Inhibiting ER Stress Weakens Neuronal Pyroptosis in a Mouse Acute Hemorrhagic Stroke Model

Chen Guang,Gao Cheng,Yan Ya’nan,Wang Tao,Luo Chengliang,Zhang Mingyang,Chen Xiping,Tao Luyang

Intracerebral hemorrhage (ICH) is a form of stroke, characterized by high morbidity and mortality and currently lacks specific therapy. ICH leads to endoplasmic reticulum (ER) stress, which can induce neurological impairment through crosstalk with programmed cell death (PCD). Pyroptosis, a newly discovered form of PCD, has received attention because of its close relationship with some certain diseases, such as traumatic brain injury and ischemic and hemorrhagic stroke. However, the relationship between ER stress and pyroptosis in ICH remains unclear. In this study, we investigated the role of ER stress in evoking neuronal pyroptosis and related mechanisms in a mouse ICH model. We used tauroursodeoxycholic acid (TUDCA) to inhibit ER stress and observed that TUDCA reduces neuronal pyroptosis and has a neuroprotective role. We explored the potential mechanisms underlying the regulation of neuronal pyroptosis by ER stress through testing the expression of interleukin-13 (IL-13). We found that ER stress inhibition alleviates neuronal pyroptosis through decreasing the expression of IL-13 after ICH. In summary, this study revealed that IL-13 is involved in ER stress–induced neuronal pyroptosis after ICH, pointing to IL-13 as a novel therapeutic target for ICH treatment.

Ce and Er Co-doped TiO2 for rapid bacteria- killing using visible light

Yawei Ren,Yajing Han,Zhaoyang Li,Xiangmei Liu,Shengli Zhu,Yanqin Liang,Kelvin Wai Kwok Yeung,Shuilin Wu

Bacterial infection and related diseases are threatening the health of human beings. Photocatalytic disinfection as a simple and low-cost disinfection strategy is attracting more and more attention. In this work, TiO 2 nanoparticles (NPs) were modified by co-doping of Ce and Er using the sol–gel method, which endowed TiO 2 NPs with enhanced visible light photocatalytic performance but not pure ultraviolet photocatalytic properties compared the untreated TiO 2 . Our results disclosed that as the doping content of Er increased, the photocatalytic activity of modified TiO 2 NPs initially increased and subsequently decreased. The same trend occurred for Ce doping. When the doping dose of Er and Ce is 0.5 mol% and 0.2 mol%, the 0.5Ce0.2Ti–O calcined at 800 °C presented the best antibacterial properties, with the antibacterial efficiency of 91.23% and 92.8% for Staphylococcus aureus and Escherichia coli , respectively. The existence of Er ions is thought to successfully turn the near-infrared radiation into visible region, which is easier to be absorbed by TiO 2 NPs. Meanwhile, the addition of Ce ions can effectively extend spectral response range and inhibit the recombination of electrons and holes, enhancing the photocatalytic disinfection activity of co-doped TiO 2 .

Yb/Er/Tm tri-doped Na3ZrF7 upconversion nanocrystals for high performance temperature sensing

Han Xia,Lei Lei,Jienan Xia,Youjie Hua,Degang Deng,Shiqing Xu

Non-contact optical thermometry based on fluorescence intensity ratio (FIR) technique has been widely researched over the past few decades. However, the reported systems exhibit two important shortcomings including the existence of a few interferential signals in addition to the required spectral bands for FIR and the absence of internal standard for reference signal. Herein, only two emission bands of Er 3+ : 4 F 9/2 → 4 I 15/2 (~673 nm) and Tm 3+ : 3 H 4 → 3 H 6 (~800 nm) are achieved in Yb/Er/Tm tri-doped Na 3 ZrF 7 nano-system. Moreover, the upconversion (UC) emission intensity of Er 3+ keeps unchanged with the rising of temperature, which is applied as reference signal; while that of Tm 3+ enhances evidently, which is applied as temperature signal. The calculated maximum absolute temperature sensitivity (S a ) and relative temperature sensitivity (S r ) are 0.17 K −1 at 393 K and 1.76% K −1 at 313 K, respectively.

