Seven-Core Tapered Optical Fiber LSPR Biosensor with CMC Structure for Ultra-Sensitive Detection of Paraoxon in Environmental Monitoring

Zhenpeng Men,Ragini Singh,Bingyuan Zhang,Santosh Kumar

In this paper, we introduce a seven-core tapered optical fiber biosensor with high sensitivity for label-free detection of paraoxon compounds that utilizes the localized surface plasmon resonance (LSPR) effect. By its unique circle-M-circle (CMC) structure surrounding the fiber surface, it achieves an obvious amplification of evanescent field at the sensor and improves sensitivity greatly. Finally, the optimized tapered shape enhances surface plasmon light interaction and hence increases sensitivity to low concentrations of paraoxon residues. Since gold nanoparticles (AuNPs) are used as an active probe for surface modification, the optical properties of Au make them have a good LSPR effect. We also employ, as auxiliary materials, tungsten disulfide (WS2) thin layers and ZnO nanowires (NWs), of which the biocompatibility advantage can reinforce together with stability for detection probes to the WaveFlex sensor. The results of the experiment show a high sensitivity for detecting paraoxon residues in agricultural products: 33.8 ng/mL and a detection limit of 6.83 μg/mL. The developed biosensor enables portability through miniaturization and also maintains its performance stability in various environmental conditions, which can be considered a powerful tool for field monitoring of paraoxon to enhance both environmental and food safety assessments. The sensor has shown favorable practical applicability due to at least desirable selectivity and repeatability. The CMC-like optical fiber-based WaveFlex biosensor being developed based on LSPR detection represents a great progress of sensor-based technology for environmental monitoring with wide application scenarios, which serves as an efficient and effective surveillance mechanism for point-of-sampling detection to the timely-onset paraoxon analysis.

Fabrication and Evaluation of Rapidly Dissolving Microneedles Loaded with Organophosphorus Hydrolase for the Treatment of Transdermal Ethyl Paraoxon Poisoning

Fengqian Cui,Xue Liang,Ming Ma,Yanan Zhai,Jing Gao

Background: Given the severe toxicity of organophosphates and the efficient catalytic hydrolysis of organophosphates by organophosphorus hydrolase (OPH) against them, a rapidly dissolving microneedle patch loaded with OPH offers a promising therapeutic strategy to provide timely detoxification conveniently. Methods: The microneedles were prepared with sulfobutylether-β-cyclodextrin (SCD) matrix to load OPHDS5, one of the mutants of OPH, and were characterized in vitro and in vivo to determine tip loading capacity, skin insertion efficiency, and protective efficacy against ethyl paraoxon poisoning. Results: Microneedles prepared with 60%w/vSCD displayed well-defined square-pyramidal tips and a mechanical strength of 4 N/needle at 1200 μm. When containing 1.2 mg OPHDS5, the microneedles enabled SD rats to completely survive a lethal challenge of ethyl paraoxon, achieving a 100% survival rate, significantly alleviating the symptoms resulting from transdermal exposure and markedly attenuating the symptoms resulting from transdermal exposure. Conclusions: A rapidly dissolving microneedle patch encapsulating OPH was successfully developed. The patch effectively penetrated the skin and exhibited robust prophylactic and therapeutic activity against ethyl paraoxon poisoning.

Smartphone-assisted bioenzyme-nanozyme-chromogen all-in-one test strip with enhanced cascade signal amplification for convenient paraoxon sensing

Hengjia Zhu,Lizhang Xu,Panwang Hu,Bangxiang Liu,Mengzhu Wang,Xiulian Yin,Jianming Pan,Xiangheng Niu

