Pulmonary bacterial infections are a common disease, with erythromycin being a first-line treatment. However, conventional erythromycin tablets and injectables exhibit limited therapeutic efficacy and are associated with severe gastrointestinal side effects. Additionally, injectable formulations suffer from poor patient compliance. Dry powder inhaler (DPI) offers superior pulmonary targeting, circumvent first-pass metabolism, and are portable, enhancing patient adherence. In this study, we successfully developed a ternary inhalable erythromycin dry powder formulation (Ery-DPI). The Quality by Design (QbD) approach was employed to establish a design space for erythromycin micronization and to optimize the formulation. The physicochemical properties and fine particle fraction of the erythromycin DPI were evaluated through in vitro experiments. The final formulation, composed of α-lactose, mannitol, and erythromycin in a ratio of 1:0.2:2 (v/v/v), exhibited an FPF of 70 ± 3%. In vivo studies demonstrated that, compared to intravenous administration, Ery-DPI achieved higher and more stable local drug exposure in the lungs. Overall, our study provides new insights into pulmonary drug delivery strategies for treating bacterial infections. Graphical

Erythromycin (ERY) is one of the most common macrolides applied in veterinary medicine to treat diseases or as a feed additive for animal growth promotion. Long-term irrational use of ERY could lead to residues in animal-derived food and the emergence of drug-resistant strains, posing potential threats to human health. In this study, a highly sensitive, specific, robust, and rapid fluorescence polarization immunoassay (FPIA) for the determination of ERY in milk has been described. Herein, to achieve high sensitivity, five tracers of ERY with different fluorescein structures were synthesized and paired with three monoclonal antibodies (mAbs). Under the optimized conditions, the combination of mAb 5B2 and tracer ERM-FITC achieved the lowest IC50value in the FPIA with 7.39 μg/L for ERM. The established FPIA was used to detect ERY in milk, revealing a limit of detection (LOD) of 14.08 μg/L with recoveries of 96.08–107.77% and coefficients of variations (CVs) of 3.41–10.97%. The total detection time of the developed FPIA was less than 5 min from the addition of samples to the result readout. All the above results showed that the proposed FPIA in this study was a rapid, accurate, and simple method for the screening of ERY in milk samples. Keywords:erythromycin;fluorescence polarization immunoassay;homogeneous detection;milk

The exposure of aquatic organisms to pollutants often occurs concomitantly with salinity fluctuations. Here, we reported the effects of erythromycin (0.250, 7.21, and 1030 μg/L) on marine invertebrate N. succinea and its intestinal microbiome under varying salinity levels (5‰, 15‰, and 30‰). The salinity elicited significant effects on the growth and intestinal microbiome of N. succinea. The susceptibility of the intestinal microbiome to erythromycin increased by 8.7- and 6.2-fold at salinities of 15‰ and 30‰, respectively, compared with that at 5‰ salinity. Erythromycin caused oxidative stress and histological changes in N. succinea intestines, and inhibited N. succinea growth in a concentration-dependent manner under 30‰ salinity with a maximum inhibition of 25%. At the intestinal microbial level, erythromycin enhanced the total cell counts at 5‰ salinity but reduced them at 15‰ salinity. Under all tested salinities, erythromycin diminished the antibiotic susceptibility of the intestinal microbiome. Two-way ANOVA revealed significant interactive effects (p < 0.05) between salinity and erythromycin on various parameters, including antibiotic susceptibility and intestinal microbial diversity. The present findings demonstrated the significant role of salinity in modulating the impacts of erythromycin, emphasizing the necessity to incorporate salinity fluctuations into environmental risk assessments.

In this work, eggshell particles, an abundant source of waste and a natural inorganic–organic composite material, were found to be well suited to stabilize O/W Pickering emulsions, because the small amount of organic components in eggshell results in a near 90 degree contact angle θ (81.03°). Erythromycin-based molecularly imprinted polymers (EM-MIPs) whose surfaces were covered in eggshell particles were successfully synthesized through developing an eggshell-stabilized Pickering emulsion methodology, and interestingly and importantly in this work, the discarded eggshell particles were employed solely to stabilize an oil-in-water emulsion for the first time. The imprinting process was conducted through radical polymerization between the functional and polymeric monomers in the presence of template molecules (EM) and initiators. Various analytical approaches including Fourier transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM), thermogravimetric analysis (TGA) and specific surface and pore size analysis were used to characterize the synthesized EM-MIPs, and the results sufficiently demonstrated the formation of approximately spherical and wrinkled imprinted polymer which showed thermal stability (especially below 250 °C) and a specific surface area of 81.3 m2 g−1. Attributed to the formation of the eggshell-stabilized Pickering emulsion structure, the as-prepared EM-MIPs not only exhibited outstanding adsorption capacities, but also showed an excellent selective adsorption effect toward EM molecules. The Langmuir isotherm model and pseudo-second-order kinetic model also strongly suggested that the chemical process was the rate-limiting step in the adsorption of EM. Moreover, we envision that the EM-MIPs and the synthesis strategy employed in this work could be utilized in a broad range of applications.

