Preparation of polyvinyl alcohol/pullulan nanofibers and Eudragit® S100/polyvinyl alcohol/pullulan core-shell nanofibers for enhanced probiotic storage and oral viability

Yangrui Wang,Yingxue Yue,Shengjun Yang,Sufang Duan,Yuning Yang,Zhaozhi Wang,Xuetong Li,Bailiang Li,Yupeng Wang

Probiotics, which play a crucial role in regulating intestinal homeostasis and maintaining host health, face significant challenges in maintaining their survival rate after processing, storage, and oral administration, posing a substantial hurdle in probiotic use. In this study, electrospinning technology was employed to prepare polyvinyl alcohol (PVA)/Pullulan (PUL) nanofibers and Eudragit® S100 (ES100)/PVA/PUL core-shell nanofibers. The effects of different PVA, PUL, and ES100 solution concentrations, applied voltage, collector distance, and feed rate on the morphology and diameter of nanofibers were investigated by scanning electron microscopy. The fibers were characterized using fourier transform infrared spectroscopy, X-ray diffraction, and thermal analysis. Additionally, the storage stability and resistance to gastrointestinal stress of the nanofibers loaded with Lactiplantibacillus plantarum KLDS1.0386 were investigated. The results indicate that the prepared PVA/PUL nanofibers and ES100/PVA/PUL core-shell nanofibers exhibit smooth surfaces, uniform diameters, and excellent thermal stability. Compared with free L. plantarum KLDS1.0386, the survival rate of PVA/PUL nanofiber-encapsulated bacteria increased by 25.07% after 21 days of storage at 4 °C and by 33.21% at 25 °C, respectively. Experiments simulating gastrointestinal digestion showed that the survival rates of PVA/PUL nanofibers and ES100/PVA/PUL nanofiber-coated L. plantarum KLDS1.0386 were 9.11% and 15.73% higher in simulated gastric juice compared to free L. plantarum KLDS1.0386, respectively. Additionally, 13.15% and 13.92% higher in simulated intestinal fluid, respectively. The nanofibers prepared in this study can be used as delivery carriers for probiotics. They enhance the storage stability and improve the tolerance of probiotics to the harsh gastrointestinal environment.

Application of Pullulan and Chitosan Multilayer Coatings in Fresh Papayas

Linyun Zhang,Chongxing Huang,Hui Zhao

In this work, some multilayer coatings (two-layer, four-layer or six-layer) based on pullulan and chitosan for protecting papayas were prepared by the layer-by-layer technique. The papayas were coated by immersion and stored at 25 °C, 50% relative humidity or up to 14 days. Uncoated and monolayer-coated papayas were used as controls. The pullulan/chitosan coatings decreased the papaya weight loss, softening, color change (b*, ΔE), and pH, retarded the fall of titratable acidity and vitamin C, and maintained respiratory rate and soluble solid contents. Sensory quality evaluation demonstrated that pullulan/chitosan coatings effectively preserved papaya flavor and overall acceptance. In general, the four-layer coatings provided the best fruit preservation. In conclusion, multilayer pullulan/chitosan coatings are efficient in maintaining the post-harvest quality and prolonging the shelf life of fresh papaya. Keywords:papaya;preservation;pullulan;chitosan;multilayer coating

An edible film of sodium alginate/pullulan incorporated with capsaicin

Shen Zhang,Feng Wei,Xiaojun Han

Capsaicin, an active ingredient in red pepper, shows great antibacterial activity. Herein, a series of sodium alginate/pullulan composite films incorporated with different concentrations of capsaicin were fabricated. Their physico-chemical properties were systematically investigated. With the increased content of capsaicin, the transmittance, elongation at break, and moisture content of the films decreased, whereas the tensile strength, water vapor permeability, and surface contact angle increased. Furthermore, the liquid culture tests revealed that the composite films exhibited good antibacterial performance against E. coli and S. aureus. The practical application of these antibacterial films for apple protection also exhibited a positive result, which makes it possible to use them in food packaging.

