BACKGROUND Conventional pesticide delivery systems often suffer from low deposition and insufficient targeted release in complex pathogenic microenvironments. Inspired by the pathogen infection process, this study aimed to develop a novel controlled-release platform capable of responding to specific infection microenvironments to achieve efficient and targeted pesticide delivery. RESULTS A dual-responsive nano-delivery platform (BZP) was successfully constructed by loading the fungicide boscalid (Bos) onto a zeolitic imidazolate framework-8 (ZIF-8) nanocarrier and utilizing pectin as a pathogen-recognizable gatekeeper. The acidic pH- and pectinase-responsive properties enabled on-demand pesticide release specifically in the infection induced by Botrytis cinerea . BZP demonstrated an 87.35% cumulative release rate of Bos under acidic conditions (pH 5.5) with pectinase, exceeding that of nanocarriers without the pectin coating (65.64%) after 168 h of treatment. The nanoparticles (73 ± 11 nm) enabled bidirectional vascular transport and fungal cell wall penetration for targeted antimicrobial action. Meanwhile, it exhibited potent antifungal activity that matched the efficacy of the boscalid wettable powder (BWP). Additionally, the pectin coating greatly improved the foliar retention of BZP. Notably, BZP showed no apparent phytotoxicity to test plants and exhibited excellent biosafety in earthworms. CONCLUSION This study provides theoretical insights for enhancing the targeted release and efficient foliar deposition of nanopesticides, aiming to improve pesticide utilization efficiency and ensure environmental safety, thereby offering promising applications in sustainable agriculture. © 2026 Society of Chemical Industry.

Diaporthe Z1-1N, the primary causal agent of soft rot disease in kiwifruit, exhibited higher pectinase activity compared to cellulase activity in both in vitro and in vivo incubation models. To gain deeper insights into the role of pectinases in the pathogenicity of this fungus, we evaluated the effects of incubation temperature (ranging from 18 to 38 °C), duration (1 to 7 days), and medium pH (4.0 to 9.0) on the activities of two crucial pectinases: polygalacturonase (PG) and polymethylgalacturonase (PMG). Our single-factor experiments revealed that the optimal conditions for maximizing PMG yield were a pH of 7.5 and a temperature of 28 °C, with peak activity occurring after three days of incubation. Notably, PG activity peaked on the fourth day under the same pH and temperature conditions. Under the optimal conditions identified through an orthogonal experimental design, PMG exhibited higher activity than PG. Further analysis showed that temperature was the most influential factor on PMG activity, followed by incubation duration and pH. The lesion size caused by the purified pectinase extracts was 50% the lesion size that caused by the fungal mycelium of Diaporthe Z1-1N. These findings underscore the significance of PG and PMG as key virulence factors in the pathogenicity of Diaporthe Z1-1N, providing a solid scientific basis for future research into the functions of these enzymes.

To reduce food contamination caused by foodborne pathogens, improve encapsulation stability and targeting efficacy of antimicrobial agents, and minimize the required dosage, we developed a pectin-coated liposome containing thymol (Thy@Lip@Pec). The pectin shell of this formulation can be enzymatically degraded by pectinase secreted by fungi, thereby triggering the on-demand release of thymol. Under the action of pectinase at 1.0 mg/mL, this formulation released 76.45 % of thymol within 6 h. Thy@Lip@Pec treatment significantly inhibited Aspergillus niger and Penicillium spp. colonies on agar plates. Moreover, while maintaining quality indicators of oranges, Thy@Lip@Pec significantly suppressed postharvest diseases caused by A. niger and Penicillium spp., reducing the decay incidences of A. niger from 80.00 % (control) to 6.67 %, and that of Penicillium spp. from 93.33 % (control) to 33.34 % in artificially inoculated oranges. This on-demand release antimicrobial material system represents an advanced and efficient strategy for food preservation.

