Background: Oxidative stress and apoptosis of neurons significantly contribute to the pathophysiological cascade of spinal cord injury (SCI). However, the role of hypoxic-preconditioned mesenchymal stem cell-derived small extracellular vesicles (H-sEVs) in promoting SCI repair remains unclear. Hence, the present study aims to investigate the regulatory effects of H-sEVs on neuronal oxidative stress and apoptotic responses following SCI.Methods: The administration of H-sEVs of SCI rats was assessed using behavioral evaluations such as Basso-Beattie-Bresnahan (BBB) scores, neuroelectrophysiological monitoring, and Catwalk gait analysis. Indices of oxidative stress (including superoxide dismutase [SOD], total antioxidant capacity [T-AOC], and malondialdehyde [MDA]) were measured. Neuronal survival was evaluated through Nissl staining, while the expression level of sirtuin 1 (SIRT1) was examined using immunohistochemical staining. Additionally, histological evaluation of lesion size was performed using hematoxylin-eosin (HE) staining. Tunel cell apoptosis staining and analysis of apoptosis-associated proteins (B-cell lymphoma-2 [Bcl2] and BCL2-Associated X [Bax]) were conducted through immunofluorescence staining and western blot, respectively. Furthermore, the model of oxidative stress was established using PC12 cells, and apoptosis levels were assessed via flow cytometry and western blot analysis. Importantly, to ascertain the critical role of SIRT1, we performed SIRT1 knockout experiments in PC12 cells using lentivirus transfection, followed by western blot. Results: Using those behavioral evaluations, we observed significant functional improvement after H-sEVs treatment. Nissl staining revealed that H-sEVs treatment promoted neuronal survival. Moreover, we found that H-sEVs effectively reduced oxidative stress levels after SCI. HE staining demonstrated that H-sEVs could reduce lesion area. Immunohistochemical analysis revealed that H-sEVs enhanced SIRT1 expression. Furthermore, Tunel cell apoptosis staining and western blot analysis of apoptosis-related proteins confirmed the anti-apoptotic effects of H-sEVs. The PC12 cells were used to further substantiate the neuroprotective properties of H-sEVs by significantly inhibiting neuronal death and attenuating oxidative stress. Remarkably, SIRT1 knockout in PC12 cells reversed the antioxidant stress effects induced by H-sEVs treatment. Additionally, we elucidated the involvement of the downstream Nrf2/HO-1 signaling pathway. Conclusion: Our study provides valuable insights into the effects of H-sEVs on neuronal oxidative stress and apoptosis after SCI. These findings underscore the potential clinical significance of H-sEVs-based therapies for SCI.

Background: Atrial fibrillation (AF) is often associated with atrial fibrosis and oxidative stress. Neferine, a bisbenzylisoquinoline alkaloid, has been reported to exert an antiarrhythmic effect. However, its impact on Angiotensin II (Ang II) infusion-induced AF and the underlying mechanism remains unclear. This study aimed to investigate whether neferine alleviates Ang II-induced AF and explore the underlying mechanisms. Methods: Mice subjected to Ang II infusion to induce AF were concurrently treated with neferine or saline. AF incidence, myocardial cell size, fibrosis, and oxidative stress were then examined. Results: Neferine treatment inhibited Ang II-induced AF, atrial size augmentation, and atrial fibrosis. Additionally, we observed that Ang II increased reactive oxygen species (ROS) generation, induced mitochondrial membrane potential depolarization, and reduced glutathione (GSH) and superoxide dismutase (SOD) levels, which were reversed to some extent by neferine. Mechanistically, neferine activated the Nrf2/HO-1 signaling pathway and inhibited TGF-β/p-Smad2/3 in Ang II-infused atria. Zinc Protoporphyrin (ZnPP), an HO-1 inhibitor, reduced the anti-oxidative effect of neferine to some extent and subsequently abolished the beneficial effect of neferine on Ang II-induced AF. Conclusions: These findings provide hitherto undocumented evidence that the protective role of neferine in Ang II-induced AF is dependent on HO-1.

