MicroRNA (miRNA) is riveting nowadays due to its close relevance to human malignancies. Exploiting a fast and convenient biosensor for sensitive and specific detecting miRNA is necessary and it has significant meaning for oncology. In this work, to understand the relationship between miRNA-21 and gastric cancer, we employ T4 RNA ligase 2 to initiate specific ligation reaction depending on the sequence of target RNA, and T7 exonuclease to catalyze the first stage cyclic enzymatic amplification method (CEAM) specifically, which also resting on the target RNA sequence , and the second stage CEAM to further amplify the response resulting from the initial target RNA. Our two-stage CEAM strategy can detect miRNA-21 as low as 0.36 fM with remarkable specificity, and most importantly, it can be used to inspect the expression level of miRNA-21 in blood serum of gastric cancer patients . This will offer new perspective for figuring out the pathways of miRNA-21 involving in cancer.

MicroRNA(miRNA) serves as an effective and viable biomarker for early diagnosis and monitoring of cancer disorders. It is highly expressed in tumor cells, including lung cancer, liver cancer and lymphoma. Herein, we propose a ratiometric electrochemical sensor for ultrasensitive detection of miRNA-21 using dual signal amplification, hybridization chain reaction and Exo III assisted-amplification. Methylene blue (MB) and Hemin are chosen as two electrochemical species. Then the ratiometric electrochemical sensor were developed, which showed favorable performance of miRNA-21 detection, and exhibited a detection concentration range from 1 fM to 10 nM. Notably, the limit of detection for this biosensor was 0.15 fM. Overall, this strategy for miRNA detection holds significant promise for early cancer screening.

MicroRNA-21(miRNA-21) can act as a novel biomarker in early stage of acute kidney injury (AKI). The development of simple but sensitive miRNA-21 detection technology is helpful for prevention and treatment of AKI. In this work, based on the cascaded signal amplifications of the target-induced hybridization chain reaction (HCR) and magnetic assisted enrichment, a highly sensitive homogeneous electrochemiluminescence (ECL) biosensor for miRNA-21 detection had been developed. Firstly, capture probe H1 (CPH1) had been modified on magnetic beads (MBs) which could capture target (miRNA-21) and trigger HCR amplification to produce long strand dsDNA in the homogeneous solution. Ru(phen) 3 2+ could be embedded into the grooves of dsDNA and played the role of luminous tag, which could be separated by magnetic easily to realize the enrichment. Then the enriched products had been dissolved in the buffer solution and the ECL signal could be detected easily. The ECL intensity had a linear correlation with the logarithm of miRNA-21 concentration in the range of 1.0 fM-1.0 nM, the detection limit was 0.14 fM. The method developed in this work showed well sensitivity, specificity, which had been applied to detect miRNA-21 in urine of AKI patients with satisfied results.

Background Medication-related osteonecrosis of the jaw (MRONJ), caused by long-term anti-resorptive therapy, leads to bone necrosis and impaired healing. This study developed a miR-21-loaded mesoporous silica nanoparticles (MSN) hydrogel to promote local bone regeneration. Methods Transcriptome sequencing of zoledronic acid (ZOL)-treated osteoclasts (OCs) revealed PDCD4 upregulation and NF-κB (p65) phosphorylation inhibition. miR-21, identified as a regulator of PDCD4, was validated in vitro. Amino-modified mesoporous silica nanoparticles (MSN-NH₂) were synthesized to carry miR-21, and embedded in a dynamic Schiff base-crosslinked hydrogel. The hydrogel's biocompatibility, sustained release, and therapeutic effect were evaluated in a rat MRONJ model via micro-CT, histology, TRAP staining, and RNA in situ hybridization (RISH). Results miR-21 reversed ZOL-induced suppression of NF-κB/p65 phosphorylation by targeting PDCD4, thereby restoring osteoclast differentiation and resorption activity. The miR-21-loaded MSN hydrogel promoted bone regeneration, increased TRAP⁺ osteoclast numbers, and elevated local miR-21 expression, while maintaining systemic safety. These findings suggest that the miR-21-loaded MSN hydrogel system exerts its therapeutic effect primarily through the miR-21/PDCD4/NF-κB signaling pathway, facilitating coordinated regulation of osteoclast-mediated bone remodeling. Conclusions The miR-21-loaded MSN hydrogel effectively restored bone remodeling and healed MRONJ defects without systemic toxicity, offering a promising localized adjunct to anti-resorptive therapies.