Monitoring of pesticide residues in food and environmental matrices is undoubtedly crucial to guarantee food safety and ecological health, yet how to realize their sensitive and convenient detection is still challenging. Herein, we propose an all-in-one test strip that elaborately integrates bioenzyme, nanozyme and chromogen together, and achieve the highly sensitive and convenient sensing of pesticide residues assisted by a smartphone. A sequential self-assembly strategy was first explored to acquire an integrative bioenzyme-nanozyme-chromogen assembly, and then the assembly was confined in a biocompatible hydrogel to construct the test strip. Thanks to both the proximity and confinement effects, a ∼1.2-fold improvement of the cascade catalytic efficiency was gained to benefit high-sensitivity detection. More importantly, since all the sensing elements, including target recognition units and signal amplification modules, were rationally integrated in the test strip, detection operation was significantly simplified, making it possible for in-field rapid analysis. Besides, the microenvironment provided by the alginate hydrogel carrier endowed the test strip with an excellent sensing stability. By taking paraoxon as a typical pesticide, high-performance detection of the target was accomplished via the smartphone-assisted all-in-one test strip. Moreover, the test strip was successfully applied for paraoxon detection in various real samples and exhibited good correlations with commercial kits, demonstrating its great prospect for practical applications. Our work not only offers a new tool for the high-sensitivity and convenient monitoring of pesticide residues, but will also inspire the development of efficient multi-enzyme sensing platforms.

Enzyme-free carbon dots@MgO nanocomposite as an efficient sensor for on-site detection and degradation of paraoxon toxins

Bo Liu,Meijie Cai,Xiangling Feng,Siyu Lu,Song Lin,Feng Tian

Serial nanocomposites consisting of blue fluorescent carbon dots (B-CDs) and MgO nanoparticles were facilely prepared. Then on-site detection and degradation of ethyl-paraoxon were achieved and evaluated. It was revealed that ethyl-paraoxon was firstly decomposed into p-nitrophenol by MgO. Then, p-nitrophenol could significantly quench the fluorescence of decorated B-CDs due to an inner filter effect (IFE). The 0.05B-CDs@MgO showed excellent comprehensive performance among the serial xB-CDs@MgO samples due to its relatively high degradation ability towards ethyl-paraoxon and potent initial fluorescence emission intensity at an excitation wavelength of 360 nm between pH 4 and 10. Finally, based on the IFE, on-site paper sensors via color recognition app on a smartphone were explored compared with a conventional fluorescence spectrometer. Both the quantitative fluorescence intensities and G/B values showed the multi-piecewise linear correlations to ethyl-paraoxon concentration up to 3000 μg/mL in river water with a limit of detection to 0.06 μg/mL and 3.48 μg/mL as well as high recovery rates between 99%-102% and 95%–116%, respectively. The in vitro cytotoxicity and phytotoxicity assessment of the 0.05B-CDs@MgO revealed dose-dependent effects. Such portable and integrated fluorescent B-CDs@MgO nanocomposites on paper arrays show excellent potential in shortening the detection time and reducing the detection cost as a new strategy for the on-site quantitative detection and degradation of ethyl-paraoxon in actual river water.

Highly sensitive electrochemical detection of paraoxon ethyl in water and fruit samples based on defect-engineered graphene nanoribbons modified electrode

Zhao Yong,Ma Yuzhi,Zhou Ruiyong,He Yong,Wu Yuntao,Yi Yinhui,Zhu Gangbing

In this work, defect-engineered graphene nanoribbon (DGNR) was prepared and used for the first time as a novel electrode material for the sensitive detection of parathion methyl (MP). The morphology, structure and component of DGNR were investigated via scanning electron microscope, transmission electron microscopy, Fourier transform infrared spectroscopy and Raman spectra; and the detection conditions for MP were optimized. Owing to the large surface area, superior conductivity, rich edge defects and active sites, DGNR modified glassy carbon electrode (GCE) shows excellent sensing performances for MP. Under optimized conditions, a considerable low detection limit of 4.3 nM and wide linearity from 0.01 to 25.0 μM were obtained. In addition, the applications of the proposed DGNR/GCE in water and fruit samples possess satisfactory recoveries from 95.7 to 106.4%. This study offers a promising application for DGNR-based materials in developing simple, low cost and highly sensitive sensing protocol for MP and other pesticides. Graphical abstract

Catalytic preference-enabled exclusive bimodal detection of methyl-paraoxon in complex food matrices using double site-synergized organophosphorus hydrolase-mimetic fluorescent nanozymes

Qingzhen Tian,Jinjin Liu,Yuxuan Long,Hao Liang,Kechen Wu,Xi Chen,Qinqin Bai,Xiangheng Niu