As emerging pollutants, antibiotics were widely detected in water bodies and dietary sources. Recently, their obesogenic effects raised serious concerns. So far, it remained unclear whether their obesogenic effects would be influenced by water- and diet-borne exposure routes. In present study, Caenorhabditis elegans , nematodes free-living in air–water interface and feeding on bacteria, were exposed to water- and diet-borne erythromycin antibiotic (ERY). The statuses of the bacterial food, inactivated or alive, were also considered to explore their influences on the effects. Results showed that both water- and diet-borne ERY significantly stimulated body width and triglyceride contents. Moreover, diet-borne ERY’s stimulation on the triglyceride levels was greater with alive bacteria than with inactivated bacteria. Biochemical analysis showed that water-borne ERY inhibited the activities of enzymes like adipose triglyceride lipase (ATGL) in fatty acid β-oxidation. Meanwhile, diet-borne ERY inhibited the activities of acyl-CoA synthetase (ACS) and carnitine palmitoyl transferase (CPT) in lipolysis, while it stimulated the activities of fatty acid synthase (FAS) in lipogenesis. Gene expression analysis demonstrated that water-borne ERY with alive bacteria significantly upregulated the expressions of daf-2 , daf-16 and nhr-49 , without significant influences in other settings. Further investigation demonstrated that ERY interfered with bacterial colonization in the intestine and the permeability of the intestinal barrier. Moreover, ERY decreased total long-chained fatty acids (LCFAs) in bacteria and nematodes, while it decreased total short-chained fatty acids (SCFAs) in bacteria but increased them in nematodes. Collectively, the present study demonstrated the differences between water- and diet-borne ERY’s obesogenic effects, and highlighted the involvement of insulin and nhr-49 signaling pathways, SCFAs metabolism and also the interaction between intestinal bacteria and the host.

Magnetic imprinted polymers (MIPs) were synthesized by Pickering emulsion polymerization and used to adsorb erythromycin (ERY) from aqueous solution. The oil-in-water Pickering emulsion was stabilized by chitosan nanoparticles with hydrophobic Fe 3 O 4 nanoparticles as magnetic carrier. The imprinting system was fabricated by radical polymerization with functional and crosslinked monomer in the oil phase. Batches of static and dynamic adsorption experiments were conducted to analyze the adsorption performance on ERY. Isotherm data of MIPs well fitted the Freundlich model (from 15 °C to 35 °C), which indicated heterogeneous adsorption for ERY. The ERY adsorption capacity of MIPs was about 52.32 μmol/g at 15 °C. The adsorption kinetics was well described by the pseudo-first-order model, which suggested that physical interactions were primarily responsible for ERY adsorption. The Thomas model used in the fixed-bed adsorption design provided a better fit to the experimental data. Meanwhile, ERY exhibited higher affinity during adsorption on the MIPs compared with the adsorption capacity of azithromycin and chloramphenicol. The MIPs also exhibited excellent regeneration capacity with only about 5.04% adsorption efficiency loss in at least three repeated adsorption–desorption cycles.

Erythromycin (EM) was encapsulated into poly(methyl methacrylate) (PMMA) and poly(methyl methacrylate- co - N -vinylcaprolactam) (P(MMA-NVCL)) microspheres in situ by oil-in-water emulsion photopolymerization under green LED irradiation. The physicochemical properties of PMMA-EM and P(MMA-NVCL)-EM microspheres were characterized by SEM, FTIR, TG-DSC and DLS. P(MMA-NVCL)-EM microspheres showed temperature sensitivity, and the EM loading content of PMMA-EM and P(MMA-NVCL)-EM microspheres reached 9.7% and 15.6%, respectively. In vitro release of EM from microspheres presented good controlled release behavior in 12 h. Further release kinetic and thermodynamic investigation revealed that the release process was mainly dominated by EM concentration, accompanied by microspheres relaxation and rupture of hydrogen bonds .

This study reports the synthesis of green-emissive carbon dots ( λ ex = 399 nm, λ em,max = 563 nm; G-CDs) using bromothymol blue and dimethylglyoxime as precursors via a solvothermal method. The as-prepared G-CDs were characterized by fluorescence spectrometry, Fourier-transform infrared spectroscopy (FTIR), and X-ray photoelectron spectroscopy (XPS). The synthesized G-CDs exhibited an excitation peak at 399 nm, an emission peak at 563 nm, and a quantum yield of 22.3 %. They demonstrated excellent salt tolerance, photostability, and excitation wavelength-independent emission behavior, indicating single-emission characteristics. Furthermore, the G-CDs showed high selectivity and sensitivity for erythromycin detection and temperature monitoring, yielding satisfactory results in both applications. Thus, G-CDs can serve as a fluorescent probe for simultaneous erythromycin detection and temperature sensing, expanding their potential in environmental fields.