Asthenozoospermia, a major cause of male infertility characterized by impaired sperm motility, is critically driven by oxidative stress and dysregulated iron metabolism, which directly induce ferroptosis in sperm cells. To address this, a novel nanotheranostic platform utilizing ferritin nanocages to encapsulate vitexin, a natural flavonoid with potent antioxidant properties and unique aggregation-induced emission (AIE) characteristics, is introduced. This ferritin nanocage-loaded vitexin (HFn@VI) system combines the iron-chelating capability of ferritin with vitexin's ability to mitigate oxidative stress, enabling precise dual-targeting of the ferroptotic pathways in spermatogenic cells. In the asthenozoospermia model mice, HFn@VI significantly improves sperm count and kinematic parameters (motility, velocity) without observable systemic toxicity, demonstrating superior therapeutic efficacy. Mechanistic studies reveal that the ferritin component elevates ferritin heavy chain 1(FTH1) levels, sequestering free iron ions and reducing cellular iron overload. Concurrently, vitexin activates the nuclear factor erythroid 2-related factor 2 (Nrf2)/heme oxygenase-1 (HO-1) pathway, enhancing glutathione peroxidase 4 (GPX4) expression to inhibit lipid peroxidation and ferroptosis. This study pioneers the application of vitexin, a natural product exhibiting AIE characteristics, as a therapeutic agent in reproductive medicine and underscores the potential of dual-target nanomedicines for precise ferroptosis intervention, establishing a new paradigm for treating male infertility through advances in material science.

Tea polyphenols (TPs) are the major bioactive extract from green tea that have been extensively reported to prevent and treat oxidative stress damage. In previous studies, TPs have been demonstrated to protect cells against oxidative injury induced by hydrogen peroxide (H2O2). However, the underlying mechanism remains unclear. The aim of the current study was to investigate whether the protective and regulatory effects of TPs on oxidative stress damage were dependent on the mammalian STE20‑like protein kinase (Mst)/nuclear factor (erythroid‑derived 2)‑like 2 (Nrf2) axis and the Kelch‑like ECH‑associated protein 1 (Keap1)/Nrf2/heme oxygenase 1 (HO‑1) pathway in RAW264.7 cells, a murine macrophage cell line. Maintaining a certain range of intracellular reactive oxygen species (ROS) levels is critical to basic cellular activities, while excessive ROS generation can override the antioxidant capacity of the cell and result in oxidative stress damage. The inhibition of ROS generation offers an effective target for preventing oxidative damage. The results of the present study revealed that pretreatment with TPs inhibited the production of intracellular ROS and protected RAW264.7 cells from H2O2‑induced oxidative damage. TPs was also demonstrated to attenuate the production of nitric oxide and malondialdehyde and increase the levels of antioxidant enzymes (superoxide dismutase, catalase and glutathione peroxidase). In addition, following TPs treatment, alterations in Mst1/2 at the mRNA and protein level inhibited the production of ROS and promoted the self‑regulation of antioxidation. TPs‑induced Keap1 gene downregulation also increased the expression of Nrf2 and HO‑1. Collectively, the results of the present study demonstrated that TPs provided protection against H2O2‑induced oxidative injury in RAW264.7 cells.

Under 980 nm excitation, Ho 3+ in Ho/Yb co-doped materials can emit light spanning the infrared to green spectral regions through down-conversion and multiphoton up-conversion processes. The multi-wavelength infrared emissions from Yb/Ho co-doped yttria-stabilized zirconia (YSZ) enable fast wavelength hopping, making it highly valuable for secure laser communication systems. However, achieving efficient and intense multi-wavelength emission remains challenging. In this work, high-quality YSZ, YSZ:Yb 3 Ho 0.1 , Lutetia-stabilized zirconia (LuSZ) and LuSZ:Yb 3 Ho 0.1 single crystals were grown by the optical floating zone method. A first-time comparative study reveals distinct photoluminescence characteristics between YSZ:Yb 3 Ho 0.1 and LuSZ:Yb 3 Ho 0.1 crystals under 980 nm excitation. Both systems show emission peaks at 2450, 2100, 1845, 1230, 1050, 650, and 538 nm from the down-conversion and up-conversion photoluminescence (DCPL and UCPL). Notably, LuSZ:Yb 3 Ho 0.1 exhibits stronger UCPL peaks at 650 and 538 nm, attributed to its lower phonon energy. In contrast, YSZ: Yb 3 Ho 0.1 shows higher DCPL intensities infrared peaks at 2450, 2100, 1845, 1230, and 1050 nm, benefiting from its higher transmittance and phonon energy. The unique strong multi-wavelength infrared emission of YSZ:Yb 3 Ho 0.1 crystal enables rapid wavelength switching, highlighting its potential for secure laser communication.

