Development of a kaolin-based Bacillus velezensis BF01 composite for application in biocontrol of postharvest strawberry diseases

Hongling Lu,Suxia Guo,Qiannan Wang,Bang An

BACKGROUND Biological control agents are promising alternatives to conventional fungicides for managing postharvest diseases but their practical use is hindered by variable effectiveness and by application challenges. One promising yet underexplored approach involves utilizing bacteria that produce volatile organic compounds (VOCs) with antifungal properties. RESULTS In this study, Bacillus velezensis BF01, a strain with significant antifungal activity, was identified and isolated. Volatile organic compounds produced by this strain inhibited the growth and conidial germination of Botrytis cinerea , Fusarium oxysporum , and Colletotrichum gloeosporioides . Gas chromatography-mass spectrometry identified 90 VOCs, with ketones, esters, nitrogen compounds, aldehydes, and alcohols as the dominant components, many of which exhibited antifungal or plant resistance-inducing properties. To enhance practical application, B. velezensis BF01 was incorporated into a kaolin-based composite, which maintained long-term viability under refrigerated conditions and inhibited fungal growth through the controlled release of volatile organic compounds. This composite significantly extended the shelf life of strawberries by reducing postharvest decay. CONCLUSION The development of a kaolin-based B. velezensis BF01 composite provides an effective and sustainable biocontrol strategy for postharvest disease management. By improving bacterial stability and facilitating the controlled release of antifungal volatile organic compounds, this approach offers a practical alternative to chemical fungicides, highlighting its potential in extending fruit shelf life and improving food safety. © 2025 Society of Chemical Industry.

Fermentation of millet bran with bacillus natto: enhancement of bioactive levels and the bioactivity of bran extract

Shimei Zhang,An Wang,Zhaoxin Lu,Fengxia Lu,Haizhen Zhao

Background Millet bran (MB), a byproduct of millet production, is rich in functional components, but has a low utilization rate. In recent years, researchers have shown that fermentation can improve the biological activities of cereals and their byproducts. This study used Bacillus natto to ferment millet bran to improve its added value and broaden the application areas of MB. The bioactive component content, physicochemical properties, and functional activities of millet bran extract (MBE) from fermented millet bran were determined. Results After fermentation, the contents of soluble dietary fiber (SDF), β-glucan, polypeptides, polyphenols, flavones, and total amino acids increased by 92.0%, 164.4%, 111.4%, 32.5%, 16.4%, and 95.4%, respectively. Scanning electron microscopy revealed that the microscopic morphology of MBE changed from complete and dense blocks to loosely porous shapes after fermentation. After fermentation, the solubility, water holding capacity, and viscosity significantly increased, and the particle size decreased. Moreover, the glucose adsorption capacity (2.1 mmol/g), glucose dialysis retardation index (75.3%), and α-glucosidase inhibitory (71.4%, mixed reversible inhibition) activity of the fermented MBE (FMBE) were greater than those of the unfermented MBE (0.99 mmol/g, 32.1%, and 35.1%, respectively). FMBE presented better cholesterol and sodium cholate (SC) adsorption properties, and the adsorption was considered inhomogeneous surface adsorption. Conclusion Fermentation increased the bioactive component content and improved the physicochemical properties of MBE, thereby improving its hypoglycemic and hypolipidemic properties. This study will not only resolve the problem of millet bran waste but also promote the development of a higher value-added application method for millet bran. This article is protected by copyright. MOF-818 nanozyme-based biosensor for specific point-of-care testing of Bacillus subtilis in complex agricultural soil