In this work, using discarded white pomelo peel as the green precursor and hydrochloric acid (HCl) as the dopant, water-soluble Cl-N-doped carbon dots (Cl-NCDs) with excellent performance were synthesized by one-step hydrothermal method. The Cl-NCDs function as both luminescent agents and catalysts, significantly enhancing the chemiluminescence intensity of the HCl-HCHO-KMnO 4 system, forming a novel HCl-HCHO-(Cl-NCDs)-KMnO 4 chemiluminescence system. The addition of erythromycin (EM) solution inhibits the chemiluminescence intensity of new system. By optimizing the conditions, we established a simple, cost-effective, and reliable chemiluminescence method for EM detection, offering a linear range of 5-100 μM and a detection limit of 0.0149 μM. In addition, this method was successfully applied to the analysis of EM in various water simples, achieving recovery rates ranging from 97.6% to 106%.

Herein, we have manufactured a newly designed bifunctional impedimetric and amperometric immunosensor for rapidly detecting erythromycin (ERY) in complicated environments and food stuffs. For this, bimetallic cobalt/cerium-layered double hydroxide nanosheets (CoCe-LDH NSs), which was derived from Co-based zeolite imidazole framework via the structure conversion, was simultaneously utilized as the bioplatform for anchoring the ERY-targeted antibody and for modifying the gold and screen printed electrode. Basic characterizations revealed that CoCe-LDH NSs was composed of mixed metal valences, enrich redox, and abundant oxygen vacancies, facilitating the adhesion on the electrode, the antibody adsorption, and the electron transfers. The manufactured impedimetric and amperometric immunosensor based on CoCe-LDH has showed the comparable sensing performance, having a wide linear detection range from 1.0 fg mL −1 to 1.0 ng mL −1 with the ultralow detection limit toward ERY. Also, the portable, visualized, and efficient analysis of ERY was then attained at the smartphone-assisted CoCe-LDH-based SPE.

Bacterial keratitis is a serious ocular infection that can impair vision or even cause blindness. The clinical use of antibiotics is limited due to their low bioavailability and drug resistance. Hence, there is a need to develop a novel drug delivery system for this infectious disease. In this study, erythromycin (EM) was encapsulated into a bifunctional polyhedral oligomeric silsesquioxane (BPOSS) with the backbone of the poly-PEG/PPG urethane (BPEP) hydrogel with the aim of improving the drug efficiency in treating bacterial keratitis. A comprehensive characterization of the BPEP hydrogel was performed, and its biocompatibility was assessed. Furthermore, we carried out the evaluation of the antimicrobial effect of the BPEP-EM hydrogel in S. aureus keratitis using in vivo mouse model. The BPEP hydrogel exhibited self-assembling and thermogelling properties, which assisted the drug loading of drug EM and improved its water solubility. Furthermore, the BPEP hydrogel could effectively bind with mucin on the ocular surface, thereby markedly prolonging the ocular residence time of EM. In vivo testing confirmed that the BPEP-EM hydrogel exerted a potent therapeutic action in the mouse model of bacterial keratitis. In addition, the hydrogel also exhibited an excellent biocompatibility. Our findings demonstrate that the BPEP-EM hydrogel showed a superior therapeutic effect in bacterial keratitis and demonstrated its potential as an ophthalmic formulation.

Bacteria of the genus Saccharopolyspora produce important polyketide antibiotics, including erythromycin A (Sac. erythraea) and spinosad (Sac. spinosa). We herein report the development of an industrial erythromycin-producing strain, Sac. erythraea HOE107, into a host for the heterologous expression of polyketide biosynthetic gene clusters (BGCs) from other Saccharopolyspora species and related actinomycetes. To facilitate the integration of natural product BGCs and auxiliary genes beneficial for the production of natural products, the erythromycin polyketide synthase (ery) genes were replaced with two bacterial attB genomic integration sites associated with bacteriophages ϕC31 and ϕBT1. We also established a highly efficient conjugation protocol for the introduction of large bacterial artificial chromosome (BAC) clones into Sac. erythraea strains. Based on this optimized protocol, an arrayed BAC library was effectively transferred into Sac. erythraea. The large spinosad gene cluster from Sac. spinosa and the actinorhodin gene cluster from Streptomyces coelicolor were successfully expressed in the ery deletion mutant. Deletion of the endogenous giant polyketide synthase genes pkeA1-pkeA4, the product of which is not known, and the flaviolin gene cluster (rpp) from the bacterium increased the heterologous production of spinosad and actinorhodin. Furthermore, integration of pJTU6728 carrying additional beneficial genes dramatically improved the yield of actinorhodin in the engineered Sac. erythraea strains. Our study demonstrated that the engineered Sac. erythraea strains SLQ185, LJ161, and LJ162 are good hosts for the expression of heterologous antibiotics and should aid in expression-based genome-mining approaches for the discovery of new and cryptic antibiotics from Streptomyces and rare actinomycetes.
