Preserving fish meat poses a significant challenge due to its high protein and low fat content. This study introduces a novel approach that utilizes a common type of lanthanide metal-organic frameworks (Ln-MOFs), EuMOFs, in combination with 5-fluorescein isothiocyanate (FITC) and methylcellulose (MC) to develop fluorescent sensor arrays for real-time monitoring the freshness of fish meat. The EuMOF-FITC/MC fluorescence films were characterized with excellent fluorescence response, ideal morphology, good mechanical properties, and improved hydrophobicity. The efficacy of the fluorescence sensor array was evaluated by testing various concentrations of spoilage gases (such as ammonia, dimethylamine, and trimethylamine) within a 20-min timeframe using a smartphone-based camera obscura device. This sensor array enables the real-time monitoring of fish freshness, with the ability to preliminarily identify the freshness status of mackerel meat with the naked eye. Furthermore, the study employed four convolutional neural network (CNN) models to enhance the performance of freshness assessment, all of which achieved accuracy levels exceeding 93 %. Notably, the ResNext-101 model demonstrated a particularly high accuracy of 98.97 %. These results highlight the potential of the EuMOF-based fluorescence sensor array, in conjunction with the CNN model, as a reliable and accurate method for real-time monitoring the freshness of fish meat.

The toxicity of environmental pollutants is often manifested through metabolic disruptions and damage to detoxification organs. However, current understanding is insufficient to explain the physiological response mechanisms of metabolically abnormal fish exposed to secondary pollutants in complex natural environments. This study established a fish model with non-alcoholic fatty liver to evaluate the effects of Zn oxide nanoparticles (nZnO) and Zn²⁺ on physiological metabolism using untargeted lipidomics and bioimaging techniques. Nile red and hematoxylin and eosin (H&E) staining indicated that increased Zn levels reduced the number of lipid droplets (LDs) and hepatocyte vacuolization in the livers of groupers. Non-targeted lipidomics, employing an unsupervised K-means clustering algorithm, identified key lipid profiles that differentiated the effects of nZnO and Zn, including TG (16:0/16:1/18:1), PC (18:2/22:6), TG (18:2/18:2/22:6), SM (d18:1/24:1), and TG (16:1/18:1/18:2). The increased content of SM (d18:1/24:0) indicated that fish liver cells internalized nZnO via lipid raft structures on the cell membrane, a process distinct from Zn ion uptake. Moreover, nZnO/Zn treatments significantly activated lipolysis regulation in fish liver experiencing oxidative stress. This study contributed to the use of non-targeted lipidomics to identify differential biomarkers of nZnO and Zn, as well as their compensatory mechanisms in metabolically abnormal fish. These findings provide novel insights into the effects of nanometal exposure on aquatic animal health in complex environments.

A facile colorimetric aptasensor based on unmodified gold nanoparticles (AuNPs) for rapid, highly sensitive and selective detection of malachite green (MG), one of triphenylmethane dyes and antibiotics, was successfully developed. Malachite green RNA aptamer having the binding ability with high affinity and specificity would be the ideal recognition receptor for MG. RNA aptamer can adsorb on the AuNPs and protect AuNPs against salt-induced aggregation. When malachite green were added in a solution, aggregation of AuNPs was specifically induced by desorption of aptamer from the surface of gold nanoparticles as the formation of aptamer-target complex. Therefore, the salt could trigger AuNP aggregate and the solution color changed from red to blue. The solution colors and absorbance were related with the concentrations of MG, and a well linear relationship in the wide range was obtained. The detection limit of MG was down to 15.95 nM. This colorimetric method was better than other traditional methods in terms of its reduced detection time, simple signal generation, high selectivity, lower detection limit, and observation with the naked eye. The proposed one-step, label-free method was successfully used to determine MG in actual fish samples. The high recoveries of MG in fish sample detection were satisfied.

