Direct oxidation of antibiotic trimethoprim by unactivated peroxymonosulfate via a nonradical transformation mechanism

Xuerui Yang,Xi Ding,Lei Zhou,Qing Zhao,Yuefei Ji,Xingbao Wang,Jean-Marc Chovelon,Guangli Xiu

Application of activated peroxymonosulfate (PMS) to generate sulfate radical or hydroxyl radical is a promising strategy for wastewater treatment, while our knowledge on the background reaction, namely, the direct interaction between PMS and target contaminants is quite limited. In this contribution, the degradation kinetics, stoichiometry, products and mechanism of the reaction between unactivated PMS and trimethoprim (TMP), one of the most commonly detected micro-pollutants in the aquatic system were investigated systematically. The results indicated that TMP was susceptible to degradation by direct PMS oxidation via a non-radical process. By recording the decay of two reactants simultaneously, the stoichiometric ratio between PMS and TMP was estimated to be approximately 1. Higher PMS levels exhibited a promotion effect on PMS decay. And the degradation was pH-dependent, basic conditions were favorable for TMP degradation, which could be well modeled based on the species-specific reactions. The two amine groups on the pyrimidine ring were identified as the reactive sites. After direct attacks by PMS, they would be oxidized to form hydroxylamine-products, namely, N 8 –OH-TMP and N 9 –OH-TMP. These two hydroxylamine-products were quite stable and resistant to further oxidation by PMS, preventing the formation of more toxic nitroso- and nitro-products. The new findings in the present work would provide beneficial information on the strategy choice for the elimination of specific pollutants, like TMP, as PMS also exhibits relatively high reactivity towards them.

Amorphous PAA-stabilized solid dispersion for enhanced antibacterial activity and bioavailability of trimethoprim

Zhang Zhenghua,Lu Ping,Niu Ming,Long Haitao,Zhao Jie,Xu Weibing,Hang Zhaojia

The clinical application of trimethoprim (TMP) is limited by the low water solubility, short half-life and low oral bioavailability. Polyacrylic acid-stabilized trimethoprim amorphous solid dispersant (PAA/TMP ASD) are prepared by spray drying technology. The PAA/TMP ASD forms microspheres with diameters from hundreds of nanometers to tens of micrometers. The payload of TMP in the ASD can be regulated by adjusting the feeding ratio. During the continuous 7-day test, the variation range of particle size and Zeta potential is both less than 10%. In gastric juice, the ASD show low cumulative release (13.1% and 30.2%) with an initial explosive phase, whereas in PBS and intestinal fluid, the equilibrium release arrives to 64.3% and 41.5%. At a concentration of 0.097 mg/mL, the in vitro inhibition rate against E. coli can reach more than 98%. The inhibition rate of the biofilm formation is 82% at the concentration of 0.2 mg/mL. The TMP/PAA ASD shortens the peak time from 3.333 to 2.0 h, increases the peak concentration from 1.012 to 1.133 µg/mL, and significantly reduces the absorption half-life from 1.387 to 0.191 h. The TMP/PAA ASD formulation prolongs the elimination half-life from 26.033 to 34.699 h, along with increases AUC and MRT values, and demonstrates higher drug exposure and a longer duration of action in vivo. This work develops an important method to enhance the oral utilization rate of TMP.

Concurrent modulation of solubility and antibacterial activity in trimethoprim-meloxicam drug-drug salt

Lin Lyv,Shutian Zhong,Yanshan Zhu,Liang Li,Zhengzheng Zhou

A novel drug-drug salt composed of trimethoprim (TMP), a broad-spectrum antibiotic, and meloxicam (Mobic), a non-steroidal anti-inflammatory drug, was synthesized and structurally characterized. The salt was analyzed using single-crystal and powder X-ray diffraction, thermal analysis, and Fourier-transform infrared spectroscopy. Structural analysis revealed a 1:1 stoichiometric composition comprising two TMP + cations and two MLX − anions in the asymmetric unit, stabilized by N H···N and N H···O hydrogen bonds, forming a tetrameric assembly within the triclinic space group P -1. Equilibrium solubility studies in pure water revealed that the salt markedly reduced TMP solubility by 98.0 % while increasing MLX solubility by 4.4-fold, demonstrating that can effectively modulate the dissolution profiles of both active pharmaceutical ingredients (APIs) simultaneously. Moreover, antimicrobial evaluation demonstrated enhanced antibacterial activity of the salt against Escherichia coli at concentrations of 0.7 and 0.8 mmol/L (P < 0.05) compared to the individual components. These results underscore the potential of drug-drug salts to fine-tune the physicochemical and biological properties of APIs, providing a promising strategy for the development of optimized combination therapies.

