BACKGROUND Chlorantraniliprole (CAP) is a diamide insecticide with high efficacy against many pest insects, including the black cutworm, Agrotis ipsilon . Agrotis ipsilon is a serious pest causing significant yield losses in crops. Glutathione- S -transferases (GSTs) belong to a family of metabolic enzymes that can detoxify a wide range of pesticides. However, little is known about the functions of GSTs in CAP tolerance in A. ipsilon . RESULTS A cDNA sequence (designated AiGSTu1 ) encoding an unclassified GST was identified from A. ipsilon . AiGSTu1 is highly expressed during the 3 rd -instar larval and the pupal stages. Most of the mRNA transcripts were found in larval Malpighian tubules. Exposure to CAP strongly enhanced AiGSTu1 expression, GST activity, hydrogen peroxide (H 2 O 2 ) and malondialdehyde levels in larvae. H 2 O 2 treatment upregulated the transcription level of AiGSTu1 , suggesting that CAP-induced oxidative stress may activate AiGSTu1 expression. The activity of recombinant AiGSTu1 was inhibited by CAP in a dose-dependent manner. Metabolism assay results demonstrated that AiGSTu1 is capable of depleting CAP. Overexpression of AiGSTu1 enhanced the tolerance of Escherichia coli cells to H 2 O 2 and the oxidative stress inducer, cumene hydroperoxide. Silencing of AiGSTu1 by RNA interference increased the susceptibility of A. ipsilon larvae to CAP. CONCLUSION The findings of this study provide valuable insights into the potential role of AiGSTu1 in CAP detoxification and will improve our understanding of CAP tolerance in A. ipsilon . © 2023 Society of Chemical Industry.

BACKGROUND Phytophthora capsici is an extremely destructive phytopathogenic oomycete that causes huge economic losses. However, due to the drug resistance risk and environmental threat of chemical fungicides, it is necessary to develop environmentally friendly biocontrol alternatives. Rhein is a major medicinal ingredient of traditional Chinese herbs, and it is widely used in the medical field. However, its inhibitory effect against phytopathogens is unknown. Herein, the antifungal spectrum of rhein and its possible action mechanism against P. capsici were investigated. RESULTS Rhein possessed broad-spectrum antifungal activity against phytopathogens, particularly P. capsici , Phytophthora infestans , Helminthosporium maydis , and Rhizoctonia solani . Rhein inhibited the mycelial growth as well as the spore germination of P. capsici with mean 50% effective concentration (EC 50 ) values of 4.68 μg mL −1 and 6.57 μg mL −1 against 117 P. capsici isolates, respectively. Rhein effectively suppressed the occurrence and spread of Phytophthora blight and significantly destroyed the cell membrane permeability and integrity of P. capsici , corroded its cell wall integrity, and damaged its morphology and ultrastructure. Moreover, rhein caused a considerable reduction in the phospholipid and cellulose contents. Genome-wide transcriptional profiling of P. capsici in response to rhein indicated significant reduction in the expression levels of genes participating in glycerolipid metabolism and starch and sucrose metabolism. Additionally, rhein strengthened the disease defense system of pepper by enhancing related enzyme activities. CONCLUSION This study demonstrated that rhein could effectively inhibit P. capsici using multiple mechanisms of action. Rhein has the potential to be an efficient alternative to control diseases caused by P. capsici . © 2023 Society of Chemical Industry.

Background Root knot nematodes cause great damage to crops worldwide. Due to the negative effects of the application of fumigant and old chemical nematicides, biological nematicides have drawn increasing attention in recent years. Here we tested the fumigant activity of the volatile organic compounds (VOCs) blends emitted from Paenibacillus polymyxa and pure commercial VOCs against M. incognita . Results In this study, we investigated whether P. polymyxa strain J2-4 could produce VOCs that exhibit nematicidal activity. In vitro assays indicated that J2-4 VOCs were highly toxic to second stage juveniles (J2s) and could inhibit egg hatching. Three-layered pot experiments showed that the number of nematodes that penetrating in cucumber roots was reduced by 69.27% after the application of J2-4 VOCs under greenhouse conditions. We identified fourteen volatiles using solid-phase micro-extraction gas chromatography–mass spectrometry. The efficacy of six commercially available VOCs, namely 2-isobutyl-3-methylpyrazine, 2,4-dimethoxybenzaldoxime, 2-dodecanone, 2-tridecanol, 2-tridecanone, and 2-tetradecanol, against M. incognita were examined. Except for 2,4-dimethoxybenzaldoxime, the remaining five VOCs showed strong direct-contact nematicidal activity against J2s of M. incognita , and only 2-isobutyl-3-methylpyrazine showed strong fumigant activity against J2s of M. incognita . In pot experiments, 2-isobutyl-3-methylpyrazine and 2-dodecanone reduced the number of root galls by about 70%, and 2-tridecanone reduced the number of root galls and egg masses by about 63% compared with controls. Conclusion Paenibacillus polymyxa strain J2-4 exhibited high fumigant activity against M. incognita. Our results provide evidence for the use of J2-4 and its VOCs as biocontrol agents in the management of root-knot nematodes. This article is protected by copyright. All rights reserved.