Sulfonamide antibiotics with superior performance are widely used in aquaculture. However, the residues of sulfonamide antibiotics in aquatic products may pose a threat to human health. In this work, a novel type of magnetic molecularly imprinted polymer (mMIP) was synthesized and coupled with ultra-high performance liquid chromatography–tandem mass spectrometry (UHPLC-MS/MS) for the selective separation and determination of 4 sulfonamide antibiotics residues in tilapia. The structure and adsorption properties of the polymers were characterized and evaluated. The key mMIP variables were investigated to determine the optimal magnetic solid phase extraction conditions. Results showed that the mMIPs had saturated adsorption capacities of 137.4, 78.7, 65.2, and 58.4 mg/g for trimethoprim, sulfadiazine, sulfamethazine, and sulfamethoxazole, respectively. Under optimized conditions, the linear range of the method for the detection of the 4 sulfonamides was 0.1–100 ng mL −1 ( R > 0.991), the detection limit was 0.015 μg kg −1 (S/N = 3), the recovery rate was 78.6–101%, and the relative standard deviation (RSD%) was 4.2–7.8%. This new rapid and ultra-sensitive pretreatment method was successfully applied to the monitoring of trace sulfonamide antibiotics residues in fish samples.

Background Due to the serious issue of ofloxacin (OFL) abuse, there is an increasingly urgent need for accurate and rapid detection of OFL. Immunoassay has become the “golden method” for detecting OFL in complex matrix beneficial to its applicability for a large-scale screening, rapidity, and simplicity. However, traditional antibodies used in immunoassay present challenges such as time-consuming preparation, unstable sensitivity and specificity, and difficulty in directional evolution. In this paper, we successfully developed an OFL detection method based on a shark-derived single-domain antibody (ssdAb) to address these issues. Results Using phage display technology and a heterologous expression system, OFL-specific clones 1O11, 1O13, 1O17, 1O19, 1O21, and 2O26 were successfully isolated and expressed in soluble form. Among all OFL-specific ssdAbs, the 1O17 ssdAb exhibited the highest binding affinity to OFL in a concentration-dependence manner. The limit of detection (IC 10 ) of 1O17 ssdAb was calculated as 0.34 ng/mL with a detection range of 3.40–1315.00 ng/mL, and its cross reactivity with other analogs was calculated to be less than 5.98 %, indicating high specificity and sensitivity. Molecular docking results revealed that 100Trp and 101Arg located in the CDR3 region of 1O17 ssdAb were crucial for OFL binding. In fish matrix performance tests, the 1O17 ssdAb did not demonstrate severe matrix interference in OFL-negative fish matrix, achieving satisfactory recovery rates ranging from 83.04 % to 108.82 % with high reproducibility. Significance This research provides a new and efficient OFL detection recognition element with significant potential in immunoassay applications, broadening the application scenarios of ssdAbs. It offers valuable insights into the structure-activity relationship between ssdAbs and small molecules, laying a theoretical foundation for the further directional modification and maturation of ssdAbs in subsequent applications.

Nuclear factor erythroid 2-related factor 2 (Nrf2) is an essential component in regulating oxidative stress. Sulforaphane (SFN) is a natural antioxidant and gene Nrf2 agonist that can increase the antioxidant capacity of the organism and reduce oxidative stress. However, research on the repair of oxidative stress damage by SFN in aquatic animals remains extremely scarce. In order to further explore the function and role of SFN in the repair of oxidative stress injury in aquatic animals, this study took Litopenaeus vannamei as the research object. We established an oxidative stress model of L. vannamei through 6% oxidized fish oil (OFO) feeding. Methods, such as RNA interference (RNAi), qPCR, histopathological analysis, and TUNEL assay, were used to detect the changes in the oxidative stress status of L. vannamei . The results showed that the expression of Nrf2 in the hepatopancreas of L. vannamei in the double-stranded RNA (dsRNA)- Nrf2 + SFN group was significantly higher than that in the dsRNA- Nrf2 group and control group at 24 h ( p < 0.05). The transcription levels of antioxidant and autophagy genes in the SFN group were significantly higher than those in the control group ( p < 0.05), and the expression of related genes in the dsRNA- Nrf2 + SFN group was also higher than that in the dsRNA- Nrf2 group. Histopathology showed that Nrf2 knockdown would aggravate hepatopancreatic apoptosis and vacuolation, while SFN treatment after Nrf2 knockdown could alleviate hepatopancreatic injury and apoptosis caused by OFO. The results indicated that SFN could repair the oxidative stress injury of L. vannamei induced by OFO by activating Nrf2 . This study investigated the role of SFN in alleviating and repairing the oxidative stress damage in L. vannamei caused by OFO, aiming to provide a theoretical basis for the research on the antioxidant effect of SFN and the regulation of the antioxidant capacity of shrimp.

