Tumor microenvironment (TME)-responsive imaging and therapy has great potential for application towards precision diagnosis and treatment of cancer. Photoacoustic (PA) imaging with the second near-infrared window (NIR-II) and photothermal therapy are important ways to address high-resolution imaging of in vivo deep tumor tissue and high-effect therapy of cancer. A gold nanocluster (AuNCs) encapsulated nanorod (AuNRs) (AuNRs@MEA/DTGA@AuNCs, hereafter AMDAs) is prepared. The AuNRs are first modified using β-mercaptoethylamine (MEA) to form AuNRs@MEA. Dithioglycolic acid (DTGA) is coupled with AuNRs@MEA to obtain AuNRs@MEA/DTGA (AMD). Finally, AuNCs are attached to AuNRs@MEA/DTGA surface to form AMDAs. In this nanocomposite materials, the encapsulation of the AuNRs by the AuNCs obscures NIR-II PA signal of AuNRs. The high expressed glutathione in the TME react with the AMDAs via redox reaction, the detachment of the AuNCs from the AuNRs surface, and detection of the NIR-II PA signal from the AuNRs, thus enabling NIR-II PA imaging of tumor tissues. In addition, due to the high intensity light absorption of AuNRs in the NIR-II region, the release of AuNRs also enhance tumor photothermal imaging and therapy. This work establishes an accurate and efficient integrated platform for nanodiagnosis and treatment of cancer.

Herbal medicines and dietary supplements are widely used worldwide. However, reports have revealed that a large number of herbal medicines are implicated in the hepatic damage, which is referred to as Herbal medicines-induced liver injury (HILI). Traditional diagnostic methods for HILI include histological and serological assessments, which fail to offer real-time and unambiguous visualization of hepatotoxicity in vivo . Herein, we constructed a second near-infrared (NIR-II) luminescent nanoprobes to achieve accurate, high-efficiency, and non-invasive detection of HILI by the assembly of lanthanide-based NIR-II luminescence nanoparticles with a chromophore with absorption band in the NIR-II region. Specifically, the NIR-II luminescence of the lanthanide nanoparticles can be effectively quenched by the chromophores on the surface of nanoparticles via an absorption competition-induced quenching effect, and subsequently recovered by the pathophysiological biomarker peroxynitrite (ONOO − ) generated in early HILI. It is demonstrated that the nanoprobes are capable for the screening of liver toxins in a noninvasive pattern, which is more convenient than the conventional diagnostic approaches. Finally, our luminescence detection approach offers an immediate testing technique for the identification of hepatotoxic components of herbal medicine such as colchicine, monocrotaline and stibene glucoside, providing a high-throughput screening strategy for herbal medicine-induced hepatotoxicity.

Chemodynamic therapy, which produces cytotoxic hydroxyl radicals through intratumoral Fenton reactions, has been extensively explored in nanomedicine for cancer treatment. However, the limited endogenous hydrogen peroxide (H 2 O 2 ) levels, which serve as the reactant, weakens the anticancer effect of the Fenton reaction. In this work, a cascade catalytically reactive nanosystem has been constructed for the treatment of epithelial and embryonal tumors with high synergetic efficacy through the integration of natural glucose oxidase (GOD) and polyethylene glycol (PEG) onto the surface of Fe 3 S 4 nanoplates (abbreviated as Fe 3 S 4 -PEG-GOD NPs). The GOD component consumes glucose in tumor and produces a large amount of H 2 O 2 , which provides rich substrates for the Fenton reaction catalyzed by the Fe catalytic center of the nanoplates. The intriguing photothermal conversion efficiency (45%) of the constructed composite nanocatalysts in the second near infrared (NIR-II) biowindow allows for photothermal treatment to substantially strengthen and synergize the anticancer effects of nanocatalysts both in vitro and in vivo. This synergy occurs through generation of a hyperthermic effect necessary for promoting the sequential catalytic reaction. The synergistic anticancer effect has been verified in both epithelial and embryonal tumor mouse models, and provides justification for the biomedical use of multifunctional nanocatalysts for versatile tumor nanotherapeutics.

