In this paper, a novel and simple electrochemiluminescent sensor of CQDs based on ZIFs electrospun carbon fiber was constructed for the detection of malathion. Carbon quantum dots (CQDs) was obtained by pyrolysis of citric acid at high temperature. ZIFs electrospun carbon fiber (ENCF-67) prepared by electrospinning combined with high temperature calcination adsorbed Au NPs to obtain ENCF-67@Au. And the sensor of Nafion/ENCF-67@Au/CQDs/GCE was constructed by sequentially modified CQDs, ENCF-67@Au and Nafion on the electrode surfaces. ENCF-67@Au was used as the fixation material of CQDs and accelerator of system because ENCF-67 had the advantage of large specific surface area and porosity. Nafion/ENCF-67@Au/CQDs/GCE obtained sensitive and stable electrochemical luminescence signal in PBS containing S 2 O 8 2- . The addition of malathion enhanced the ECL signal of the sensor with a linear increase in the range of 1.0 × 10 -12 − 1.0 × 10 -7 M and the limit of detection 3.3 × 10 -13 M (S/N = 3). The sensor had good selectivity for malathion, which provided a simple and sensitive method for the detection of malathion in fruits and vegetables in real life and had potential applications.

The widespread use of malathion (MA) poses significant risks to environmental safety and human health, necessitating the development of highly sensitive detection platforms. Herein, a novel composite was first synthesized by integrating phytic acid (PA) doped polyaniline (PANI) with a bimetallic Cu and Zn zeolitic imidazolate framework (CZ-ZIF), denoted as PCZ-ZIF. Subsequently, the PCZ-ZIF composite was immobilized onto a fluorine-doped tin oxide (FTO) electrode to construct an ultra-sensitive electrochemical sensor (designated as PCZ-ZIF/FTO) for the precise detection of MA. PA bridges PANI and CZ-ZIF via hydrogen bonding and P–O–M coordination. The incorporation of Cu 2+ into the ZIF framework provides a strong affinity toward the P S bond in MA, while the doping of PANI significantly accelerates the interfacial electron transfer. Consequently, the PCZ-ZIF/FTO sensor exhibits exceptional electrocatalytic activity through a specific two-electron reduction process. Under optimal conditions, the sensor achieves a broad linear detection range from 1 ppt to 10 ppb and an ultra-low limit of detection (LOD) of 0.210 ppt. Furthermore, the device demonstrates outstanding reproducibility, excellent storage stability, and robust anti-interference capabilities against non-target species. Practical applicability was successfully validated in surface water and groundwater samples, yielding recoveries in agreement with standard high-performance liquid chromatography (HPLC) methods. Our work presents a highly reliable and promising strategy for the trace-level monitoring of organophosphorus pesticides in real environments.

The degradation process of malathion in the acetyl peroxyborate (APB) solution of different APB/malathion molar ratio and in the carbonate-activated APB (APB/CO 3 2− ) solution of different pH was studied by 31 P NMR technology . In the APB solution, all malathion could be degraded in 47.5 min when the molar ratio of APB/malathion was 60. CO 3 2− could effectively activate APB to degrade all malathion in 10 min at pH of 10 when APB/malathion was 10, which was obviously higher than in APB solution. 1 O 2 , •O 2 − , •OH and carbon-centered radicals (RC•) could be produced in the APB/CO 3 2− solution, and the degradation of malathion was mainly affected by RC•. The degradation mechanism of malathion in the APB/CO 3 2− solution was proposed based on the research results of malathion degradation process by 31 P NMR and active species quenching test, which involves two steps: the first step is the oxidation of malathion to malaoxon by RC•, and the second step is the hydrolysis of malaoxon to dimethyl phosphate via hydroxyl anions nucleophilic addition.