Electrochemical sensing performance of Eu-BTC and Er-BTC frameworks toward Sunset Yellow

Yajuan Cai,Xiaoyu Li,Kangbing Wu,Xiaofeng Yang

Two lanthanide metal-organic frameworks (Ln-MOFs) using 1,3,5-benzenetricarboxylic acid (H 3 BTC) and Ln(III) nitrate hydrate (Ln = Eu, Er) as precursors have been prepared through a one-step solvothermal approach. The as-synthesized compounds were characterized by X-ray diffraction, X-ray photoelectron spectroscopy and scanning electron microscopy, confirming that the targeted Ln-MOFs were successfully synthesised, with a straw-sheaf appearance. Their sensing properties were studied using cyclic voltammetry and electrochemical impedance spectroscopy , and the results showed that the prepared Ln-BTCs possessed larger electrochemical-response areas and stronger electron-transport capability, especially the Er-BTC. The oxidation behaviour of Sunset Yellow was investigated, and it was found that the Er-BTC exhibited a superior enhancement effect. Chronocoulometry was used to prove that Er-BTC more strongly adsorbs Sunset Yellow, resulting in a larger electrochemical response signal. The novel Sunset Yellow sensor exhibited a linear response in the range of 0.2–100 nM with a low limit of detection (0.05 nM, S/N = 3), and was applied in drink samples. The results were validated by high-performance liquid chromatography , indicating this sensor is a promising tool for Sunset Yellow detection.

Mn2+-doped NaYF4:Yb/Er upconversion nanoparticle-based electrochemiluminescent aptasensor for bisphenol A

Guo Xiaofei,Wu Shijia,Duan Nuo,Wang Zhouping

A novel aptasensor labeled with Mn 2+ -doped NaYF 4 :Yb/Er upconversion nanoparticles (NaYF 4 :Yb,Er/Mn UCNPs) was employed in electrogenerated chemiluminescence (ECL) for the sensitive detection of bisphenol A (BPA). The ECL aptasensor was assembled by immobilizing the thiolated aptamers of BPA covalently on a gold nanoparticle (AuNPs)-modified electrode and pairing with complementary DNA labeled with NaYF 4 :Yb,Er/Mn UCNPs. The ECL aptasensor can not only rapidly and accurately detect BPA concentrations from 0.05 to 100 ng/mL with a detection limit of 0.037 ng/mL but also provides a new platform for ECL applications based on the use of upconversion nanoparticles as a promising alternative material. The NaYF4:Yb,Er/Mn UCNPs combining with the BPA aptamer serving as recognition elements create a ECL platform for the sensitive detection of bisphenol A. The change in ECL signals induced by aptamer-target interactions was measured and a significant decrease in intensity was found on interaction with BPA in the concentration range of 0.05 to 100 ng/mL.

Hydrothermal Synthesis of Monodispersed BaGdF5:Yb/Er Nanoparticles for CT and MR Imaging

Rongjin Sun,Ting Yin,Peng Huang,Guo Gao,Joseph George Shapter,Yulan Shen,Jingjing Zhang,Daxiang Cui

In this paper, we report the development of a facile one-pot hydrothermal method for the controlled synthesis of monodispersed BaGdF 5 :Yb/Er nanoparticles for computed tomography (CT) and magnetic resonance (MR) imaging. The as-prepared nanoparticles have uniform size with a diameter of 20–25 nm. MTT tests show that the cell viability surpasses 90% even with a concentration of nanoparticles of 500 µg/mL, suggesting that the as-prepared BaGdF 5 :Yb/Er nanoparticles possess low toxicity. For both BaGdF 5 :Yb/Er nanoparticles and iopromide, the Hounsfield unit (HU) values increase with the their concentration. The slope of BaGdF 5 :Yb/Er nanoparticles is about 26.47, which is much higher than that of iopromide (16.98), indicating a better CT imaging effect. Interestingly, we find that the synthesized BaGdF 5 :Yb/Er nanoparticles exhibit paramagnetism. Our in vitro and in vivo imaging experiments demonstrate that the synthesized monodispersed BaGdF 5 :Yb/Er nanoparticles can serve as effective contrast agents for CT and MR imaging.

SigmaR1 is an auxiliary translocon factor with lipid-binding activity that regulates protein and lipid droplet homeostasis

Hu Xuewen,Zhou Tiantian,Cui Tiansi,Du Yuanjiao,Song Chunyu,Chang Weiping,Xiong Juan,Qin Jichao,Deng Lin,Ji Wei-Ke

Sigma Non-Opioid Intracellular Receptor 1 (SigmaR1) is a member of the sigma family of receptors that interacts with a variety of psychotomimetic drugs and is involved in a wide range of cellular and physiological functions. Despite its increasing importance in human physiology and disease, the subcellular localization of SigmaR1 and its molecular function remain poorly defined. Using endogenous tagging and cell fractionation, we show that SigmaR1 is a type II integral ER membrane protein that is specifically enriched at ER sheets. A short region at the N-terminus of SigmaR1 promotes its ER-sheet localization. Importantly, our biochemical studies demonstrate that SigmaR1 directly interacts with components of the translocon complex including TRAPα and Nicalin. In addition, we found that a β-barrel at the C-terminal of SigmaR1 binds phosphatidylcholine (PC), and the binding of PC strengthens the association of SigmaR1 with the translocon complex. SigmaR1 knockout systematically impaired cellular protein and lipid homeostasis, resulting in accumulation of lipid droplets in hepatocytes. Collectively, we propose that SigmaR1 is an auxiliary translocon factor that binds lipids to regulate protein and lipid droplet homeostasis, which may underlie the broad and vital roles of SigmaR1 in physiology and disease.

