Rapid and sensitive detection of picric acid (PA), a highly toxic and water-mobile nitroaromatic pollutant, is essential for environmental safety and public health protection. Herein, we fabricate a dual-emission ratiometric fluorescent probe by immobilizing high-load red-emitting carbon dots (R-CDs) onto NH₂-functionalized MIL-53(Al), forming an R-CDs/NH 2 -MIL-53(Al) composite for selective aqueous-phase PA sensing. R-CDs were hydrothermally synthesized with citric acid and neutral red as co-precursors. The loading efficiency was maximized by systematically optimizing precursor molar ratios and dilution conditions. The composite exhibits distinct selective fluorescence quenching in the presence of PA. Mechanistic characterizations, including fluorescence lifetime decay measurements and spectral overlap analysis, verify that the fluorescence quenching originates from two synergistic quenching pathways, namely competitive light absorption and fluorescence resonance energy transfer (FRET), rather than nonspecific physical interactions. This ratiometric sensing platform achieves a wide linear detection range of 0.1–70 μM and an ultralow limit of detection (LOD) of 32.9 nM. Spike-and-recovery assays using real environmental water samples (tap water, commercial bottled mineral water, and Jialing River surface water) produced recoveries of 99.7%–105.1%, with relative standard deviations (RSD) below 5.86% ( n = 3). Furthermore, a smartphone-enabled visual quantitative method based on RGB color channel analysis was constructed, enabling portable on-site detection without large-scale laboratory instrumentation.

A new type of fluorescence “off-on” probe was designed for L-Cysteine (L-Cys) based on the fluorescence resonance energy transfer (FRET) between negatively charged amino-capped porous silicon nanoparticles (SiNPs) and positively charged citrate-stabilized Au nanoparticles (AuNPs). In this proposed FRET immunosensor, novel water-soluble amino-conjugated porous SiNPs in ethanol with excellent photoluminescence properties act as the energy donor. Excellent quenching efficiency between SiNPs-ethanol and citrate-stabilized AuNPs by electrostatic interaction via FRET provides an ideal “off-state” (turn-off). The addition of L-Cys leads to releasing the adsorbed AuNPs from the surface of SiNPs and hence the fluorescence emission of SiNPs-ethanol is restored (turn-on), which means the coordination ability of the thiols with AuNPs is stronger than that of the electrostatic interaction. The fluorescence intensity of SiNPs-AuNPs in ethanol is sensitive to L-Cys, and such a restored fluorescence is proportional to the concentration of L-Cys. The method will broadly benefit the development of a new thiol biosensor based on nanostructured porous materials, and the proposed procedure is also expected to develop a variety of functional nanoparticles to form other novel kinds of nanosensors.Keywords:“off-on” fluorescence biosensor;porous SiNPs;ethanol solution;AuNPs;L-Cysteine

Förster resonance energy transfer (FRET) is a powerful tool to study single molecular conformation and/or distance changes between two fluorophores, which is mainly depended on the combinations and spatial distance between the donor and acceptor. The present study has built a FRET assembly by using gold nanoclusters capped by glutathione (AuNCs@GSH) and carbon dots (CDs) prepared by polyethyleneimine (PEI) through supramolecular interactions. The assembly is characterized in detail by employing Zeta potential, UV–vis absorption and fluorescence spectra, the luminescent decay curves and TEM images, which indicate the AuNCs are adhered to the surface of CDs. In addition, the transfer efficiency between them has been calculated to be 65.7%, which is sufficient enough for practical applications, and particularly, the quantum yield (QY) of AuNCs@GSH is increased from 3.07% to 5.35% via the assembly. Moreover, the FRET assembly is used as a ratiometric probe for cysteine detection in showing a limit of detection (LOD) at 100 nM, a very high selectivity to cysteine among amino acids as well as a strong anti-interference on them, especially, it was feasible for cysteine detection in fetal calf serum, which showed very high potential in the diagnose of cysteine-related disease.

The authors describe an aptasensor based on time-resolved fluorescence resonance energy transfer (TR-FRET) for the identification of Salmonella typhimurium ( S. typhimurium ). Aptamer-functionalized nanoparticles (NPs) is used as the energy donor, and a complementary oligonucleotide (cDNA) labeled with carboxyfluorescein (FAM) acts as the acceptor. The detection scheme is based on the hybridization between aptamer and cDNA, upon which photonic energy is transferred from NPs to FAM unless aptamer interacts with S. typhimurium . Due to the highly specific recognition ability of aptamer and the strong fluorescence intensity of NPs, the method is highly sensitive and selective for S. typhimurium . Under the optimal conditions, at excitation wavelength of 273 nm, a delay time of 100 μs and a gating time of 1 ms, the integrated time-resolved fluorescence intensity ratio (FAM 520 /Tb 489 ) is linear in the 100 to 10 6 cfu·mL −1 range, and the limit of detection is as low as 25 cfu·mL −1 . The assay was applied to the analysis of eggs and chicken meat for S. typhimurium , and the results were consistent with those of a plate-counting method.

