal bromate formation, attributed to the capacity of sulfite to reduce active bromine intermediates into bromine ions.

Conventional Anthelmintic Concentration of Deltamethrin Immersion Disorder in the Gill Immune Responses of Crucian Carp

Hao Wu,Xiping Yuan,Jinwei Gao,Min Xie,Xing Tian,Zhenzhen Xiong,Rui Song,Zhonggui Xie,Dongsheng Ou

Current treatment strategies for parasitic infectious diseases in crucian carp primarily rely on chemotherapy. As a commonly used antiparasitic agent, deltamethrin (DEL) may have the potential adverse effects on external mucosa of fish such as gills. In this study, 180 healthy juvenile crucian carp (Carassius auratus) (average weight: 8.8 ± 1.0 g) were randomly divided into three groups for 28 days, which were immersed in 0 μg/L, 0.3 μg/L, and 0.6 μg/L of DEL, respectively. The results of histological analysis revealed that severe hyperplasia in the secondary lamellae of gills was observed, and the number of goblet (mucus-secreting) cells increased significantly after DEL immersion. TUNEL staining indicated that the number of apoptotic cells increased in crucian carp gill. At the molecular level, the mRNA expression analysis revealed significant upregulation of apoptosis (caspase 3, caspase 8, and bax), autophagy (atg5 and beclin-1), and immune response (lzm, muc5, il-6, il-8, il-10, tnfα, ifnγ, tgfβ, tlr4, myd88, and nf-kb), whereas tight junction-related genes (occludinandclaudin12) were downregulated after DEL immersion, suggesting that DEL immersion altered innate immunity responses and promoted mucus secretion. Moreover, tandem mass tag (TMT)-based proteomics revealed that a total of 428 differentially expressed proteins (DEPs) contained 341 upregulated DEPs and 87 downregulated DEPs with function annotation were identified between the control and DEL groups. Functional analyses revealed that the DEPs were enriched in apoptotic process, phagosome, and lysosome pathways. Additionally, DEL immersion also drove gill microbiota to dysbiosis and an increase in potentially harmful bacteria such asFlavobacterium. Overall, this study showed that DEL elicited shifts in the immune response and changes in the surface microbiota of fish. These results provide new perspectives on the conventional anthelmintic concentration of DEL immersion disorder of the gill immune microenvironment in crucian carp and theoretical support for future optimization of their practical application. Keywords:deltamethrin;gill barrier;TMT-based proteomics;gill microbiota;cell apoptosis

Development of a recombinant Bacillus ligniniphilus L1 laccase-rice straw biochar composite for efficient deltamethrin bioremediation

Rahman Mujeeb ur,Shah Junaid Ali,Haroon,Gul Hansa,Nasreen Zahida,Shoaib Muhammad

This study reports the development of a recombinant Bacillus ligniniphilus  L1 laccase–rice straw biochar composite and its application in deltamethrin bioremediation. The L1-laccase was heterologously expressed in E. coli BL21 (DE3), purified to a specific activity of 186 U/mg, and covalently immobilized on NaOH- and glutaraldehyde-modified rice straw biochar. Molecular docking against the AlphaFold2-predicted structure (UniProt E9PZ36) yielded a binding affinity of − 6.9 kcal/mol, with one hydrogen bond to MET216 (2.77 Å) and hydrophobic contacts with MET197, MET202, and MET210. However, 100 ns molecular dynamics simulations confirmed a conformationally stable complex (RMSD ~0.42 nm, radius of gyration ~2.21 nm, SASA ~215 nm 2 ). Immobilization shifted the activity optima from pH 7.5 and 45 °C (free) to pH 8.0 and 50 °C (immobilized) and markedly improved stability: the immobilized enzyme retained 74% of its initial activity after 6 days at 4 °C and 18% after 8 catalytic cycles. The apparent Kₘ increased from 0.38 mM (free) to 0.54 mM (immobilized), with kcat values of 178 s⁻ 1 and 130.7 s⁻ 1 , respectively. Under an aggressive deltamethrin load (1000 mg/L), the immobilized laccase achieved 92.3% degradation within 18 h at 50 °C, compared with 68.7% for the free enzyme, producing two major phenolic metabolites (tR = 1.854 and 2.436 min) and one minor unidentified product. These results establish a cost-effective, reusable, and scalable biocatalyst for the bioremediation of pyrethroid-contaminated environments.

