As one of the most toxic environmental pollutants, cadmium (Cd) has lastingly been considered to have negative influences on plant growth and productivity. Recently, increasing studies have shown that low level of Cd exposure could induce hormetic effect which benefits to plants. However, the underlying mechanisms of Cd-triggered hormesis are poorly understood. In this study, we found that Cd stress treatment showed a hormetic effect on peppermint and Cd treatment with 1.6 mg L-1 concertation manifested best stimulative effects. To explore the hormesis mechanisms of Cd treatment, comparative transcriptome analysis of peppermint young plants under low (1.6 mg L-1) and high (6.5 mg L-1) level of Cd exposure at 0 h, 24 h and 72 h were conducted. Twelve of differentially expressed genes (DEGs) were selected for qRT-PCR validation, and the expression results confirmed the credibility of transcriptome data. KEGG analysis of DEGs showed that the phenylpropanoid biosynthesis and photosynthesis were important under both low and high level of Cd treatments. Interestingly, GO and KEGG analysis of 99 DEGs specifically induced by low level of Cd treatment at 72 h indicated that these DEGs were mainly involved in the pathway of phenylpropanoid biosynthesis and their functions were associated with antioxidant activity. The expression pattern of those genes in the phenylpropanoid biosynthesis pathway and encoding antioxidant enzymes during 72 h of Cd exposure showed that low level of Cd treatment induced a continuation in the upward trend but high level of Cd treatment caused an inverted V-shape. The changes of physiological parameters during Cd exposure were highly consistent with gene expression pattern. These results strongly demonstrate that low level of Cd exposure constantly enhanced antioxidant activity of peppermint to avoid oxidative damages caused by Cd ion, while high level of Cd stress just induced a temporary increase in antioxidant activity which was insufficient to cope with lasting Cd toxicity. Overall, the results presented in this study shed a light on the underlying mechanisms of the Cd-mediated hormesis in plant. Moreover, our study provided a safe method for the efficient utilization of mild Cd-contaminated soil as peppermint is an important cash plant.

Although the introduction of glycerosomes has enriched strategies for efficient transdermal drug delivery, the inclusion of cholesterol as a membrane stabilizer has limited their clinical application. The current study describes the development and optimization of a new type of glycerosome (S-glycerosome) that is formed in glycerol solution with β-sitosterol as the stabilizer. Moreover, the transdermal permeation properties of lappaconitine (LA)-loaded S-glycerosomes and peppermint oil (PO)-mediated S-glycerosomes (PO-S-glycerosomes) are evaluated, and the lipid alterations in the stratum corneum are analyzed via lipidomics. The LA-loaded S-glycerosomes prepared by the preferred formulation from the uniform design have a mean size of 145.3 ± 7.81 nm and an encapsulation efficiency of 73.14 ± 0.35%. Moreover, the addition of PO positively impacts transdermal flux, peaking at 0.4% (w/v) PO. Tracing of the fluorescent probe P4 further revealed that PO-S-glycerosomes penetrate deeper into the skin than S-glycerosomes and conventional liposomes. Additionally, treatment with PO-S-glycerosomes alters the isoform type, number, and composition of sphingolipids, glycerophospholipids, glycerolipids, and fatty acids in the stratum corneum, with the most notable effect observed for ceramides, the main component of sphingolipids. Furthermore, the transdermal administration of LA-loaded PO-S-glycerosomes improved the treatment efficacy of xylene-induced inflammation in mice without skin irritation. Collectively, these findings demonstrate the feasibility of β-sitosterol as a stabilizer in glycerosomes. Additionally, the inclusion of PO improves the transdermal permeation of S-glycerosomes, potentially by altering the stratum corneum lipids. Graphical Abstract

Emulsion-based carriers have been widely studied for the co-encapsulation of active ingredients with different partition coefficient (Log P). However, it is challenging to encapsulate active ingredients with low Log P values in emulsified oil droplets. In this study, (-)-epigallocatechin gallate (EGCG, 160 µg/g*emulsion, Log P 2.38) and resveratrol (Res, 320 µg/g*emulsion, Log P 2.57), as models with low Log P, were co-encapsulated in emulsified oil droplets for the first time by peppermint essential oil (PEO) mediation. During storage process, EGCG and Res consistently remain co-located inside the emulsified oil droplets. However, both EGCG and Res migrated towards the inner layer of the interface of emulsified oil droplets along with PEO. Res exhibited better stability compared to EGCG, while the degradation of EGCG occurred within the emulsified oil droplets. Furthermore, when co-localized with Res, the degradation rate of EGCG slowed down. PEO did not degrade during storage, and over 98 % of PEO was encapsulated within the emulsified oil droplets. This is the crucial reason for maintaining the stability of SC-stabilized emulsion and achieving high encapsulation efficiency of active ingredients. This provides a green and simple strategy for co-encapsulation of active ingredients with Log P lower than 3.0 within emulsified oil droplets.