An Electrochemical Sensor for Trimethoprim Based on a Magnetic Molecularly Imprinted Carbon Paste Electrode

Peng Liu,Ruiying Zhang,Liyan Zheng,Qiue Cao

In order to achieve simple, rapid, and highly sensitive detection of trimethoprim (TMP), a magnetic molecularly imprinted carbon paste electrode (MCPE) was prepared by drop-coating magnetic molecularly imprinted polymer (MIP@Fe3O4@MWNTs) on the surface of reduction graphene oxide (rGO)/MCPE doped with Fe3O4@MWNTs. The introduction of multi-walled carbon nanotubes (MWNTs) and rGO served as dual signal-amplification materials, which can improve the response sensitivity of the sensor. In addition, the magnetic interaction between the substrate electrode and the molecularly imprinted material was beneficial to increasing the stability of the sensor. As expected, the electrochemical sensor not only showed sensitivity and selectivity for the detection of TMP, but it also possessed good stability. The detection range for TMP was 4.0 × 10−9~5.0 × 10−4mol/L, and the detection limit was 1.2 × 10−9mol/L. The response performance varied within 10% when the sensor was placed for more than 2 months and used more than 60 times. The spiked recoveries of TMP in environmental water samples, urine samples, and pharmaceuticals (drugs) were between 91% and 110%, and the relative standard deviation (RSD) was within 5%.Keywords:trimethoprim;molecular imprinting technique;electrochemical sensor

Quantifying the Relative Roles of Different Microorganisms on Trimethoprim Degradation by Nitrifying Activated Sludge

Pei-Kun Yuan,Run-Ze Xu,You Ma,Yu Wu,Han Meng,Guo-Xiang Wang,Li-Min Zhang,Xian-Qiu Zhang,Guo-Zheng Jia,Yan Xiao,Wei-Hua Li,Wen-Ming Xie

The cometabolism of ammonia-oxidizing bacteria (AOB) in nitrifying activated sludge can effectively degrade antibiotics. However, the roles of other microorganisms in antibiotic removal processes remain uncharacterized. In this study, the effects of AOB, nitrite oxidizing bacteria (NOB), and heterotrophs on the removal of the antibiotic trimethoprim (TMP) at environmentally relevant concentrations in an enriched nitrifying activated sludge are quantified. AOB is found to be the most efficient for TMP removal, which is related with ammonium concentration. TMP removal is increased from 48.0% to 64.0% as the ammonium concentration increased from 50 to 150 mg L −1 . However, with a further increase in ammonium concentration to 250 mg L −1 , TMP removal is decreased to 45.6%. The percentages of TMP removed by AOB, NOB, and heterotrophs are quantified by batch experiments, and are 78.7%, 18.6%, and 2.7%, respectively. The results demonstrate that in nitrifying activated sludge system, the contribution of NOB and heterotrophs on TMP removal should also be considered.

Degradation of trimethoprim in the simulated solar light/periodate system: Process and mechanism analysis

Yuyue Huang,Qingjie Xie,Huijuan Wang,Muhammad Imran Nawaz,Huihui Zhang,Shilin Song,Kun Hu,Qiangshun Wu

Trimethoprim (TMP), a kind of emerging pollutant, has attracted considerable attention. To achieve an effective degradation of the TMP, a simulated solar light/periodate (SSL/PI) system was set up by using the Xe lamp as the SSL source. The critical parameters affecting the degradation, including the PI content, the TMP concentration as well as the initial pH, were analyzed. The obtained results show that the TMP removal could reach 92 % after 50 min of treatment under the condition of 4 mM PI addition while lower TMP concentrations showed higher degradation percentages. The efficient TMP degradation in the SSL/PI system was achieved in a wide range of the solution pH. The quenching test proved that IO 3 , IO 4 , 1 O 2 , OH, and O 2 − were critical reactive species for the TMP degradation in the SSL/PI system and the electron spin resonance analysis proved the generation of 1 O 2 and OH. Three possible pathways of the TMP degradation were proposed according to the LC-MS detection and Pathway II might be the specific one according to the density functional theory (DFT) calculation. Toxicity evaluation showed that the toxicity of the initial TMP solution reduced after the treatment, which confirmed the effectiveness of the PI activation by the SSL.

