Heavy rare earth elements (HREEs) have extensive applications in critical industries. Xenotime (YPO 4 ) is an important phosphatic mineral rich in HREEs. The application of 2-hydroxy-3-naphthyl hydroxamic acid (NHA) as a collector in the flotation of xenotime has rarely been been explored previously. This study reveals the effect of NHA on the flotation behavior and adsorption mechanism of xenotime. The high-grade rare earth concentrate with significant recovery is achieved at the optimized flotation conditions. Techniques such as Fourier transform-infrared spectra (FTIR), X-ray photoelectron spectroscopy (XPS), and first-principles calculations were employed to clarify the adsorption mechanism of NHA on xenotime. These studies disclose that chemical adsorption occurs through the interaction between N-bonded and carbonyl O atoms, creating a bidentate bond with an Y 3+ ion on the xenotime surface. These findings underscore the potential of NHA for selective flotation of xenotime, providing valuable insight into xenotime processing.

Object The aim of this study was to investigate the effects of aligned poly (lactic-co-glycolic acid)/nano-hydroxyapatite (PLGA/nHA) fiber scaffolds with micro-/nanostructures on the osteogenic differentiation potential of human periodontal ligament stem cells (hPDLSCs). Methods Aligned/random PLGA and PLGA/nHA fiber scaffolds with micro-/nanostructures were created by electrospinning. Comprehensive characterization of the scaffolds with scanning electron microscopy (SEM), X-ray diffraction, Fourier transform infrared spectroscopy, mechanical testing, thermogravimetric analysis and hydrophilicity was performed. In addition, they were observed if they could promote the osteogenic differentiation of hPDLSCs by RT-qPCR, SEM, alkaline phosphatase staining and alizarin red staining. The subcellular distribution of YAP protein between the nucleus and cytoplasm was detected by YAP immunofluorescence staining. Results Aligned PLGA/nHA fiber scaffolds had a better mechanical strength, thermal stability, hydrophilicity than random fiber scaffolds. Furthermore, aligned PLGA/nHA scaffolds also promoted cell adhesion, obvious cytoskeletal elongation and enhanced osteogenic differentiation of hPDLSCs was observed by the upregulation of the key genes (OCN, RUNX2, OPN) expression, increased the activity of alkaline phosphatase, and positive for alizarin red. Meanwhile, the a-PLGA/nHA scaffolds showed the strongest nuclear YAP fluorescence intensity. Conclusion The synergistic effect of fiber arrangement and nHA can be used to increase the osteogenic differentiation of hPDLSCs and the composite scaffold developed may have translational potential for alveolar bone regeneration.

3D-printed porous scaffolds have emerged as a potential solution for treating critical-sized bone defects, yet localized inflammatory imbalance leads to suboptimal therapeutic outcomes. This study utilized nHA/PA66 composite material as the foundation, employing APF additive manufacturing technology to fabricate a three-dimensional porous scaffold (HP) suitable for local bone filling, and functionalized the porous scaffold through modification with polydopamine and magnesium ions (Mg 2+ ) (HPDM). First, the physicochemical properties and biosafety of the HPDM scaffold were verified, followed by a comparative analysis of its effects on macrophage phenotype transformation, vascular endothelial cell differentiation, and pre-osteoblast differentiation differences. The HPDM scaffold achieves coordinated balance in the transformation between M1 and M2 macrophages through sustained release of polydopamine and Mg 2+ . Mg 2+ plays a crucial role in inflammatory regulation by downregulating the NF-κB signaling pathway. Furthermore, several experiments demonstrated that the HPDM scaffold regulates orderly inflammatory responses to promote intercellular interactions, stimulating angiogenesis and osteogenic regeneration. In New Zealand rabbits' femoral condyle bone defect model, the HPDM porous scaffold achieved significant vascularized bone regeneration. This study confirms that the functionalized HPDM porous scaffold prepared using nHA/PA66 as the base material has significant potential in regulating immune responses and enhancing vascularized bone regeneration.