Wu Chaoqian,Wang Yanlin,Li Nana,Zhang Haoqing,Ge Tida,Yu Shaoning

Bacillus subtilis (BS) effectively suppresses soilborne pathogens and enhances crop resistance through various mechanisms. However, its efficacy is significantly influenced by effective inter-root colonization rates, among other factors, necessitating precise quantitative monitoring for optimal application. Conventional detection methods are often time-consuming and technically demanding, limiting their application in the field. To address this challenge, we developed a dual-recognition biosensing platform. Specifically, tridentate-functionalized aptamer was utilized for rapid BS enrichment and secondary recognition achieved using fragment crystallizable mannose-binding lectin (FcMBL)-modified nanozyme (MOF-818). The rigid scaffold of MOF-818 ensured superior catechol oxidase activity and environmental stability, facilitating sensitive and robust online quantification of BS in complex soil matrices. The linear detection range of the platform was 10 2 –10 7  CFU/mL, and the detection limit was 12 CFU/mL. This work provides a reliable tool for real-time monitoring of BS in agricultural systems, facilitating its efficient use in biocontrol and soil health management. Enhanced dimethyl phthalate phytoremediation efficiency by C4-HSL-assisted Bacillus sp. YT2: Roles of rhizosphere microbiome-plant interaction

Xiaoxian Cai,Ke Chen,Lixue Zheng,Huijun Guan,Tianlong Hao,Wenqing Zhou,Jingjing Yang,Lihua Qi,Chunfeng Guan

The widespread accumulation of organic pollutants in agricultural soils, such as dimethyl phthalate (DMP), poses serious risks to human health and ecological safety. The application of plant growth-promoting rhizobacteria (PGPR) to improve phytoremediation efficiency for organic pollutant-contaminated soils has gained increasing attention. However, synergistic PGPR-plant signaling and competitive root colonization of PGPR directly affect the remedial efficiency of the PGPR-plant system for soil organic pollutants. Acyl-homoserine lactones (AHLs), one of the quorum sensing (QS) signals, can enhance microbial cooperation and promote plant growth, thereby probably facilitating phytoremediation processes for pollutant degradation. In this study, the potential capacity of AHLs to assist PGPR in the phytoremediation of DMP-polluted soils was explored, with a focus on their effects on DMP removal, plant physiological functions, and rhizosphere microbial community structure. The results showed that AHLs (C 4 -HSL)-assisted PGPR ( Bacillus YT2) treatment significantly increased DMP removal to 92.60 % and improved plant growth. Compared to the individual AHLs or PGPR treatments, the AHLs-assisted PGPR more effectively enhanced plant photosynthesis, osmoregulation, and antioxidant capacity, reduced oxidative damage, and improved soil enzyme activity and nutrient availability. Additionally, the AHLs-assisted PGPR treatment modulated microbial community in the rhizosphere, increasing the richness of pivotal functional taxa such as Xanthomonadaceae and Acidobacteriota. AHLs might directly promote DMP degradation by regulating the QS system and enhancing plant-bacterial communication, while also indirectly supporting plant growth through stress tolerance enhancement. In conclusion, AHLs-mediated enhancement of PGPR functionality provides a promising approach for remediating DMP-contaminated soils and offers a theoretical basis for developing composite microbial formulations.

AFB1 Microbial Degradation by Bacillus subtilis WJ6 and Its Degradation Mechanism Exploration Based on the Comparative Transcriptomics Approach

Peizhou Yang,Wenjing Wu,Danfeng Zhang,Lili Cao,Jieshun Cheng

Aflatoxin pollution poses great harm to human and animal health and causes huge economic losses. The biological detoxification method that utilizes microorganisms and their secreted enzymes to degrade aflatoxin has the advantages of strong specificity, high efficiency, and no pollution inflicted onto the environment. In this study,Bacillus subtilisWJ6 with a high efficiency in aflatoxin B1degradation was screened and identified through molecular identification, physiological, and biochemical methods. The fermentation broth, cell-free supernatant, and cell suspension degraded 81.57%, 73.27%, and 8.39% of AFB1, respectively. The comparative transcriptomics analysis indicated that AFB1led to 60 up-regulated genes and 31 down-regulated genes inB. subtilisWJ6. A gene ontology (GO) analysis showed that the function classifications of cell aggregation, the organizational aspect, and the structural molecule activity were all of large proportions among the up-regulated genes. The down-regulated gene expression was mainly related to the multi-organism process function under the fermentation condition. Therefore,B. subtilisWJ6 degraded AFB1through secreted extracellular enzymes with the up-regulated genes of structural molecule activity and down-regulated genes of multi-organism process function. Keywords:aflatoxin B1;Bacillus subtilis;microbial degradation;comparative transcriptomics;biological detoxification