A highly selective and ultra-sensitive colorimetric assay has been developed for detecting the magnesium ion (Mg 2+ ) based on pectinase (PE)-protected Au nanoparticles (PE-AuNPs). This is the first study reporting the use of pectinase for green synthesizing gold nanoparticles. Aggregation of PE-AuNPs was induced immediately following the addition of Mg 2+ ion under Tris-HCl buffer at pH 4.0, yielding a color change from red to blue, and the characteristic surface plasmon resonance (SPR) peak of PE-AuNPs was red-shifted to 665 nm. The effects of parameters such as pH, the amount of PE-AuNPs, and incubation time on the sensitivity of colorimetric assay were investigated in detail. The Mg 2+ -induced aggregation of PE-AuNPs could be monitored by both the naked eye and UV–vis spectrophotometry. The lowest detection concentration with the naked eye is 0.52 μM. A linear relationship between the absorbance ratio ( A
665 / A
523 ) and Mg 2+ concentration was observed in two ranges of Mg 2+ concentration by UV–vis spectrophotometry, including 5.34 × 10 −7 μM to 51.4 μM and 5.12 × 10 2 μM to 3.19 × 10 3 μM. The Mg 2+ detection limit was determined to be 4.0 × 10 −9 μM with UV–vis spectrophotometer. The proposed colorimetric assay possesses a highly selective response for Mg 2+ over other metal ions. This method has been successfully applied to determine the Mg 2+ ion in some water samples.

Kiwifruit, a climacteric-fruit susceptible to rapid deterioration, attains considerable value upon transformation into kiwifruit wine, thereby increasing its added value. However, a noteworthy challenge arises from its high pectin content, which negatively affects byproduct quality, necessitating pectinase treatment during processing. Commercial pectinases exhibit heightened pectin esterase activity, giving rise to safety apprehensions due to augmented methanol content during kiwifruit pectin hydrolysis. In this study, Bacillus velezensis W17-6 was screened for its high pectinase activity coupled with low pectin esterase activity. The resultant crude pectinase named PBV was acquired through fed-batch-fermentation and partial purification. Subsequently, PBV was deployed in the fermentation of Xuxiang kiwifruit wine, and its impact on overall quality was assessed through diverse analyses. The results indicated that Xuxiang kiwifruit wine with PBV treatment outperformed counterparts treated with commercial pectinases. PBV treatment not only yielded superior clarity (83.75% ± 0.47%) and juice yield (81.60% ± 0.47%) but also increased the total flavonoids content in Xuxiang kiwifruit wine by 1.2 times compared to commercial pectinase. Notably, the methanol content of PBV-treated Xuxiang kiwifruit wine was significantly lower at 36.13 ± 1.83 mg/L than that of wines treated with two commercial pectinases. PBV-treated kiwifruit wine also exhibited elevated concentrations of volatile compounds such as phenethyl alcohol, ethyl undecanoate, lauric aldehyde, isobutyric acid, and myrcene. Impressively, PBV-treated kiwifruit wine surpassed other wines in both electronic and artificial sensory analyses. In summary, PBV exhibits substantial potential in the production of low-methanol, high-quality kiwifruit wine, enhancing the flavor and nutritional value while minimizing methanol production and improving the safety profile.

In this study, a novel type of stimuli-responsive support materials was synthesized successfully by atom transfer radical polymerization (ATRP) and ring opening polymerization (ROP) of N-isopropylacrylamide (NIPAAm), tert -butyl methacrylate (tBMA) and ε-caprolactone (CL). β-Cyclodextrin (β-CD) and azobenzene (Azo) were employed as the starting groups to form PCL-hv-PNIPAAm by host–guest inclusion complexation, while PCL-PMAA was converted by simple hydrolysis of PCL-PtBMA with trifluoroacetic acid. Both PCL-PMAA and PCL-hv-PNIPAAm formed the microspheres by self-assembly. Their characteristics were confirmed by NMR, TEM, GPC, DLS, etc. Then, the microspheres were used for the immobilization of pectinase by electrostatic interaction. The optimal immobilization reaction parameters were 10 U/mL for pectinase at 25 °C for 4 h at pH = 6. Compared with free pectinase, the immobilized enzymes were found to exhibit better tolerance of variations in pH and temperature, as well as improved storage stability. Furthermore, the relative activity of the immobilized pectinase was controlled at approximately 13% when the temperature was higher than the LCST of PNIPAAm. With different light irradiation, there is approximately a 15% influence on the relative activity of the immobilized pectinase. This new enzyme support material has good environmental adaptability and can make the activity of immobilized enzymes be tailored predictably and enable the enzyme to be used as a biocatalyst for various applications in the food industry.