Metal–Organic Framework (UiO-66)-Based Pesticide Delivery System for Effective Deltamethrin Loading and Sustained Pesticide Release

Minghui Wan,Saijie Song,Wenli Feng,Xiaoyuan Wang,Jian Shen

A pesticide delivery system (PDS) is an effective strategy to overcome the low efficiency of conventional pesticide formulations. UiO-66, a metal–organic framework (MOF), is a suitable nanocarrier for pesticide delivery due to its large surface area, good stability, and nontoxicity. In this work, we developed deltamethrin-loaded UiO-66 (DM@UiO-66) as a PDS for sustained DM release and enhanced pest control. DM was loaded on UiO-66 via a physical adsorption method, forming a stable DM@UiO-66 nanoformulation (loading capacity 1049.6 mg/g). Unlike the burst release of free DM, DM@UiO-66 exhibits sustained pesticide release performance, which can maintain effective insecticidal concentration for a long time. Biological assay results indicate that DM@UiO-66 possesses better insecticidal activity than free DM, and the nanoformulation is safe for maize seeds and seedlings. In addition, DM@UiO-66 contains no toxic organic solvents or additives, which makes it environmentally friendly. Thus, the UiO-66-based PDS will hold a great future in agriculture and pest control fields.

Robust deltamethrin biodegradation by an alginate-chitosan encapsulated whole-cell biocatalyst with high p-NPA hydrolyzing activity

Hao Liu,Qianwei Li,Daoqing Liu,Chunmao Chen

Deltamethrin (DEL) is a highly potent and persistent pyrethroid insecticide, and its widespread accumulation poses significant ecological risks. While microbial biodegradation targeting ester bond cleavage offers a promising green remediation strategy, free bacterial cells often exhibit poor operational stability and are vulnerable to environmental stresses under realistic conditions. To address this challenge and develop robust biocatalysts, we initially screened for a strain possessing high esterolytic potential using a rapid p -nitrophenyl acetate (p-NPA) hydrolysis assay, identifying Stenotrophomonas sp. Klm2. To enhance its physiological resilience for practical application, whole Klm2 cells were encapsulated within a sodium alginate/chitosan (SA/CTS) dual-network hydrogel composite. Characterization revealed that structural synergy between the biopolymers improved network integrity, providing a protective microenvironment that significantly bolstered the biocatalyst's tolerance to thermal fluctuations, pH shifts, and chemical toxicity compared to free counterparts. The encapsulated Klm2@SA/CTS exhibited remarkable stability, retaining 90.2% of its initial catalytic activity after 7 days of storage at 25 °C and maintaining performance over 8 reuse cycles, with only a 25.9% total activity loss. Crucially, under high DEL loading conditions (200 mg/L), the encapsulated cells outperformed free cells by a wide margin. This enhanced performance was driven by a demonstrated synergistic mechanism coupling rapid physical adsorption onto the hydrogel matrix with sustained intracellular biochemical degradation. This study highlights how rationally engineering the biopolymer microenvironment modulates mass transfer and microbial stability, establishing a reusable, physiologically robust whole-cell platform for efficient pesticide detoxification.