Function-driven design of Bacillus kochii and Filobasidium magnum co-culture to improve quality of flue-cured tobacco

Xinying Wu,Wen Cai,Pengcheng Zhu,Zheng Peng,Tianfei Zheng,Dongliang Li,Jianghua Li,Guanyu Zhou,Juan Zhang,Guocheng Du

Flue-cured tobacco (FCT) is an economical raw material whose quality affects the quality and cost of the derived product. However, the time-consuming and inefficient spontaneous aging is the primary process for improving the FCT quality in the industry. In this study, a function-driven co-culture with functional microorganisms was built in response to the quality-driven need for less irritation and more aroma in FCT. The previous study has found that Bacillus kochii SC could degrade starch and protein to reduce tobacco irritation and off-flavors. The Filobasidium magnum F7 with high lipoxygenase activity was screened out for degrading higher fatty acid esters and terpenoids to promote the aroma and flavor of FCT. Co-cultivation with strain SC and F7 obtained better quality improvement than mono-culture at an initial inoculation ratio of 1:3 for 2 days, representing a significant breakthrough in efficiency and a reduction in production costs compared to the more than 2 years required for the spontaneous aging process. Through the analysis of microbial diversity, predicted flora functions, enzyme activities and volatile compositions within the mono- and co-cultivation, our study showed the formation of a function-driven co-culture between two strains through functional division of labor and nutritional feeding. Herein, the function-driven co-culture via bioaugmentation will become an increasingly implemented approach for the tobacco industry.

Protein Hydrolysates from Pleurotus geesteranus Modified by Bacillus amyloliquefaciens γ-Glutamyl Transpeptidase Exhibit a Remarkable Taste-Enhancing Effect

Xiaozhou Xia,Yu Fu,Liang Ma,Hankun Zhu,Yong Yu,Hongjie Dai,Jiadong Han,Xin Liu,Zhengfang Liu,Yuhao Zhang

Long-term high salt intake exerts a negative impact on human health. The excessive use of sodium substitutes in the food industry can lead to decreased sensory quality of food. γ-Glutamyl peptides with pronounced taste-enhancing effects can offer an alternative approach to salt reduction. However, the content and yield of γ-glutamyl peptides in natural foods are relatively low. Enzyme-catalyzed synthesis of γ-glutamyl peptides provides a feasible solution. In this study, Pleurotus geesteranus was hydrolyzed by Flavourzyme to generate protein hydrolysates. Subsequently, they were modified by Bacillus amyloliquefaciens γ-glutamyl transpeptidase to generate γ-glutamyl peptides. The reaction conditions were optimized and their taste-enhancing effects were evaluated. Their peptide sequences were identified by parallel reaction monitoring with liquid chromatography–tandem mass spectrometry and analyzed using molecular docking. The optimal conditions for generation of γ-glutamyl peptides were a pH of 10.0, an enzyme condition of 1.2 U/g, and a reaction time of 2 h, which can elicit a strong kokumi taste. Notably, it exhibited a remarkable taste-enhancing effect for umami intensity (76.07%) and saltiness intensity (1.23-fold). Several novel γ-glutamyl peptide sequences were found by liquid chromatography–tandem mass spectrometry, whereas the binding to the calcium-sensing receptor was confirmed by molecular docking analysis. Overall, γ-glutamyl peptides from P. geesteranus could significantly enhance the umami and salt tastes, which can serve as promising taste enhancers.

