Gentamicin decorated phosphatidylcholine-chitosan nanoparticles against biofilms and intracellular bacteria

Yuanhao Qiu,Dan Xu,Guoqing Sui,Dongdong Wang,Ming Wu,Lipeng Han,Haibo Mu,Jinyou Duan

Biofilms and intracellular bacteria often cause a series of overwhelming public health issues due to their strong drug resistance. Hence, chitosan nanoparticles (CS NPs), phosphatidylcholine and gentamicin were used to synthesize a novel nanodrug delivery system (GPC NPs). Dynamic light scattering (DLS) demonstrated that the surface zeta-potential of GPC NPs was −19.5 mV. The morphology of GPC NPs was observed by scanning electron microscopy (SEM). The gentamicin adsorption and release behaviors of GPC NPs were also investigated. The GPC NPs could effectively damage and remove the biofilm formed by pathogens through permeation of the antibiotic into the biofilm. In addition, the nanoparticles were readily engulfed by macrophages which facilitated the killing of intracellular bacteria and had neglectable cytotoxicity. Our study indicated that GPC NPs could be used as a promising nanoantibacterial agent against biofilms and intracellular bacteria.

Adsorption of aniline on aminated chitosan/graphene oxide composite material

Zhang Yi,Li Huajie,Li Mingchun,Xin Meihua

On the basis of aminated chitosan, graphene oxide was blended to prepare ACS/GO composite material. The composite material was characterized by SEM and X-ray diffraction, and applied to the adsorption of aniline from aqueous solution and cigarette smoke. The results showed that when ACS/GO composite material was prepared, graphene oxide was more dispersed and no agglomeration occurred, the surface area of ACS/GO was 7.460 m 2 /g. The adsorption of aniline by ACS/GO could meet both the Freundlich adsorption model and the Langmuir adsorption model in water. Meanwhile, the kinetics data of the adsorption of aniline by ACS/GO was fit better into the pseudo-second-order equation. The addition of GO could help to improve the adsorption performance of ACS on aniline in cigarette smoke.

Tuning the Biodegradability of Chitosan Membranes: Characterization and Conceptual Design

Shuyu Shi,Xin Liu,Weiyi Li,Zhuo Li,Guoquan Tu,Baolin Deng,Chongxuan Liu

Sustainable development entails the revival of biopolymer-based membranes for water treatment with the expectation of addressing concerns relevant to the depletion of fossil-based resources and their negative impacts on the environment. However, it is challenging to tune the biodegradability of biopolymer-based membranes such that the filtration performance would be maintained during the service life while reducing the environmental footprint. This study established a conceptual framework of achieving tunable biodegradability by incorporating chitosan membranes with silver nanoparticles. In particular, the membranes were fabricated on the basis of dissolving chitosan in an aqueous urea/alkali solution, which enabled a green approach for forming various porous substructures via phase inversion. The effects of biodegradation on chitosan membranes were investigated in the context of correlating the filtration performance with the geometrical variations in the polymer network. A series of characterization experiments were implemented to corroborate that the degree of protecting the porous substructures against enzymatic digestion should be a function of the silver loading, whereas the service life would be dependent on the silver-leaching rate. Moreover, it was revealed that optimizing the coagulation conditions could be an effective way to control the silver release during the filtration process.

Pengjie Lin,Liangling Liu,Guanghua He,Tian Zhang,Ming Yang,Jinzhong Cai,Lihong Fan,Shengxiang Tao

Hydrogel wound dressing is a type of hydrophilic polymer, which has been widely studied and applied in biomedical field. In this study, a simple and non-toxic method was developed to prepare a new type of composite hydrogel, which was formed through the Schiff-base reaction between the aldehyde of Oxidized Hydroxyethyl Cellulose (OHEC) and the amino of Carboxymethyl Chitosan (CMCS). Hence, a series of tests toward this new composite hydrogel which contained its structure and performance was applied. Statistics achieved from those tests showed that this composite hydrogel comprised of some high-quality properties such as suitable gelation time, good swelling ability, suitable water evaporation rate, good blood compatibility and biocompatibility. Considering these properties, this hydrogel has a potential to be explored as wound dressing.

Eugenol nanocapsules embedded with gelatin-chitosan for chilled pork preservation

Qian Wang,Li Zhang,Wu Ding

Chilled pork is widely consumed in China. However, various contaminants during storage directly lead to a decline in the quality of chilled pork products. An extract of natural plant sources, eugenol (Eug) exerts good antibacterial and antioxidant effects. Nanometerization was used in this study to improve the insoluble and volatile characteristics of Eug. Eug nanocapsules embedded with gelatin/chitosan (Eug-Gel-CS NPs) were used to preserve chilled pork. Results indicated that Eug-Gel-CS NPs could effectively inhibit increases in the pH, total volatile basic nitrogen (TVB-N), and thiobarbituric acid-reactive substances (TBARS) of chilled pork than that of the Eug group ( p  < 0.05). The L * and a * values of the Eug-Gel-CS NPs group were significantly higher than those of the Eug and gelatin-chitosan (CS-Gel) groups ( p  < 0.05). The total number of colonies (TBC) showed that the storage period of the Eug-Gel-CS NPs group could be extended to 15 d, which was significantly different from that of the CK group (8 d) ( p  < 0.05). The Eug-Gel-CS NPs also effectively delayed the decline in the water- holding capacity (WHC), springiness, and cohesiveness of the chilled pork. Therefore, Eug-Gel-CS NPs exert good antiseptic, antibacterial, and antioxidative effects on preserved chilled pork.