Bone-targeted therapies have been the choice of treatments for cancer metastases in bone to minimize skeletal morbidity and preserve patients' quality of life . Rhein is of particular interest due to its high bone affinity. Here we reported a novel Rhein- polyethylene glycol (PEG)-nano hydroxyapatite (nHA) conjugate to deliver doxorubicin (DOX) and Phosphorus-32 ( 32 P) simultaneously for enhanced cancer chemo-radiotherapy. The synthetic Rhein-PEG-nHA conjugates were sphere in shape with an average diameter of ~120 nm . Their morphology, drug release and bone affinity were confirmed in vitro . The release profiles of DOX depend on pH condition , but 32 P exhibited good stability. Rhein-PEG-nHA also showed high bone affinity in vivo , and the tumor volume decreased after the [email protected] and 32 [email protected] treatments. Most importantly, the DOX/ 32 [email protected] showed the strongest inhibition on the growth of bone metastases of breast cancer. We revealed the potential of Rhein-PEG-nHA in combined chemo-radiation treatment for bone metastases of breast cancer.

Amidst the relentless innovation in materials science and bone tissue engineering, the quest for next-generation bone graft materials with bespoke functionalities has emerged as a pivotal research domain. This study produced and characterized a novel bone healing material. First, we explained how three-armed polylactic acid (3s-PLA) was created in supercritical carbon dioxide (ScCO 2 ) and utilized as a matrix material. Following that, we described how the solution blending approach was employed to create three-armed polylactic acid/chitosan/nanohydroxyapatite (3s-PLA/CS/nHA) composites. The composites were then drug-loaded with prednisone acetate as a model drug utilizing the supercritical impregnation technique. Ultimately, the Ritger-Peppas model was substantially followed by the drug release of the drug-loaded composites when the in vitro drug-release kinetics of the drug-loaded materials were investigated. The porous structure of 3s-PLA was demonstrated by differential scanning calorimetry (DSC), Fourier transform infrared spectroscopy (FTIR), and nuclear magnetic resonance (NMR) hydrogen spectroscopy. Morphologic studies using scanning electron microscopy (SEM) revealed the homogeneous distribution of drug in the matrix of the mixtures as well as the porous structure of 3s-PLA and 3s-PLA/CS/nHA. The hydrophilicity of 3s-PLA/CS/nHA was examined by the use of water contact angle (WCA), revealing that the material in question has a hydrophilic water contact angle of 45.69°. Furthermore, research was conducted using one-way tests and investigations to characterize in vitro drug-release carrier materials under various drug conditions and temperatures. The carrier material consistently released up to 84.8% of prednisone acetate over the course of 72 h, demonstrating good control over prolonged release, according to the results. Graphical abstract

Introduction Bone scaffold is expected to possess excellent mechanical and biological properties similar to natural bone tissues. In this study, we aimed to prepare a biomineralized Col and hydroxyapatite composite scaffold consisting of biomimetic bone components and multi-level bionic bone structure to strengthen its mechanical properties. Methods We prepared a Col/nano-hydroxyapatite biological composite scaffold with multi-level structure (from nanofibers to micron bionic bone motif to bionc bone scaffold) of biomimetic bone tissue, and biomineralized the scaffold in simulated body fluid (SBF) preheated to 37 °C. X-ray diffraction (XRD), Fourier transform infrared (FTIR) spectroscopy and Scanning electron microscope, were used to characterize the biomineralized products. Results Morphological study confirmed in situ deposition of nHA in the multi-scale hierarchical structure of the biomineralized scaffold. We explored the biomineralization nucleation mechanism of the scaffolds at the atomic level based on the first principles and the mechanisms for growth of mineralized nHA crystal array in its multi-scale structure, and how the double multiscales structure strengthened the mechanical properties of the material. Conclusions This synthetic bone scaffold, with bionic bone composition and double multi-level interface reinforcement, provides a new strategy for synthesizing bioactive bone scaffolds with enhanced biomechanical properties.

