Detoxification of DON-induced hepatotoxicity in mice by cold atmospheric plasma

Ruonan Ma,Yongqin Fan,Xudong Yang,Chunli Liu,Junhu Wan,Cui Xu,Ruixue Wang,Junxia Feng,Zhen Jiao

Deoxynivalenol (DON) is one of the most common mycotoxins distributed in food and feed, which causes severe liver injury in humans and animals. Cold atmospheric plasma (CAP) has received much attention in mycotoxin degradation due to the advantages of easy operation, high efficiency, and low temperature. So far, the majority of studies have focused on the degradation efficiency and mechanism of CAP on DON, while there is still little information available on the hepatotoxicity of DON after CAP treatment. Herein, this study aimed to investigate the effect of CAP on DON-induced hepatotoxicity both in vitro and in vivo and its underlying mechanisms. The results showed that 120-s CAP treatment achieved 97 % degradation of DON. The vitro hepatotoxicity of DON in L02 cells was significantly reduced with CAP treatment time. Meanwhile, CAP markedly alleviated DON-induced liver injury in mice including the balloon-like degeneration of liver tissues and elevation of AST and ALP level. The underlying mechanism for CAP detoxification of DON-induced hepatotoxicity was further elucidated. The results showed that DON caused severe oxidative stress in cells by suppressing the antioxidant signaling pathway of Nrf2/HO-1/NQO-1, consequently leading to mitochondrial dysfunction and cell apoptosis, accompanied by cellular senescence and inflammation. CAP blocked DON inhibition on the Nrf2/HO-1/NQO-1 signaling pathway through the efficient degradation of DON, accordingly alleviating the oxidative stress and liver injury induced by DON. Therefore, CAP is an effective method to eliminate DON hepatotoxicity, which can be applied in the detoxification of mycotoxin-contaminated food and feed to ensure human and animal health.

Detoxification of Cr(VI) using biochar-supported Cu/Fe bimetallic nanoparticles

Fengli Shao,Shaoyu Zhou,Jiaxin Xu,Qiong Du,Jianqiu Chen,Jingge Shang

Nanoscale zero-valent iron (nZVI) has been proven to be more effective for the treatment of hexavalent chromium(Cr(VI)) when modified with the transition metal Cu. In this study, biochar-supported Cu/Fe bimetallic nanoparticles (Cu-nZVI/BC) were prepared and used for Cr(VI) removal from aqueous solution. The effects of the pH value, the initial concentration of Cr(VI), coexisting ions and humic acids (HA) were investigated. The compositional structures of Cu-nZVI/BC were analyzed by transmission electron microscopy, scanning electron microscopy, X-ray diffraction and X-ray photoelectron spectroscopy. The maximum removal efficiency of Cr(VI) was 50.57% (126.41 ± 3.75 mg g –1 ) for Cu-nZVI/BC at pH 2. The adsorption kinetic data were in agreement with the pseudo-second- order model, which confirmed that the adsorption step was a chemical adsorption and reduction process. The presence of Ca 2+ and NO 3 – had an inhibitory effect on Cr(VI) removal, while dHA increased the removal efficiency at high concentrations. This study proves that Cu-nZVI/BC removal of solution Cr(VI) is feasible and shows significant potential in practical applications.

Sustainable Enzyme-Assisted Coagulation for Decolorization and Detoxification of Acid Dye

Na-won Baek,Shin-hee Lee

This study proposes an enzyme-assisted coagulation (EAC) system as a novel, nature-inspired, energy-free approach for the treatment of acid dyes in textile wastewater. The EAC system combines enzymatic oxidation with natural polymer-based coagulation, proceeding under mild conditions without the need for external energy input. In this study, horseradish peroxidase (HRP) catalyzed the oxidative breakdown of the dye, while chitosan served as a representative biopolymer coagulant, facilitating the aggregation of degradation products through electrostatic and hydrogen-bonding interactions. Analytical techniques, including UV–vis spectroscopy, FTIR, HPLC, and GC-MS, confirmed the effective degradation and coagulation of dye byproducts. Furthermore, acute toxicity assays using Daphnia magna demonstrated that the EAC system substantially reduced ecotoxicity compared with enzymatic or coagulation treatments alone. This flexible and sustainable platform is applicable to a wide range of acid dyes, offering a scalable, low-carbon alternative to conventional dye wastewater treatment methods.