Polygalae Radix, a traditional Chinese medicine for insomnia and memory disorders, is highly susceptible to fungal contamination and mycotoxin production (especially byAspergillus flavus) during storage, compromising its safety and efficacy. Therefore, in this study, high-throughput sequencing was employed to evaluate the dynamic changes in fungal communities during the storage of Polygalae Radix and to analyze common mycotoxin-producing genera. Furthermore, the inhibitory effects of peppermint essential oil (PEO) onA. flavuswere assessed through fumigation treatments, combined with colony counting and quantification of aflatoxins. Results showed the following: (1) Storage for 1–3 months significantly altered the fungal structure, promoting saprophytic and pathogenic fungi (e.g.,Wallemia,Paraphoma,Didymella,Cladosporium…) and increasing the relative abundance of mycotoxin producers likePenicillium,Aspergillus, andFusarium(notably,Penicilliumincreased from 0.28–2.33% to 5.39–80.43%). Additionally,A. flavus, capable of producing aflatoxins, was detected in samples stored for two months (RM2). (2) Antifungal tests demonstrated that PEO significantly inhibited the common fungi in Polygalae Radix. At 10 μL/g, it suppressed fungal growth and significantly reduced aflatoxin B1(AFB1) and total aflatoxins (AFT, including AFB1, AFB2, AFG1, and AFG2) levels (p< 0.05). At 10 μL/g, AFB1and AFT were reduced to undetectable levels. PEO can serve as a green and effective protective strategy to inhibitA. flavusduring the storage of Polygalae Radix and control aflatoxin contamination.

Deep eutectic solvents (DESs) have shown great potential in the sustainable fabrication of nanoemulsions. However, co-assembled nanoemulsion systems that integrate both hydrophilic DES (HL-DES) and hydrophobic DES (HB-DES) (referred to as HL/HB-DES-NE) have yet to be reported. In this study, we designed and fabricated a novel HL/HB-DES-NE system and utilized it for encapsulating peppermint essential oil (PMEO). The formulation was systematically optimized, and the resulting HL/HB-DES-NE was characterized for particle size, particle size distribution, encapsulation efficiency, stability, anti-inflammatory activity and antibacterial activity. Firstly, a hydrophobic DES was prepared through eutectic mixing of thymol and polyglyceryl-4-caprate, while a hydrophilic DES was synthesized using betaine and glycerol. The optimal formulation is PMEO/thymol:polyglyceryl-4-caprate (2:1, mol/mol)/betaine:glycerol (1:3, mol/mol)/H₂O [0.275/0.25/0.94/3.76, v/v/v/v] which achieved a particle size of approximately 55.80 nm. The nanoemulsions exhibited good anti-inflammatory and antibacterial activities and maintained remarkable stability, showing no significant changes in particle size, distribution, anti-inflammatory activity and antibacterial activity after 14 days of storage at 4 °C, 20 °C, 37 °C, and 55 °C. The proposed co-assembly strategy, which leverages the unique properties of both hydrophilic and hydrophobic DESs, offers a green alternative to traditional surfactants and provides a flexible and scalable platform for the design of high-performance nanoemulsions. The findings not only provide important theoretical and practical foundations for the development of essential oil-based antimicrobial and anti-inflammatory agents in cosmetics, pharmaceuticals, and industrial applications, but also propose a novel strategy for the development of next-generation DES-based formulations across diverse industries.

In this study, we report for and first use of soy protein isolate (SPI), chitosan quaternary ammonium salt (HACC), and peppermint essential oil (PMO) to prepare emulsions. Under different SPI/HACC ratios, emulsions with an oil-phase ratio of 60% were produced using the high-speed shearing machine (i.e., 1:0, 1:1, 2:1, and 5:1). The outcome of rheological experiment demonstrated that the addition of HACC boosted the viscosity and improved gel network structure of emulsions. The stability and microstructure of the emulsion also improved as the content of HACC was increased. Also, the evaporation rate of essential oil from the emulsions was examined. The release duration of PMO was also significantly extended by the emulsions, and its capacity to inhibit the growth of three plant pathogenic fungi was enhanced. Therefore, this study suggested that HACC can greatly improved the emulsification stability of SPI as well as essential oil emulsion stabilized by SPI/HACC composite with a ratio of 1:1 had the best slow release effect. The PMO emulsions that were created had great promise for embedding volatile, water-insoluble, and micromolecular chemicals.