Pesticide sensing crucially safeguards food safety and public health against environmental and health hazards. While oxidoreductase-type nanozymes (peroxidase and oxidase) have been widely used in optical pesticide detection, their susceptibility to redox interference as well as poor target specificity limits practical applications. To overcome the deficiencies, here we developed Ca 2+ -chelated 2-aminoterephthalic acid on nanosized ceria (Ca-ATPA@CeO 2 ) as an organophosphorus hydrolase mimic. This design integrates stable fluorescence and dual-site catalytic activity to specifically detect methyl-paraoxon (MP) in complex food matrices. The synergy between Ca 2+ (hard Lewis acid) and CeO 2 creates dual active sites to catalyze MP hydrolysis into yellow p -nitrophenol (pNP), and the latter quenches nanozyme fluorescence via inner filter effect. The system enables cross-validated quantification of MP in complex samples, eliminating redox interference through target-specific catalysis. The bimodal “on” colorimetric (pNP color signal) and “off” fluorescence (nanozyme fluorescence intensity) detection achieved linear ranges within 1–200 μM, providing detection limits of 1.43 μM and 0.087 μM, respectively. Our work proposes a reliable strategy for selective MP detection that can avoid redox interference, also providing a simple yet efficient design of high-activity fluorescent hydrolase mimics with broadened applications in food safety analysis and beyond.

Direct and specific detection of methyl-paraoxon using a highly sensitive fluorescence strategy combined with phosphatase-like nanozyme and molecularly imprinted polymer

Xuan Zhang,Nan Hao,Shucheng Liu,Kai Wei,Changchang Ma,Jianming Pan,Sheng Feng

Methyl paraoxon (MP) is a highly toxic, efficient and broad-spectrum organophosphorus pesticide , which poses significant risks to ecological environment and human health. Many detection methods for MP are based on the enzyme catalytic or inhibition effect. But natural biological enzymes are relatively expensive and easy to be inactivated with a short service life . As a unique tool of nanotechnology with enzyme-like characteristics, nanozyme has attracted increasing concern. However, a large proportion of nanozymes lack the intrinsic specificity, becoming a main barrier of constraining their use in biochemical analysis. Here, we use a one-pot reverse microemulsion polymerization combine the gold nanoclusters (AuNCs) with molecularly imprinted polymers (MIPs), polydopamine (PDA) and hollow CeO 2 nanospheres to synthesize the bright red-orange fluorescence probe (CeO 2 @PDA@AuNCs-MIPs) with high phosphatase-like activity for selective detection of MP. The hollow structure possesses a specific surface area and porous matrix, which not only increases the exposure of active sites but also enhances the efficiency of mass and electron transport . Consequently, this structure significantly enhances the catalytic activity by reducing transport distances. The introduced MIPs provide the specific recognition sites for MP. And Ce (III) can excite aggregation induced emission of AuNCs and enhance the fluorescent signal. The absolute fluorescence quantum yield (FLQY) of CeO 2 @PDA@AuNCs-MIPs (1.41 %) was 12.8-fold higher than that of the GSH-AuNCs (0.11 %). With the presence of MP, Ce (IV)/Ce (III) species serve as the active sites to polarize and hydrolyze phosphate bonds to generate p -nitrophenol (p-NP), which can quench the fluorescent signal through the inner-filter effect. The as-prepared CeO 2 @PDA@AuNCs-MIPs nanozyme-based fluorescence method for MP detection displayed superior analytical performances with wide linearities range of 0.45–125 nM and the detection limit of 0.15 nM. Furthermore, the designed method offers satisfactory practical application ability. The developed method is simple and effective for the in-field detection.

Cu-C3N4 nanoenzyme-based freezing-dried bioactive capsule integrated with 3D-printed smartphone platform for visual detection of organophosphorus pesticides paraoxon in scallion

Guorui Chang,Sirui Li,Yunqi Wang,Qingxia Ran,Qing Tan,Shuli Gou,Haiying Du,Shuxia Xu