As an important biomarker for early diagnosis of cancers, sensitive detection and high-resolution imaging of microRNA-21 in cancer cells have become important and challengeable. In this work, highly sensitive detection and spatial imaging of intracellular microRNA-21 were realized by the reduced signal background through vertical polarization excitation with a polarizer. The lateral local surface plasmon resonance property of gold nanorods (AuNRs) displayed a pronounced green color with low scattering intensity, which was adjusted to red color with strong scattering intensity when the core–satellite gold nanoparticle (AuNP) assembly was constructed on the side of AuNRs through a catalyzed hairpin assembly (CHA) circuit in the presence of microRNA-21. This unique approach allows for effectively reducing the strong background signal to improve the sensitivity of detection. Additionally, the proposed strategy can not only realize the sensitive detection of microRNA-21 with the limit of detection as low as 2 pM (3σ) but also achieve the high spatial imaging of cancer cells, which provided a specific strategy for the construction and imaging of intracellular imaging probes. It is believed that the simple and sensitive approach on the basis of lateral local surface plasmon resonance property of anisotropic AuNRs with excellent sensitivity combined with high spatial imaging holds promising potentials to visualize intracellular microRNAs with low abundance.

A sensitive and selective electrochemical method for microRNA-21 (miRNA-21) detection was developed. This detection tactic was based on graphene, gold nanoparticles (AuNPs), locked nucleic acid (LNA) incorporated hairpin DNA switching and enzymatic signal amplification. Graphene and AuNPs can improve the electrode conductivity and increase the electrode active surface. The LNA incorporated hairpin DNA probe was modified with biotin at its 5′-end and –SH at its 3′-end. After the probe hybridized with target miRNA-21, its loop-and-stem structure was unfolded to force the biotin away from the electrode surface. Through the specific interaction between biotin and streptavidin-horseradish peroxidase (streptavidin-HRP), miRNA-21 can be quantitatively realized by electrochemical detection of the enzymatic product of benzoquinone in the presence of hydrogen peroxide and hydroquinone. The determination conditions, such as probe concentration, probe immobilization time, hybridization time and applied potential, were optimized. This assay allowed the detection of miRNA-21 in the range of 1–5000 pM with a detection limit of 0.4 pM (S/N = 3). Based on the locked nucleic acid incorporated probe with hairpin structure, the sensor can well discriminated complementary and base mismatched sequences. Successful attempts were made in miRNA-21 expression analysis of human HeLa cells and normal human hepatic L02 cells.

Detection of biomarkers at the cellular level can provide more accurate and comprehensive information that is important for early diagnosis of diseases and evaluation of new drugs. However, the interference of a large number of components in cells and the requirement of high sensitivity bring great challenges for their detection. Herein, a robust and enzyme-free electrochemical platform was proposed for microRNA-21 (miRNA-21) detection by integrating the efficient separation of magnetic nanobeads (MBs) with the multisignal amplification of strand displacement amplification (SDA) and electrochemically mediated atom transfer radical polymerization (eATRP). The eATRP is capable of de novo growth of a number of electroactive polymers on the electrode surface for signal amplification. Compared to simple hybridization, SDA and eATRP can enhance the signals by ∼35-fold, achieving high signal-to-noise ratio for low-abundant target detection. Owing to their superparamagnetism and strong magnetic response ability, MBs endow the method with excellent specificity and anti-interference ability to detect miRNA-21 in cells. Using MBs as capture carriers, SDA and eATRP for signal amplification, and gold nanoflower (AuNF)-modified electrodes as working electrodes, as low as 0.32 aM miRNA-21 was detected. Furthermore, the successful detection of miRNA-21 in cells indicated the great prospect of this method in the early diagnosis of cancers, life science research, and single-entity electrochemical detection.

The efficient and quantitative detection of miRNA-21 plays a critical role in early cancer diagnosis, as well as in the screening and treatment of early-stage infectious diseases. In this study, we developed an electrochemical biosensor for the detection of miRNA-21 in human serum samples. The sensor is constructed based on carboxyl-modified cobalt-doped nitrogen-doped carbon (Co-NC–COOH), a dual-output toehold-mediated strand displacement (TMSD) reaction, and gold nanoparticles (Au NPs) to achieve electrocatalytic signal amplification toward acetaminophen (AP). Co-NC–COOH possesses abundant active and catalytic sites, along with a porous surface morphology, which facilitates electron transfer and enhances the loading capacity of the capture probe (CP). In the presence of miRNA-21, the dual-output TMSD reaction is initiated, resulting in the release of a large amount of product strand (P). Subsequently, P hybridizes with the CP immobilized on the electrode surface, enabling the attachment of Au NPs to the electrode and thereby achieving significant electrochemical signal amplification. The results demonstrate that the proposed biosensor exhibits excellent analytical performance, including a wide linear detection range (0.1 fM–50 nM), an ultralow detection limit (63 aM), and satisfactory spike recovery rates (95.18%–103.59%). Moreover, the biosensor shows good stability and high specificity, providing a reliable platform for the precise quantification of miRNA-21 in human serum.