Understanding electrical transport mechanisms and key thermal stability factors in high-temperature thermosensitive ceramics is essential for developing highly stable temperature sensors under extreme thermal conditions. This study develops the high-entropy pyrochlore stannates (La 0.2 Nd 0.2 Sm 0.2 Gd 0.2 M 0.2 ) 2 Sn 2 O 7 (M = Eu, Dy, Ho, or Yb) via solid-state synthesis, demonstrating exceptional negative temperature coefficient (NTC) behavior across 300 – 1400 °C. Hall measurements and X-ray photoelectron spectroscopy (XPS) analyses reveal a dual redox mechanism for these ceramics: partial reduction of Sn 4+ to Sn 2+ at B-sites and rare-earth ions (Sm/Eu/Yb) to RE 2+ at A-sites, generating hole-dominated p-type conductivity. The materials exhibit outstanding thermal stability, with resistance drift below 1.96% after 500 hours at 1400 °C, attributed to the entropy-stabilized structure that effectively suppresses lattice disorder transformation. Aging studies further demonstrate that high oxygen vacancy concentrations lead to resistance drifts, as oxygen filling at elevated temperatures oxidizes RE 2+ to RE 3+ , reducing charge carriers and ultimately increasing resistivity. This study elucidates for the first time the hole-dominated conduction mechanism arising from dual-site cation valence changes (Sn/RE) and the regulation effect of oxygen vacancies on high-temperature stability, which provides an important theoretical basis and material design strategy for the development of the next-generation high-temperature NTC sensors with excellent stability.

Two novel low-firing KBaHo(MoO 4 ) 3 (KBHM) and KBaTm(MoO 4 ) 3 (KBTM) ceramics were synthesized using a conventional solid-state method. Rietveld refinement results of the X-ray diffraction (XRD) have verified that the KBHM and KBTM ceramics are classified within a monoclinic crystal system with C2/c space group. The correlations among structural characteristics, bond features and microwave dielectric properties were comprehensively studied utilizing bond valence, packing fraction, Raman spectroscopy, global instability index, and Phillips−Van Vechten−Levine (P−V−L) theory. The compressive effects exerted by the K + and Ba 2+ mainly contributed to the higher ε theo compared to the ε corr . According to the analysis grounded in P−V−L, the lattice energy and bond energy associated with the Mo−O bond played a substantial role in influencing the Q × f and τ f . Furthermore, the Q × f values had relation to the material's density and its packing fraction. The excellent microwave dielectric properties ( ε r = 8.74, τ f = −30.4 ppm/°C and Q × f = 42, 071 GHz) were obtained in the KBHM ceramic sintered at 800 °C. By comparison, KBTM ceramic sintered at 810 °C exhibited superior properties ( ε r = 8.81, Q × f = 52, 946 GHz and τ f = −31.6 ppm/°C). Moreover, KBTM ceramic can be co-fired with Ag without chemical reactivity, which makes it a promising candidate for low temperature co-fired ceramic (LTCC) technology.

Panaxadiol saponin (PND) is a latent targeted drug for the treatment of aplastic anemia (AA). In this study, we examined the effects of PND on ferroptosis in iron-overload AA and Meg-01 cells. We utilized RNA-seq to analyze differentially expressed genes in iron-induced Meg-01 cells treated with PND. The effects of PND or combined with deferasirox (DFS) on iron deposition, labile iron pool (LIP), several ferroptosis events, apoptosis, mitochondrial structure, as well as ferroptosis-, Nrf2/HO-1-, and PI3K/AKT/mTOR pathway-related markers in iron-induced Meg-01 cells were examined by Prussian-blue staining, flow cytometer, ELISA, Hoechst 33342 staining, transmission electron microscope, and Western blot assays, respectively. Additionally, an AA mice model with iron overload was established. Then, the blood routine was assessed, and the number of bone marrow-derived mononuclear cells (BMMNCs) in mice was counted. Also, serum iron, ferroptosis events, apoptosis, histology, T lymphocyte percentage, ferroptosis-, Nrf2/HO-1-, and PI3K/AKT/mTOR signaling-related targets in primary megakaryocytes of AA mice with iron overload were assessed by commercial kits, TUNEL staining, hematoxylin and eosin (H&E) staining, Prussian blue staining, flow cytometer, and qRT-PCR analysis, respectively. PND suppressed iron-triggered iron overload, and apoptosis, and ameliorated mitochondrial morphology in Meg-01 cells. Importantly, PND ameliorated ferroptosis-, Nrf2/HO-1-, and PI3K/AKT/mTOR signaling-related marker expressions in iron-induced Meg-01 cells or primary megakaryocytes of AA mice with iron overload. Moreover, PND ameliorated body weight, peripheral blood cell counts, the number of BMMNCs, and histological injury in the iron-overload AA mice. Also, PND improved the percentage of T lymphocytes in the iron-overload AA mice. PND attenuates ferroptosis against iron-overload AA mice and Meg-01 cells via activating Nrf2/HO-1 and PI3K/AKT/mTOR pathway and is a promising novel therapeutic candidate for AA.