The development of ecofriendly food packaging materials is crucial for addressing environmental concerns and enhancing food preservation. This study aimed to develop an antibacterial water-resistant film using steam-exploded bamboo leaf carboxymethyl cellulose (CMC) combined with cinnamaldehyde (CIN) and cellulose nanocrystals (CNCs). It was demonstrated that ultrasonic treatment (300 W; 0, 10, 20, 30, 40, and 50 min) enhanced the film matrix uniformity. Incorporating 3 % CNCs improved the tensile strength (26.95 % increase) and hydrophobicity. The composite film significantly extended the fish shelf life by 4–6 days ( p < 0.05), as validated by biochemical and sensory analyses. This study validates bamboo leaf-derived CMC as a sustainable base for multifunctional packaging films, providing improved mechanical strength, moisture resistance, and antimicrobial performance. The CMC–CIN–CNCs film demonstrates significant potential for ecofriendly food preservation, addressing both environmental challenges and food safety needs.

Globally, billions of tons of food are lost or wasted annually due to microbial contamination. Hydrogels are a new type of fresh-keeping material because of their porous three-dimensional network structure, offering enhanced loading capacity. However, traditional hydrogel packaging materials often suffer from limitations such as poor frost resistance, lack of catalytic antibacterial activity, and low factory application rate. In this study, firstly, carbon dots (CPCDs) catalyzing oxygen under visible light irradiation with broad-spectrum antibacterial activity (The minimum photocatalytic antibacterial concentration of H. alvei and P. fluorescens was 62.5 μg/mL) were synthesized. A hydrogel was rapidly formed using polyvinyl alcohol (PVA) and gelatin within 14 min of one-time freezing at −20 °C through hydrogen bonds, and excellent photocatalytic antibacterial activity and timely preservation were achieved by loading CPCDs. The low swelling (3.0) of the hydrogel and a release rate of <20% reduced the loss of CPCDs, thereby achieving a sustained antibacterial effect. Additionally, In the DSC experiment, no significant peak was observed between −80 °C and − 20 °C. The hydrogel was not crystallized and maintained its flexibility, which proved that the hydrogel had good frost resistance. The hydrogel could be completely peeled off from the surface of the Oncorhynchus mykiss fillet with a force of <0.5 N without any residue. The hydrogel showed no significant cytotoxicity and could extend the shelf life of O. mykiss fillet by >3 days. These findings underscored the potential of the developed hydrogels as efficient materials for the rapid preservation of frozen seafood.

In the present study, we constructed an electrochemical sensor using three materials, namely, cerium dioxide (CeO 2 ) composite platinum (Pt) and polymer 3-amino-5-mercapto-1,2,4-triazole (PTA), prepared by hydrothermal method, to detect xanthines and uric acid. Scanning electron microscopy (SEM), X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), and Fourier infrared spectroscopy (FT-IR) were used to characterize the morphology, elemental composition, and structure of materials. The results demonstrated that the PTA/CeO 2 @Pt composites were prepared. The electrochemical behavior of xanthines and uric acid on PTA/CeO 2 @Pt composite electrodes was assessed by cyclic voltammetry (CV) and differential pulsed voltammetry (DPV). The results demonstrated that the dropping amount was 20 µL under a phosphoric acid buffer solution (pH 6). The peak current of xanthine (XA) demonstrated two good linear relationships with concentration, the linear regression equation was I XA = 0.3265C XA + 8.1967 R 2 = 0.994 in the concentration range of 10–138 µmol/L, whereas it was I XA = 3.9884C XA + 51.661 R 2 = 0.992 in the concentration range of 139–330 µmol/L. The detection limit was 10.36 × 10 –7 M. The freshness of fish meat was evaluated, and the correlation analysis between evaluation results and results of xanthine detection in fish meat by PTA/CeO 2 @Pt electrochemical sensor demonstrated that r = 0.945 and P < 0.01 had a good correlation. The sensor could be used to determine the xanthine content in fish to reflect the freshness of fish, thus displaying good application prospects.

