Moderation of cross linkage of sodium alginate-polyethylene oxide films loaded with natamycin for treatment of Aspergillus fumigatus keratitis

Chi Menghui,Zhang Leyuan,Deng Bing,Lin Jing,Wang Qian,Zhang Lina,Yin Min,Gu Lingwen,Li Daohao,Zhao Guiqiu,Li Cui

The blindness rate of fungal keratitis is high, however, traditional therapy, like eye drops, has poor bioavailability. To make the traditional treatment more effective, new drug-loading system was explored, which can attach to the ocular surface to prolong the release time of natamycin (NATA). Sodium alginate (SA) has attractive properties of biocompatibility, biodegradability, which have been exploited to be natural-origin polymer of drug release. Because of special egg box structure of SA, the sodium ions in the structure can be exchanged with divalent cations through crosslinking, which could control the pore size inside the material and release rate of the loaded drug. Here, we utilized the composite of sodium alginate and polyethylene oxide (PEO) with natamycin loaded through crosslinking with calcium ion ethanol aqueous solution to delay drug release and treat fungal keratitis. The results from experiments proved that the membrane with the slowest rate of drug release was the group with the ratio of ethanol to water 2:1, and the 1% natamycin-loaded films could effectively inhibit the growth of Aspergillus fumigatus . SA-PEO membranes could reduce the inflammatory response. Conclusively, NATA-SA-PEO films could be considered a useful approach to prolong ocular natamycin maintenance and improve the outcome of fungal keratitis. Graphical

Apo-Transferrin Modified Natamycin-Loaded Liposomes Improve the Prognosis of Fungal Keratitis via Enhanced Antifungal and Anti-Inflammatory Activities

Luwen Hu,Junjie Luan,Xing Liu,Zhaoyang Zhang,Zhenglu Tong,Yutian Qu,Zhongtai Jiang,Jing Lin

Background The proliferation and establishment of infections by pathogenic microorganisms require essential nutrients, such as host-derived iron. The sequestration of iron by the host represents an ancient defense mechanism against microbial invasion. Apo-Transferrin (Apo-Tf) is a promising and novel antimicrobial agent with intrinsic iron sequestration properties and therefore merits clinical investigation. Given the characteristics of natamycin (NAT) such as its low aqueous solubility and significant ocular irritation, we developed apo-Tf decorated liposomes encapsulating NAT for the treatment of fungal keratitis (FK).Methods NAT was encapsulated into liposomes using the lipid film hydration method. Natamycin-loaded transferrin-conjugated liposomes (NAT@TF-LP) were synthesized via the post-insertion technique and characterized through zeta-sizer analysis, morphology and Fourier transform infrared (FTIR) spectroscopy. The encapsulation efficiency, loading capacity, and in vitro release profile were determined by ultraviolet spectrophotometry. The cytotoxicity, ocular surface irritation, and systemic safety of liposomal formulations were evaluated in corneal epithelial cells and mice. In vitro studies investigated the antifungal properties of NAT@TF-LP, while in vivo experiments assessed eye surface retention and therapeutic efficacy in an FK mouse model. Results The prepared liposomes exhibited a well-defined nanostructure with uniform dimensions and apo-Tf was effectively conjugated to the surface, as verified by the emergence of characteristic amide bands in FTIR analysis. The loading efficiencies of NAT@LP and NAT@TF-LP were 5.6% and 3.53% respectively, with encapsulation efficiencies above 96%. Both formulations facilitated sustained NAT release and prolonged ocular surface retention. The in vitro release of NAT from NAT@TF-LPs followed a diffusion-controlled profile, exhibiting an excellent fit to both Higuchi and Korsmeyer–Peppas models. Additionally, they exhibited good biosafety profiles in both cytotoxicity and ocular irritation assays. In vivo, NAT@TF-LPs demonstrated superior therapeutic efficacy, resulting in a 15.4-fold reduction in corneal fungal burden (approximately 1.19-log reduction) compared to the free NAT group, alongside a marked decrease in the protein expression of pro-inflammatory cytokines, including IL-1β (65.0%), IL-6 (59.0%), and TNF-α (73.0%), with a 69.3% enhancement in anti-inflammatory IL-10 levels. Conclusion The apo-Tf functionalized liposomal system improves NAT ocular bioavailability and exhibits augmented antifungal and therapeutic efficacies. This enhancement is attributed to the combined benefits of the liposomal platform and the inherent antimicrobial properties of apo-Tf. This study highlights the promising potential of NAT@TF-LPs for the pharmacotherapy of FK.

