Fucoidan is a fucose-rich sulfated polysaccharide that has gained attention owing to its various biological activities. In this study, fucoidan was isolated from Saccharina japonica using an enzyme-assisted method, and its antioxidant and anti-hepatocarcinoma effects were evaluated. The fucoidan was a 112.8 kDa polysaccharide comprising seven monosaccharides: fucose, xylose, glucuronic acid, rhamnose, glucose, mannose, and galactose. The main chain residues were (1 → 3)-α-L-Fuc p and (1 → 4)-α-L-Fuc p units with sulfate groups at the C-2/C-4 positions of the (1 → 3)-α-L-Fuc p residues. S. japonica fucoidans showed excellent antioxidant potency with values of 1.02 mg TE/g and 5.39 mg TE/g for the ABTS and FRAP assays, respectively. Additionally, they exerted antitumor efficacy and low systemic toxicity in H22 tumor-bearing mice, with a tumor inhibition rate of 42.93%. Furthermore, it significantly inhibited tumor angiogenesis and reduced pro-inflammatory cytokines levels (IL-1β, IL-6, and TNF-α). Our results suggest that fucoidan isolated from S. japonica possesses potent antioxidant and anticancer properties and may be used as a potential agent for hepatocellular carcinoma treatment.

The smart oral administration Insulin device has the potential to improve glycemic management. It can reduce the risk of hypoglycemia associated with exogenous Insulin (INS) therapy while also avoiding many of the disadvantages associated with subcutaneous injections. Furthermore, diabetes mellitus (DM) is an endocrine illness characterized by inflammation, and it is critical to minimize the amount of inflammatory markers in diabetic patients while maintaining average blood glucose. In this study, a responsive nanosystem vitamin B12-Fucoidan-Concanavalin A (VB12-FU-ConA NPs) with anti-inflammatory action was developed for smart oral delivery of Insulin. Con A has high sensitivity and strong specificity as a glucose-responsive material. Fucoidan has anti-inflammatory, immunomodulatory, and hypoglycemic functions, and it can bind to Con A to form a reversible complex. Under high glucose conditions, free glucose competitively binds to Con A, which swells the nanocarrier and promotes Insulin release. Furthermore, in the low pH environment of the gastrointestinal tract, positively charged VB12 and anionic fucoidan bind tightly to protect the Insulin wrapped in the carrier, and VB12 can also bind to intestinal epithelial factors to improve transit rate, thereby promoting INS absorption. In vitro tests showed that the release of nanoparticles in hyperglycemic solutions was significantly higher than the drug release in normoglycemic conditions. Oral delivery of the nanosystems dramatically lowered blood glucose levels in type I diabetic mice (T1DM) during in vivo pharmacodynamics, minimizing the risk of hypoglycemia. Blood glucose levels reached a minimum of 8.1 ± 0.4 mmol/L after 8 h. Administering the nanosystem orally notably decreased the serum levels of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in diabetic mice. The nano delivery system can be degraded and metabolized in the intestinal tract after being taken orally, demonstrating good biodegradability and biosafety. In conclusion, the present study showed that VB12-FU-ConA nanocarriers are expected to be a novel system for rationalizing blood glucose.

While 3D bioprinting serves as a powerful tool in the field of tissue engineering, there is still a lack of natural biomaterial inks that simultaneously combine high mechanical performance with multiple biofunctionalities. Here, a single-component natural bioink with high strength and multi-biofunctionality was developed through the simple degradation and methacrylation of natural fucoidan. Hydrothermal degradation significantly decreased the natural fucoidan solution's viscosity by 99.9 %, meeting the necessary viscosity for Digital Light Processing (DLP) 3D printing. Meanwhile, various biofunctionalities of low molecular weight fucoidan obtained through degradation, such as antimicrobial and antioxidant properties, were developed. The resulting bioink exhibited good mechanical performance (compression modulus of 311 kPa), antimicrobial properties (antibacterial rates of 95.5 % and 97.9 % against E. coli and S. aureus , respectively), and antioxidant properties (intracellular ROS inhibition rates of 94.7 %). Using DLP 3D bioprinting, all printed products showed high shape fidelity with exceptional viability and activity of the encapsulated cells. Due to the unique sulfate structure resembling the natural components of chondroitin sulfate, the in vivo tests revealed its efficacy in promoting cartilage defect repair. In conclusion, the novel bioink blending high mechanical performance with multiple biofunctionalities, shows great potential in the 3D printing of tissue and organ regeneration.