Improving the Production of Riboflavin by Introducing a Mutant Ribulose 5-Phosphate 3-Epimerase Gene in Bacillus subtilis

Yang Bin,Sun Yiwen,Fu Shouying,Xia Miaomiao,Su Yuan,Liu Chuan,Zhang Chunzhi,Zhang Dawei

Ribulose 5-phosphate (Ru5P) and guanosine 5′-triphosphate (GTP) are two key precursors of riboflavin, whereby Ru5P is also a precursor of GTP. Ribulose 5-phosphate 3-epimerase (Rpe) catalyzes the conversion of ribulose 5-phosphate into xylulose 5-phosphate. Inactivation of Rpe can reduce the consumption of Ru5P, enhancing the carbon flux toward riboflavin biosynthesis. Here we investigated the effect of mutation of rpe and other related genes on riboflavin production, physiological and metabolic phenotypes in Bacillus subtilis LY (BSLY). Introducing single nucleotide deletion (generated BSR) or nonsense mutation (generated BSRN) on the genomic copy of rpe, resulting in more than fivefold increase of riboflavin production over the parental strain. BSR process 62% Rpe activity, while BSRN lost the entire Rpe activity and had a growth defect compared with the parent strain. BSR and BSRN exhibited increases of the inosine and guanine titers, in addition, BSRN exhibited an increase of inosine 5′-monophosphate titer in fermentation. The transcription levels of most oxidative pentose phosphate pathway and purine synthesis genes were unchanged in BSR, except for the levels of zwf and ndk, which were higher than in BSLY. The production of riboflavin was increased to 479.90 ± 33.21 mg/L when ribA was overexpressed in BSR. The overexpression of zwf, gntZ, prs, and purF also enhanced the riboflavin production. Finally, overexpression of the rib operon by the pMX45 plasmid and mutant gnd by pHP03 plasmid in BSR led to a 3.05-fold increase of the riboflavin production (977.29 ± 63.44 mg/L), showing the potential for further engineering of this strain.

Enhanced extracellular raw starch-degrading α-amylase production in Bacillus subtilis through signal peptide and translation efficiency optimization

He Li,Dongbang Yao,Jingru Ying,Xudong Han,Xuecheng Zhang,Xiuju Fang,Zemin Fang,Yazhong Xiao

A strategy that combines signal peptide (SP) optimization and translation efficiency improvement was used to enhance the extracellular production of raw starch-degrading α-amylase AmyZ1 from Pontibacillus sp. ZY in Bacillus subtilis . The optimal SP YpuA screened from 173 B. subtilis SPs mediated a 1.28-fold higher AmyZ1 activity than the original SP amyQ . Then, strategies of translation efficiency improvement, including optimizing the Shine-Dalgarno-like (SD-like) sequence, constructing a dual 5′-untranslated region (dual-UTR), optimizing the spacer region of the UTR, and optimizing the 5′-proximal coding sequence, were used to enhance AmyZ1 production. The resulting recombinant strain (BZYACO6) containing SD-like3, dUTR2, and an optimized 5′-proximal coding sequence (O6) produced the highest extracellular α-amylase activity of 2974.0 U/mL in shake flask, 2.07-fold higher than that of the original strain BZd34 (1437.6 U/mL). After fermentation optimization, the BZYACO6 strain produced an extracellular AmyZ1 activity of 3915.2 U/mL in shake flask and 25,070 U/mL in 3-L fermenter, representing the highest level of recombinant raw starch-degrading α-amylase production reported to date. Furthermore, this work provides a strategic reference for enhancing the extracellular production of other recombinant proteins in B. subtilis .

The Synergistic Effect of High Pressure CO2 and Nisin on Inactivation of Bacillus subtilis Spores in Aqueous Solutions