Cryogenic magnetocaloric effects of NaLnF4 (Ln = Gd, Tb, Dy, Ho, Er, Tm, and Yb)

Liu Peng,Ren Weijun,Guo Qing,Zhang Zhao,Zhao Xueting,Zhang Zhidong,Dai Wei,Li Bing

Cryogenic refrigeration technology based on magnetocaloric effects plays a critical role in a variety of technological applications. In this paper, we report the cryogenic magnetocaloric effects of a series of sodium-rare earth fluoride samples (Ln = Gd, Tb, Dy, Ho, Er, Tm, Yb) synthesized by the solid-state reaction method. These compounds all crystallize in a hexagonal crystal structure. Down to 2 K, no magnetic ordering was detected, while all compounds show negative Curie–Weiss temperatures indicative of strong antiferromagnetic coupling. Magnetic fields effectively suppress the magnetic fluctuations, leading to a maximum magnetic entropy change of −56 J kg −1  K −1 in NaGdF 4 at the magnetic field change from 0 to 50 kOe. These series of compounds are potentially excellent magnetic refrigerants at low temperatures.

Dengzhan Shengmai capsule ameliorates cognitive impairment via inhibiting ER stress in APP/PS1 mice

Hui-Han Ma,Jia-Yi Zheng,Yu-Hui Qiu,Shan Su,Fang-Mei Lu,Guang-Liang Wu,Shi-Jie Zhang,Ye-Feng Cai

Ethnopharmacological relevance Alzheimer's disease (AD) is a common type of neurodegenerative disease with the β-amyloid plaques (Aβ) deposition. Previously, Dengzhan Shengmai capsule (DZSM) has been shown to reduce the pathology associated with AD, but the underlying mechanism is unclear. Aim of study This study investigated the potential mechanisms of DZSM against AD. Materials and methods The six-month-old wild-type male mice and APP/PS1 double transgenic male mice were administered 0.9 % saline or DZSM for 8 weeks by gavage. Open field test, new object recognition test, and Morris Water maze test were used to assess spatial learning and memory. Aβ plaques in brains were visualized using ThT staining. Nissl staining, TUNEL staining, and Western blot analyses were used to detect the neuronal function and apoptosis level. The superoxide dismutase (SOD), glutathione peroxidase assay kit (GSH-Px), and malondialdehyde (MDA) kits were performed to assess oxidative stress levels. Then, immunofluorescence and Western blot analysis were applied to evaluate ER stress pathway protein levels. Finally, HT22 cells were treated by Aβ 1-42 with or without DZSM medicated serum. Cell viability was assessed using the CCK-8 assay, and Western blot analysis was applied to evaluate ER stress pathway protein levels. Results Open filed test, new object recognition test and Morris Water maze test showed that DZSM restored cognitive disorders in APP/PS1 mice. Immunohistochemistry and Thioflavin T staining results indicated that DZSM reduced Aβ plaques in the brain. Deeper and denser Nissl bodies were found in APP/PS1 mice after DZSM administration. Besides, APP/PS1 mice treated with DZSM showed a lower level of TUNEL and Bax/Bcl-2 ratio. DZSM improved the acetylcholine (ACh), choline acetyltransferase (ChAT), superoxide dismutase (SOD), and glutathione peroxidase (GSH-Px) activity while reducing acetylcholinesterase (AChE) and malondialdehyde (MDA) activity. In addition, the levels of ER stress pathway containing Phospho-PKR-like ER kinase (P-PERK), phosphorylate eukaryotic initiation factor 2 (P-eIF2α), activating transcription factor 4 (ATF4), glutamine-rich protein 1 (QRICH1), phosphorylate inositol-requiring protein 1α (P-IRE1α), the spliced form of X-box binding protein 1 (XBP1s), activating transcription factor-6 (ATF6) and C/EBP homologous binding protein (CHOP) were decreased by DZSM. CCK-8 results indicated that DZSM medicated serum played cytoprotective effects on Aβ 1-42 -induced HT22 cells. Western blot results suggested DZSM possibly inhibited ER stress pathways in Aβ 1-42 -induced HT22 cells. Conclusion The potential protective mechanism of DZSM on cognitive impairment in AD might be related to ER stress pathways.