Environmentally Friendly Cellulose/Lignin Aerogel as Adsorbent for Efficiently Adsorptive Removal of Trimethoprim from Water

Jiahao Liu,Jun Zhang,Na Li

An environmentally friendly aerogel (CE/LN), composed of cellulose (CE) and lignin (LN) was synthesized via one-pot cross-linking method at room temperature, followed by freeze-drying. The CE/LN aerogel exhibited good mechanical properties and abundant functional groups, making it an effective adsorbent for the adsorption and removal of the antibiotic trimethoprim (TMP) from water. This study systematically investigated the effects of solution pH, aerogel dose, contact time, initial TMP concentration, temperature, ionic strength, and various water sources on TMP adsorption. The CE/LN aerogel demonstrated a higher TMP adsorption capacity of TMP than the CE aerogel across the entire pH range of 2.0–10.0. The adsorption process was well described by the Langmuir isotherm model and pseudo-second-order kinetic model. The CE/LN 6% aerogel achieved the highest adsorption capacity, with a theoretically maximal TMP uptake of 0.402 mmol/g at pH 6.0. However, high concentration of NaCl significantly reduced TMP uptake on the CE/LN 6% aerogel due to the electrostatic shielding effect. After five cycles of adsorption, the CE/LN 6% aerogel maintained a high adsorption capacity, retaining 80% of its initial uptake. Combined with the adsorption performance, the Fourier transform infrared spectrometry and X-ray photoelectron spectroscopy analyses spectroscopy analysis before and after TMP adsorption, it can be concluded that the adsorption mechanism involved hydrogen bonding, electrostatic interactions, and π–π electron donor–acceptor interactions. Thus, the CE/LN aerogel, with its simple preparation method, good adsorption capacity, and reusability, shows great potential for widespread application in the removal of antibiotics from water.

Biofilm enhanced the mitigations of antibiotics and resistome in sulfadiazine and trimethoprim co-contaminated soils

Zhi Mei,Fang Wang,Yuhao Fu,Yu Liu,Syed A. Hashsham,Yu Wang,Jean Damascene Harindintwali,Qingyuan Dou,Marko Virta,Xin Jiang,Yu Deng,Tong Zhang,James M. Tiedje

Reducing antibiotic levels in soil ecosystems is vital to curb the dissemination of antimicrobial resistance genes (ARGs) and mitigate global health threats. However, gaps persist in understanding how antibiotic resistome can be suppressed during antibiotic degradation. Herein, we investigate the efficacy of a biochar biofilm incorporating antibiotics-degrading bacterial strain ( Arthrobacter sp. D2) to mitigate antibiotic resistome in non-manured and manure-amended soils with sulfadiazine (SDZ) and trimethoprim (TMP) contamination. Results show that biofilm enhanced SDZ degradation by 83.0% within three days and increased TMP attenuation by 55.4% over 60 days in non-manured soils. In the non-manured black soil, the relative abundance of ARGs increased initially after biofilm inoculation. However, by day 30, it decreased by 20.5% compared to the controls. Moreover, after 7 days, biofilm reduced TMP by 38.5% in manured soils and decreased the total ARG abundance by 19.0%. Thus, while SDZ degradation did not increase sulfonamide resistance genes, TMP dissipation led to a proliferation of insertion sequences and related TMP resistance genes. This study underscores the importance of antibiotic degradation in reducing related ARGs while cautioning against the potential proliferation and various ARGs transfer by resistant microorganisms.

Degradation of trimethoprim in aqueous by persulfate activated with nanosecond pulsed gas-liquid discharge plasma

Jian-Ping Liang,Xiong-Feng Zhou,Zi-Lu Zhao,De-Zheng Yang,Wen-Chun Wang

The persulfate activation by nanosecond pulsed gas-liquid discharge (NPG-LD) is employed to degrade the trimethoprim (TMP) in water. The results show that persulfate addition enhances the degradation of TMP by NPG-LD through an obvious synergetic effect. With treatment time of 50 min, the high removal efficiency and energy yield reach 94.6% and 0.57 gkWh −1 in air NPG-LD with the addition of persulfate, respectively, which is 13.5% and 0.09 gkWh −1 higher than that in solo air NPG-LD, respectively. Correspondingly, the calculated synergetic factor achieves 1.62, indicating the synergetic effect is established. The activation mechanism of persulfate by NPG-LD is analyzed by the measurement of reactive species and the effects of radical scavenger addition on TMP removal. It is found that the synergetic effect between NPG-LD and persulfate is attributed to the increased production of OH, H 2 O 2 , and . Besides, the TMP degradation by NPG-LD and persulfate synergetic system is influenced by discharge working gas, pulse voltage, addition dosage of persulfate, initial TMP concentration, and initial pH value. Subsequently, the degradation pathway of TMP is analyzed using LC-MS/MS.