BACKGROUND The bites and blood sucking of bed bugs ( Cimex spp.) (Hemiptera: Cimicidae) pose a serious threat to human physical and mental health. Application of an effective repellent can prevent or reduce bed bug bites. Previous studies on repellent screening mainly focused on Cimex lectularius L. In this study, we investigated the repellent effect of two safe food additives, ethyl anthranilate (EA) and butyl anthranilate (BA), against Cimex hemipterus (F.), and also explored the role of antennae and mouthparts on C. hemipterus perception of repellents. RESULTS Both EA and BA had a strong repellent effect against tropical bed bugs and their repellency was similar or lower than that of N , N -diethyl-3-methyl benzoyl amide, depending on whether or not a CO 2 source was present. EA had higher repellency than BA and exhibited repellency to C. hemipterus for 3 days when a CO 2 source was present. C. hemipterus avoided resting on 20% EA- and BA-treated harborages. Applying 20% EA and BA on rabbit skin significantly reduced the blood intake of C. hemipterus within 2 h. C. hemipterus could perceive EA and BA after their antennae or mouthparts or both antennae and mouthparts were removed. CONCLUSION Both EA and BA had strong repellency against C. hemipterus , with EA being more repellent. Ablation of antennae and mouthparts did not affect the perception of EA and BA by C. hemipterus . © 2023 Society of Chemical Industry.

BACKGROUND Nectar plants provide extra nourishment for parasitoids, which can utilize floral volatiles to locate nectar-rich flowers. A promising strategy is to screen potential floral species based on the wasps' olfactory preferences for nectar sources, and to ensure their suitability for both natural enemies and targeted pests. Cotesia vestalis (Haliday) is a dominant parasitoid of the oligophagous pest Plutella xylostella , which poses a significant threat to cruciferous vegetables globally. However, the chemical cues in plant–parasitoid complexes mediating Cotesia vestalis to locate nectar food resources and the positive effect of nectar plants on the Cotesia vestalis population are poorly understood. RESULTS The results showed that Fagopyrum esculentum was the most attractive plant that attracted Cotesia vestalis , not Plutella xylostella in 44 flowering plants from 19 families. 1,2-Diethyl benzene and 1,4-diethyl benzene, identified from the floral volatiles from F. esculentum in full bloom, were found to elicit dose-dependent electrophysiological responses and attract Cotesia vestalis adults, demonstrating their potential as semiochemicals. Moreover, the age-stage, two-sex life table revealed that feeding on nectar food increased the efficacy of Cotesia vestalis adults against Plutella xylostella . CONCLUSION In summary, the findings provide insights into the chemical ecology of plant–parasitoid complexes and support the potential use of F. esculentum as insectary plants in habitat manipulation against Plutella xylostella by supplying natural nectar food for the Cotesia vestalis population. Our results suggest an attract and reward strategy based on an attractant for Cotesia vestalis to control Plutella xylostella , or the development of volatile-based artificial food for Cotesia vestalis . © 2023 Society of Chemical Industry.