Study on low concentration deltamethrin toxicity mediated by phospholipase D in Chinese mitten crab (Eriocheir sinensis) ovary

Qi Sun,Jun Dong,Zehui Su,Xuanyun Huang,Xiaoning Gao,Kun Hu,Xiaoling Gong

This study evaluates the impact of environmentally relevant, low-concentration deltamethrin exposure to Eriocheir sinensis ovaries. Our findings revealed that even at a concentration of 0.05 µg/L, deltamethrin exposure can induce significant ovarian toxicity through a 5-day exposure, with gradual amplification detected with time, demonstrating the toxicity amplification effect. Hematoxylin and Eosin staining revealed that low-concentration deltamethrin exposure produces pathological damage consistent with acute toxicity—yolk granules were dissolved and oocyte membranes were ruptured. High-throughput RNA-sequencing data indicated that the acute and low-concentration exposure groups involved completely different pathways and molecular functions, suggesting distinct mechanisms for their toxic effects. Following the identification of phospholipase D (PLD) as a potential core factor regulating the toxicity amplification effect of low concentration deltamethrin, we delved into subsequent mechanism studies using quantitative real-time PCR, immunofluorescence and enzyme-linked immunosorbent assay. Through the GnRH signaling pathway, increased PLD indirectly stimulates augmented estradiol secretion, subsequently inducing apoptosis by upregulating Cathepsin D, which can activate the key executioners of apoptosis—caspases (CASP3 and CASP7). In conclusion, low-concentration deltamethrin exposures can induce significant ovarian damage through apoptosis mediated by the upregulation of PLD in the ovaries of Eriocheir sinensis at environmentally relevant concentrations, which lays the preliminary theoretical groundwork for further elucidating the mechanism of toxicity amplification effect of pesticide exposure at low concentrations.

Bioaccessibility and Intestinal Transport of Deltamethrin in Pacific Oyster (Magallana Gigas) Using Simulated Digestion/NCM460 Cell Models

Jiao Yadan,Liu Chune,Feng Chunsong,Regenstein Joe M.,Luo Yongkang,Tan Yuqing,Hong Hui

Deltamethrin (DEL) can be introduced into the food chain through bioaccumulation in Pacific oysters, and then potentially threaten human health. The objective of this study was to investigate the bioaccessibility of DEL in oysters with different cooking methods after simulated digestion. DEL content in different tissues of oysters going from high to low were gills, mantle, viscera, and adductor muscle. Bioaccessibility of DEL in oysters decreased after steaming (65%) or roasting (51%) treatments compared with raw oysters (82%), which indicated that roasting can be used as a recommended cooking method for oysters. In the simulated digestion process, the concentration of DEL in the digestive juice and the bioaccessibility of DEL were affected by the pH in the gastric phase. And the transport efficiency of DEL through the monolayer molecular membrane of NCM460 cells ranged from 35 to 45%. These results can help assess the potential harm to consumers of DEL in shellfish. Furthermore, it provides a reference for the impact of lipophilic toxins in seafood.

Integration of metabolomics and transcriptomics reveals the toxicological mechanism of deltamethrin exposure in Chinese mitten crab Eriocheir sinensis

Zhiruo Fang,Yu Yao,Liping Cao,Jiancao Gao,Quanjie Li,Zhijuan Nie,Yi Sun,Gangchun Xu,Jinliang Du