Fish gelatin (FG) is a biopolymer with favorable biocompatibility, mechanical strength, and water retention, making it ideal for films, hydrogels, and bio-based applications. This study aimed to comparatively investigate the FG based intelligent films and hydrogels for monitoring shrimp freshness. Intelligent films and hydrogels were prepared using FG as a macromolecular matrix, incorporating ferulic acid (FA)-crosslinked anthocyanin (ACN) as an indicator, and enhanced with polysaccharides (κ-carrageenan (κC), xanthan glucomannan (XG), and konjac glucomannan (KGM)). UV–Vis spectroscopy and molecular docking analyses confirmed that FA-ACN copigmentation improved light stability and enhanced molecular interactions. Prepared intelligent films and hydrogels were characterized by evaluating mechanical strength, water retention, color stability, and sensitivity to freshness indicators. Results showed the addition of polysaccharides significantly enhanced mechanical strength and water retention of films, while the combination of FA and κC improved mechanical properties, water resistance, and color stability, and FA-FG-κC hydrogel had the strongest gel strength, reaching 1211.29 g. The intelligent hydrogels demonstrated superior sensitivity, with color changes closely correlated (R 2 > 0.9) to both TVB-N and TVC levels, thus accurately reflecting shrimp freshness during storage. Compared with films, hydrogels offered improved responsiveness and reliability. This work presents a sustainable, real-time monitoring strategy for aquatic products, contributing to food safety and environmental sustainability. These findings support the potential of FG-based intelligent hydrogels in quality monitoring of seafood during cold chain storage.

A series of novel dummy molecularly imprinted polymers (DMIPs) were prepared as highly class-selective sorbents for fluoroquinolones. A non-poisonous dummy template, daidzein, was used for the first time to create specific molecular recognition sites for fluoroquinolones in the synthesized polymers. The influence of porogen polarity on dummy molecular imprinting effect was studied. The DMIP prepared using dimethylsulfoxide−acetonitrile (1:1.8, v/v) as porogen achieved the highest imprinting factors (IF) for fluoroquinolones over a range of IF 13.4–84.0. This DMIP was then used for selective extraction of eight fluoroquinolones (fleroxacin, ofloxacin, norfloxacin, pefloxacin, ciprofloxacin, lomefloxacin, enrofloxacin and gatifloxacin) from fish samples based on dummy molecularly imprinted matrix solid-phase dispersion (DMI-MSPD). The extracted fluoroquinolones were subsequently analyzed by high-performance liquid chromatography (HPLC) equipped with a fluorescence detector (FLD). The developed method had acceptable recoveries (64.4−102.7%) and precision (RSDs: 1.7−8.5%, n = 5) for determination of fluoroquinolones in fish samples fortified at levels of 10 and 100 ng g −1 . The limits of detection (LODs) for identification of eight fluoroquinolones ranged between 0.06 and 0.22 ng g −1 . The results demonstrated great potential of the optimized method for sample preparation in routine analysis of trace fluoroquinolones in fish samples.

Residues of eight perfluorinated compounds (PFCs) were investigated in 15 fish samples from the Danjiangkou reservoir and Hanjiang river (Xiangyang and Zhongxiang sections). The total concentrations of PFCs in fish muscles ranged from 2.01 to 43.8 ng g − 1 dry weight. The mean concentration of total PFCs from related muscles showed the following trend in various regions: Zhongxiang section < Xiangyang section < Danjiangkou reservoir. Perfluorooctane sulphonate (PFOS) was the dominant PFC in the fish liver samples from the Danjiangkou reservoir. The calculated hazard ratio (HR) of PFCs, for all fish muscle samples, was less than 1.0, and could be classified at safe levels for the general population. However, yellow croaker fish from the Danjiangkou reservoir and Hanjiang river–Xiangyang section had HRs of 0.2, indicating that frequent consumption of this contaminated fish may pose an unacceptable risk to human health.