Introduction: Drugs and biocompatible nanoparticles have raised significant potential in advancing the bone regeneration. Electrospinning technology enables the full realization of the value of drugs and nanoparticles. Methods: In this study, we have successfully fabricated core–sheath nanofibers solely composed of polycaprolactone (PCL) polymer. Simvastatin (SIM) was confined to the core of the nanofibers while nanohydroxyapatite (nHA) was loaded on the nanofiber surface. Results: All the prepared nanofibers exhibited a cylindrical micromorphology, and the core–sheath structure was exploited using a Transmission Electron Microscope. X-ray pattern results indicated that SIM was in an amorphous state within nanofibers, while Fourier Transform InfraRed spectroscopy showed excellent chemical compatibility among SIM, nHA, and PCL. The actual loading of nHA within the nanofiber was determined by a thermogravimetric test due to the high melting point of nHA. Core–sheath nanofibers could release SIM for 672 h, which was attributed to the core–sheath structure. Furthermore, nanofibers loaded with SIM or nHA had a positive impact on cell proliferation, with the core–sheath nanofibers displaying the most favorable cell proliferation behavior. Discussion: Such a synergistic facilitation strategy based on materials and nanostructure may encourage researchers to exploit new biomedical materials in future.

Balancing mechanical strength, controllability of degradation and bioactivities, remains a significant challenge for absorbable barrier membranes in guided bone regeneration (GBR) for the treatment of bone defects. To overcome these limitations, a novel absorbable synthetic PGA-TMC/PTMC/nHA (PGTTH) barrier membrane was developed using electrospinning technology to prolong barrier duration and promote rapid bone repair. The PGTTH membrane integrates three functional components in a one-step fabrication process: a poly (glycolic acid-co-trimethylene cabonate) (PGA-TMC) copolymer matrix providing mechanical integrity, polytrimethylene carbonate (PTMC) imparting flexibility and a slow, linear degradation profile to ensure long-term structural stability, nano-hydroxyapatite (nHA) delivering bioactive ions to enhance osteogenesis and biomineralization. To systematically evaluate the performance of the PGTTH membrane, mechanical characterization and in vitro degradation tests confirmed that the membrane exhibits appropriate mechanical properties and a slow degradation profile, thereby demonstrating its ability to maintain structural space over an extended period. Furthermore, in vitro and in vivo studies demonstrated that the PGTTH membrane significantly promotes cell adhesion, proliferation, and osteogenic differentiation. In conclusion, this study not only presents a promising GBR barrier membrane material with potential for clinical application, but also offers a new design strategy and experimental basis for developing absorbable membranes that combine excellent barrier functionality with osteoinductive capacity.

Guided bone regeneration (GBR) technology has been widely used for the regeneration of periodontal bone defects. However, the limited mechanical properties and bone regeneration potential of the currently available GBR membranes often limit their repair effectiveness. In this paper, serum-derived growth factor lysophosphatidic acid (LPA) nanoparticles and dopamine-decorative nanohydroxyapatite (pDA/nHA) particles were double-loaded into polylactic-glycolic acid/polycaprolactone (PLGA/PCL) scaffolds as an organic/inorganic biphase delivery system, namely, PP-pDA/nHA-LPA scaffolds. Physicochemical properties and osteogenic ability in vitro and in vivo were performed. Scanning electron microscopy and mechanical tests showed that the PP-pDA/nHA-LPA scaffolds had a 3D bionic scaffold structure with improved mechanical properties. In vitro cell experiments demonstrated that the PP-pDA/nHA-LPA scaffolds could significantly enhance the attachment, proliferation, osteogenic differentiation, and mineralization of MC3T3-E1 cells. In vivo, the PP-pDA/nHA-LPA scaffolds exhibited great cytocompatibility and cell recruitment ability in 2- and 4-week subcutaneous implantation experiments and significantly promoted bone regeneration in the periodontal defect scaffold implantation experiment. Moreover, LPA-loaded scaffolds were confirmed to enhance osteogenic activities by upregulating the expression of β-catenin and further activating the Wnt/β-catenin pathway. These results demonstrate that the biphase PP-pDA/nHA-LPA delivery system is a promising material for the GBR.