The Mineralization and Detoxification of PFOA Using a Sn/ZnO2@Ti4O7Anode

Yang Zehong,Wang Zihao,Qiu Qiuhong,Luo Yuzhan,Wang Chenglong,Huang Yaobin,Zhuang Tao,Zheng Weijin,Chen Liang,Lan Liying,Liu Guangli,Fang Jiasheng,Li Yanliang,Lv Sihao,Wang Wenlong,Qiu Yongfu,Zhuo Qiongfang

A Sn/ZnO 2 @Ti 4 O 7 anode was prepared using a combination of hydrothermal synthesis and discharge plasma sintering methods for the electrochemical degradation of perfluorooctanoic acid (PFOA). The introduction of Sn/ZnO 2 significantly increased the electrochemical active surface area (ECSA) and the oxygen evolution potential (OEP) of Ti 4 O 7 . A 97.6% degradation rate of PFOA can be achieved using a Sn/ZnO 2 @Ti 4 O 7 anode, at a current density of 40 mA/cm 2 , with the initial PFOA concentration of 10 mg/L, at a pH value of 5.69, and within 120 min. Radical quenching experiments indicated that direct oxidation was the main mechanism for PFOA degradation, contributing 82.5%. The degradation efficiencies of PFOA remained above 88% in Dongjiang and Zhujiang rivers (DJR and ZJR) matrixes, confirming the system’s resistance to complex matrix. The total organic carbon (TOC) removal ratio was 56.8%, and the defluorination ratio was 48% after electrolysis of 6 h. After 36 cycles of degradation, the degradation rate was 95.6%. The toxicity assessments indicated that the degradation intermediates were significantly less toxic than the parent PFOA. Graphical

Highly efficient detoxification of dinitrotoluene by transgenic switchgrass overexpressing bacterial nitroreductase

Kunlong Su,Zhenying Wu,Yuchen Liu,Shanshan Jiang,Dongmei Ma,Yan Wang,Chunxiang Fu

Dinitrotoluene (DNT) has been extensively used in manufacturing munitions, polyurethane foams and other important chemical products. However, it is highly toxic and mutagenic to most organisms. Here, we synthesized a codon-optimized bacterial nitroreductase gene, NfsI, for plant expression. The kinetic analysis indicates that the recombinant NfsI can detoxify both 2,4-DNT and its sulfonate (DNTS), while it has a 97.6-fold higher catalytic efficiency for 2,4-DNT than DNTS. Furthermore, we overexpressed NfsI in switchgrass ( Panicum virgatum L.), which is a multiple-purpose crop used for fodder and biofuel production as well as phytoremediation. The 2,4-DNT treatment inhibited root elongation of wild-type switchgrass plants and promoted reactive oxygen species (ROS) accumulation in roots. In contrast, overexpression of NfsI in switchgrass significantly alleviated 2,4-DNT-induced root growth inhibition and ROS overproduction. Thus, the NfsI overexpressing transgenic switchgrass plant removed 94.1% 2,4-DNT after 6 days, whose efficiency was 1.7-fold higher than control plants. Moreover, the comparative transcriptome analysis suggests that 22.9% of differentially expressed genes induced by 2,4-DNT may participate in NfsI-mediated 2,4-DNT detoxification in switchgrass. Our work sheds light on the function of NfsI during DNT phytoremediation for the first time, revealing the application potential of switchgrass plants engineered with NfsI .

Enhanced sulfamethazine detoxification by a novel BiOCl (110)/NrGO/BiVO4 heterojunction

Xiaofan Yang,Yutao Xu,Saraschandra Naraginti,Xueyu Wei

The emerging contaminants removal from the environment has recently been raised concerns due to their presence in higher concentrations. Over usage of emerging contaminant such as sulfamethazine poses serious threat to the aquatic and human health as well. This study deals with rationally structured a novel BiOCl (110)/NrGO/BiVO 4 heterojunction which is used to detoxify sulfamethazine (SMZ) antibiotic efficiently. The synthesised composite was well characterized and the morphological analysis evidenced the formation of heterojunction consisted of nanoplates BiOCl with dominant exposed (110) facets and leaf like BiVO 4 on NrGO layers. Further results revealed that the addition of BiVO 4 and NrGO tremendously increased the photocatalytic degradation efficiency of BiOCl with the rate of 96.9% (k = 0.01783 min −1 ) towards SMZ within 60 min of visible light irradiation. Furthermore, heterojunction energy-band theory was employed to determine the degradation mechanism of SMX in this study. The larger surface area of BiOCl and NrGO layers are believed to be the reason for higher activity which facilitates the excellent charge transfer and improved light absorption . In addition, SMZ degradation products identification was carried out by LC-ESI/MS/MS to determine the pathway of degradation. The toxicity assessment was studied using E. coli as a model microorganism through colony forming unit assay (CFU), and the results indicated a significant reduction in biotoxicity was observed in 60 min of degradation process. Thus, our work gives new methods in developing various materials that effectively treat emerging contaminants from the aqueous environment.

