Penicillium expansum is a major pathogen causing blue mold rot in fruits and vegetables. Given the increasing concerns over the use of chemical fungicides in postharvest treatments, this research explores the effects of pulsed light (PL) on the growth and patulin production of P. expansum . Our results exhibited that PL significantly suppressed spore germination and mycelial growth while impairing the cell membrane integrity. Mechanistic studies revealed that PL treatment caused disruption of antioxidant defense systems and the excessive accumulation of reactive oxygen species (ROS), leading to the oxidative damage and mitochondrial dysfunction. Moreover, PL treatment significantly downregulated the expression of key genes associated with P. expansum growth and patulin biosynthesis. Specifically, the expression levels of global regulators CreA and LaeA were decreased by 39 % and 62 %, and 47 % and 82 % after PL30 and PL90 treatments, respectively. Additionally, genes within the patulin biosynthesis gene cluster, including patA , patK , patN , and patL , were significantly downregulated. In apples with PL90-treated P. expansum , the rot onset was delayed by 4 d. On day 7, PL90 reduced lesion diameter by 87.9 %, decay incidence by 67 %, and patulin content by 95 %. The findings indicate that PL is an effective technology for controlling postharvest diseases in fruits, making it a promising alternative to chemical treatments.

The relationship between human health and patulin (PAT) in the diet is a complex and intertwined one. The development of a sensing approach for the field detection of patulin is crucial, as the current approach lacks real-time detection capabilities and is costly in terms of material and technology. This paper presents a portable ratiometric fluorescence sensor that can be used to rapidly, accurately, and efficiently detect patulin in food items at the point of origin. The sensor employs a combination of sulfhydryl functionalized gold nanoclusters (MUA-AuNCs) and blue emission carbon dots (B-CDs), which have been engineered to serve as highly effective “on-off” nanoprobes. The modified sulfhydryl ( SH) groups present on the gold clusters serve as specific recognition sites for patulin binding. The probes exhibit a discernible shift in hue, from orange-red to blue. The sensitivity detection limit (LOD) for patulin was found to be 0.019 μM, with a substantial linear correlation observed in the range of 0–2.2 μM. The objective of the combined chromatographic test strip and color recognition platform was to facilitate the sensitive, accurate, and real-time detection of patulin in foodstuffs, which is of paramount importance for the prevention of early disease. To facilitate rapid and straightforward preliminary testing of food security, it is anticipated that the integrated chromatographic strip ratiometric fluorescence sensing platform will be developed into portable home detection equipment.

Patulin (PAT) is a widespread fruit toxin. Trace-level PAT exposure can cause serious harm to human health. Herein, a multimodal PAT aptasensor was designed based on Ru(bpy) 3 2+ -based metal organic framework composited hydrogel (RuMOF@hydrogel) and versatile banana peel-derived carbonized polymer dots (BPPDs). RuMOF@hydrogel modified magnetic-electrode exhibited excellent anodic and cathodic electrochemiluminescence (ECL) emission and stability. Meanwhile, the BPPDs could enhance anodic ECL of RuMOF@hydrogel, and also show excellent fluorescence (FL) and photothermal (PT) properties. With the aid of PAT-triggered hybridization chain reaction and magnetic separation, ECL, FL, and PT responses could be recorded concurrently. The detection limit can reach as low as 0.25 fg mL −1 . The ratiometric ECL quantitation ensured the sensitivity and accuracy of this assay. And visual FL and portable PT modes contributed to the utility. Furthermore, this aptasensor demonstrated better performances than HPLC in fruit products and the protocol can be extended to determine various contaminants in foods.

A novel surface-enhanced Raman scattering (SERS) probe was developed for the selective determination of patulin using gold nanobipyramids as the SERS substrate. Patulin undergoes a selective chemical reaction with sulfhydryl groups of 4-mercaptobenzoic acid (4-MBA) to form new compounds. A substantial change of the SERS spectrum of 4-MBA was observed and a new peak at 1641 cm−1 appeared. A significant increase in the intensity of the SERS signal at 1641 cm−1 was observed upon addition of more targets. This feature was exploited for sensitive quantification of patulin in aqueous solution by ratioing the intensities of the new peak at 1641 cm−1 and the peak of 4-MBA at 1586 cm−1. This method displayed excellent selectivity for patulin. Under optimal conditions, the ratioed signal intensity is related to the concentration of patulin in the range between 10 μg L−1 and 3000 μg L−1, with a detection limit of 6 μg L−1. This method was successfully used for the determination of patulin in juice.