A novel label-free aptamer surface-enhanced Raman scattering (SERS) sensor for trace malathion residue detection was proposed. In this process, the binding of malathion molecule with aptamer is identified directly. The silver nanoparticles modified with positively charged spermine served as enhancing and capture reagents for the negatively charged aptamer. Then, the silver nanoparticles modified by aptamer were used to specifically capture the malathion. The SERS background spectra of spermine, aptamer, and malathion were recorded and distinguished with the spectrum of malathion–aptamer. To enhance the characteristic peak signal of malathion captured by the aptamer, the aggregate reagents (NaCl, KCl, MgCl 2 ) were compared and selected. The selectivity of this method was verified in the mixed-pesticide standard solution, which included malathion, phosmet , chlorpyrifos-methyl, and fethion. Results show that malathion can be specifically identified when the mixed-pesticide interferences existed. The standard curve was established, presenting a good linear range of 5 × 10 − 7 to 1 × 10 − 5 mol·L − 1 . The spiked experiments for tap water show good recoveries from 87.4% to 110.5% with a relative standard deviation of less than 4.22%. Therefore, the proposed label-free aptamer SERS sensor is convenient, specifically detects trace malathion residues, and can be applied for qualitative and quantitative analysis of other pesticides.

A highly sensitive dual-signal aptamer sensor based on polydopamine-gold nanoparticles (PDA-AuNPs) and exonuclease I (Exo I) was developed for detection of malathion. Compared with traditional sensing elements, aptamer has many advantages, such as high affinity, superior specificity, strong stability and easy modification. The electrodeposition synthesis of PDA-AuNPs gave excellent biocompatibility and electrical conductivity on the sensor. With the addition of malathion, the specific interaction between malathion and its aptamer forced the aptamer to detach from the electrode surface and induced the capture probe to form a hairpin structure on the electrode surface. Exo I was added to motivate the autocatalytic target cycling which remarkably increased the current change of electrochemical signal over 2 times. Therefore, the promising strategy gave rise to an optional dual-signal current readout in both the signal-on of Fc and the signal-off of Tn. In this work, the prepared biosensor exhibited high sensitivity to malathion via the combination of dual-signal design and autocatalytic target cycling amplification. Under the optimized conditions, the proposed sensor showed a wide linear range from 0.5 to 600 ng/L malathion. It also exhibited excellent specificity, acceptable repeatability and good stability. The application of real samples obtained satisfactory recovery results, demonstrating a promising potential in food safety analysis.

Due to the diversification and complexity of organophosphorus pesticide residues brings great challenges to the detection work. Therefore, we developed a dual-ratiometric electrochemical aptasensor that could detect malathion (MAL) and profenofos (PRO) simultaneously. In this study, metal ions , hairpin-tetrahedral DNA nanostructures (HP-TDN) and nanocomposites were used as signal tracers, sensing framework and signal amplification strategy respectively to develop the aptasensor. Thionine (Thi) labeled HP-TDN (HP-TDNThi) provided specific binding sites for assembling Pb2+ labeled MAL aptamer (Pb2+-APT1) and Cd2+ labeled PRO aptamer (Cd2+-APT2). When the target pesticides were present, Pb2+-APT1 and Cd2+-APT2 were dissociated from the hairpin complementary strand of HP-TDNThi, resulting in reduced oxidation currents of Pb 2+ (I Pb 2+ ) and Cd 2+ (I Cd 2+ ), respectively, while the oxidation currents of Thi (I Thi ) remained unchanged. Thus, I Pb 2+ /I Thi and I Cd 2+ /I Thi oxidation current ratios were used to quantify MAL and PRO, respectively. In addition, the gold nanoparticles (AuNPs) encapsulated in the zeolitic imidazolate framework (ZIF-8) nanocomposites ( [email protected] ) greatly increased the catch of HP-TDN, thereby amplifying the detection signal. The rigid three-dimensional structure of HP-TDN could reduce the steric hindrance effect on the electrode surface , which could greatly improve the recognition efficiency of the aptasensor for the pesticide. Under the optimal conditions, the detection limits of the HP-TDN aptasensor for MAL and PRO were 4.3 pg mL-1 and 13.3 pg mL-1, respectively. Our work proposed a new approach to fabricating a high-performance aptasensor for simultaneous detection of multiple organophosphorus pesticides, opening a new avenue for the development of simultaneous detection sensors in the field of food safety and environmental monitoring.