Efficient Electrocatalytic Hydrodechlorination and Detoxification of Chlorophenols by Palladium–Palladium Oxide Heterostructure

Yu Wang,Qian-Yuan Wu,Min-Yong Lee,Yu-Jia Nong,Wen-Long Wang,Jörg E. Drewes

Electrocatalytic hydrodechlorination is a promising approach for simultaneous pollutant purification and valorization. However, the lack of electrocatalysts with high catalytic activity and selectivity limits its application. Here, we propose a palladium–palladium oxide (Pd-PdO) heterostructure for efficient electrocatalytic hydrodechlorination of recalcitrant chlorophenols and selective formation of phenol with superior Pd-mass activity (1.35 min–1 mgPd–1), which is 4.4 times of commercial Pd/C and about 10–100 times of reported Pd-based catalysts. The Pd-PdO heterostructure is stable in real water matrices and achieves selective phenol recovery (>99%) from the chlorophenol mixture and efficient detoxification along chlorophenol removal. Experimental results and computational modeling reveal that the adsorption/desorption behaviors of zerovalent Pd and PdO sites in the Pd-PdO heterostructure are optimized and a synergy is realized to promote atomic hydrogen (H*) generation, transfer, and utilization: H* is efficiently generated at zerovalent Pd sites, transferred to PdO sites, and eventually consumed in the dechlorination reaction at PdO sites. This work provides a promising strategy to realize the synergy of Pd with different valence states in the metal–metal oxide heterostructure for simultaneous decontamination, detoxification, and resource recovery from halogenated organic pollutants.

Elimination kinetics and detoxification mechanisms of microcystin-LR during UV/Chlorine process

Xinran Zhang,Jun He,Shuqi Xiao,Xin Yang

Microcystin-LR (MC-LR), a toxin produced by cyanobacteria, is very toxic and poses a threat to public health when entering water treatment works. In this study, UV/chlorine process, as an advanced oxidation process (AOP), has been demonstrated for effective elimination of MC-LR levels and associated toxicity. At a chlorine dose of 3.0 mg L −1 and UV fluence of 125 mJ cm −2 , MC-LR (initial concentration 1.0 μM) was reduced by 92.5%, which was much higher than 20.3% removal under UV irradiation alone and 65.1% removal during dark chlorination. Enhanced degradation was attributed by hydroxyl radicals (HO ) and reactive chlorine species (RCS), mainly Cl 2 - and ClO . Increasing chlorine doses or lowering pH favored MC-LR removal. Increased bicarbonate and natural organic matter concentrations inhibited MC-LR removal, but bromide ions enhanced MC-LR removal instead. MC-LR elimination rates in natural waters were roughly two times smaller than those in ultrapure water. The reactive radicals promoted hydroxylation of both diene of Adda moiety and double bond of Mdha moiety in MC-LR. UV exposure enhanced the dechlorination of chloro-MC-LR via the cleavage of C Cl bond. The toxicity was evaluated by a protein phosphatase (PP2A) inhibition assay. At a chlorine dose of 3.0 mg L −1 and UV fluence of 125 mJ cm −2 , the toxicity of the treated water was reduced by 75.0%, which was also higher than 25.7% and 46.7% removal under UV irradiation alone and during dark chlorination, respectively. These results highlight UV/chlorine is an efficient AOP for MC-LR degradation and detoxification.

Role of multiple detoxification enzyme genes in Nitenpyram resistance of Nilaparvata lugens

Liyi Liu,Xiongwei Zhang,Xueya Zheng,Dan Zhang,Xiangying Zhao,Yuxuan Chen,Shenglan Wen,Shuxiang Wang,Xiaolei Zhang,Junkai Li

The brown planthopper (BPH), Nilaparvata lugens , is a major insect pest of rice. Nitenpyram, a neonicotinoid insecticide, is a key compound for its control. Understanding the underlying resistance mechanisms is critical for developing effective management strategies. In this study, a nitenpyram-resistant (NR) N. lugens strain was established and characterized. Synergism tests and detoxification enzyme assays collectively demonstrate that enhanced metabolic detoxification plays a pivotal role in nitenpyram resistance in N. lugens . Quantitative RT-PCR (qRT-PCR) analysis revealed the significant overexpression of multiple candidate genes in the NR strain, including an ABC transporter gene ( ABCG1 ), four P450 genes ( CYP6ER1 , CYP3115A1 , CYP417A2 , and CYP418A1 ), four CarE genes ( CarE2 , CarE7 , CarE15 , and CarE26 ), and a UGT gene ( UGT 2B9 ). Additionally, molecular docking and molecular dynamics simulations further corroborated the high binding affinity and structural stability of these enzymes (ABCG1, CarE7, CarE15, CYP6ER1, CYP3115A1, CYP417A2, CYP418A1) and nitenpyram. Functional validation via RNA interference (RNAi) showed that silencing of ABCG1 , CarE15 , CYP3115A1 , CYP418A1 , and CYP6ER1 significantly increased nitenpyram susceptibility, confirming their functional role in resistance. Collectively, these findings elucidate a metabolic resistance mechanism mediated by multiple detoxification enzyme genes upregulation and provide a scientific basis for optimizing resistance management strategies to prolong the efficacy of nitenpyram in rice ecosystems.