Pullulan-stabilized Soybean Phospholipids/Cinnamaldehyde emulsion for Flammulina velutipes preservation

Xue Shao,Ben Niu,Xiangjun Fang,Weijie Wu,Ruiling Liu,Honglei Mu,Haiyan Gao,Hangjun Chen

Fresh mushrooms ( Flammulina velutipes ) are very perishable and easily brown; also they undergo postharvest loss of nutritive constituents. In this study, cinnamaldehyde (CA) emulsion was prepared by using soybean phospholipids (SP) as emulsifier and pullulan (Pul) as stabilizer. The effect of emulsion on the quality of mushroom during storage was also studied. The experimental results indicated that the emulsion obtained by adding 6 % pullulan was found to the most uniform and stable, which is beneficial to its application. Emulsion coating maintained the storage quality of Flammulina velutipes. The incorporation of CA emulsion into the coating system showed a positive effect on inhibiting the accumulation of reactive oxygen species, resulting from improving the effectiveness of delaying active free radical scavenging enzymes . The shelf life of mushrooms coated with emulsion was significantly prolonged, which indicates its potential application in food preservation.

Enhancement of a recombinase-aided amplification assay using betaine and pullulan

Jinrong Wang,Guowei Song,Yue Ming,Jing Pan,Ruiqing Zhang,Guohao Fan,Xinxin Shen,Xuejun Ma,Lixin Li

Background Nucleic acid amplification enhancers suitable for use in a recombinase-aided amplification (RAA) assay were studied for the first time, and amplification of a long-fragment (509 bp) was initially explored. Methods Using recombinant plasmids and clinical samples, RAA fluorescence and basic methods were used to evaluate the efficacy. The fluorescence method was evaluated by threshold time and fluorescence value, and the basic method was characterized by 2% agarose gel electrophoresis. Results Taking a previously established RAA assay for HPV18 as an example, we demonstrated that the addition of 0.2 M, 0.4 M, and 0.6 M betaine and 10% pullulan could enhance the RAA. The new RAA assays with betaine and pullulan were named B-RAA and P-RAA, respectively. Using the B-RAA and P-RAA fluorescence methods, the threshold time values could be shortened by 1.72–2.32 minutes and 2.60 minutes, respectively, and the fluorescence values could be enhanced by 8847.25–9094.37 mv and 5250 mv, respectively. Using the basic method, the sensitivity could be increased 10-fold. We successfully amplified a long-fragment of 509 bp using a P-RAA assay with a sensitivity of 10 2 copies/µL (compared with 10 3 copies/µL in the RAA assay). Conclusions Thus, we concluded that betaine and pullulan are effective additives to enhance the sensitivity of RAA assays.

Oxidized pullulan/carboxymethyl chitosan hydrogel with salidroside for enhanced wound healing

Yuting Fu,Anji Chen,Ziyi Fu,Qingjuan Zhang,Wuqiang Deng,Liuyun Jiang,Xiang Hu

Polysaccharide-based hydrogels with integrated bioactive functionality are highly desirable for wound dressing applications. Here, we report an injectable, self-healing hydrogel (OPC@Sal) fabricated via dynamic Schiff-base crosslinking between oxidized pullulan (OPL) and carboxymethyl chitosan (CMCS), with simultaneous incorporation of salidroside as a therapeutic agent. The gelation kinetics, mechanical properties, and microstructure were finely tuned by varying the OPL concentration at a fixed oxidation degree. The optimized OPC@Sal hydrogel exhibited rapid self-healing, excellent injectability, favorable biocompatibility, moisture retention, exudate absorption, and sustained salidroside release, along with pronounced antibacterial activity against E. coli and S. aureus . In vitro, the hydrogel effectively scavenged intracellular reactive oxygen species and suppressed the expression of pro-inflammatory cytokines (TNF-α, IL-6, IL-1β) in LPS-stimulated macrophages. Based on the established in vitro effects, network pharmacology analysis was performed as a mechanistic interpretation tool, identifying key targets (TNF-α, IL-6, CASP3, IL-1β) through which salidroside exerts its multi-target regulation of wound healing. In a mouse full-thickness skin defect model, OPC@Sal significantly accelerated wound closure, promoted collagen deposition, and enhanced re-epithelialization. Collectively, this work establishes a fully polysaccharide-derived hydrogel platform that synergistically integrates structural functionality with intrinsic bioactivity. The molecular engineering of oxidized pullulan and carboxymethyl chitosan, combined with network pharmacology-guided active compound selection, offers a sustainable and scalable strategy for developing advanced wound dressings.