The bioaccumulation, metabolism, tissue-specific distribution and toxicity of the widely used organophosphorous pesticide malathion to zebrafish were investigated on an enantiomeric level for evaluating the environmental risks. The metabolites were also monitored and evaluated. Malathion was metabolized by zebrafish very fast with the half-life of 0.12 d and showed a middle accumulation capacity in zebrafish with bioaccumulation factor (BCF) of 12.9 after a 15-d exposure. Brain could enrich higher concentration of malathion than other tissues. The metabolites malaoxon, malathion/malaoxon monocarboxylic acid (DMA), malathion/malaoxon dicarboxylic acid (DCA), dimethylthiophosphate (DMTP) and dimethyldithiophosphate (DMDTP) were found, in which DMTP and DCA were in higher level, indicating the metabolism was mainly induced by carboxylesterase degradation. The accumulation of malathion and malaoxon was stereoselective in zebrafish tissues, exhibiting S-enantiomer preferentially enriched. The acute toxicity test showed rac-malathion was low toxic to zebrafish, which was 1.2 and 1.6 folds more toxic than S-malathion and R-malathion respectively. Malaoxon was highly toxic to zebrafish and approximately 32 times more toxic than malathion. The toxicity of other metabolites was lower than malathion. Malathion could cause an apparent developmental toxicity to zebrafish embryo, including bradycardia, hatchability reduction and deformity, and abnormal movement patterns in zebrafish larva. Chronic toxicity indicated that malathion and malaoxon induced oxidative damage and neurotoxicity in the liver, brain and gill of zebrafish, and malaoxon exhibited a relatively high injury to the zebrafish brain. The results can provide information for the comprehensive assessment of the potential risk of malathion to aquatic organisms and highlight the necessity of consideration of stereoselectivity and metabolites when systemically evaluating pesticides.

A dual-mode sensing platform has been developed for ultrasensitive malathion (MAT) detection based on bifunctional rGO@ZnO/Zn-Fe 3 O 4 composites (reduced graphene oxide-supported ZnO nanorods and Zn-doped Fe 3 O 4 microspheres). The rGO@ZnO/Zn-Fe 3 O 4 nanozyme is fabricated via a one-pot hydrothermal strategy and integrates favorable electrochemiluminescence (ECL) emission and intrinsic peroxidase-mimicking activity to realize dual-signal readout. In the K 2 S 2 O 8 ECL system, Au NPs@UiO-66 serves as a coreactant booster to further magnify the ECL intensity of the composite. Meanwhile, the nanozyme catalyzes the H 2 O 2 -mediated oxidation of 3,3',5,5'-tetramethylbenzidine (TMB) to produce a colorimetric response. MAT suppresses both ECL and colorimetric responses by occupying Fe 2+ /Fe 3+ catalytic sites and scavenging of reactive radicals. The sensor achieves limits of detection as low as 2.8 × 10 –13 mol/L under ECL mode and 1.32 × 10 –6 mol/L under colorimetric mode. In practical testing of MAT-spiked vegetable samples, the sensor exhibits acceptable recoveries and low relative standard deviations, demonstrating its practicability for field food safety supervision and rapid pesticide residue screening. A photoelectrochemical (PEC) sensor was constructed for the detection of non-electroactive malathion. It is based on the use of a hierarchical CuO material derived from a Cu-BTC metal-organic framework (where BTC stands for benzene-1,3,5-tricarboxylic acid). The modified CuO was obtained by calcination of Cu-BTC at a high temperature (300 °C) and possesses a high photocurrent conversion efficiency. Under irradiation with visible light and in the presence of malathion, the formation of the CuO-malathion complex on the CuO gave rise to an increase in steric hindrance. This results in a decrease in photocurrent. This novel PEC detection method has a lower detection limit of 8.6 × 10 −11 mol L −1 and a wide linear range (1.0 × 10 −10 ~ 1.0 × 10 −5 mol L −1 ).