Application of Pullulan and Chitosan Multilayer Coatings in Fresh Papayas

Linyun Zhang,Chongxing Huang,Hui Zhao

In this work, some multilayer coatings (two-layer, four-layer or six-layer) based on pullulan and chitosan for protecting papayas were prepared by the layer-by-layer technique. The papayas were coated by immersion and stored at 25 °C, 50% relative humidity or up to 14 days. Uncoated and monolayer-coated papayas were used as controls. The pullulan/chitosan coatings decreased the papaya weight loss, softening, color change (b*, ΔE), and pH, retarded the fall of titratable acidity and vitamin C, and maintained respiratory rate and soluble solid contents. Sensory quality evaluation demonstrated that pullulan/chitosan coatings effectively preserved papaya flavor and overall acceptance. In general, the four-layer coatings provided the best fruit preservation. In conclusion, multilayer pullulan/chitosan coatings are efficient in maintaining the post-harvest quality and prolonging the shelf life of fresh papaya. Keywords:papaya;preservation;pullulan;chitosan;multilayer coating

Chitosan-functionalized graphene oxide as adjuvant in HEV P239 vaccine

Qianyu Bai,Zhiwen Wang,Yina AN,Jijing Tian,Zhilin Li,Yifei Yang,Yanjun Dong,Mingyong Chen,Tianlong Liu

Searching appropriate adjuvants for vaccine is a potent method to intense the immune efficacy. In the present study, we developed a novel Hepatitis E virus (HEV) vaccine by utilizing chitosan modified nano-graphene oxide (GO-CS) as an adjuvant to support HEV antigen P239 protein (GO/CS/P239). The characterization of GO/CS/P239 was observed by atomic force microscope. The safety of GO/CS/P239 was measured by CCK-8 method, hemolysis test and acute challenge test. The anti-HEV titers and cytokines production were analyzed by double antibody sandwich ELISA . As the results showed, by contrast with a vaccine that contained only the P239 protein, GO/CS/P239 vaccine can promote immune cells to produce more IgG antibodies and cytokines, which were able to stimulate the organism to produce stronger both cellular and humoral immunity. Collectively, GO/CS/P239 particles have been demonstrated to be safe both in vitro and in vivo, and can facilitate sufficient immune response to protect organisms from virus infection , which suggested that our exploration offers a promising alternative vaccine that can control HEV infection.

Photothermal antibacterial MoS2 composited chitosan hydrogel for infectious wound healing

Yue Wang,Kun Liu,Kai Huang,Wenying Wei,Ye Huang,Honglian Dai

Pathological bacterial infection poses a serious threat to public health security. The excessive use of antibiotics has resulted in a serious decline in treatment effect and bacterial resistance. For the treatment of infected wounds, we compounded dopamine-assisted exfoliated molybdenum disulfide (MoS 2 @PDA) into lipoic acid modified chitosan (LAMC) to obtain a composite hydrogel dressing (LAMC-MoS 2 @PDA). LAMC-MoS 2 @PDA hydrogels exhibited excellent photothermal conversion ability and the LAMC-MoS 2 @PDA2 group (0.3 wt%) has a photothermal conversion efficiency of 26.29 %. Meanwhile, they showed good biocompatibility and ROS scavenging activity in vitro. Photothermal therapy usually utilizes photothermal agents to convert near-infrared light into heat energy for bacterial cell membrane destruction and bacterial protein inactivation. Under the near-infrared light irradiation, the antibacterial ratio of LAMC-MoS 2 @PDA hydrogels against Staphylococcus aureus and Escherichia coli reached nearly 100 %, and the morphology of the bacteria showed obvious contraction and cleavage. The hydrogels also showed an excellent antibacterial effect and wound healing promotion in the infected wound of rats. In particular, the LAMC-MoS 2 @PDA2 (+) group (with NIR) showed almost complete wound closure after 14 days, indicating that the LAMC-MoS 2 @PDA hydrogels have great potential in clinical anti-infected treatment.

