This study evaluated the effects of esculetin on heat stress-induced splenic immunosuppression in broilers. Esculetin significantly reduced rectal temperature and respiratory rate in heat-stressed broilers ( P < 0.05), improved growth performance, serum biochemical parameters, and the spleen index, and attenuated heat stress-induced splenic histopathological damage and HSP70 expression. Concurrently, esculetin elevated serum levels of immunoglobulins (IgA, IgM, and IgG) and upregulated the gene expression of anti-inflammatory cytokines, while downregulating pro-inflammatory cytokine genes (IL-1β, IL-6, and TNF-α). Network pharmacology analysis, molecular docking, and in vivo validation demonstrated that the immunomodulatory effects of esculetin were primarily mediated through targeting NF-κB, TNF-α, and IL-6, thereby modulating the NF-κB/MAPK signaling pathway. These findings provide a scientific basis for the application of esculetin as a functional feed additive to alleviate heat stress-induced immune dysfunction in poultry.

A fully printed high-performance nanomaterial-modified electrochemical sensor. • Sensor array enables co-detection of esculetin and quercetin in Chinese herbs. • EC combined with SPE enables highly sensitive assaying target in complex samples. • MXene/MWCNTs decorated SPSA exhibits low resistance and excellent activity. Viola tianschanica Maxim is a crucial medicinal herb in traditional Chinese medicine. However, current rapid quality evaluation methods for Chinese medicine herbs face significant limitations, thereby hindering the industrial development of cultivation and processing. Here, a fully screen-printed sensor array (SPSA) decorated with Ti 3 C 2 T x /MWCNTs was prepared and applied for the determination of key bioactive compounds (esculetin and quercetin) listed in the Chinese Pharmacopoeia. Ti₃C₂Tₓ and MWCNTs were directly formulated into a printable ink, enabling a single-step fabrication of the nanomaterial-modified sensor interface via screen-printing. This approach largely avoids the cumbersome multi-step modification procedures and the resulting batch-to-batch performance variations inherent in the conventional fabrication of electrochemical sensors. The composite exhibits a pronounced synergistic effect: MXene with abundant terminal functional groups effectively adsorbs the analytes, while the MWCNTs network provides rapid electron transfer pathways. Electrochemical studies revealed that both esculetin and quercetin undergo an adsorption-controlled process on the electrode surface, following a quasi-reversible two-proton, two-electron transfer mechanism and exhibiting excellent electrochemical activity in acidic buffer systems. The SPSA demonstrated a linear response range of 2–200 μmol/L for both analytes, with a detection limit (LOD) of 10 nmol/L (S/N = 3). In addition, with the assistance of solid phase extraction for purifying and enriching the analytes in herbal samples, the SPSA exhibited good selectivity, robustness, and accuracy. No significant difference was observed between the results obtained by the SPSA and those obtained by conventional HPLC-PDA, confirming the high practical value of this strategy for rapid quality assessment in Chinese medicine cultivation, processing, and trade. Objective Encapsulation of esculetin into DSPE-MPEG2000 carrier was performed to improve its water solubility and oral bioavailability, as well as enhance its anti-inflammatory effect on a mouse model of ulcerative colitis that was induced with dextran sulphate sodium (DSS).Methods We determined the in-vitro and in-vivo high-performance liquid chromatographic (HPLC) analysis method of esculetin; Esculetin-loaded nanostructure lipid carrier (Esc-NLC) was prepared using a thin-film dispersion method, wherein a particle size analyser was used to measure the particle size (PS) and zeta potential (ZP) of the Esc-NLC, while a transmission electron microscope (TEM) was employed to observe its morphology. Also, HPLC was used to measure its drug loading (DL), encapsulation efficiency (EE) and the in-vitro release of the preparation, as well as investigate the pharmacokinetic parameters. In addition, its anti-colitis effect was evaluated via histopathological examination of HE-stained sections and detection of the concentrations of tumour necrosis factor-alpha (TNF-α), interleukin (IL)-1 beta (β), and IL-6 in serum with ELISA kits. Results The PS of Esc-NLC was 102.29 ± 0.63 nm with relative standard deviation (RSD) of 1.08% (with poly-dispersity index-PDI of 0.197 ± 0.023), while the ZP was −15.67 ± 1.39 mV with RSD of 1.24%. Solubility of esculetin was improved coupled with prolonged release time. Its pharmacokinetic parameters were compared with that of free esculetin, wherein the maximum concentration of the drug in plasma was increased by 5.5 times. Of note, bioavailability of the drug was increased by 1.7 times, while the half-life was prolonged by 2.4 times. In the anti-colitis efficacy experiment, the mice in Esc and Esc-NLC groups exhibited significantly reduced levels of TNF-α, IL-1β, and IL-6 in their sera comparable to the DSS group. Colon histopathological examination revealed that mice with ulcerative colitis in both Esc and Esc-NLC groups displayed improved inflammation, amid the Esc-NLC groups having the best prophylactic treatment effect. Conclusion Esc-NLC could ameliorate DSS-induced ulcerative colitis by improving bioavailability, prolonging drug release time and regulating cytokine release. This observation confirmed the potential of Esc-NLC to reduce inflammation in ulcerative colitis, albeit the need for follow-up research to verify the application of this strategy to clinical treatment of ulcerative colitis.