Establishing Plasma Concentration Methodology for Trimethoprim/Sulfamethoxazole: Application in Therapeutic Drug Monitoring of Critically Ill Patients

Xiangxiang Fu,Jiao Fu,Haibiao Gong,Lili Zhong,FangLing Wang,MengYu Mei,Bingli Zhang,Wenya Xu,Xian Zhang,Min Wang,Wanyang Sun,Lei Yang

This study aimed to develop a high-throughput and high-sensitivity method for simultaneously determining trimethoprim (TMP), sulfamethoxazole (SMX), and its metabolite acetyl- N -sulfamethoxazole (NSMX) in critically ill patients' plasma. The samples were precipitated using methanol and then purified using the Oasis PRiME HLB solid-phase extraction column. The mobile phase consisted of a mixture of 40% 0.1% formic acid aqueous solution and 60% acetonitrile, with a flow rate of 0.35 mL/min. Analyses were conducted using UPLC-Q-Exactive Plus Orbitrap mass spectrometry in positive ion mode. With only 50 μL of plasma required for accurate quantification, this method was applicable for therapeutic drug monitoring (TDM). Validation demonstrated precision and accuracy within 15% of nominal values. The samples remained stable for 12 h at 4°C and −20°C and for 15 days at −20°C and −80°C, which ensured the reliability of the method during sample storage and handling under different environmental conditions. The method can be applied for concentration monitoring and pharmacokinetic studies, enabling TDM and dose adjustments for critically ill patients. This approach will enhance patient outcomes and reduce medical resource waste, offering reliable sensitivity and specificity to support precision medicine in critical care.

Insight into the interaction between trimethoprim and soluble microbial products produced from biological wastewater treatment processes

Runze Xu,Fang Fang,Longfei Wang,Jingyang Luo,Jiashun Cao

Soluble microbial products (SMPs), dissolved organic matter excreted by activated sludge, can interact with antibiotics in wastewater and natural water bodies. Interactions between SMPs and antibiotics can influence antibiotic migration, transformation, and toxicity but the mechanisms involved in such interactions are not fully understood. In this study, integrated spectroscopy approaches were used to investigate the mechanisms involved in interactions between SMPs and a representative antibiotic, trimethoprim (TMP), which has a low biodegradation rate and has been detected in wastewater. The results of liquid chromatography-organic carbon detection-organic nitrogen detection indicated that the SMPs used in the study contained 15% biopolymers and 28% humic-like substances (based on the total dissolved organic carbon concentration) so would have contained sites that could interact with TMP . A linear relationship of fluorescent intensities of tryptophan protein-like substances in SMP was observed ( R 2 >0.99), indicating that the fluorescence enhancement between SMP and TMP occurred. Fourier-transform infrared spectroscopy and X-ray photoelectron spectroscopy indicated that carboxyl, carbonyl, and hydroxyl groups were the main functional groups involved in the interactions. The electrostatic and π-π interactions were discovered by the UV-vis spectra and 1 H nuclear magnetic resonance spectra. Structural representations of the interactions between representative SMP subcomponents and TMP were calculated using density functional theory, and the results confirmed the conclusions drawn from the 1 H nuclear magnetic resonance spectra. The results help characterize SMP–TMP complexes and will help understand antibiotic transformations in wastewater treatment plants and aquatic environments.

Adsorption–Desorption Behaviors of Enrofloxacin and Trimethoprim and Their Interactions with Typical Microplastics in Aqueous Systems

Zhichao Li,Xiao Meng,Xiaoyong Shi,Chunyue Li,Chuansong Zhang

Microplastics can transfer antibiotics in water through adsorption and desorption, causing adverse effects on the water environment. Therefore, understanding the interaction between microplastics and antibiotics is important in order to assess their impact on the environment. In this study, the adsorption–desorption behaviors of two commonly used antibiotics [enrofloxacin (ENR) and trimethoprim (TMP)] in aquaculture and their interactions with three typical microplastics [polystyrene (PS), polyvinyl chloride (PVC), and polyethylene (PE)] were investigated through laboratory experiments. The results showed that the adsorption capacity of the three microplastics was 1.229–1.698 mg/g for ENR and 1.110–1.306 mg/g for TMP, correlating with the octanol–water partition coefficients (logKow) of antibiotics. Due to the larger specific surface areas and special functional groups of microplastics, the antibiotic adsorption capacity of PS and PVC was higher than that of PE. The adsorption behavior followed pseudo-second-order kinetics and a Freundlich isotherm model, indicating a non-uniform surface with multilayer adsorption. A thermodynamic analysis showed that these were all spontaneous endothermic adsorptions. X-ray photoelectron spectroscopy (XPS) and Fourier-transform infrared spectroscopy (FTIR) analyses indicated that the adsorption mechanism was dominated by physical adsorption, involving π–π conjugation, halogen bonds, hydrogen bonding, and electrostatic interactions. High salinity and alkaline environments were conducive to desorption, and the ENR and TMP desorption rates of the microplastics ranged from 20.65% to 24.95%. This indicates that microplastics adsorbed with antibiotics will desorb antibiotics when entering the seawater system, thereby affecting marine ecosystems. These findings reveal the interaction mechanism between microplastics and aquaculture antibiotics in aqueous systems, providing theoretical support for environmental protection and sustainable development.
