The cell membrane, consisting of a phospholipid bilayer, is an important defense system of lactic acid bacteria (LAB) against adverse conditions. However, this membrane gets damaged during the process of spray drying of LAB into powder. In this study, two strains of Lactobacillus bulgaricus L9–7 and L4–2-12 with significantly different survival rates of about 22.49% and 0.43% after spray drying were explored at the cell membrane level. A total of 65 significantly different lipid species were screened from the cell membranes of two strains, with cardiolipin (CL) 15:1_22:6_24:0_28:0 being the crucial lipid species affecting membrane resistance. Finally, the KEGG enrichment analysis revealed that glycerophospholipid metabolism was the most predominant pathway, and eleven lipid species were annotated, including CL. Overall, this paper provides valuable insights into enhancing the heat tolerance of LAB.

Coal fly ash (CFA) disposal poses environmental concerns, yet offers a source for valuable metal recovery. This study introduces a sustainable heterotrophic bioleaching approach utilizing the citrus peel autoclave liquor as a novel and economical carbon source. Hydrothermal pretreatment of citrus peel yielded a liquor rich in glucose (14.9 g/L), fructose (18.4 g/L), and organic acids like citric, gluconic, and malic acid. This liquor effectively supported the growth of two previously verified strains with outstanding bioleaching performance ( Thermotoga sedimenticola and Lactobacillus pentosus ). L. pentosus demonstrated superior metal extraction from CFA, achieving efficiencies of 66.2 % for Zn, 40.1 % for Sr, and 11.8 % for V at 3 wt% pulp density. The innate buffering capacity of the citrus peel autoclave liquor was essential for maintaining a near-neutral pH at high pulp densities (20 wt%), counteracting the high alkalinity of CFA and sustaining L. pentosus ' bioleaching activity. Conversely, under similar high pulp density, pure sugar media failed to support the growth of L. pentosus or effectively leach metals due to the extreme alkalinity. This research successfully validates citrus peel autoclave liquor as an effective and economical substrate for valuable metal recovery from CFA, offering a promising "waste-to-value" strategy and an alternative to conventional methods for CFA bioleaching.

This present study was designed to test the protective role of two Lactobacillus plantarum strains, E680 and ZY08, against alcoholic liver disease (ALD) in C57BL/6 mice. The ALD mouse model was established by exposing the mice to a Lieber-DeCarli ethanol liquid diet. The two probiotic strains (10 9 cfu/day) were administered by oral gavage, respectively. Our data showed that L. plantarum ZY08, but not E680, intervention significantly mitigated alcohol-related hepatic steatosis, liver injury, intestinal barrier, and it alleviated plasma endotoxin (LPS) levels, and affected hepatic genes relating to lipid metabolism. Furthermore, Lactobacillus plantarum ZY08 effectively restored intestinal flora homeostasis via recovering flora abundance, including Blautia, Oscillibacter , Lachnoclostridium and Intestimonas , and consequently elevated intestinal short-chain fatty acid (SCFA) content. More importantly, removing intestinal microorganisms through ABX gavage markedly abolished the beneficial aspects of Lactobacillus plantarum ZY08, indicating that the regulative role of Lactobacillus plantarum ZY08 contributed to its protective role against ALD. Overall, Lactobacillus plantarum ZY08 is a potential candidate for mitigating alcohol-induced hepatic steatosis and liver injury.

Scope Chronic liver diseases are clinically silent and responsible for significant morbidity and mortality worldwide. Jujube has displayed various biological activities. Here, the therapeutic effect of Lactobacillus acidophilus ( L. acidophilus )-fermented jujube juice (FJJ) and the possible mechanism against chronic liver injury (CLI) in mice are further studied. Methods and results After the CCl 4 -induced CLI mice are separately treated with L. acidophilus (LA), unfermented jujube juice (UFJJ), and FJJ, FJJ but not LA or UFJJ suppresses the liver index. By using H&E staining, immunofluorescence staining, RT-PCR, and western blotting, it is shown that LA, UFJJ, and FJJ intervention ameliorate hepatocyte necrosis, inhibit the mRNA levels of pro-inflammatory (NLRP3, Caspase-1, IL-1β, and TNF-α) and fibrosis-associated factors (TGF-β1, LXRα, and MMP2). Also, FJJ displays significant protection against mucosal barrier damage in CLI mice. Among the three interventions, FJJ exhibits the best therapeutic effect, followed by UFJJ and LA. Furthermore, FJJ improves dysbiosis in CLI mice. Conclusions This study suggests that FJJ exhibits a protective effect against CCl 4 -induced CLI mice by inhibiting apoptosis and oxidative stress, regulating liver lipid metabolism, and improving gut microecology. Jujube juice fermentation with L. acidophilus can be a food-grade supplement in treating CLI and related liver diseases.