Acetylacetone alleviates tetracycline stress in Chlorella vulgaris: Detoxification mechanisms and adaptive response

Jiamin Xu,Shuyue Wang,Peng Peng,Shujuan Zhang

Microalgae-based system represents a promising and eco-friendly solution for mitigating antibiotic pollution. However, their efficacy is often compromised by antibiotics stress. Here, we demonstrate that acetylacetone (AA), a natural small molecule with potential to regulate microalgal growth and metabolism, significantly alleviates tetracycline (TC) toxicity in Chlorella vulgaris , enhancing TC removal in aquaculture wastewater from 1 mg L -1 to 0.01 mg L -1 . The AA-treated effluent showed no significant developmental toxicity to wheat and negligible antibiotic resistance gene propagation. Proteomic and metabolomic analyses revealed that AA restores cellular homeostasis in microalgae by counteracting TC-induced oxidative stress and metabolic dysregulation, thereby sustaining removal performance under stressful conditions. This stress-regulation mechanism enables robust microalgae-based antibiotic degradation while preserving aquatic ecological safety, providing a sustainable strategy for antibiotic-containing wastewater treatment.

Pipeline electrode assembly reactor for in-transit electro-detoxification of medical wastewater

Hailong Tong,Baiming Sun,Shuzhao Pei,Yan Zhang,Nanqi Ren,Yuan Yao,Shijie You

High-risk toxic medical wastewater containing pharmaceutical residues and pathogens is highly toxic to aquatic ecosystems and public health. Herein, we developed a pipeline electrode assembly reactor (PEAR) by embedding a tubular concentric membrane electrode (TCME) into stainless steel pipeline for in-transit electro-detoxification of medical wastewater. The TCME configuration with synergistic alignment of electric field, flow field, and concentration field enhanced mass transfer by providing a uniform and enlarged electroactive interface. Based on a porous titanium suboxide (TiSO) anode and stainless-steel pipeline cathode, simulations and experiments revealed that forced convection reduced the viscous boundary layer by 99.9% to increase the mass transfer coefficient by 5.3–18.3 times (up to 4.8 × 10 −5  m s −1 ) and active site density by 42.3% at flow rate of 150 mL min −1 . The intensified interfacial transport facilitated coupled direct electron transfer and radical-mediated oxidation, which achieved efficient degradation of carbamazepine (CBZ; k  = 0.19 min −1 ) and 5.9-log inactivation of Bacillus anthracis endospores through coat disruption and intracellular oxidation. The PEAR could remove ≥95% COD with low energy consumption (0.4–1.8 kWh m −3 ) over 30 days of continuous operation, demonstrating excellent stability, scalability and energy efficiency. This study provides an integratable, scalable and chemical-free strategy for in-transit electro-detoxification of high-risk toxic medical wastewater.

Biodegradation and detoxification of chlorimuron-ethyl by Enterobacter ludwigii sp. CE-1

Xiong Pan,Saige Wang,Nan Shi,Hua Fang,Yunlong Yu

The application of the herbicide chlorimuron-ethyl has a lasting toxic effect on some succession crops. Here, a bacterium capable of utilizing chlorimuron-ethyl as the sole source of nitrogen was isolated from the contaminated soil and was identified as Enterobacter ludwigii sp. CE-1, and its detoxification and degradation of the herbicide were then examined. The biodegradation of chlorimuron-ethyl by the isolate CE-1 was significantly accelerated with increasing concentration (1–10   mg/l) and temperature (20–40   °C). The optimal pH for the degradation of chlorimuron-ethyl by the isolate CE-1 was pH 7.0. A pathway for the biodegradation of chlorimuron-ethyl by the isolate CE-1 was proposed, in which it could be first converted into 2-amino-4-chloro-6-methoxypyrimidine and an intermediate product by the cleavage of the sulfonylurea bridge and then transformed into saccharin via hydrolysis and amidation. The plant height and fresh weight of corn that had been incubated in nutrient solution containing 0.2   mg/l of chlorimuron-ethyl significantly recovered to 83.9% and 83.1% compared with those in the uninoculated control, although the root growth inhibition of chlorimuron-ethyl could not be alleviated after inoculation for 14 d. The results indicate that the isolate CE-1 is a promising bacterial resource for the biodegradation and detoxification of chlorimuron-ethyl.