A dual-stimuli-responsive support material for pectinase immobilization through ionic bonding was prepared. Specifically, polystyrene-b-polymethylacrylic (PS-b-PMAA), light- and pH-sensitive polystyrene-(5-propargylether-2-nitrobenzyl bromoisobutyrate)-b-poly(diethylamino)ethyl methacrylate-b-poly(polyethylene glycol methacrylate) (PS-ONB-PDEAEMA-b-PPEGMA) were synthesized through atom transfer radical polymerization, click chemistry, and hydrolysis. The two parts could self-assemble into the micelles in an aqueous solution. The micelles shrunk at a higher pH, and their size reduced under UV irradiation. The stimuli-responsive properties of micelles were characterized by dynamic light scattering and transmission electron microscopy. It has been found that this support was able to adsorb 10 U/mL of immobilized pectinase (approximately 223 mg/g) at pH 5.0 and 60 °C for 60 Min. Meanwhile, the highest relative activity of immobilized pectinase was up to approximately 95% at pH 5.0 and 60 °C. The immobilized pectinase retained more than 50% of the initial activity after eight cycles. The relative activity of the pectinase immobilized on the supports without UV irradiation was approximately 3% lower than that after UV irradiation at 60 °C, indicating that tailoring of enzyme activity was achieved by changing environmental conditions. Apparently, the original enzymatic support material had a great application prospect on enzyme immobilization.

Pectins or polygalacturonic acids (PGA) were originated from alkaline peroxide bleaching of mechanical pulps and considered as the dominant troublesome substance, which seriously decreased the runability of the paper machine in process water closure. To find wide application of enzymes in lowering PGA concentration in papermaking industries, cross-linked chitosan beads were prepared. Results showed that the PGA-absorption capability of chitosan beads was greatly affected by its cross-linking degree. The activity of immobilized pectinase on cross-linked chitosan beads were also investigated and the highest activity of binary immobilized pectinase on cross-linked chitosan was achieved using 1.00% of activating reagent or 0.005% of glutaraldehyde. Cationic demand of PGA solutions was obviously lowered by increasing the temperature of enzymatic treatment. GPC analysis showed a sharp decrease in the molecular weight of PGA after enzymatic treatment. Besides, operational stability of the immobilize pectinase was analyzed to evaluate its application potential.

Matrix binding poses a challenge to accurate pesticide residue detection in fruits and vegetables. This study investigated the distribution, binding status, and detection accuracy of neonicotinoid insecticides thiamethoxam (TMX) and clothianidin (CLO) in eggplant. To address the limitations posed by matrix binding, a pectinase-assisted extraction method was developed, which significantly enhanced the detectable levels of pesticides compared to the standard method and cellulase-assisted extraction. In situ Raman mapping and scanning electron microscopy revealed a strong correlation between the pectin distribution and the pesticide residues observed near the epidermal surface. Spectroscopic, nuclear magnetic resonance, and computational chemistry analyses demonstrated that TMX formed covalent bonds with pectin, while CLO interacted through hydrogen bonding. Notably, thermal processing promoted the formation of amide bonds, reducing detectable TMX levels. In summary, the optimized pectinase-assisted extraction method increased detectable TMX and CLO levels in positive samples by 25–40%, providing an effective approach for overcoming matrix binding effects.