Multidrug-resistant Staphylococcus aureus (MRSA) seriously endanger human health. The development of efficient methods to eliminate the infections and monitor the treatment process are of great significance. Near-infrared-II (NIR-II) photoacoustic (PA) imaging and photothermal therapy (PTT) are highly integrated theranostic platforms with superior performance including a low imaging background, increased tissue penetration depth, and high photothermal threshold. Herein, we report an activatable near-infrared II (NIR-II) phototheranostic strategy using miniature Au/Ag nanorods (NRs) for the photochemical synergistic therapy of MRSA infections and in situ monitoring of the treatment progress. Au/Ag NRs were efficiently activated by ferricyanide solution and allowed to continuously release free Ag + to eliminate MRSA, triggering NIR-II photothermal and PA performance enhancement. The activated NIR-II photothermal effect in turn accelerated the release of free Ag + from Au/Ag NRs for the synergistic elimination of gram-positive Staphylococcus aureus and promoted wound healing. No photothermal damages or free Ag + -induced side effects were observed in treated mice. After synergistic treatment, a 20-fold NIR-II PA signal increase with a maximum signal-to-noise measurement of 9.5 was observed between the implanted site and normal tissue, enabling sensitive monitoring of Ag + release process. The prepared Au/Ag NRs were stable and biocompatible, showing great potential for activatable NIR-II phototheranostic application.

Exploiting two-dimensional nanomaterials as photo-based theranostic agents is promising for the highly efficient ablation of deep-tissue-buried tumors. However, they are limited by their poor absorption in the second near-infrared-light (NIR-II) bio-window (1000–1300 nm) and intrinsic nonbiodegradability. Herein, defect-rich sulfur-doped Ni(OH)2 (S-Ni(OH)2) nanosheets decorated with bovine serum albumin (BSA) as a novel theranostic agent is developed, which can accomplish multimodal-imaging-guided photothermal ablation of mouse cancers in the NIR-II bio-window. Sulfur doping extends the absorption spectra of Ni(OH)2 nanosheets from the visible to NIR-II bio-window, affording highly efficient photothermal conversion (58.20% for 1064 nm), entailing it to become an excellent contrast agent for photoacoustic imaging. Further, because of their intrinsic paramagnetic property, they can be applied for magnetic resonance imaging. Owing to the abundant defective sites in S-Ni(OH)2 nanosheets, they exhibit response to the tumor microenvironment, resulting in effective biodegradation and excretion from the body. In vivo toxicity experiments indicated that S-Ni(OH)2-BSA NSs delivered no appreciable toxicity and good biocompatibility. This work provides an avenue for the rational design of effective theranostics agents.

Timely detection of hepatic oxidative stress is effective in preventing the onset and progression of liver disease. However, real-time and dynamic monitoring of oxidative stress in vivo remains challenging. Herein, we report a H 2 O 2 -responsive dual-peak ratiometric NIR-II fluorescent probe (Er@Ag 2 S:Pb), made by combining H 2 O 2 -responsive fluorescent Pb-doped Ag 2 S quantum dots (Ag 2 S:Pb QDs) with a fluorescence-stabilized reference substrate NaYF 4 :Yb/Er @NaYF 4 . By monitoring the change in emitted I 1550 /I 1220 intensities the probe can be used to sense and quantify H 2 O 2 concentrations ranging from 5 to 100 μM with excellent specificity. Experimental studies using mice models have shown that this probe can be used to monitor of hepatic H 2 O 2 -related oxidative stress in real time and non-invasively. Thus, the developed probe provides a powerful ratiometric NIR-II fluorescence imaging tool for timely monitoring of oxidative stress in vivo .