Facile fabrication of 3D graphene–multi walled carbon nanotubes network and its use as a platform for natamycin detection

Xiaopeng Yang,Xiang Yu,Yan Heng,Fei Wang

In this work, a three dimensional graphene-multiwalled carbon nanotubes (3DG-MWCNTs) network is successfully fabricated on the surface of glassy carbon electrode (GCE) using pulse potential method (PPM) in a graphene oxide-multi walled carbon nanotubes (GO-MWCNTs) homogeneous dispersion. Scanning electron microscopy (SEM) images and electrochemical experiments confirmed the 3D structure of interpenetrated graphene-MWCNTs network and excellent electrochemical performance. The 3DG-MWCNTs network was presented to develop a sensitive voltammetric sensor for detection of natamycin . Systematic electrochemical tests demonstrate that the 3DG-MWCNTs network modified electrode can effectively increase response to the oxidation of natamycin due to the large accessible surface area as well as its excellent conductivity. Under optimized conditions, the voltammetric sensor showed a linear relationship in the range of 5.0 × 10 −8 –2.5 × 10 −6  mol L −1 and a low detection limit of 1.0 × 10 −8  mol L −1 . Moreover, the proposed method was also applied for detection of natamycin content in real samples (commercial red wine and beverage).

Temperature and osmotic pressure dual-stimuli-responsive ophthalmic gels embedded with HPCD-modified nanoparticles for prolonged-release delivery of natamycin

Lin Bixia,Fan Fangfei,Xu Hui,Li Xin,Li Yufei,Qin Chunya,Zhang Xiaoyun,Zhang Shiqiang,Gao Yang,Niu Lirong

Fungal keratitis (FK) is a severe ocular disease that may cause blindness in severe cases. The only FDA-approved first-line treatment drug is natamycin (NAT). However, NAT bioavailability is less than 2% due to special environment of the eye. To address this issue, hydroxypropyl-β-cyclodextrin (HPCD) was applied in this study to form a hydrophilic coating on the surface of nanoparticles. The FTIR analysis suggested interactions between HPCD and the nanoparticle components, which may contribute to the enhanced colloidal stability. As a result, the nanoparticles with a hydrophilic membrane (F2) demonstrated more superior stability than the plain nanoparticle (F1). Furthermore, compared to the NAT suspension, surface wetting of F2 enhanced significantly, the cumulative drug release reduced by 27.34% at 72 h, isolated corneal permeability increased 2.89-fold, as well as protein binding was upgraded 6.1-fold at 30 s. Subsequently, F2 was incorporated into a temperature- and osmolarity dual-responsive ophthalmic gel (Tem-Osm gel). The Tem-Osm gel behaved a shear-thinning non-Newtonian fluid, which exhibited excellent biocompatibility and higher ocular bioavailability. Moreover, the in vitro antifungal experiments demonstrated that F2 and Tem-Osm gel exhibited more superior antifungal activity, lower MIC/EC50 values and larger inhibition zones than the NAT suspension. Overall, the study indicates that the novel combination of nanoparticle and multi-responsive ocular gel sustained-release delivery system holds promising potential as a therapeutic approach for fungal keratitis.