This paper reports the syntheses of a series of polychromatic pigments with the general formula LiAl 1- x M x (MoO 4 ) 2 (M = Fe, Pr, Ho, Nd, Er, Ce; x = 0–0.4) and the characterization of near-infrared reflectance properties. The synthesized pigments exhibited green (Pr), lavender (Nd), pink (Er) and different yellows (Fe, Ho, Ce). The XRD patterns of typical LiAl 1- x Fe x (MoO 4+δ ) 2 samples show that the prepared samples are triclinic lithium aluminum molybdate structures with space group P -1.The iron in the sample mainly existed in the form of Fe 3+ , which led to the directional growth of grains with different morphologies. The near-infrared solar reflectance (R*%) of LiAl 1- x Fe x (MoO 4 ) 2 pigments reached 94.19%. As the doping concentration of rare earth was increased, the color of the pigments gradually deepens. By introducing rare earth elements Pr, Ho, Nd, Er, and Ce into the LiAl(MoO 4 ) 2 matrix, the color of the pigment was successfully changed from white to green, lavender, pink, and yellow, indicating that doping can control the color of the pigment. Moreover, the prepared pigments had high R*% (>87%), indicating that the pigment samples have excellent thermal insulation properties and can be used as a new type of reflective thermal insulation coatings.

Luminescence thermometry has recently been gaining popularity in science and technology fields. However, most reported works have used the fluorescence intensity ratio (FIR) to obtain the temperature based on thermally coupled energy levels (TCLs), which have hindered the sensitivity of the technique and limited its applications. Here, NaYF4:Yb/Tm@NaYF4:Yb/Ho microcrystals were prepared using a hydrothermal method. Based on the non-thermally coupled energy levels (NTCLs) of the double luminescent ionic centers Tm3+ and Ho3+, the materials exhibited temperature-dependent luminescence in the range of 304 K to 574 K. The maximum sensitivities were found to be Sa = 0.135 K−1 at 574 K and Sr = 1.101% K−1 at 484 K based on red (Tm3+) and green (Ho3+) emission bands. This study has provided a promising new strategy to break the TCL restriction and has shown the material to have potential applications in luminescence thermometry.

Cadmium (Cd) is a non-essential heavy metal that is highly toxic to testicle. Selenium (Se) is known to possess antagonistic effects against Cd toxicity, yet the precise mechanisms through which Se counteracts Cd-induced testicular damage in chickens through Nuclear factor erythroid 2-related factor 2/Heme oxygenase-1 (Nrf2/HO-1) signaling pathway, oxidative stress (OS), apoptosis, and inflammation remained unclear. In the present study, the experimental model of chicken testis was established by incorporating CdCl 2 and Na 2 SeO 3 into the dietary intake. After 60 days, chickens from each group were euthanized, and testicular and serum samples were subsequently collected. Ultrastructural assessment revealed that Se supplementation significantly mitigated the testicular damage induced by Cd. Se effectively suppressed the Cd-induced elevation in ROS, MDA, and H 2 O 2 levels, while also preventing the downregulation of CAT, GSH, and T-AOC levels. Furthermore, Se administration ameliorated the reduction in the expression levels of Nrf2, HO-1, and Bcl-2 induced by Cd, and counteracted the overexpression of Caspase-3, Bax, Cyt-c, and Caspase-9, TNF-α, IL-2, IL-6, and IL-1β. Meanwhile, immunofluorescence data demonstrated that Se attenuated the Cd-induced decrease in Nrf2 and HO-1 expression and the upregulation of IL-6 expression. In conclusion, this study elucidated that Se might mitigate Cd-induced oxidative stress in chicken testicles through the stimulation of the Nrf2/HO-1 signaling pathway, thereby inhibiting apoptosis and inflammation, and was beneficial in reducing Cd-induced testicular injury.

Acute liver injury is a type of liver diseases, and it has raised concerns worldwide due to the lack of effective therapies. The aim of this study is to investigate the protective effects of nootkatone (NOOT) on carbon tetrachloride (CCl4)-caused acute liver injury in mice. Mice were randomly divided into control, CCl4model, NOOT, and NOOT (5, 10, and 20 mg/kg/day) plus CCl4groups, respectively. Mice in the CCl4plus NOOT groups were orally administrated with NOOT at 5, 10, and 20 mg/kg/days for seven days prior to 0.3% CCl4injection at 10 mL/kg body weight, respectively. Our results showed that NOOT supplementation significantly ameliorated CCl4-induced increases of serum AST and ALT levels, hepatocyte necrosis, inflammatory response, oxidative stress, and caspases-9 and -3 activities in the livers of mice. Moreover, NOOT supplementation significantly upregulated the expression of Nrf2 and HO-1 mRNAs but downregulated the expression of NF-κB mRNAs and the levels of IL-1β, IL-6, and TNF-α proteins in the liver tissues, compared to those in the CCl4model group. In conclusion, for the first time, our results reveal that NOOT could offer protective effects against CCl4-caused oxidative stress and inflammatory response via the opposite regulation of Nrf2/HO-1 pathway and NF-κB pathway. Keywords:nootkatone;oxidative stress;acute liver injury;Nrf2/HO-1 pathway;NF-κB pathway