Multiplexed fluorescence imaging is mainly applied to cellular imaging. However, the application of an analogous technique to mammals remains challenging due to the presence of substantial scattering and autofluorescence in biological tissues in the visible (350–700 nm) and near-infrared I region (NIR-I, 700–900 nm). Herein, we report multiplexing excitation and single-channel detection in the near-infrared II region (NIR-II) with two fluorescent probes for dual-color imaging in vivo. The first one, FNIR-1090 was constructed on Pr-1099 derivative by attaching with 1-Ethylpiperazine through the SRN1 displacement reaction. The second one, IR-765 was synthesized by a one-step reaction between the Vilsmeier–Haack reagent with 2-(3,5,5-trimethylcyclohex-2-enylidene) malononitrile (TEMO). It is noteworthy that the absorption spectrum of FNIR-1090 is almost completely separated from that of IR-765, which facilitates dual-color in vivo imaging. The two dyes can be used for high-contrast dual-channel vivo imaging of different tissues. All these prominent performances show the great promise of FNIR-1090 and IR-765 in non-invasive, real-time, and dual-color NIR-II imaging (>900 nm) in vivo.

Developing an effective dressing against bacterial infection and synchronously addressing wound complications, such as bleeding, long-term inflammation, and reinfection, are highly desirable in clinical practice. In this work, a second near-infrared (NIR-II) responsive nanohybrid consisting of imipenem encapsulated liposome with gold-shell and lipopolysaccharide (LPS)-targeting aptamer, namely ILGA, is constructed for bacteria elimination. Benefiting from the delicate structure, ILGA exhibits strong affinity and a reliable photothermal/antibiotic therapeutic effect toward multidrug-resistant Pseudomonas aeruginosa (MDR-PA). Furthermore, by incorporating ILGA with a thermosensitive hydrogel poly(lactic-co-glycolic acid)–polyethylene glycol–poly(lactic-co-glycolic acid) (PLGA-PEG-PLGA), a sprayable dressing ILGA@Gel was prepared, which enables a quick on-demand gelation (10 s) for wound hemostasis and offers excellent photothermal/antibiotic efficacy to sterilize the infected wound. Additionally, ILGA@Gel provides satisfactory wound-healing environments by reeducating wound-associated macrophages for inflammation alleviation and forming a gel layer to block exogenous bacterial reinfection. This biomimetic hydrogel reveals excellent bacteria eradication and wound recovery effectiveness, demonstrating its promising potential for managing complicated infected wounds.

Nanozymes mediated catalytic therapy can produce toxic reactive oxygen species (ROS) and destroy the metabolic balance of tumor cells, providing a new direction for cancer treatment. However, the catalytic efficiency of a single nanozyme is limited by the complexity of the tumor microenvironment (hypoxia, GSH overexpression, etc.). In order to overcome these problems, we designed flower-like Co-doped FeSe2 (Co–FeSe2) nanozymes by a simple wet chemistry method. Co–FeSe2 nanozymes not only exhibit high POD and OXD-mimicking activities for facile kinetics, but also effectively consume over-expressed glutathione (GSH), inhibiting the consumption of generated ROS and destroying the metabolic balance of the tumor microenvironment. These catalytic reactions trigger cell death through apoptosis and ferroptosis dual pathways. More importantly, under the NIR II laser irradiation, the catalytic activities of Co–FeSe2 nanozymes are boosted, confirming the photothermal and catalytic synergistic tumor therapy. This study takes advantage of self-cascading engineering that offers new ideas for designing efficient redox nanozymes and promoting their clinical translation.