This study investigated the toxicological mechanism of deltamethrin on Chinese mitten crab Eriocheir sinensis juveniles in fresh water. We first conducted an acute toxicity test, followed by laboratory methods to detect changes in immune-related indices in terms of antioxidant enzyme markers, lipid metabolism-related genes, and autophagy-related and apoptosis genes. The acute toxicity (96-h LC50) of deltamethrin to E. sinensis was 7.195 μg/L. After 48 h of exposure, serum showed elevated immune-related indices ( P  < 0.05) for alkaline phosphatase (AKP), acid phosphatase (ACP), aspartate aminotransferase (AST), alanine aminotransferase (ALT), complement components C3 and C4, and the key pro-inflammatory cytokines interleukin-6, interleukin-1β, and tumor necrosis factor alpha (TNF-α). In hepatopancreas at 48 h, indicators related to the antioxidant system, namely superoxide dismutase (SOD) and glutathione (GSH), were significantly elevated, whereas nitric oxide and total antioxidant capacity (T-AOC) were decreased ( P  < 0.05). In contrast, lipid metabolism indices for triglyceride (TG), total cholesterol (TC), and malondialdehyde (MDA) were increased ( P  < 0.05). Transcriptomics and metabolomics revealed that exposure to deltamethrin disrupted the lipid metabolic process in the hepatopancreas mainly by altering fatty acid synthesis, amino acid metabolism, immune signaling, and autophagy activation, while the exposure increased the content of phospholipids and cholesterol but decreased the levels of amino acids and palmitoleic acid. Quantitative genetics revealed significantly aberrantly expressed ( P  < 0.05) lipid metabolism-related genes, including acc1 , fasn , scd1 , and pnpla2 , all key genes involved in lipid accumulation. Deltamethrin exposure also significantly altered ( P  < 0.05) gene expression levels for Toll-like receptor ( tlr ), myeloid differentiation factor 88 ( myd88 ), crustin1 , anti-lipopolysaccharide factor isoform 3 ( alf3 ), tumor necrosis factor alpha ( tnf-α ), and NF-κB transcription factor relish . Furthermore, deltamethrin activated the toll-like receptor/major myeloid differentiation response gene 88/nuclear factor kappa-light-chain-enhancer of activated B cells (TLR/MyD88/NF-kB) signaling pathway, which activates a nonspecific immune response in E. sinensis . Additionally, carnitine palmitoyltransferase 1 A ( cpt1a ), cytochrome c ( cyt-c ), adenosine 5′-monophosphate (amp)-activated protein kinase ( ampk ), the autophagosomal protein microtubule-associated protein 1 light chain 3c ( lc3c ), and the autophagy-related proteins beclin1 , atg5 , atg12 were significantly induced ( P  < 0.05) in the adenosine monophosphate-activated protein kinase/rapamycin (AMPK/mTOR) signaling pathway. These changes resulted in excess free radicals, causing oxidative stress in the mitochondrial membrane, promoting mitochondrial autophagy. The results confirm that deltamethrin exposure can induce hepatopancreatic injury by promoting mitochondrial autophagy, activating an immune response, and inhibiting lipid metabolism. Overall, this study provides multi-level information to reveal the toxic effects of deltamethrin on E. sinensis .

An Investigation of the Intermolecular Interactions and Recognition Properties of Molecular Imprinted Polymers for Deltamethrin through Computational Strategies

Lei Xie,Nan Xiao,Lu Li,Xinan Xie,Yan Li

Deltamethrin (DM) is a toxic pesticide that is nonetheless widely used to control insect pests in agricultural production. Although the number of DM molecularly imprinted polymers (MIPs) is increasing in many scientific applications, the theoretical aspects of the participating intramolecular forces are not fully understood. This paper aims to explore the intermolecular interactions between the template molecule DM and the functional monomer acrylamide (AM) through density functional theory (DFT), analysis of hydrogen nuclear magnetic resonance (1H-NMR), Fourier transform infrared spectroscopy (FTIR), and adsorption thermodynamics. The results indicated that there is strong hydrogen bonding between O19 of DM and H9 of AM, suggesting that it is the preferable site for the binding of the target molecule. The existence of interaction sites was found to play an important role in the recognition process. The results from selective adsorption experiments showed that the DM MIPs exhibited the highest adsorption capacity for DM (Q = 75.72 mg g−1) as compared to the five structural analogs. Furthermore, the recovery rates of spiked DM from various teas using the DM MIPs as solid-phase extraction filler also possessed a high value (all greater than 83.68%), which enables them to be used as separate and recognition functional materials.