Rapid detection of pesticide residues in vegetables has recently attracted much attention. Herein, a Cu-modified graphitic carbon nitride nanomaterial (Cu-C 3 N 4 ) with peroxidase (POD)-like property was synthesized to construct an enzyme-based colorimetric sensing platform for the detection of organophosphorus pesticides paraoxon . Cu-C 3 N 4 could catalyze H 2 O 2 produced by reaction of acetylcholine with acetylcholinesterase and choline oxidase, and then further oxidized 3,3,5,5-tetramethylbenzidine (TMB) to produce a blue colored product. Presence of organophosphorus pesticide paraoxon (PA) restrained the activity of acetylcholinesterase AChE, triggering a remarkably decreased amount of H 2 O 2 towards TMB oxidation, which led to the inhibition of oxidation reaction. After that, the reaction solutions were freeze-dried in a portable bioactive capsule with addition of trehalose as stabilizer for improved enzymes activities . On the basis of the variation in gray-scale values of blue colored TMB solutions using an application, a smartphone-based 3D printing platform was obtained with a linear range from 100 to 33,000 nM (R 2 =   0.994) and a detection limit of 13 nM (3 σ/k ). This smartphone-based 3D-printed colorimetric platform combined with a bioactive capsule offers a novel approach with the potential for rapid detection of PA in vegetables.

Preparation of aerogel Mg(OH)2 nanosheets by a combined sol–gel-hydrothermal process and its calcined MgO towards enhanced degradation of paraoxon pollutants

Liu Shisheng,Wei Xiaohui,Lin Song,Guo Minjie

The preparation of MgO nanoparticles with relatively small sizes and highly active surface areas is still a great challenge nowadays. The principal objective of this work was to investigate and optimize the calcined conditions of MgO with smaller sizes and higher surface areas, which contributed to its better degradation ability towards paraoxon pollutants. Firstly, the aerogel Mg(OH) 2 nanosheets with an increased surface area of 349.0 m 2 /g was prepared via a combined sol–gel-hydrothermal method. Then the calcined MgO nanoparticles with different morphology, size dimensions, and specific surface areas were obtained at different calcination temperatures from 400 to 800 °C. The optimized average grain size of MgO obtained at 600 °C was achieved to 8.7 ± 3.2 nm with a relatively high surface area up to 231.4 m 2 /g. Meanwhile, such as-prepared MgO nanoparticles showed an excellent performance towards paraoxon degradation, and the maximum degradation amount of paraoxon-ethyl was up to 57.8 mg/g@90 min at room temperature. The kinetics of degradation was consistent with the pseudo-second-order model ( R 2  > 0.99). Such excellent degradation capacity was derived from more generations of superoxide radical (•O 2 − ) sites as revealed by using the nitro blue tetrazolium (NBT) as a probe. The results indicated that the MgO nanoparticles reported herein could be an effective candidate for the environmental remediation of organophosphorus toxin pollutants due to their facile scale-up, low cost, eco-friendly characteristic, and high removal efficiency.

Biomimetic engineering of active Zn-doped macrocyclic polyamine carbon dots as organophosphorus hydrolases-like nanozymes for colorimetric and fluorescence on-site detection of methyl-paraoxon

Xiaoyu Deng,Feng Cao,Yanli Li,Lijun Wang,Xianggui Chen,Jun Li,Zhouyu Wang,Xiaoqi Yu

Organic phosphate esters compounds, especially methyl-paraoxon (MP), pose a serious threat to the environment and human health. The development of robust organophosphorus hydrolases (OPHs)-like nanozymes to mimic natural enzymes is paramount for on-site monitoring MP. Herein, we reported a biomimetic engineering of highly active OPHs-like nanozymes through Zn-doped macrocyclic polyamine carbon dots (DOTA(Zn)-CDs). XPS confirmed Zn N coordination, while FTIR revealed macrocyclic polyamine frameworks with Zn-O/N coordination shifts, together mimicking the histidine‑zinc centers of native OPHs. This design induced a remarkable improvement in OPHs-like activity, demonstrated by a superior catalytic efficiency and a significant enhancement in adsorption affinity for MP hydrolysis, as confirmed by kinetic assays and density functional theory calculations. DOTA(Zn)-CDs nanozymes served as a dual-mode sensing platform, enabling the enhancement of detection performance. This system achieved a low detection limit of 1.55 μM and 2.57 μM for colorimetric and fluorescence detection of MP, alongside exceptional selectivity against interfering species and promising stability for 35 days. Finally, a portable DOTA(Zn)-CDs-based gelatin cube sensor was successfully constructed for practical, on-site detection. This work provides a fundamental strategy for designing efficient OPHs-like nanozymes by biomimetic active site engineering and underscores their significant potential for rapid environmental, agricultural and food detection.