The therapeutic effects of photothermal therapy (PTT) are dependent on the photothermal conversion efficiency of photothermal agents (PTAs) in tumors and the subsequent activation of the antitumor immune system. However, the insufficient tumor accumulation of current PTAs and the inevitable recruitment of tumor-associated macrophages (TAMs) could further compromise the antitumor activities of PTT. To address these issues, a biomimetic photothermal nanoplatform [email protected] is developed for the targeted remodeling of TAMs, which promotes the antitumor immunity of PTT. Au nanorods with second near-infrared (NIR-II) absorptions are fabricated to serve as PTAs to induce immunogenic cell death in tumor cells. The ferric hydroxide shell coated on Au nanorods can release iron ions to repolarize M2-like TAMs into the tumoricidal M1 phenotype via P38 and STAT1-mediated signaling pathways . Moreover, the surface decoration of platelet membranes endows biomimetic nanoplatform with enhanced tumor targeting ability for precise tumor ablation and TAM regulation. Consequently, [email protected] under NIR-II laser irradiation exhibits significantly higher inhibitory effects in a poor immunogenic 4T1 tumor-bearing mouse model with a 50% complete remission rate compared to conventional PTT (0%). By simultaneously reversing the immunosuppressive tumor microenvironment, this biomimetic nanoplatform offers a promising strategy for enhancing the antitumor efficacy of PTT. Statement of significance The therapeutic effects of current photothermal therapy (PTT) are hindered by the insufficient tumor accumulation of conventional photothermal agents and the recruitment of immunosuppressive tumor-associated macrophages (TAMs) after PTT. Herein, we report a biomimetic iron-based second near-infrared (NIR-II) photothermal nanoplatform ( [email protected] ) for targeted TAMs reprogramming and NIR-II mediated anti-tumor immunity. [email protected] can actively target the tumor site with the help of surface-decorated platelet membranes. Meanwhile, iron ions would be released from [email protected] in acidic lysosomes to reprogram TAMs into tumoricidal M1-like macrophages, which promotes the antitumor responses elicited by NIR-II PTT, thereby contributing to remarkable tumor inhibitory effects, with 50% higher complete remission rate than that of conventional PTT.

Herein, we reported nitroxide radical-modified CuS nanoparticles (CuS–NO˙ NPs), and they exhibited a typical absorption peak at 1182 nm. Due to such a long wavelength absorbance, CuS–NO˙ NPs exhibited excellent therapeutic outcome and low damage to normal tissues. Besides, we simultaneously achieved CuS–NO˙ NPs for MRI and CT dual-modal imaging, which successfully provided a new strategy for imaging-guided tumor treatment, thus increasing potential clinical applications for cancer treatment.

Phosphor conversion light-emitting diode (PC-LED) technology has become a focal point of research for portable near-infrared light sources in recent years. One of the crucial aspects is to develop phosphor materials that can be effectively stimulated by commercial available blue light. Unfortunately, efficient phosphors in the NIR-II that can be excited by commercial blue light are particularly scarce. In this study, Mg 4 Ta 2 O 9 :0.04Cr 3+ ,0.03Yb 3+ ,0.07Li + (MTO:0.04Cr 3+ ,0.03Yb 3+ ,0.07Li + ) phosphor was synthesized using the high temperature solid phase method, and its emission spectrum covered the first and second near-infrared windows (700–1200 nm). The internal quantum efficiency (IQE) and external quantum efficiency (EQE) are 88.7 % and 45.6 %, respectively, due to the synergistic effect of energy transfer and Li-ion modification. Additionally, the doping of Yb 3+ ions and Li + ions enhances the thermal stability of luminescence. The thermal stability of MTO:0.04Cr 3+ ,0.03Yb 3+ ,0.07Li + and MTO:0.04Cr 3+ ,0.03Yb 3+ is 9.81 % and 14.43 % higher, respectively, than that of MTO:0.04Cr 3+ ,0.03Yb 3+ and MTO:0.04Cr 3+ . To evaluate its potential applications, we developed a prototype PC-LED utilizing MTO:0.04Cr 3+ ,0.03Yb 3+ ,0.07Li + phosphor as an emission emitter which demonstrated advantages in biomedical imaging, night vision, and non-destructive testing.