Emulsification/internal gelation is an effective encapsulation method to protect probiotics from adverse environment. In this paper, microcapsules of Lactobacillus acidophilus CGMCC1.2686 were prepared by emulsification and gelation of alginate solutions with slow release of Ca 2+ ions from calcium carbonate (CaCO 3 ) and calcium disodium ethylenediaminetetraacetate (Ca-EDTA). Physical characterization of the two types of microcapsules showed that alginate-Ca-EDTA microcapsule was more uniform in size than alginate-CaCO 3 , with a span factor of 0.96 and 1.20, respectively. Mechanical measurements demonstrated that alginate-CaCO 3 microcapsule was more robust and elastic, which was supported by its denser structure as observed by electron scanning microscopy. Bacteria encapsulation yields in the two microcapsules were similar, 37.9% for alginate-CaCO 3 and 36.9% for alginate-Ca-EDTA. However, alginate-CaCO 3 microcapsule exhibited much higher cell survivals in both simulated gastric juice and bile salts solution (22.2% and 2.6*10 −2 %, respectively) than alginate-Ca-EDTA (7.1% and 0%, respectively), indicating a more effective protection of L. acidophilus CGMCC1.2686. The protection efficiency was discussed in relation to the mechanical properties of the microcapsules and the calcium sources used.

Probiotics are considered to have the potential to be beneficial to health when consumed in adequate amounts. Maintaining excellent probiotic activity during food storage and gastrointestinal transport is a key challenge. In this study, a stable water-in-water (W/W) Pickering emulsion consisting of hydroxypropyl methylcellulose (HPMC) and dextran (DEX) was constructed. Cellulose nanocrystals (CNCs) were used as the solid stabilizers of the emulsion. The obtained emulsions had excellent long-term stability with no phase separation within 60 days. In addition, the pH and ionic strength had little influence on the stability performance of the emulsions. The W/W Pickering emulsion system was applied to the probiotic encapsulation, and the emulsion was microencapsulated by spray drying technique. The microscopy images showed that Lactobacillus plantarum tended to be enriched in the dispersed phase of W/W Pickering emulsions. The viable cell content of obtained microcapsules was 2.22 × 10 10 CFU/g after spray drying. Furthermore, the viable cell content in emulsion microcapsules was 3.38 × 10 6 CFU/g after the simulated digestion process in artificial gastrointestinal fluid. The microcapsules containing Lactobacillus plantarum also showed excellent stability during the storage. The viable cell content of microcapsules was 1.76 × 10 9 CFU/g at 4 °C after 90 days of storage. This study could provide a potential method by using W/W Pickering emulsion as a system for encapsulating probiotics, which would provide protection for probiotics during spray drying and gastrointestinal digestion.