Chitosan (CTS) and chitosan oligosaccharides (COS) have been widely applied in food industry due to their bioactivities and functions. However, CTS and COS with positive charges could interact with proteins, such as whey protein isolate (WPI), influencing their digestion. Interaction among CTS/COS, FUC, and WPI/enzymes was studied by spectroscopy, chromatography, and chemical methods in order to reveal the role of FUC in relieving the inhibition of protein digestibility by CTS/COS and demonstrate the action mechanisms. As shown by the results, the addition of FUC increased degree of hydrolysis (DH) and free protein in the mixture of CTS and WPI to 3.1-fold and 1.8-fold, respectively, while raise DH value and free protein in the mixture of COS and WPI to 6.7-fold and 1.2-fold, respectively. The interaction between amino, carboxyl, sulfate, and hydroxyl groups from carbohydrates and protein could be observed, and notably, FUC could interact with CTS/COS preferentially to prevent CTS/COS from combining with WPI. In addition, the addition of FUC could also relieve the combination of CTS to trypsin, increasing the fluorescence intensity and concentration of trypsin by 83.3 % and 4.8 %, respectively. Thus, the present study demonstrated that FUC could alleviate the inhibitory effect of CTS/COS on protein digestion.

Doxorubicin (Dox), a chemotherapeutic agent frequently used to treat cancer, elicits cardiotoxicity, a condition referred to as Dox-induced cardiotoxicity (DIC), and ferroptosis plays a contributory role in its pathophysiology. Fucoidan, a polysaccharide with various biological activities and safety profile, has potential therapeutic and pharmaceutical applications. This study aimed to investigate the protective effects and underlying mechanisms of fucoidan in DIC. Echocardiography, biomarkers of cardiomyocyte injury, serum creatine kinase, creatine kinase isoenzyme and lactate dehydrogenase, as well as histological staining results, revealed that fucoidan significantly reduced myocardial damage and improved cardiac function in DIC mice. Transmission electron microscopy; levels of lipid reactive oxygen species, glutathione, and malondialdehyde; ferroptosis-related markers; and regulatory factors such as glutathione peroxidase 4 (GPX4), transferrin receptor protein-1, ferritin heavy chain-1, heme oxygenase-1 in the heart tissue were measured to explore the effect of fucoidan on Dox-induced ferroptosis. These results suggested that fucoidan could inhibit cardiomyocyte ferroptosis caused by Dox. In vitro experiments revealed that silencing nuclear factor-erythroid 2-related factor 2 (Nrf2) in cardiomyocytes reduced the inhibitory effect of fucoidan on ferroptosis. Hence, fucoidan has the potential to ameliorate DIC by inhibiting ferroptosis via the Nrf2/GPX4 pathway.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease that primarily affects the colon and rectum and is characterized by diffuse, superficial inflammation of the colonic mucosa. The excessive production of reactive oxygen species (ROS) plays a pivotal role in immune dysregulation and the pathogenesis of UC. However, current treatment options are limited by poor absorption, rapid metabolism, and systemic side effects due to the lack of targeted delivery. Therefore, there is an urgent need for therapies capable of specifically scavenging ROS. In this study, we developed a ROS-responsive nanoscavenger system, Fucoidan-TK-Resveratrol@Curcumin (FTR@Cur), based on the marine-derived polysaccharide fucoidan, and loaded with the natural compound curcumin (Cur) for the treatment of UC. The nanoscavenger was constructed using fucoidan (Fuc) as the hydrophilic backbone, while resveratrol (Res) was conjugated via a ROS-sensitive thioketal (TK) linker, enabling targeted ROS clearance. The resulting FTR@Cur nanoparticles had a mean particle size of 92.17 nm, a zeta potential of −22.2 mV, and an encapsulation efficiency and drug loading of 69.0% and 5.5%. FTR@Cur exhibited excellent ROS-scavenging ability and biocompatibility, effectively modulating the inflammatory and immune microenvironment in the colon. This system enhanced colonic barrier function, reduced inflammation, and offers a promising therapeutic strategy for UC.