Patulin (PAT) is a potentially harmful mycotoxin to human health and is known to contaminate apple juice. In this work, we developed a ratiometric fluorescence aptasensor using tetrakis(4-carboxyphenyl)porphyrin (H 2 TCPP)-treated octahedral UiO-66-NH 2 (defined as UiO-66-TCPP) to detect PAT. This 2-aminoterephthalic acid and H 2 TCPP functionalized metal-organic framework showed multiple adsorption effects (hydrogen bonding and π–π stacking) on the aptamer (Apt) and served as a quenching material. When the target PAT bound specifically to the Apt, the fluorescence of the 6-carboxyfluorescein-labeled Apt would recover, and the fluorescence of the H 2 TCPP ligand remained unchanged. This ratiometric fluorescence property improved the accuracy of PAT detection. Moreover, the introduction of the H 2 TCPP ligand enhanced the quenching efficiency of UiO-66-NH 2 , thus improving the sensitivity of the fluorescent aptasensor (UiO-66-TCPP vs. UiO-66-NH 2 : 0.0162 ng/mL vs. 1.8 ng/mL). In addition, we used UiO-66-TCPP to detect PAT in apple juice samples. This work provides a good paradigm for the construction of ratiometric fluorescence aptasensors with high sensitivity and accuracy.

Patulin is a toxin that can be found in damaged or moldy fruits, especially apples. Very high detection sensitivity can be achieved using the SERS (Surface Enhanced Raman Scattering) technique, which provides information about molecular vibrations. However, unexpected desorption of target toxins from the surface of the SERS substrate and interference from other substances pose a major challenge. Here we investigate a competitive binding strategy for reliable SERS analysis of PAT. Core-Raman molecule-shell (CMS) structures and programmable, specific DNA interactions are utilized to regulate Raman signal intensity without affecting the embedded Raman molecules. Specfically, aptamer-functionalized CMS structures are combined with metal-organic scaffold nanoparticles that possess complementary DNA sequences. In the presence of PAT, binding of the aptamer to the toxin leads to degradation of the hybrid nano assembly, which can be measured by a change in the SERS signal. This SERS strategy enables turn-off sensing in apple juice over a wide range (10 pg/mL to 1 μg/mL), with a detection limit as low as 0.02 ng/mL. Analysis of real samples gives results consistent with high-performance liquid chromatography (HPLC) test. The excellent selectivity and sensitivity of the SERS sensor demonstrate its potential for a wide range of SERS sensor applications.

A novel dual-dummy templates imprinted electrochemical sensor has been fabricated for the detection of patulin. Herein, 2-oxindole (2-oxin) and 6-hydroxynicotinic acid (6-HNA) as the dummy templates, 4-aminothiophenol as functional monomer, and ionic liquid (IL) as electropolymerization electrolyte are employed to prepare molecularly imprinted polymer (MIP) film. 2-Oxin and 6-HNA have multiple groups and the obtained MIP possesses different types of imprinted sites, thereby achieving a better recognition capacity than that of single-dummy imprinted film. ILs can regulate the density of molecularly imprinted film and facilitate effective molecular recognition. The composite of PtPd decorated N-doped porous carbon has good conductivity and large surface area, and can amplify the signal. With the aid of electrochemical probe [Fe(CN) 6 ] 3-/4- (0.16 V vs. SCE) patulin can be detected. Under the optimal conditions, this sensor shows a detection range from 0.01 to 10 μg L −1 , with a detection limit of 7.5 × 10 −3 μg L −1 ( S / N = 3). Two spiked juice samples were analyzed by this method, and the recovery ranges from 94 to 99.8% with RSD values of 2.4–4.6% ( n = 3), indicating that this method can be applied for the detection of patulin in real samples. Patulin (PAT) is an unsaturated lactone mycotoxin primarily produced by Penicillium expansum and Aspergillus clavatus . Given the potential health risks and economic losses associated with PAT, the rapid detection of PAT using fluorescent aptasensors is of significant importance in evaluating food safety. However, it easily increases the cost and complexity caused by signal labeling. We combined TCPP/BDC-NH 2 mixed ligands functionalized Zr metal–organic frameworks (Zr-MOF mix ) and terminated three-stranded DNA gates (ttsDNA gates) to fabricate a label-free fluorescent aptasensor for PAT detection. The Zr-MOF mix system was synthesized via a one-pot strategy and could be used to address the problem of pore size limitation and increase the loading amounts of dyes. TtsDNA gate was integrated into the Zr-MOF mix system to control the release of dyes, exhibiting a high signal-to-background ratio. The single-stranded aptamer region in ttsDNA gate situated away from the surface of the Zr-MOF mix , resulting in a natural release of dyes in the absence of PAT. While binding to PAT resulted in target-induced conformational changes that helped form the hairpin structure of the aptamer. This structure hindered the release of dyes from the pores of Zr-MOF mix , thus reducing the fluorescence signals intensity. The stimuli-responsive DNA-gated material provides a platform for PAT analysis under conditions of a low limit of detection (0.871 pg/mL). Furthermore, the excellent specificity and anti-interference of the fluorescent aptasensor make the system suitable for the analysis of apple juice samples. This label-free strategy is cheaper and simper compared with labeled detection, especially for the development of multi-target-detection.