Ferulic Acid-in-CD-MOF-1 and Natamycin Nanocapsules as a Dual-Functional System for Antioxidant and Antifungal Litchi Preservation

He Wanjun,Lv Danyu,Wu Qingxiao,Xu Huanyu,Guan Yongguang,Zeng Xin-An

Litchi ( L. chinensis Sonn.) is a perishable fruit that is vulnerable to oxidative browning and fungal infection. Herein, ferulic acid and natamycin were chosen as the antioxidant and antimycotic agents, respectively. To improve their water solubility, we used CD-MOF-1 to encapsulate ferulic acid. Meanwhile, natamycin was encapsulated with gum arabic and pullulan. The most homogeneous and stable natamycin nanocapsules were fabricated through stirring 0.5 mg/mL natamycin using 2.5% (w/v) gum arabic-pullulan (1:1, w/w, 1.25% each) in water. The main encapsulation mechanisms of ferulic acid and natamycin were electrostatic interactions between the carboxyl groups of ferulic acid and K + ions from CD-MOF-1 and hydrophobic interactions between the macrocyclic lactone ring of natamycin and the protein part of gum arabic, respectively. In addition, pullulan was indispensable for stabilizing natamycin. The antioxidant and antifungal composite sol presented 99.85% oxidation inhibition and 79.06% mycelial growth inhibition and effectively delayed the oxidative browning and fungal decay on litchi pericarps.

Dual-Layer Natamycin and Boric-Acid-Reinforced PVA/Chitosan by 3D Printing and Electrospinning Method: Characterization and In Vitro Evaluation

Büsra Oktay,Fatih Ciftci,Azime Erarslan,Esma Ahlatcıoğlu Özerol

This study presents the development and comprehensive characterization of biopolymer-based nanofibrous composites composed of polyvinyl alcohol (PVA), chitosan (CS), boric acid (BA), and a natural antifungal agent natamycin (NAT), designed for therapeutic applications. A dual-layer 3D-fiber composite (PVA/CS/BA_PVA/NAT) was successfully fabricated using a layer-by-layer 3D bioprinting technique and electro-spinning, integrating BA into the core matrix and NAT into the outer layer. Mechanical tests revealed a significantly improved elastic modulus of 763.04 ± 14.54 MPa and the highest ultimate tensile stress (50.45 ± 2.58 MPa) among all samples. Despite a moderate strain at break (11.77 ± 0.49%), the composite preserved sufficient elasticity suitable for biological interfaces. Morphological assessment via SEM confirmed the successful deposition of continuous and bead-free nanofibers, with controlled fiber alignment and reduced average fiber diameters, especially in the BA-incorporated structure. The dual-layered system displayed enhanced uniformity and structural coherence. The drug release analysis demonstrated sustained NAT delivery over a 90 min period. Kinetic modeling showed a high correlation with the Korsmeyer–Peppas model (R2> 0.99), suggesting diffusion-controlled release, supported by the Korsmeyer–Peppas model’s Fickian diffusion exponent. In contrast, zero- and first-order models exhibited weaker fits, underscoring the relevance of a matrix-based release mechanism governed by the layered configuration. Crucially, antifungal assays againstCandida albicansrevealed substantial bioactivity. The PVA/CS/BA_PVA/NAT formulation achieved the largest inhibition zone (1.64 ± 0.13 cm), significantly outperforming single-layer controls such as PVA/CS/BA (1.25 ± 0.08 cm) and PVA/CS_PVA/NAT (1.43 ± 0.08 cm), while neat PVA exhibited no inhibition. These results confirm the synergistic antifungal efficacy of BA and NAT within the dual-layer structure. Together, these findings highlight the potential of the 3D-printed PVA/CS/BA_PVA/NAT composite as a mechanically robust, morphologically optimized, and bioactive platform for antifungal therapy and wound-healing applications.