We investigated the effects of 2-Octen-1-ylsuccinic anhydride (OSA) modification and ultrasound on the structure, interface, and emulsification ability of fucoidans (FUCs). A modified polysaccharide emulsion system containing melatonin with high stability was established. The results demonstrated that the interfacial tension of the modified FUCs decrease gradually and the amphiphilicity was improved. The 9 %-OSA-FUCs emulsions ultrasonicated at 400 W presented the smallest volume-average droplet diameter (1.34 ± 0.03 μm) and the storage stability were also remarkably improved by the processing method. Meanwhile, the emulsion system enhances melatonin bioaccessability, effectively shielding it from the gastrointestinal environment and ensuring stable delivery to the small intestine for optimal absorption. At the same time, the emulsion system improves the bioaccessability of melatonin (55.32 % > 24.20 % of melatonin) and effectively delay the release of FFA in first 10 min (17.72 % < 21.98 %). This study provides insights into rational modification strategies for functional polysaccharide-based emulsifiers and designing delivery systems for chemically labile hydrophobic bioactive components.

Fucoidan, a marine-derived sulfated polysaccharide, exhibits a broad spectrum of biological activities. However, its high molecular weight and viscosity often restrict these benefits. In this study, a new fucoidanase-producing microbial strain Kosakonia oryzendophytica LH-58 was isolated from freshwater for the first time. Three low-molecular-weight fucoidans were synthesized through enzymatic hydrolysis, and their antioxidant, hypoglycemic, hypolipidemic, and anticoagulant properties were evaluated in vitro . With the hydrolysis of fucoidan by fucoidanase, there was no significant change in the composition of monosaccharides, and the sulfate content decreased slightly. Interestingly, reduced molecular weight enhanced antioxidant activity, though anticoagulant efficacy diminished. In vitro experiments demonstrated that a higher molecular weight of fucoidan significantly prolonged activated partial thromboplastin and thrombin times. The fraction F1, with a weight-average molecular weight exceeding 10 kDa, displayed superior hypoglycemic and lipid-lowering effects compared to its counterparts. The IC 50 values for F1's inhibition of α-amylase and α-glucosidase were 0.81 mg/mL and 0.94 mg/mL, respectively, while its binding rates to sodium taurocholate and sodium glycocholate were 50.11% ± 2.32% and 54.58% ± 2.71%, respectively. This study also offers a methodological reference for the swift identification of functional polysaccharides to prevent atherosclerosis.

Chemotherapy is a common method for tumor treatment. However, the non-specific distribution of chemotherapeutic drugs causes the death of normal cells. Nanocarriers, particularly mesoporous carriers, can be modified to achieve targeted and controlled drug release. In this study, mesoporous polydopamine (MPDA) was used as a carrier for the antitumor drug doxorubicin (DOX). To enhance the release efficiency of DOX in the tumor microenvironment, which contains high concentrations of glutathione (GSH), we used N,N-bis(acryloyl)cysteamine as a cross-linking agent to encapsulate the surface of MPDA with fucoidan (FU), producing MPDA-DOX@FU-SS. MPDA-DOX@FU-SS was characterized via transmission electron microscopy, thermogravimetric analysis, and X-ray photoelectron spectroscopy (XPS), and its antitumor efficacy in vitro was investigated. The optimal conditions for the preparation of MPDA were identified as pH 12 and 20 °C, and the optimal MPDA-to-FU ratio was 2:1. The DOX release rate reached 47.77% in an in vitro solution containing 10 mM GSH at pH 5.2. When combined with photothermal therapy, MPDA-DOX@FU-SS significantly inhibited the growth of HCT-116 cells. In conclusion, MPDA-DOX@FU-SS may serve as a novel, highly effective tumor suppressor that can achieve targeted drug release in the tumor microenvironment. Keywords:mesoporous polydopamine;antitumor;nanomedicine;photothermal synergistic chemotherapy