To sensitively monitor trace-level of toxic patulin (PAT), an ultrasensitive PAT ratiometric aptasensor based on the dual-potential electrochemiluminescence (ECL) of Ru(bpy) 3 2+ was first proposed. Noteworthily, Ru(bpy) 3 2+ -doped trimetallic nanocube ( [email protected] ) innovatively integrated the luminophore and cathode coreaction accelerator (CCA), which could generate strong cathodic ECL in the existence of low concentration of K 2 S 2 O 8 . Simultaneously, anthocyanin-derived carbon quantum dots (anth-CQDs) prepared from purple potato skins was first served as a green anodic coreactant. And SiO 2 -coated anth-CQDs ( [email protected] 2 ) exhibited excellent performance for enhancing anodic ECL of [email protected] Based on this, a novel ternary ECL system was established. In the presence of PAT, the ECL intensity ratio of anode to cathode ( I ECL-A / I ECL-C ) was significantly increased, and a low detection limit of 0.05 pg mL −1 was obtained. Moreover, when proposed method and high performance liquid chromatography (HPLC) were simultaneously applied to series of fruit products, the obtained results were completely consistent, reflecting its practicability.

Penicillium expansum is a major pathogen critical for the blue mold decay and patulin production in various fruits and vegetables, particularly apples, creating an urgent need to identify safer and more effective compounds for its management. In this study, two pathogenic factors of P. expansum , PeBgl1 and PeSCP, were first time targeted for virtual screening with a database (a combination of TCM Database @ Taiwan and food ingredients database) containing 20,219 molecules, and octyl gallate (OG) was identified as a common candidate inhibitor with high binding energy. Therefore, we investigated the inhibitory impacts of OG against P. expansum and patulin production in vitro and in apples. It’s revealed that OG significantly inhibited the spore germination and mycelia growth by inducing oxidative stress and compromising cell membrane integrity, as well as restrained patulin biosynthesis via suppressing the expression level of patulin synthesis genes including patE, patL , and patK. Molecular docking and dynamics simulation indicated that OG could affect the stability and function of PeBgl1 and PeSCP mainly via Van der Waals forces and hydrogen bonds. qRT-PCR analysis further demonstrated that OG directly inhibited the expression of genes encoding for PeBgl1 and PeSCP. Finally, in vivo application of OG on apples demonstrated that OG can effectively trigger the defensive enzyme system of apples and control the blue mold decay caused by P. expansum . Overall, this study emphasizes the potential of OG as a promising natural compound for the management of blue mold decay and patulin production in the post-harvest fruits and vegetables.

The removal of patulin in phosphoric acid buffer solution by cysteine was investigated. Cysteine could effectively decrease the patulin concentration at high acidic condition (pH 3.0–5.0) with the help of high temperature greater than 90 °C. Three removal mechanisms of patulin by cysteine under high acidic and high temperature conditions were deduced. Reaction temperature, pH of reactive media, molar ratio between cysteine and patulin, and reaction time were all the obvious factors influencing the removal efficiency of patulin, and the increase of any one factor could significantly improve the removal efficiency of patulin. The removal process of patulin could be simulated by the zero-order kinetic model, logarithmic model, and pseudo-first-order kinetic model, respectively, and the corresponding correlation coefficients ( R 2 ) were all greater than 0.90. Presently, this method can only be applied for the removal of patulin in contaminated water from washing fruits in juice processing industry due to the high treatment temperature more than 120 °C and the long detoxification time greater than 1 h.

Patulin, an emerging mycotoxin with high toxicity, poses great risks to public health. Considering the poor antibody production in patulin immunization, this study focuses on the four-dimensional data-independent acquisition (4D-DIA) quantitative proteomics to reveal the immune response of patulin in rabbits. The rabbit immunization was performed with the complete developed antigens of patulin, followed by the identification of the immune serum. A total of 554 differential proteins, including 292 up-regulated proteins and 262 down-regulated proteins, were screened; the differential proteins were annotated; and functional enrichment analysis was performed. The differential proteins were associated with the pathways of metabolism, gene information processing, environmental information processing, cellular processes, and organismal systems. The functional enrichment analysis indicated that the immunization procedures mostly resulted in the regulation of biochemical metabolic and signal transduction pathways, including the biosynthesis of amino acid (glycine, serine, and threonine), ascorbate, and aldarate metabolism; fatty acid degradation; and antigen processing and presentation. The 14 key proteins with high connectivity included G1U9T1, B6V9S9, G1SCN8, G1TMS5, G1U9U0, A0A0G2JH20, G1SR03, A0A5F9DAT4, G1SSA2, G1SZ14, G1T670, P30947, P29694, and A0A5F9C804, which were obtained by the analysis of protein–protein interaction networks. This study could provide potential directions for protein interaction and antibody production for food hazards in animal immunization.