Fabrication of antimicrobial PCL/EC nanofibrous films containing natamycin and trans-cinnamic acid by microfluidic blow spinning for fruit preservation

Menglu Wu,Zi-An Deng,Chaoyi Shen,Zhichao Yang,Zihan Cai,Di Wu,Kunsong Chen

Fruit is susceptible to various postharvest pathogens; thus, the development of multifunctional preservation materials that can achieve the broad-spectrum inhibition of different pathogens is a current research hotspot. Here, microfluidic blow spinning was used to create a biodegradable polycaprolactone/ethyl cellulose (PCL/EC) nanofibrous film that incorporated two naturally-sourced compounds, natamycin and trans -cinnamic acid, resulting in multi-microbial inhibition. The PCL/EC-based film had a smooth and even morphology, indicating the favorable integration of PCL and EC. After the incorporation of ingredients, the film exhibited good inhibitory activity against Escherichia coli , Staphylococcus aureus , and Botrytis cinerea , and it had finer fiber diameters, higher permeability, and antioxidant properties. We further demonstrated that strawberries that were padded with the film had good resistance to Botrytis cinerea . Also, the film did not interference with the qualities of the strawberries during storage. The study demonstrates a promising application for multi-antimicrobial and bio-friendly packaging materials in postharvest fruit preservation.

Fabrication of colloidal stable gliadin-casein nanoparticles for the encapsulation of natamycin: Molecular interactions and antifungal application on cherry tomato

Xiaochuan Wu,Qiuyun Hu,Xianrui Liang,Sheng Fang

Natamycin was encapsulated in gliadin-casein nanoparticles (G-C NPs) to control black rot in cherry tomato against Alternaria alternata . The G-C NPs with a mean particle diameter of 211 ± 4 nm were prepared using an anti-solvent method. The composite NPs showed better stability against neutral pH, ion, and storage than gliadin NPs. The quenching of gliadin by natamycin was static with a constant of 5.99 × 10 −12 M −1 ∙S −1 , and was spontaneous with a free energy of −23.5 kJ∙M −1 at 298 K. Both hydrophobic stacking and hydrogen bonds between natamycin and gliadin were found as the major driven force in the formation of the complex. The NPs kept the antifungal activity of natamycin with improved photostability. The NPs coatings exhibited better results than natamycin in controlling black rot on cherry tomato. This study shows the potential of the G-C NPs as all-natural delivery systems for natamycin in post-harvest treatments.

Enhancing Water Solubility and Stability of Natamycin by Molecular Encapsulation in Methyl-β-Cyclodextrin and its Mechanisms by Molecular Dynamics Simulations

Fang Sheng,Peng Xinling,Liang Xianrui,Shen Jian,Wang Jian,Chen Jie,Meng Yuecheng

In this study, the antifungal compound natamycin was encapsulated in methyl- β -cyclodextrin (heptakis(2,6-di-O-methyl)- β -cyclodextrin, Me- β -CD) to improve its aqueous solubility and stability. The aqueous solubilities of natamycin in the presence of β -CD, 2-hydroxypropyl- β -CD, 6-O- α -maltosyl- β -CD, and Me- β -CD were compared. The Me- β -CD showed the best result to increase the solubility of natamycin in aqueous. The pH stability of natamycin was improved by the formation of inclusion complex with Me- β -CD, especially at acidic conditions. The degradation of natamycin under UV-light exposure followed first-order kinetics with half-life times ( t 1/2 ) of 59.2 and 157.5 min in pure form and Me- β -CD inclusion complex, respectively. The in vitro antifungal activities of natamycin/Me -β -CD complex against Aspergillus niger food pathogen were evaluated. The results demonstrated that the natamycin/Me -β -CD complex could effectively improve the aqueous solubility and photostability of natamycin without compromising in antifungal activities. Finally, the molecular inclusion mechanisms and geometrical configurations of the natamycin/Me -β -CD complex were studied using molecular dynamics simulations. This research may lead to the development of more effective inclusion-based delivery systems to encapsulate and protect lipophilic antimicrobial agents for food applications.