Helicoverpa armigera Hübner (Lepidoptera: Noctuidae) is a globally devastating pest and has evolved with varying levels of resistance to a broader spectrum of insecticides. CarEs are major enzymes involved in insecticide detoxification and metabolic resistance. Four CarE genes were identified and cloned from H. armigera, and the expression pattern analyses indicated that they were predominantly expressed in the detoxifying tissues and larval feeding periods. The insecticide inductive assays further suggested that these CarE genes with expressions were significantly upregulated after beta-cypermethrin, lambda-cyhalothrin, malathion, and chlorpyrifos exposures. The purified recombinant proteins of both CarE016B and CarE016C by bacterial expression exhibited significantly higher catalytic efficiency toward α-naphthyl acetate and also showed relatively stronger binding with both β-cypermethrin and λ-cyhalothrin compared to the CarE006C and CarE015A. In vitro metabolism assay with HPLC suggests that CarE016B and CarE016C have the ability to metabolize β-cypermethrin, λ-cyhalothrin, and malathion with varying hydrolase activities. GC-MS/MS analysis identified 3-phenoxybenzaldehyde (3-PBAld) as the metabolite of both β-cypermethrin and λ-cyhalothrin. Homology modeling and molecular docking analyses further indicate that these three types of insecticides could be anchored into the active pocket of both CarEs. Collectively, these results demonstrate that both CarE016B and CarE016C play a critical role in the detoxification of pyrethroid and organophosphate insecticides in H. armigera. This study provides the foundations for a comprehensive understanding of the role of the CarEs family in insecticide detoxification and resistance in H. armigera.

Evidence suggests that exposure to organophosphate pesticides increases the risk of neurodegenerative diseases, but the mechanisms remain unclear. This study investigated the effects of malathion on Alzheimer’s disease (AD)-like symptoms at environmentally relevant concentrations using wild-type (WT) and APP/PS1 transgenic mouse models. Results showed that malathion exposure induced AD-like cognitive impairment, amyloid-β (Aβ) accumulation, and neuroinflammation in WT mice, with worsened symptoms in APP/PS1 mice. Mechanistic studies revealed that malathion induced AD-like gut microbiota dysbiosis (reduced Lactobacillus and Akkermansia, and increased Dubosiella), causing gut barrier impairment and tryptophan metabolism disruptions. This resulted in a significant increase in indole derivatives and activation of the colonic aryl hydrocarbon receptor (AhR), promoting the kynurenine (KYN) pathway while inhibiting the serotonin (5-HT) pathway. Increased neurotoxic KYN metabolites (3-hydroxykynurenine and quinolinic acid) triggered gut and systemic inflammation, upregulating hippocampal IL-6 and IL-1β mRNA levels and thereby causing neuroinflammation. Gut tryptophan metabolism disruptions caused hippocampal neurotransmitter imbalances, reducing the levels of 5-HT and its derivatives. These effects promoted AD progression in both WT and APP/PS1 mice. This study highlights the crucial role of the microbiota-gut-brain axis in AD-like cognitive impairment induced by malathion exposure, providing insights into the neurodegenerative disease risks posed by organophosphate pesticides.

An aptasensor with dual-outputs was developed for malathion detection. Fe-MOF was synthesized to design favorable signal probes for catalytic amplification. Owing to the excellent peroxidase-like activity of Fe-MOF, the redox reaction was catalyzed to produce the dual-outputs of colorimetric and electrochemical. In this sensing strategy, malathion was captured by the aptamer on sensing interface, leading to the release of signal probe. Thanks to the catalytic amplification of Fe-MOF and the high capture effect of aptamer, the aptasensor produced a sensitive response for malathion. Based on the dual-signals of absorbance and current, the detection method for malathion was developed ranging from 10 ng/mL to 500 ng/mL. The detection limit of malathion was 5.8 ng/mL for colorimetric output and 4.6 ng/mL for electrochemical output. Furthermore, the aptasensor exhibited high specificity and good repeatability in malathion detection. Finally, the aptasensor was applied to detect malathion in fruit and vegetable samples with satisfactory recovery.