This paper reported a facile method for fabricating CdTe quantum dots decorated fluorescent mesoporous silica nanoparticles containing poly(p-phenylenevinylene) by electrostatic interaction in aqueous solution. The resulted fluorescent hybrid nanomaterials showed the dual-emission centered at 500 nm and 717 nm, which can be used as a fluorescent probe to detect Cu 2+ in water through fluorescence resonance energy transfer. Cu 2+ can be captured by the amino groups of the polyethyleneimine to form an absorbent complex, resulting in a strong quenching of red-emission from CdTe quantum dots on the surface of nanoparticles via energy transfer. However, the green-emission of poly(p-phenylenevinylene) remained. Therefore the addition of Cu 2+ induced the fluorescent evolution of the nanoprobe from red to green. The nanoprobe provided an efficient platform for the sensing of Cu 2+ with a detection limit of 31.2 nM. Compared with pure quantum dots, the nanoprobe can distinguish Cu 2+ from Hg 2+ .

In vitro diagnostic is a crucial component of healthcare systems for early diagnosis of diseases and follow-up therapy, which generally makes clinical diagnosis faster, easier, and painless for patients. Herein, we report a dual-signaling nanoplatform for ratiometric absorption and time-resolved fluorescence resonance energy transfer based on L-cysteine-mediated aggregated gold nanoparticles and long afterglow nature of persistent luminescence nanoparticles. With this nanoplatform, we have achieved high-throughput sequential detection of L-cysteine and insulin in human serum without matrix interference. The proposed nanoplatform shows excellent linearity and precision for the determination of L-cysteine in the range of 10 nM to 5.5 μM with the limit of detection (LOD) of 2.2 nM and for the detection of insulin in the range of 12 pM to 3.44 nM with LOD of 2.06 pM. The developed dual-signaling nanoplatform has been successfully applied for the sequential determination of L-cysteine and insulin in human serum.

A dual-channel ratiometric nanoprobe is described for detection and imaging of microRNA. It was prepared from MoS 2 quantum dots (QDs; with blue emission and excitation/emission peaks at 310/398 nm) which acts as both the gene carrier and as a donor in fluorescence resonance energy transfer (FRET). Molecular beacons containing loops for molecular recognition of microRNA and labeled with carboxyfluorescein (FAM) were covalently attached to the MoS 2 QDs and serve as the FRET acceptor. In the absence of microRNA, the nanoprobe exhibits low FRET efficiency due to the close distance between the FAM tag and the QDs. Hybridization with microRNA enlarges the distance between QD and beacon. This results in an enhancement of the FRET efficiency of the nanoprobe. The ratio of green and blue fluorescence (I 520 /I 398 ) increases linearly in the 5 to 150 nM microRNA concentration range in both aqueous solution and diluted artificial cerebrospinal fluid. The limit of detection (LOD) is as low as 0.38 nM and 0.52 nM, respectively. Other features of this nanoprobe include (a) excellent resistance to nuclease-induced false positive signals and (b) the option to use it for distinguishing different cell lines by in-situ imaging of intracellular microRNAs.
Schematic of a dual-channel photoluminescence nanoprobe for the determination of microRNA-21 (miR-21) by monitoring the microRNA-triggered enhancement of the fluorescence resonance energy transfer (FRET) efficiency between MoS 2 QDs and carboxyfluorescein-labeled molecular beacons.

Effective use of near-infrared (NIR) light for photocatalysis in the solar full-spectrum remains a major challenge. Herein, LuF 3 : Yb 3+ /Tm 3+ @Lu 6 O 5 F 8 : Yb 3+ /Tm 3+ (LYT@LOYT), which could convert NIR light into UV–visible light, was combined with BiOI to form a novel core-shell structured LuF 3 : Yb 3+ /Tm 3+ @Lu 6 O 5 F 8 : Yb 3+ /Tm 3+ @BiOI (LYT@LOYT@BiOI) upconversion photocatalyst. LOYT shell layer was grown in situ on LYT by high-temperature calcination in air, resulting in a 2.36-fold enhancement of blue light emission from the upconversion luminescent core. The tight core-shell wrapping formed by the solvothermal method achieved an efficient Förster resonance energy transfer (FRET) between LYT@LOYT and BiOI. Meanwhile, the formation of heterogeneous structure also accelerated the separation and transfer of photogenerated electron-hole pairs. As a result, under NIR and Vis-NIR irradiation, the degradation rates of LYT@LOYT(20)@BiOI for bisphenol A (BPA) were 1.24 and 1.11 times of LYT(20)@BiOI, and 1.57 and 1.30 times of BiOI, respectively. The enhanced photocatalytic activity was attributed to the expanded photoresponse range, enhanced upconversion emission, efficient energy transfer process, and excellent electron-hole pairs separation transfer efficiency. h + and ⋅O 2 - were shown to be active species in the degradation process. This work provides a useful reference for the rational design of solar full-spectrum responsive photocatalysts which can efficiently utilize NIR light.