Arterial thrombosis is a critical thrombotic disease that poses a significant threat to human health. However, the existing clinical treatment of arterial thrombosis lacks effective targeting and precise drug release capability. In this study, we developed a system for targeted delivery and on-demand release in arterial thrombosis treatment. The carrier was constructed using chitosan (CS) and fucoidan (Fu) through layer-by-layer assembly, with subsequent surface modification using cRGD peptide. Upon encapsulation of urokinase-type plasminogen activator (uPA), the resulting therapeutic drug delivery system, uPA-CS/Fu@cRGD, demonstrated dual-targeting abilities towards P-selectin and αIIbβ3, as well as pH and platelet-responsive release properties. Importantly, we have demonstrated that the dual targeting effect exhibits higher targeting efficiency at shear rates simulating thrombosed arterial conditions (1800 s −1 ) compared to single targeting for the first time. In the mouse common iliac artery model, uPA-CS/Fu@cRGD exhibited great thrombolytic capability while promoting the down-regulation of coagulation factors (FXa and PAI-1) and inflammatory factors (TNF-α and IL-6), thus improving the thrombus microenvironment and exerting potential in preventing re-occlusion. Our dual-target and dual-responsive, fucoidan-based macrovesicle represent a promising platform for advanced drug target delivery applications, with potential to prevent coagulation tendencies as well as improving thrombolytic and reducing the risk of re-occlusion.

Cervical cancer remains one of the leading causes of mortality among women, and immunotherapy targeting the cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) pathway holds promise for its treatment. This study has developed nanoparticles based on fucoidan (Fu/CA NPs), successfully loading them with caffeic acid (CA) for application in cervical cancer therapy. In vitro experiments revealed that Fu/CA NPs significantly inhibited the proliferation of cervical cancer HeLa cells (by 65.73 ± 4.06 %) and induced apoptosis through the accumulation of reactive oxygen species and mitochondrial damage. Furthermore, treatment with Fu/CA NPs activated the cGAS-STING pathway, attributed to the cytoplasmic release of mitochondrial DNA (mtDNA) and the induction of DNA double-strand breaks (dsDNA) by Fu/CA NPs. In vivo results confirmed that Fu/CA NPs suppressed solid tumor growth (by 67.8 %), with even more pronounced antitumor effects observed when combined with cisplatin (96.5 %), a phenomenon also associated with the activation of the cGAS-STING pathway. Excitingly, the combination of Fu/CA NPs and cisplatin alleviated cisplatin-induced nephrotoxicity, as indicated by a decrease in blood urea nitrogen (BUN) by 53.27 % and serum creatinine (SCr) by 74.93 %. In summary, our research presents a potential therapeutic avenue for cervical cancer treatment, particularly highlighting the synergistic benefits of combining Fu/CA NPs with cisplatin.

Pulmonary embolism remains the third leading cause of human mortality after malignant tumors and myocardial infarction. Commonly available thrombolytic therapeutic agents suffer from the limitations of very short half-life, inadequate targeting, limited clot penetration, and a propensity for severe bleeding. Inspired by the trident, we developed the armor-piercing microcapsule (MC), fucoidan-urokinase-S-nitrosoglutathione-polydopamine@MC (FUGP@MC), which exhibited a triple combination of photothermal, mechanical and pharmacological thrombolysis for the therapeutic treatment of acute pulmonary embolism (APE). Briefly, the outermost fucoidan layer was utilized for targeting to the APE area. Programmed APE treatment was triggered by near-infrared (NIR) light irradiation. Photothermal thrombolytic therapy was carried out by photothermal conversion of polydopamine. The photothermal conversion broke the S-nitroso bond in S-nitrosoglutathione (GSNO) and produced large amounts of nitric oxide (NO) for mechanical thrombolysis, which subsequently disrupted the interfacial structure of microcapsule to stimulate the release of the urokinase (UK), leading to a triple synergistic thrombolytic effect. The results demonstrated that the embolization residual rate of FUGP@MC (contained ≈ 1452.5 IU/kg UK) group was significantly lower than that of UK (10,000 IU/kg) group (6.35 % VS 16.78 %). Remarkably, FUGP@MC demonstrated a reliable in vivo biosafety proficiency. In summary, trident-inspired armor-piercing microcapsule FUGP@MC reveals a potential avenue for advancing pulmonary embolism therapeutics and promises to be a safer alternative candidate to current drug approaches.