Pulsed electric field coupled pectinase (PEF-PE) increased 50.79% oil yield. • PEF-PE enhanced the oxidation stability and nutritional value of olive oil. • PEF-PE destroyed cell wall by reducing polysaccharide content and crystallinity. • PEF-PE treated olive paste had the lowest viscosity and optimal viscoelasticity. • PEF-PE promoted cell wall roughening and oil droplet aggregation. Superior olive oil requires advanced extraction techniques to ensure both extraction efficiency and quality. This study comparatively evaluated the effects of different combining sequences of pulsed electric field (PEF) and pectinase (PE) on olive oil extraction. Among the combined treatments, PEF followed by PE treatment (PEF-PE) showed the best performance, increasing oil yield by 50.79% compared with the control group, while maintaining high phenolic content (106.01 μg/g) and favorable oxidation-related quality parameters. Cell wall compositional analysis showed that PEF-PE reduced pectin, hemicellulose, and cellulose contents by 51.05%, 39.79%, and 37.93%, respectively, and decreased cellulose crystallinity by 17.01%. Rheological and microscopic observations further showed that PEF-PE reduced paste viscosity and viscoelasticity, increased cell wall surface roughness, and promoted oil droplet aggregation. These results suggest that PEF-PE is an effective pretreatment sequence for olive oil extraction and provide a preliminary reference for further evaluation. The pulp and paper industry faces growing challenges arising from fiber shortages and environmental concerns. High yield pulping technology has emerged as a key solution, but the high lignin content in high yield pulp (HYP) restricts its brightness, posing a significant challenge for the bleaching process. Although traditional strong alkaline hydrogen peroxide (H 2 O 2 ) bleaching of HYP is effective, it results in fiber yield loss and the generation of anionic impurities, thereby bringing about additional environmental burdens. In contrast, weakly alkaline H 2 O 2 bleaching is more environmentally friendly and preserves a high pulp yield, but it struggles to achieve the desired brightness due to the reduced reactivity of lignin. This work addresses these challenges by investigating the effect of xylanase-pectinase (XP) pretreatment combined with weakly alkaline H 2 O 2 bleaching to improve both bleaching efficiency and environmental benefits in the bleaching of poplar chemi-thermomechanical pulp (CTMP). The kinetic analysis reveals that the XP pretreatment significantly enhanced the efficiency of weakly alkaline H 2 O 2 bleaching, accelerating lignin removal and increasing pulp brightness. The XP pretreatment effectively reduced the electrical conductivity, cationic demand, and chemical oxygen demand of the bleaching effluent. The mechanism studies demonstrate that the XP pretreatment effectively cleaved lignin-carbohydrate complex linkages and degraded hemicelluloses side chains, significantly increasing the accessibility and reactivity of the residual lignin. The integration of XP pretreatment with weakly alkaline H 2 O 2 bleaching can offer a sustainable and efficient process for poplar CTMP upgrading, simultaneously enhancing the bleaching efficiency and mitigating the environmental footprint.

Enzymatic hydrolysis using pectinase is critical for producing high-yield and quality sea buckthorn juice. This study determined the optimal temperature, time, and enzyme dosage combinations to guide manufacturers. A temperature of 60 °C, hydrolysis time of 3 h, and 0.3% enzyme dosage gave 64.1% juice yield—25% higher than without enzymes. Furthermore, monitoring physicochemical properties reveals enzyme impacts on composition. Higher dosages increase soluble solids up to 15% and soluble fiber content by 35% through cell wall breakdown. However, excessive amounts over 0.3% decrease yields. Pectin concentration also declines dose-dependently, falling by 91% at 0.4%, improving juice stability but needing modulation to retain viscosity. Electrochemical fingerprinting successfully differentiates process conditions, offering a rapid quality control tool. Its potential for commercial inline use during enzymatic treatment requires exploration. Overall, connecting optimized parameters to measured effects provides actionable insights for manufacturers to boost yields, determine enzyme impacts on nutrition/functionality, and introduce novel process analytical technology. Further investigations of health properties using these conditions could expand sea buckthorn juice functionality.