The aim of the present study was to characterize the fermentation properties of Lactobacillus rhamnosus GG (LGG; ATCC 53103) in vitro, and investigate the effects of inoculation with LGG on fermentation quality of whole plant corn silage (WPCS) and fermented total mixed ration (FTMR). The antifungal activity of LGG against 4 prevalent fungi isolated from corn stalk was determined using the Oxford cup method. The binding capacity of LGG to aflatoxin B 1 , zearalenone and deoxynivalenol was examined by in vitro adsorption experiments using a batch mode in phosphate buffered saline. The antioxidant activity of LGG in vitro was evaluated by determining the resistance to hydrogen peroxide, free radical scavenging capacity and reducing activity. Three batches of whole plant corn and total mixed ration were collected separately. Each batch of raw material was divided into equal portions and applied to 2 treatments: (1) control with distiller water (CON), and (2) treatment inoculated with LGG (LGG). After 48 d of ensiling, the chemical composition, ruminal degradability, microbial community and mycotoxin concentrations of WPCS and FTMR were determined. The results showed that lactic acid was the predominant component among organic acids in LGG culture process, with a rapid generated occurring at 4 h after inoculation and reaching a stable state at approximately 48 h. Lactobacillus rhamnosus GG could effectively inhibit the growth of 4 prevalent fungi isolated from corn stalk, the diameter of inhibition zone was higher for Mucor circinelloides than that of the other moulds ( P = 0.016). With increasing of LGG cell amount, the binding capacity to 3 mycotoxins increased ( P < 0.01); the heat and acid treated LGG also enhanced the binding capacity to mycotoxins ( P < 0.01). Lactobacillus rhamnosus GG displayed tolerance to hydrogen peroxide (0.8 mmol/L) for 8 h at 37 °C (96%, viability), but the viability decreased sharply (52.39%, viability) under the concentration of H 2 O 2 increased to 1.2 mmol/L ( P < 0.001). The free radical scavenging activity and reducing activity of viable LGG were greatest, followed by cell-free extracts and inactivated cells ( P < 0.001). Ensiling of WPCS and FTMR inoculated with LGG decreased dry matter loss ( P < 0.05), increased lactic acid and acetic acid production ( P < 0.05), enhanced some nutrients ruminal degradability ( P < 0.05). Meanwhile, the inoculation of LGG reduced the counts of yeast, mold and coliform bacteria ( P < 0.05) and, more importantly, alleviated mycotoxin contamination ( P < 0.05). These results indicated that LGG could be a novel inoculant to improve the quality and safety of WPCS and FTMR.

In this study, the microbial interactions among co-culture of Lactobacillus delbrueckii DPUL-F36, Lacticaseibacillus paracasei DPUL-F115, and Kluyveromyces marxianus 39 in fermented milk were evaluated during 13 days of refrigerated storage (4 ± 0.5 °C), in terms of titratable acidity, viable cell counts and metabolites. The fermented milk involving K.marxianus 39 showed a maximum lactobacilli count (4.8 × 10 9 CFU/mL) and titratable acidity (151.0 ± 1.7 °T). K. marxianus 39 could enrich the flavor of the fermented milk and improve the sensory quality of the fermented milk on the 5th and 9th day of storage. The contents of aspartic acid, glycine, alanine, valine, methionine, leucine, lysine, and tyrosine increased significantly, while the contents of valine, methionine, leucine, phenylalanine, and lysine in milk fermented by Lactobacillus delbrueckii DPUL-F36, Lacticaseibacillus paracasei DPUL-F115, and Kluyveromyces marxianus 39 were significantly lower than those in milk fermented by Lactobacillus delbrueckii DPUL-F36, Lacticaseibacillus paracasei DPUL-F115 during storage. The higher concentrations of acetic acid, nonanoic acid, acetoin, 2-nonanone, and 2-undecanol were observed in the fermented milk involved with K. marxianus 39. Our results showed K. marxianus 39 has the potential to be a starter culture for fermented dairy products.

This present study was designed to explore the protective role of Lactobacillus plantarum ZJUIDS14 against Non-alcoholic Fatty Liver Disease (NAFLD) in a high-fat-diet (HFD)-induced C57BL/6 mice model. The probiotic (109 CFU/every other day) was administered by oral gavage for 12 weeks. We found that L. plantarum ZJUIDS14 intervention significantly alleviated HFD related hepatic steatosis, liver damage, insulin resistance, and increased hepatic expression of peroxisome proliferator activated receptor α (PPAR-α) while stimulating the activation of AMP-activated protein kinase (AMPK). Furthermore, L. plantarum ZJUIDS14 improved mitochondrial function as reflected by an increase in dynamin related protein 1 (DRP1) and a decrease of proteins associated with oxidative phosphorylation (OXPHOS) after the treatment. Additionally, mice from the L. plantarum ZJUIDS14 group had a restored intestinal flora and homeostasis involving Coprostanoligenes group, Ruminococcaceae UCG-014, Allobaculum, Ruminiclostridium 1, and Roseburia. Meanwhile, these five genera exhibited a significant (negative or positive) association with ileum inflammation mRNA levels and SCFA contents, by Spearman’s correlation analysis. In general, our data demonstrated that L. plantarum ZJUIDS14 mitigates hepatic steatosis and liver damage induced by HFD. Specifically, they strengthened the integrity of the intestinal barrier, regulated gut microbiota, and improved mitochondrial function. Our data provide an experimental basis for L. plantarum ZJUIDS14 as a promising candidate to prevent NAFLD.