Algae-inspired chitosan-pullulan-based multifunctional hydrogel for enhanced wound healing

Taishan Liu,Huan Lei,Linlin Qu,Chenhui Zhu,Xiaoxuan Ma,Daidi Fan

Chronic wounds caused by hyperglycaemia, hypoxia and bacterial infections are common complications in diabetic patients and chronic wound repair is extremely challenging in clinical practice. A series of hydrogels QPMP with good antioxidant and antimicrobial functions were prepared based on quaternized chitosan (QCS), oxidized pullulan polysaccharide (OP), dopamine-coated polypyrrole (PPY@PDA), and Chlorella vulgaris . The Schiff base cross-linking between the quaternized chitosan (QCS) and oxidized pullulan polysaccharide (OP) constitutes the basic skeleton of the hydrogel, and imparts a certain antimicrobial ability to the hydrogel. Chlorella vulgaris continuously produced oxygen under light conditions to relieve wound hypoxia and promote wound healing. The incorporation of PPY@PDA gave the hydrogel near-infrared (NIR) irradiation-assisted bactericidal activity and antioxidant activity, and as a conductive hydrogel, the hydrogel can be used to sense wound exudate and temperature changes, which can help to achieve the integration of diagnosis and treatment of wound healing. Most importantly, in a chronic wound model, the QPMP hydrogel was more effective in controlling the level of wound inflammation and promoting collagen deposition, angiogenesis, and early wound closure compared to the HeraDerm dressing. Therefore, this conductive oxygen-producing hydrogel is extremely beneficial for chronic wound healing in diabetes.

Pullulan/zein nanofiber film with synergistic antibacterial effects for strawberry preservation

Zihuan Yuan,Tailong Dong,Xinmeng Zhang,Xiufeng Liu,Wei Shao

PZ/CT nanofiber film for fruit preservation was prepared. • Synergistic antibacterial effects of CO/THY were presented. • The shelf-life of strawberries was extended to 8 days. • High DPPH free radical scavenging (98.2%) was obtained. This study fabricated a pullulan/zein-based (PZ/CT) edible nanofiber film co-loaded with cornmint oil (CO) and thymol (THY) for strawberry preservation. The synergistic antibacterial activity of CO and THY, the electrospinning encapsulation, as well as the physicochemical, functional, and preservation properties of the nanofiber films were characterized. The PZ/CT nanofibers exhibited uniform morphology with an average diameter of 143 nm. The film presented a water contact angle of 82.8°, a water vapor permeability of 0.83 × 10 −10  g·m −1 ·s −1 ·Pa −1 , a tensile strength of 1.28 MPa, and an elongation at break of 36.5%. It showed UV-shielding capacity and achieved a DPPH radical scavenging rate of 98.2% within 0.5 h. At 25 mg/mL, the film showed inhibition rates of 99.8% against Escherichia coli ATCC 25922, 99.7% against Staphylococcus aureus ATCC 6538, and 100% against Listeria monocytogenes ATCC 19119. Under ambient storage conditions (25 ± 2 °C), PZ/CT packaging extended the shelf life of strawberries to 10 days by retarding microbial growth, weight loss, and quality deterioration. These results indicate that the electrospun PZ/CT nanofiber film could serve as a promising biodegradable active packaging material for fruit preservation.

Pullulan dialdehyde crosslinked gelatin hydrogels with high strength for biomedical applications

Liru Zhang,Jie Liu,Xuejing Zheng,Ali Zhang,Xianliang Zhang,Keyong Tang

Herein the construction of a strong gelatin hydrogel is presented by using pullulan dialdehyde (PDA) as a macromolecular crosslinker. The resultant PDA crosslinked gelatin hydrogels (G-PDA) exhibit extremely high mechanical strength, manifested in the achieved optimal compressive stress of 5.80 MPa at 80% strain, which is up to 152 times higher than pure gelatin hydrogel. The G-PDA were characterized by Fourier transform infrared (FTIR) spectroscopy and scanning electron microscopy (SEM). The extent of crosslinking was determined by ninhydrin assay. The results suggested that the synergistic effect of dual-crosslinking, which is composed of short- and long-range covalent crosslinking and thermoreversible physical crosslinking, may played a key role in enhancing the load-bearing capacity of ensuing hydrogels. The swelling and enzymatic degradation of G-PDA are gradually limited with increasing PDA concentration. The result from MTT assay demonstrated that G-PDA is non-cytotoxic against MC3T3 cells, regardless of the concentrations of PDA.