Large bone defects present significant clinical challenges, with distraction osteogenesis (DO) requiring prolonged treatment periods and yielding suboptimal outcomes. Calcitonin gene-related peptide (CGRP) demonstrates potent bone-forming activity but suffers from rapid degradation and a short half-life, limiting its therapeutic applications. This study engineered sustained-release CGRP microspheres using poly(D,L-lactide-co-glycolide)/nano-hydroxyapatite/graphene oxide (PLGA/nHA/GO) composite matrices via W/O/W double emulsion-solvent evaporation method to address these limitations. The fabricated microspheres exhibited uniform spherical morphology (51.15 ± 0.40 μm), high encapsulation efficiency (86.14 ± 2.5 %), and sustained CGRP release over 42 days. In vitro studies compared four groups: control (untreated), blank microspheres, free CGRP solution, and CGRP-loaded microspheres. CGRP microspheres significantly enhanced rat bone marrow mesenchymal stem cell proliferation, migration capacity, and osteogenic differentiation compared to all other treatment groups. Mechanistic investigations confirmed activation of the cAMP/PKA/CREB signaling pathway with upregulation of osteogenic transcription factors (Runx2, Osterix) and bone matrix proteins (osteopontin, osteocalcin). In a rat femoral distraction osteogenesis model, CGRP microspheres demonstrated superior bone regeneration compared to control, blank microspheres, and free CGRP groups: substantially increased bone mineral density, significantly improved biomechanical properties, and accelerated bone formation. Histological analysis confirmed enhanced bone maturation and integration This engineered sustained-release system represents a promising therapeutic platform for enhancing bone regeneration through targeted molecular pathway activation, offering significant potential for clinical translation in orthopedic applications.

The poor mechanical performance of the aerogels hampered their potential applications in bone tissue engineering. To reinforce their mechanical properties, an electrospinning nanofibers-incorporated aerogels were fabricated in this study. Here, silk fibroin solution is blended with chitin in various proportions, and then electrospun to produce the hybrid nanofibers. The silk fibroin/chitin hybrid nanofibers were incorporated into the SF-based composite aerogels to enhance their mechanical properties. nHA was deposited onto the surface of the nanofiber-incorporated aerogels via in situ biomineralization, leading to the nanofibers/nHA-reinforced composite aerogels. The as-prepared composite aerogels exhibit favorable mineralization abilities through immersing in simulated body fluid. SEM observation suggested that the mineralization degree of the SF-based aerogels was positively related with the content of electrospinning nanofibers in the composite aerogels. The results obtained from the compress stress-strain curves revealed that the incorporation of nanofibers and in vitro biomineralization of nHA both enhanced the mechanical performance of the composite aerogels. The osteogenic ability of the prepared aerogels was also improved after incorporation of NF SF-0.75%CT and nucleation of nHA in SBF. Therefore, the composite aerogels prepared by the incorporation of NF SF-0.75%CT and immersing into SBF exhibited an enhanced mechanical properties and improved osteogenic ability.

Background Prostate cancer (PCa) ranks second in the incidence of all malignancies in male worldwide. The presence of multi-organ metastases and tumor heterogeneity often leads to unsatisfactory outcomes of conventional radiotherapy treatments. This study aimed to develop a novel folate-targeted nanohydroxyapatite (nHA) coupling to deliver adriamycin (Doxorubicin, DOX), 32P, and 99mTc simultaneously for the diagnosis and treatment of prostate-specific membrane antigen (PSMA) positive prostate cancer. Methods The spherical nHA was prepared by the biomimetic method and characterized. Folic acid (FA) was coupled to nHA with polyethylene glycol (PEG), and the grafting ratio of PEG-nHA and FA-PEG-nHA was determined by the thermogravimetric analysis (TGA) method. In addition, 32P, 99mTc, and DOX were loaded on nHA by physisorption. And the labeling rate and stability of radionuclides were measured by a γ-counter. The loading and release of DOX at different pH were determined by the dialysis method. Targeting of FA-PEG-nHA loaded with 99mTc was verified by in vivo SPECT imaging. In vitro anti-tumor effect of 32P/DOX-FA-PEG-nHA was assessed with apoptosis assay. The safety of the nano-drugs was verified by histopathological analysis. Results The SEM images showed that the synthesized nHA was spherical with uniform particle size (average diameter of about 100nm). The grafting ratio is about 10% for PEG and about 20% for FA. The drug loading and the delayed release of DOX at different pH confirmed its long-term therapeutic ability. The labeling of 32P and 99mTc was stable and the labeling rate was great. SPECT showed that FA-PEG-nHA showed well in vivo tumor targeting and less damage to normal tissues. Conclusion FA-targeted nHA loaded with 32P, 99mTc, and DOX may be a new diagnostic and therapeutic strategy for targeting PSMA-positive prostate cancer tumors, which may achieve better therapeutic results while circumventing the severe toxic side effects of conventional chemotherapeutic agents.