Various attributes of micelles , such as PEG density and particle size, are considered to be related to blood clearance. The structural stability of micelles is another key attribute that will affect the in vivo fate. This study employed fluorescence resonance energy transfer (FRET) analysis to guide the preparation of polymeric micelles with different structural stability. Micelles prepared using copolymers with longer hydrophobic blocks showed higher structural stability; emulsification was a better method than nanoprecipitation to prepare stable micelles. The fast chain exchange kinetics and the high-water content of micellar cores explained the low structural stability of those micelles. Moreover, this study highlighted the importance of structural stability that affected blood clearance in concert with PEG length and particle size. One-third of the small and stable micelles were detected in the blood 24 h after injection. While unstable micelles would be cleared from the circulation within 4 h. Notably, there would be a threshold of structural stability. Micelles with structural stability below this threshold were quickly cleared even if they possessed a longer PEG length and a smaller size. In contrast, higher structural stability allowed polymeric micelles to maintain higher integrity in vivo and enhance tumor accumulation and anti-tumor efficacy. In conclusion, this study systematically analyzed the importance of the structural stability of micelles on the in vivo fate.

In this work, a versatile cellulose nanocrystal‑carbon dots (CNC-CDs) architecture is prepared and applied in environmental remediation and biological sensing. The citric acid (CA) is employed to extract CNCs and in-situ synthesize blue-emissive CDs. The carboxyl-rich surface of CNC-CDs enables the chelation of Hg 2+ and fluorescence quenching of CDs, realizing the detection and adsorption of Hg 2+ with a film comprised of the architecture. The limit of detection (LOD) is measured as 12.8 nM with satisfying retention efficiency. The versatility of CNC-CDs is demonstrated by the fabrication of a fluorescence resonance energy transfer (FRET) sensor using m-tetrakis(4-sulfonatophenyl)porphine (TSPP) as FRET acceptor and Fe 3+ receptor. Fluorescence of the sensor is responsive to Fe 3+ in a ratiometric manner, which significantly contributes to the visualized and sensitive Fe 3+ detection (6.9 nM). In combination with excellent biocompatibility, the FRET sensor is capable of imaging intracellular Fe 3+ .

A dual-emission ratiometric fluorometric aptasensor is presented for highly specific detection of adenosine. An adenosine binding aptamer (ABA) was split into two halves (termed as ABA1 and ABA2). ABA1 was covalently bound to blue-emitting carbon dots (with excitation/emission maxima at 365/440 nm) as responsive fluorophore (referred to as ABA1-CDs). ABA2 was linked to red-emitting silica-coated CdTe quantum dots (with excitation/emission maxima at 365/613 nm) acting as internal reference and referred to as ABA2-QDs@SiO 2 . Upon addition of graphene oxide, the fluorescence of ABA1-CDs is quenched. After subsequent addition of ABA2-QDs@SiO 2 and different amounts of adenosine, the blue fluorescence is recovered and causes a color change from red to royal blue. The method represents a ratiometric turn-on assay for visual, colorimetric and fluorometric determination of adenosine. The limit of detection is as low as 2.4 nM in case of ratiometric fluorometry. The method was successfully applied to the determination of adenosine in (spiked) human urine. Recoveries range from 98.8% to 102%.
Adenosine binding aptamer1-carbon dots (ABA1-CDs) can absorb on graphene oxide (GO) via π stacking. This causes fluorescence to be quenched by fluorescence resonance energy transfer (FRET). After addition of ABA2-silica-coated quantum dots (ABA2-QDs@SiO 2 ) and adenosine, binding of adenosine to two pieces of aptamers forms a complex (ABA1-CD/adenosine/ABA2-QD@SiO 2 ) which dissociates from GO. As a result, fluorescence is recovered.

A label-free fluorescent aptasensor for specific and ultrasensitive monitoring ochratoxin A(OTA) was developed using the specific aptamer of OTA(OSA) as recognition element, an aggregation-induced emission(AIE) molecule(a 9,10-distyrylanthracene with two ammonium groups, DSAI) as a fluorescent probe, and graphene oxide(GO) as a quencher. In the absence of OTA, the AIE probe DSAI and OSA complex(DSAI/OSA) is adsorbed on the GO surface, and the fluorescence of DSAI will be quenched efficiently via the fluorescence resonance energy transfer(FRET) from DSAI to GO. Upon the OTA addition, a more stable complex(OSA-OTA) is formed and released from GO. Meanwhile, DSAI and OSA-OTA can form a new complex(DSAI/OSA-OTA), then the fluorescent signal of DSAI recovers gradually. Therefore, by introducing GO and DSAI, the fluorescence signal of DSAI can be easily turned from “off” to “on” after the addition of OTA, and the ultrasensitive detection of OTA by monitoring the change of the fluorescence signal of DSAI can be readily realized. The detection limit of the assay can reach 0.324 nmol/L with a linear detection range of 10—200 nmol/L. And the aptasensor exhibits high selectivity for OTA against other analogues. Moreover, it has been successfully applied for the detection of OTA in red wine samples.