Sensitive determination of natamycin based on a new voltammetric sensor: a single-walled carbon nanotube composite poly(L-serine) film modified electrode

Zhuo Ye,Lingxi Yang,Jianguo Wen,Baoxian Ye

A novel voltammetric sensor, based on a single-walled carbon nanotube (SWNT) composite poly(L-serine) film modified electrode, was fabricated by simple dipping–drying and electrodeposition methods and used for highly sensitive determination of natamycin. At this sensor, the electrochemical response of natamycin was improved greatly due to the synergistic effect of SWNTs and poly(L-serine). A new voltammetric method for the determination of natamycin was proposed with a detection linear range of 6 × 10−8 mol L−1 to 6 × 10−6 mol L−1 and a detection limit of 4 × 10−8 mol L−1. Furthermore, the electrode reaction of natamycin was investigated in detail and some dynamic parameters were calculated for the first time. Using the present method, the residual quantity of natamycin in available claret and beverage samples was evaluated with good satisfactory recovery.

Effect of radio frequency energy combined with natamycin on Aspergillus niger survival and quality of dried shiitake mushroom with different moisture content

Lihui Zhang,Jialing Zhu,Shaojin Wang,Long Chen,Zihan Song,Lei Zhang,Haile Ma

Effectively controlling molds in dried shiitake mushrooms is important for food industry to maintain the product quality. To improve the edible safety of shiitake mushrooms, this study applied radio frequency (RF) energy combined with natamycin to inactivate Aspergillus niger spores, and evaluate the quality changes after the treatment. The results showed that the population of A. niger spores in shiitake mushrooms with different moisture contents (MCs) could achieve a 5-log (CFU/g) reduction after the combined treatment at different temperatures. Weibull distribution ( R 2  = 0.978 to 1.000) provided a better fit for the inactivation kinetics of A. niger spores than Linear model ( R 2  = 0.954 to 0.990) and Logistic one ( R 2  = 0.953 to 0.988). Meanwhile, the three-dimensional inactivation curves were constructed based on the sample MCs, RF heating time, and holding time. In addition, the MCs of treated shiitake mushrooms decreased by 4.12%–7.66% (w.b.) after natural cooling, achieving synchronous drying. There was no significant difference in textural properties of shiitake mushrooms after the combined treatment. The changes in color, structure, rehydration ratio, and total sugar content were affected by the treatment time and temperatures. As one of the main aroma substances, the content of sulfur-containing compounds in shiitake mushrooms after treatment increased compared to that of the control samples. This study indicates that the combination of RF energy and natamycin is feasible for inactivating pathogens in shiitake mushroom and ensuring the safety of their consumption.

Preparation of copper sulfate/natamycin/carboxymethylcellulose/agar film for synergistic antibacterial application in fruit via causing cuproptosis of Escherichia coli O157: H7

Jinyue Sun,Han-Xiao Zhuang,Jia-Neng Pan,Qian-Jun Shen,Hui-Ling Shen,Wen-Wen Zhou

This study has investigated cuproptosis caused by copper sulfate (CuSO 4 ) in Escherichia coli O157: H7 and its mechanism. Meanwhile, the mechanism of CuSO 4 combined with natamycin (Nata) against E. coli O157: H7 planktonic and biofilm cells were also investigated. Finally, we prepared an agar-based film, researched its properties, and used it for packaging cherry tomatoes and purple kale. The results indicated that CuSO 4 could cause cuproptosis in E. coli O157: H7 and lead the genes related to Fe-S cluster protein ( hyfA , sufB , xdhC , and narY ) to down-regulate and cuproptosis-related proteins to change. Then, CuSO 4 combined with Nata could synergistically decrease E. coli O157: H7 bacterial counts in planktonic condition and biofilm by synergistically affecting the membrane potential, cytoplasm leakage, permeability of outer membrane and inner membrane. After that, CuSO 4 and Nata could change the mechanical properties, color and optical characteristics of agar-based film, and decrease the water vapor permeability and oxygen permeability value. Finally, CuSO 4 @Nata@Carboxymethylcellulose@Agar film effectively lowered the counts of E. coli O157: H7 in cherry tomatoes and purple kale during the storage.