Ocean acidification aggravates the toxicity of deltamethrin in Haliotis discus hannai: Insights from immune response, histopathology and physiological responses

Xiaojing Lv,Qinyou Deng,Lizhu Chen,Xin Wang,Yijing Han,Guiqing Wu,Yongliang Liu,Haiyue Sun,Xuan Li,Jinxia He,Xiangquan Liu,Dinglong Yang,Jianmin Zhao

Ocean acidification (OA) and other environmental factors can collectively affect marine organisms. Deltamethrin (DM), a type II pyrethroid insecticide, has been widely detected in coastal and estuarine areas, while little attention has been given to the combined effects of DM and OA. In this study, Haliotis discus hannai was exposed to three pH levels (8.1, 7.7 and 7.4) and three DM nominal concentrations (0 μg/L, 0.6 μg/L and 6 μg/L) for 14 and 28 days. The results indicated that experimental acidification and/or DM exposure led to impaired immune function and pathological damage. Additionally, acidified conditions and DM exposure induced oxidative stress, and gills are more sensitive than digestive glands. With increasing p CO 2 and DM nominal concentrations, superoxide dismutase (SOD) activity decreased, whereas catalase (CAT) and glutathione S-transferase (GST) activities increased in the gills. Moreover, the expression levels of Toll-like receptor (TLR) pathway-related genes were upregulated after exposure. Integrated biomarker response (IBR) analysis proved that acidified conditions and/or DM detrimentally affected the overall fitness of H. discus hannai , and co-exposure to experimental acidification and DM was the most stressful condition. This study emphasizes the necessity of incorporating OA in future pollutant environmental assessments to better elucidate the risks of environmental disturbance.

Mitigative effects of Vitamin C on deltamethrin-induced acute toxicity in crucian carp by modulating the AhR/Nrf2 signaling pathways

Hao Wu,Jinwei Gao,Dong Liu,Xiangrong Liu,Feiming Chen,Jiayuan Qin,Yunfei Ye,Zhenzhen Xiong,Jiayu Wu,Can Yang,Min Xie,Rui Song,Dongsheng Ou

In recent years, deltamethrin (DEL) pollution poses a significant threat to aquaculture; however, the protective role of vitamin C (VC) against acute toxicity in crucian carp has not been well understood. This study provides compelling evidence that exposure to 2.0 mg/L waterborne VC significantly mitigates DEL-induced 96-hour mortality in crucian carp ( p < 0.001). Importantly, this research represents the first documentation of the protective effects of waterborne VC in fish, which is mediated through the modulation of the aryl hydrocarbon receptor (AhR)/nuclear factor erythroid 2-related factor 2 (Nrf2) signaling pathway—a mechanism not previously reported in crucian carp. The observed protective effect is attributed to the enhancement of antioxidant and anti-inflammatory responses via this signaling pathway. Exposure to DEL at 6.2 μg/L resulted in severe oxidative stress, characterized by significantly increased malondialdehyde (MDA) levels and depletion of catalase (CAT), glutathione (GSH), and superoxide dismutase (SOD) in the liver and intestine ( p < 0.01). Histopathological damage, including hepatic vacuolation, shortened villi, and inflammation, as well as elevated reactive oxygen species (ROS), was observed. Co-exposure to VC normalized antioxidant markers, reduced MDA and ROS levels, and alleviated tissue damage ( p < 0.05). Mechanistically, VC counteracted the suppression of AhR/Nrf2 by DEL, leading to the statistically significant upregulation of antioxidant genes ( gst, sod, cat ) and suppression of pro-inflammatory cytokines ( il1β, il6, tnfα ) ( p < 0.05). VC exhibited tissue-specific regulation by enhancing hepatic Nrf2 defenses and co-activating intestinal AhR/Nrf2, as confirmed by Western blot analysis. Our study shows that waterborne VC effectively protects against acute DEL toxicity under current experimental conditions, suggesting its potential as an eco-friendly emergency intervention in aquaculture during sudden pesticide exposure. Further research is needed to confirm its effectiveness in complex pond systems and various fish species.