Fatty liver is one of the most pervasive liver diseases worldwide. Probiotics play an important role in the progression of liver disease, but their effects on host regulation are poorly understood. This study investigated the protective effects of lactobacillus gasseri (L. gasseri) against high-cholesterol diet (HCD)-induced fatty liver injury using a zebrafish larvae model. Liver pathology, lipid accumulation, oxidative stress and hepatic inflammation were evaluated to demonstrate the changes in a spectrum of hepatic injury. Moreover, multiple indexes on host gene expression profiles were comprehensively characterized by RNA screening. The results showed that treatment withL. gasseriameliorated HCD-induced morphological and histological alterations, lipid regulations, oxidative stress and macrophage aggregation in the liver of zebrafish larvae. Furthermore, the enrichment of the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway revealed that the core pathways ofL. gasseriregulation were interleukin-17 (IL-17) signaling, phosphoinositide 3-kinase (PI3K)-AKT signaling pathway, the regulation of lipolysis and adipocytes and fatty acid elongation and estrogen signaling. The genes at key junction nodes, hsp90aa1.1, kyat3, hsd17b7, irs2a, myl9b, ptgs2b, cdk21 and papss2a were significantly regulated byL. gasseriadministration. To conclude, the current research extends our understanding of the protective effects ofL. gasseriagainst fatty liver and provides potential therapeutic options for fatty liver treatment. Keywords:fatty liver;Lactobacillus gasseri;high cholesterol diet;zebrafish;IL-17

Background: Lactobacillus fermentation has become a typical food processing method with the development of the modern food industry. Aims: This study aimed to assess the effects of Lactobacillus acidophilus NCUF202.2 fermentation on Artemisia selengensis Turcz (AST) in relieving hyperuricemia (HUA) and the mechanisms involved. Results: The fermented Artemisia selengensis Turcz extracts (FASTE) could significantly increase the content of free polyphenol and enhance the inhibitory effect on xanthine oxidase (XOD) in vitro. The result of ultrafiltration coupled with UPLC-MS/MS screened out the components that might be combined with XOD in the caffeoylquinic acids (CQAs) of Artemisia selengensis Turcz extracts (ASTE) as 5-CQA, 3-CQA, 4-CQA, 1-CQA, 3,4-diCQA, 3,5-diCQA, 1,5-diCQA and 4,5-diCQA as well as caffeic acid (CA) of FASTE except the above. Our results also showed that ASTE and FASTE could alleviate HUA in rats. In contrast, FASTE had a better ability to reduce serum adenosine deaminase (ADA) activity than ASTE. Furthermore, FASTE could restore catalase (CAT) in HUA model rats in vivo, and superoxide dismutase (SOD) activity to a better degree, thereby inhibiting the production of excess malondialdehyde (MDA). Under the intervention of ASTE and FASTE, the levels of serum pro-inflammatory and anti-inflammatory factors in rats tended to be normal. However, FASTE could increase the expression of the uric acid secretion protein OAT1 and decrease the expression of reabsorption proteins URAT1 and GLUT9 in model rats, thereby reducing the serum uric acid (SUA) level in model rats. ASTE and FASTE can increase the abundance of beneficial bacteria, such as Bifidobacterium and Akkermansiaceae. Conclusion: Overall, part of diCQAs and CQAs was decomposed after fermentation. FASTE had a more substantial HUA-relieving effect than ASTE. It is expected to be applied to functional foods and contribute to the research and development of uric acid lowering functional foods.

Sour bamboo shoot is a popular traditional fermented vegetable in southern China. The current study aims to clarify the relationship among physicochemical properties, flavor, and microbial compositions of sour bamboo shoots during fermentation. Four organic acids (OAs) were considered to be the main acids in sour bamboo shoots. At least 7 volatile compounds (OAV >1) were identified as the characteristic aroma substances, especially p -cresol, which accounted for the sour odor of sour bamboo shoots. The microbial community showed that Firmicutes and Proteobacteria were the dominant phyla, while Lactococcus and Lactobacillus were the dominant genera in sour bamboo shoots. Correlation analysis indicated that Firmicutes positively correlated with p -cresol, phenol , 2-methoxy-phenol, malic acid , total titratable acidity (TTA), and some OAs. Lactococcus was positively correlated with phenol, 2-methoxy-phenol, benzyl alcohol, TTA, some OAs, and amino acids (AAs). This study provided a theoretical basis for screening indigenous aroma-producing microorganisms as the starter of sour bamboo shoots.