Oxidized chitosan modified electrospun scaffolds for controllable release of acyclovir

Mehdihasan I. Shekh,Jhaleh Amirian,Florian J. Stadler,Bing Du,Yanxia Zhu

Developing a novel scaffold carrier with a sustained and controllable release profile of drug is essential to promote the effective transdermal delivery for acyclovir (ACY). In this work, electrospun polyacrylonitrile nanofibers (PAN NFs) was chemically modified with oxidized chitosan (OC). The modified fibrous scaffold was further loaded with the ACY for drug released investigation. FT-IR and NMR results revealed that the conversion of the functional group for each step has successfully occurred on the surface of the fibers. Through the in-vitro drug release and kinetic study, it demonstrated that ACY could be sustainably and controlled released from the OC modified scaffold following the Korsmeyer-Peppas model with a Fickian diffusion mechanism. The human adipose-derived stem cells and the blood combability evaluation confirmed the obtained scaffold possessed excellent cell biocompatibility and hemocompatibility. It could be concluded that the resultant OC modified scaffold based on electrospun PAN NFs opened a new potential option for the topical/transdermal drug delivery of ACY.

Chitosan-polyvinyl alcohol-diatomite hydrogel removes methylene blue from water

Yanfei Gao,Pingxiong Cai,Lei Zhong,Ruixian Zhang,Xueyi Hou,Xiuxiu Ren,Junzhong Wang,Xiaokun Chu,Yanyue Lu,Zeguang Zhou

Dye pollution in the aquatic environment can harm ecosystems and human health. Here, we developed a new green adsorbent by applying an improved drying process. Diatomite was embedded in a network structure formed between chitosan and polyvinyl alcohol without using any crosslinking agent to prepare chitosan-polyvinyl alcohol-diatomite hydrogel beads through alkali solidification. The beads were tested for removing a cationic dye (methylene blue (MB)) from water. The structure of the adsorbent beads was analysed using scanning electron microscopy, energy-dispersive spectroscopy, X-ray diffraction, X-ray photoelectron spectroscopy, and Fourier-transform infrared spectroscopy. The adsorption capacity was investigated, and the results indicated excellent MB adsorption properties. The adsorbents had a rough surface and high swelling capacity of 66.9 g/g. The maximum MB adsorption capacity was 414.70 mg/g, and the adsorption followed the Freundlich isothermal and quasi-second-order kinetic models. The adsorption was an endothermic spontaneous process governed by both intra-particle and external diffusion processes. The proposed adsorption mechanisms involved hydrogen bonding and electrostatic interactions. These adsorbent beads have considerable application potentials owing to their high adsorption capacity, green composition, and non-polluting nature.

Sustainable fabrication of chitosan membranes with optimized performance for ultrafiltration

Xin Liu,Zhujun Zou,Dong Wang,Dongyu Du,Zhuo Li,Weiyi Li

A key to the sustainable development of membrane technology is the green fabrication of biopolymer membranes, whose filtration performance is comparable to that made from petroleum-based polymers. In this study, chitosan membranes were fabricated via wet precipitation of the alkaline dope; both the composition and temperature of the coagulation bath were regulated to optimize the filtration performance for ultrafiltration. It is revealed by comparing the morphological characterization with the filtration-performance evaluation that the use of pure ethanol as the coagulant at a subzero temperature would be in favor of the formation of a laminated substructure, which can enhance the water permeance not at the cost of decreasing the rejection of micron-sized particles and macromolecules. Optical coherence tomography (OCT) was employed to in-situ characterize the film formation in an effort to understand how the variation in coagulating conditions could change the geometrical and topological characteristics of the resulting membrane. The OCT-based characterization highlights the key role of the Liesegang phenomenon in forming the hierarchical substructures, while resolving the synergistic effects emanating from the ethanol-induced film shrinkage and the structural preservation in the subzero environment.

Supramolecular Responsive Chitosan Microcarriers for Cell Detachment Triggered by Adamantane

Lixia Huang,Yifei Jiang,Xinying Chen,Wenqi Zhang,Qiuchen Luo,Siyan Chen,Shuhan Wang,Fangqing Weng,Lin Xiao

Supramolecular responsive microcarriers based on chitosan microspheres were prepared and applied for nonenzymatic cell detachment. Briefly, chitosan microspheres (CSMs) were first prepared by an emulsion crosslinking approach, the surface of which was then modified with β-cyclodextrin (β-CD) by chemical grafting. Subsequently, gelatin was attached onto the surface of the CSMs via the host–guest interaction between β-CD groups and aromatic residues in gelatin. The resultant microspheres were denoted CSM-g-CD-Gel. Due to their superior biocompatibility and gelatin niches, CSM-g-CD-Gel microspheres can be used as effective microcarriers for cell attachment and expansion. L-02, a human fetal hepatocyte line, was used to evaluate cell attachment and expansion with these microcarriers. After incubation for 48 h, the cells attached and expanded to cover the entire surface of microcarriers. Moreover, with the addition of adamantane (AD), cells can be detached from the microcarriers together with gelatin because of the competitive binding between β-CD and AD. Overall, these supramolecular responsive microcarriers could effectively support cell expansion and achieve nonenzymatic cell detachment and may be potentially reusable with a new cycle of gelatin attachment and detachment. Keywords:chitosan;microcarrier;cell culture;cell detachment;host–guest interaction