Rao Lei,Wang Yongtao,Chen Fang,Liao Xiaojun

The inactivation effects of high pressure CO2 + nisin (simultaneous treatment of HPCD and nisin, HPCD + nisin), HPCD→nisin (HPCD was followed by nisin), and nisin→HPCD (nisin was followed by HPCD) treatments on Bacillus subtilis spores in aqueous solutions were compared. The spores were treated by HPCD at 6.5 or 20 MPa, 84–86°C and 0–30 min, and the concentration of nisin was 0.02%. Treated spores were examined for the viability, the permeability of inner membrane (IM) using flow cytometry method and pyridine-2, 6-dicarboxylic acid (DPA) release, and structural damage by transmission electron microscopy. A synergistic effect of HPCD + nisin treatment on inactivation of the spores was found, and the inactivation efficiency of the spores was HPCD + nisin > HPCD→nisin or nisin→HPCD. Moreover, HPCD + nisin caused higher IM permeability and DPA release of the spores than HPCD. A possible action mode of nisin-enhanced inactivation of the spores was suggested as that HPCD firstly damaged the coat and cortex of spores, and nisin penetrated into and acted on the IM of spores, which increased the damage to the IM of spores, and resulted in higher inactivation of the spores.

Characterization of a novel carboxylesterase from Bacillus velezensis SYBC H47 and its application in degradation of phthalate esters

Lin Huang,Di Meng,Qiaopeng Tian,Shaolan Yang,Huaxiang Deng,Zhengbing Guan,Yujie Cai,Xiangru Liao

Recently, residual plasticizer phthalate esters (PAEs) in the different environments pose a serious health threat to humans and mammals. Biodegradation has been considered a promising and eco-friendly way to eliminate PAEs. In this study, a gene ( baces04 ) encoding the novel PAEs hydrolase , carboxylesterase (BaCEs04), was screened from the genome of Bacillus velezensis SYBC H47 via bioinformatics analysis. Then, baces04 was cloned and expressed in Escherichia coli BL21 (DE3). BaCEs04 belonged to the esterase family VI. It contained a conserved domain (Gly 159 -His 160 -Ser 161 -Leu 162 -Gly 163 ) and a typical serine hydrolase catalytic site (Ser 161 -Asp 204 -His 261 ). The characterization of BaCEs04 showed that the activity was optimal at 60°C and pH 7.5. This enzyme also displayed high resistance to metal ions, organic solvents , and detergents. After treatment with BaCEs04 for 5 h, the degradation ratio of four different 1 mM PAEs, including dimethyl phthalate, diethyl phthalate, dipropyl phthalate, and dibutyl phthalate, was 32.4%, 50.5%, 77.9%, and 86.8%, respectively. The degradation products of four PAEs were identified as their corresponding monoalkyl phthalates. This is the first report that family VI esterase displaying PAE-hydrolysis activity. This study also proved that BaCEs04 could be used as an ideal candidate for the application in bioremediation and industry.

Mechanistic insights into extracellular reductive dechlorination of hexachloroethane by common non-electroactive bacteria Bacillus subtilis and Escherichia coli

Sadiq Naveed,Xinwei Zhou,Dongqiang Zhu

Extracellular reductive dechlorination of chlorinated aliphatic hydrocarbons (CAHs) by non-electroactive bacteria is rarely investigated. In this study, we demonstrated that common bacteria Gram-positive Bacillus subtilis (B. subtilis) and Gram-negative Escherichia coli (E. coli) in aqueous suspensions (1.2 × 108 cells per mL) could dechlorinate a model CAH compound hexachloroethane (HCE, initial 40 µmol L−1) at ratios of 83.7% and 54.1% respectively to pentachloroethane (PCE) and tetrachloroethylene (TCE) after 56 h incubation at pH 7.0 and 30 °C. The majority of the parent compound (HCE) and reaction products (PCE and TCE) present in the extracellular matrix indicated the predominance of extracellular reduction. Removing EPS from bacteria suppressed HCE reduction, while adding extra dissolved EPS slightly enhanced HCE reduction. The enhanced reduction of HCE by low-molecular-weight EPS (<3 kDa) relative to bulk EPS of B. subtilis and its counterpart high-molecular-weight EPS (>3 kDa) indicated that low-molecular-weight EPS were more enriched in active reducing agents. Additionally, dechlorination of HCE by electrochemically reduced glutathione and disodium anthraquinone-2,6-disulfonate or juglone suggested that thiol groups and quinones were involved as reducing agents and electron mediators, respectively. These findings underscore a previously unknown process of extracellular reductive dechlorination of CAHs by common non-electroactive bacteria and the key role played by EPS.