Multifunctional nano-materials that can be used to monitor the expression of specific biomarkers and serve as vehicles for controlled drug delivery are highly desirable. Herein, we report a new DNA-hybrid-gated core–shell upconversion nanoprobe (UCNP@MOF/DOX) for fluorescence analysis of microRNA-21 (miR-21), which also triggers the release of drug loaded in the probes for on-demand anti-cancer treatment. The nanoprobe is built on the merits of ultraviolet-visible light of upconversion nanoparticles (UCNPs) excited by near-infrared (NIR) and extraordinary loading capability of metal–organic frameworks (MOFs) for drug delivery. Controlled release of doxorubicin (DOX) from the nanoprobe by miR-21 underwent the following two-stage kinetics: a fast release stage specifically triggered by miR-21 and proportional to miR-21 concentration and a slow stage observed in both gated and ungated nanoprobes due to collapse of the UIO-66-NH2 coatings via ligand exchange with phosphates. In addition, the nanoprobe showed good selectivity, a linear response towards miR-21 ranging from 4 nM to 500 nM, and a limit of detection in 4 nM, which precluded unintended payload leakage due to low-abundance endogenous miR-21 expression in normal cells. Moreover, based on a dual-targeted delivery system constituted by AS1411-mediated recognition and responsive release of DOX, a specific cytotoxic efficacy was observed in MCF-7 cells. The present work provides a smart and robust nanoprobe for real-time detection of miRNA and dual-responsive drug delivery in tumor cells.

The cathodic electrochemiluminescence (ECL) behaviour of nontoxic MoS2 quantum dots (QDs) was studied for the first time using potassium peroxydisulfate as the co-reactant. Ag-PAMAM NCs, serving as difunctional tags for quenching and enhancing ECL of MoS2-reduced graphene oxide composites, were introduced into the ECL detection system for signal amplification. By modulating the interparticle distance between MoS2 QDs and Ag-PAMAM NCs, the ECL quenching from resonance energy transfer and the ECL enhancement from surface plasma resonance were realized. Coupling the good ECL performance of MoS2 QDs with the excellent ECL quenching and enhancement effects of Ag-PAMAM NCs, a novel MoS2 QDs-based ECL biosensing platform for sensitive detection of microRNA-21 was achieved with a detection limit of 0.20 fmol L−1 (S/N = 3). This method was successfully applied to the determination of microRNA-21 in human serum samples with recoveries of 90.0–110.0%, suggesting great potential for its applications in biological and chemical analysis.

Signal amplification strategies increase the complexities of biosensors while improving the response signals. Herein, a novel electrochemical biosensor was developed based on a DNA hydrogel for sensitive analysis using microRNA-21 (miRNA-21) as a detection model. Poly C sequences combined with C–Ag(I)–C hydrogel formed a DNA hydrogel by the unique interaction between the cytosines and silver ions. Thus, with a three-way conjunction structure of DNA, this C–Ag(I)–C hydrogel was constructed as a novel biosensor for the detection of miRNAs. With the assistance of this hydrogel, numerous silver ions gathered around DNA strands, which would amplify the signal. Under these conditions, the silver ions produced distinct square wave voltammetry oxidation peak currents. This electrochemical biosensor we designed exhibited a great linear relationship for the logarithm of the concentration of miRNA-21 from 1 fM to 100 pM with a detection limit of 0.117 fM. Furthermore, our sensors were able to differentiate miRNA-21 from its homologous family with satisfactory responsiveness in the dilute bovine serum system.

MicroRNA-21 (miRNA-21) is a significant biomarker which is closely related to some kinds of diseases, such as cancer, cardiovascular disease and kidney disease. Therefore, the detection of miRNA-21 is of great importance and can provide essential information for disease diagnosis. In this study, we report a facile, sensitive assay for miRNA-21 detection using personal glucose meters (PGM). Biotinylated DNA strand linked invertase (Inv) is conjugated on the surface of streptavidin-coated magnetic beads (MBs) to form a MBs–DNA–Inv complex. Target miRNA-21 in the detection system is captured by the MBs–DNA–Inv probe through DNA/RNA hybridization. The duplex-specific nuclease (DSN) enzyme specifically cleaves the DNA to recycle the target miRNA and release invertase, thereby triggering the dual signal amplification and ensuring high sensitivity. Besides, we establish a linear relationship between PGM and different concentrations of miRNA-21 in the range of 10 to 200 pM. The limit of detection is 1.8 pM, which is more sensitive than some of the previous reports. In addition, the biosensor exhibits excellent sequence selectivity and single-base mutation can be discriminated. Moreover, the expression of miRNA-21 is confirmed in urine from mice by our method, which is in good accordance with the qRT-PCR result. Therefore, a dependable, low-cost strategy for the detection of miRNA has been established and it meets the latest analytical demands for miRNA determination that is suitable for the public.