Highly Enhanced OER Performance by Er-Doped Fe-MOF Nanoarray at Large Current Densities

Yan Ma,Yujie Miao,Guomei Mu,Dunmin Lin,Chenggang Xu,Wen Zeng,Fengyu Xie

Great expectations have been held for the electrochemical splitting of water for producing hydrogen as a significant carbon-neutral technology aimed at solving the global energy crisis and greenhouse gas issues. However, the oxygen evolution reaction (OER) process must be energetically catalyzed over a long period at high output, leading to challenges for efficient and stable processing of electrodes for practical purposes. Here, we first prepared Fe-MOF nanosheet arrays on nickel foam via rare-earth erbium doping (Er0.4Fe-MOF/NF) and applied them as OER electrocatalysts. The Er0.4Fe-MOF/NF exhibited wonderful OER performance and could yield a 100 mA cm−2current density at an overpotential of 248 mV with outstanding long-term electrochemical durability for at least 100 h. At large current densities of 500 and 1000 mA cm−2, overpotentials of only 297 mV and 326 mV were achieved, respectively, revealing its potential in industrial applications. The enhancement was attributed to the synergistic effects of the Fe and Er sites, with Er playing a supporting role in the engineering of the electronic states of the Fe sites to endow them with enhanced OER activity. Such a strategy of engineering the OER activity of Fe-MOF via rare-earth ion doping paves a new avenue to design other MOF catalysts for industrial OER applications.Keywords:rare-earth doping;MOFs;large current;electronic structure

Resveratrol promotes lysosomal function via ER calcium-dependent TFEB activation to ameliorate lipid accumulation

Shao Rong,Shi Jiahui,Du Kaili,Wang Na,Cai Weijie,Liu Siyu,Ding Zongxian,Wang Yihan,Li Dan

Abnormal lipid accumulation is associated to the development of metabolic diseases such as hepatic steatosis and lipid storage diseases. Pharmacological agents that can attenuate lipid accumulation therefore have therapeutic potentials for these diseases. Resveratrol (RSV), a natural active substance found in fruits and nuts, has been reported to effectively reduce the intracellular lipid accumulation, but the underlying mechanisms of RSV remain elusive. Here, we show that RSV triggers an endoplasmic reticulum (ER)- Ca2+ signaling that activates transcriptional factor EB (TFEB), a master transcriptional regulator of autophagic and lysosomal biogenesis. Moreover, RSV activates protein phosphatase 2A (PP2A), which binds and dephosphorylates TFEB, promoting its nuclear translocation and the expression of TFEB target genes required for autophagosome and lysosomal biogenesis. Notably, genetic inhibition of TFEB significantly ameliorates RSV-mediated lipid clearance. Taken together, these data link RSV-induced ER calcium signaling, PP2A and TFEB activation to promote autophagy and lysosomal function, by which RSV may trigger a cellular self-defense mechanism that effectively mitigate lipid accumulation commonly associated with many metabolic diseases.

VMP1 restricts STING trafficking and activation through controlling CDIPT recruitment to ER membrane curvature

Zhou Chen,Bai Dongsheng,Zhao Jiawei,Yin HongJie,Du Jiaying,Gu Chunyang,Wang Yuxiang,Lin Binyan,Lu Na,Zhao Yue

The cGAS-STING signaling serves as a central hub for innate immunity by orchestrating type I interferon production and inflammatory cascades in response to cytosolic DNA. A critical rate-limiting step in this pathway involves STING exit from the endoplasmic reticulum (ER), a process initiated by COPII-mediated trafficking that requires ER membrane curvature formation. However, the molecular mechanism that drives this process is poorly understood. In this study, we identify VMP1, an ER-resident transmembrane protein, as a negative regulator that constrains STING ER exit and downstream signaling activation. VMP1 controlled ER PtdIns(3)P accumulation, which was essential for ER membrane curvature formation. Depletion of Vmp1 triggered CDIPT-dependent PtdIns(3)P synthesis through a non-canonical pathway independent of VPS34 stability and MTMR3 expression. Furthermore, VMP1-mediated macroautophagy degraded Rab10 GTPase, thereby suppressing CDIPT recruitment to the ER membrane. Myeloid-specific Vmp1 knockout mice exhibited augmented antiviral defense against DNA virus infection but displayed unaltered susceptibility to RNA virus infection. These results suggest that VMP1 controls STING trafficking by modulating the Rab10-CDIPT axis to restrict ER PtdIns(3)P accumulation and limit ER membrane curvature formation required for COPII vesicle biogenesis.