Tocopheryl pullulan-based self assembling nanomicelles for anti-cancer drug delivery

Jingyun Wang,Shuang Cui,Yongming Bao,Jishuang Xing,Wenbo Hao

Amphiphilic α-tocopherol pullulan polymers (PUTC1, PUTC2, and PUTC3) with different degrees of substitution were synthesized as new carriers for anticancer drugs. The polymers easily self-assembled into nanomicelles through dialysis method. The critical micelle concentrations (CMCs) were 38.0, 8.0, and 4.3   mg/L for PUTC1, PUTC2, and PUTC3, respectively. 10-Hydroxycamptothecin (HCPT) used as a model drug was successfully loaded into the PUTC nanomicelles. Transmission electron microscopy images demonstrated that HCPT-loaded PUTC nanomicelles were almost spherical and had sizes ranging within 171.5–257.8   nm that increased with increased HCPT-loading content, as determined by dynamic laser scattering. The highest encapsulation efficiency of HCPT in PUTC nanomicelles reached 98.3%. The in vitro release of HCPT from PUTC micelles demonstrated sustained release for over 80   h. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assays showed that blank PUTC micelles were nontoxic to normal cells and that the HCPT-loaded PUTC2 nanomicelles showed higher cytotoxicity than the free drug, which was attributed to the enhanced cellular uptake of drug-loaded nanomicelles. Biodistribution experiments showed that PUTC micelles provided an excellent approach to rapid drug transport into cell nuclei. Moreover, the cellular uptake of micelles was found to be an energy-dependent and actin polymerization-associated endocytic process by endocytosis inhibition experiments. These results suggested that PUTC nanomicelles had considerable potential as a drug carrier for drug intracellular delivery in cancer therapy.

PTX-loaded, polysorbate 80-functionalized brain-targeting pullulan nanoparticles for drug delivery

Huabing Yuan,Lu Han,Hao Deng,Yue Tan,Yi Yang,Yi Liu,Jiaxin Ying,Mengqi Fang,Hui Wei,Zhihe Tao

Brain-targeted drug delivery remains a major challenge in pharmaceutical research. In this study, four types of polysorbate 80 and cholesterol-modified pullulan polymers (CHPP) were synthesized and self-assembled into nanoparticles (CHPP NPs) for brain drug delivery. The properties of these NPs, including brain targeting efficiency, were systematically evaluated to investigate the influence of polymer modification on their performance. The hydrophobicity of the polymers increased with the degree of cholesterol substitution, which in turn led to a reduction in nanoparticle size. Furthermore, higher hydrophobicity contributed to an increased drug loading capacity for paclitaxel (PTX) and a more sustained drug release profile. In vivo fluorescence imaging revealed that formulations with higher levels of polysorbate 80 and cholesterol modification exhibited significantly enhanced brain targeting efficiency compared to other variants. The PTX-loaded CHPP NPs (PTX-CHPP NPs) demonstrated potent cytotoxicity and inhibitory effects against SJ-GBM2 glioma cells in vitro, underscoring their potential as a promising platform for brain-targeted drug delivery. These findings provide valuable insights for the rational design of brain-targeting nanocarriers.

Pullulan-spermine enhance the tolerance of probiotics and immune stimulation of macrophages

Zihui Meng,Qinghui He,Litong Mu,Jiaying Feng,Fei Zhang,Jiayi Wu,Li Zhou,Qingang Hu,Xuna Tang,Yanan Li

The application of probiotics as adjunctive therapy in colorectal cancer treatment is hindered by the paucity of strains with specialized functions and the instability of their in vivo efficacy. The design of innovative and simple encapsulation strategies to enhance their stability and efficacy of probiotics has garnered substantial interest. This study investigated four Bifidobacterium longum strains from human feces for tolerance and cytotoxicity, and then synthesized a cationic polysaccharide, pullulan-spermine (PS), for probiotic encapsulation. The results indicated that the encapsulation by PS hold superior protective capacity and elevated the level of TNF-α and IL-12. In vivo studies further confirmed the retention capacity and safety of this probiotic-PS complex. Generally, this research presents an effective probiotic encapsulation strategy that could enhance macrophage immune responses, offering novel insights for probiotic-based therapies in major diseases like colon cancer treatment.