BACKGROUND Root-knot nematodes (RKNs) are the highly damaging pests for various crops, and the prevalence of RKNs has posed serious threats to worldwide agricultural harvest, severely affecting global food security and ecosystem health. Traditional pesticide systems on controlling RKNs generally cause environmental hazards and phytotoxicity due to the excessive use of pesticides resulted from low utilization efficiency. And effective approaches with biosafe and efficient features are highly demanded to break away from the dilemma caused by RKNs. RESULTS In this research, a nanopesticide system with root-targeted delivery function was developed to achieve effective prevention and control of RKNs. The nanocarriers (MSN-KH560-Gly) and the obtained nanopesticides (EB@MSN-KH560-Gly) were proved to be biosafe. Also, this nanopesticide system demonstrated sustained release behavior. The grafting of glycine (Gly) significantly improved the pesticide contents translocating to cucumber roots (about 304.7%). Besides, such root-targeted delivery function resulted in no root nodule in cucumber plants after the foliar application of these nanopesticides (prevention rate of 100%). In addition, the root nodule numbers of the infected cucumber plants decreased by 71.67%. CONCLUSION Foliar application of these Gly-functionalized nanopesticides achieved effective prevention and control of RKNs due to the root-targeted delivery property inherent in this nanopesticide system, and such root-targeted delivery strategy opens a novel and efficient method to protect crops from RKN invasion and thus facilitates the development of sustainable agriculture. © 2023 Society of Chemical Industry.

BACKGROUND The diamondback moth, Plutella xylostella , has developed resistance to almost all insecticides used for its control. The ‘push–pull’ method has been shown as an effective control strategy to address this resistance challenge of P. xylostella . The key focus of the strategy is the identification of attractive or repellent volatile components. The aim of this study was to identify attractive volatile compounds released from host plants. Identified compounds were applied in the biological control of this pest. RESULTS Nine active compounds released into the headspace of seven cruciferous plant species were identified using gas chromatography–electroantennographic detection and gas chromatography–mass spectrometry. Electroantennographic detection-active compounds included five green leaf volatiles (hexanal, trans -2-hexen-1-ol, cis -3-hexen-1-ol, cis -3-hexenyl acetate, and 1-penten-3-ol), three isothiocyanates (isopropyl isothiocyanate, allyl isothiocyanate, and butyl isothiocyanate), and nonanal. Except for nonanal, all the identified green leaf volatiles and isothiocyanates elicited strong electrophysiological and behavioral responses in P. xylostella . The strongest attractive compounds, trans -2-hexen-1-ol and isopropyl isothiocyanate, were further evaluated in oviposition and field-trapping assays. Results showed that they both lured female moths to lay eggs, and were highly attractive to P. xylostella adults in field, especially when used in combination with yellow and green sticky boards. However, a blend of the two compounds showed no synergistic effect, but rather an antagonistic effect. CONCLUSIONS Green leaf volatiles and isothiocyanates were identified as key olfactory cues for host selection of P. xylostella . Trans -2- hexen-1-ol and isopropyl isothiocyanate were identified as candidate attractive compounds to serve in a ‘push–pull’ strategy for P. xylostella control. © 2023 Society of Chemical Industry.

BACKGROUND Insect RDL (resistant to dieldrin) receptors are essential pentameric ligand-gated chloride channels that mediate the neuroinhibitory effect of GABA, the chief inhibitory neurotransmitter in the central nervous system. These receptors serve as primary targets for various insecticides, including noncompetitive antagonists (NCAs) such as cyclodiene organochlorines and phenylpyrazoles, as well as allosteric modulators like meta-diamides and isoxazolines. This study focuses on a newly discovered A2′N mutation within the RDL receptors, identified in fipronil-resistant planthoppers. Despite in vitro electrophysiological studies have proposed its role in conferring target-site resistance, in vivo genetic functional validation of this mutation remains unexplored. RESULTS Our research employed toxicity bioassays, assessing various Rdl genotypes against a spectrum of insecticides, including fipronil, α-endosulfan, broflanilide, and isocycloseram. Results revealed distinct resistance profiles for A2′N and A2′S mutants, indicating different binding interactions of RDL receptors with NCAs. Significantly, the A2′N heterozygote showed substantial resistance to fipronil, despite its homozygous lethality. Molecular modeling and docking simulations further supported these findings, highlighting unique binding poses for fipronil and α-endosulfan. CONCLUSION This study confirmed that A2′N mutation of the RDL GABA receptor confer high resistance to fipronil in vivo . The observed resistance in A2′N mutants is likely attributable to a steric hindrance mechanism, wherein the introduction of larger side chains hampers fipronil binding, even in a heterozygous state. © 2023 Society of Chemical Industry.

BACKGROUND Sclerotinia stem rot caused by Sclerotinia sclerotiorum seriously endangers oilseed rape production worldwide, and the occurrence of fungicide-resistant mutants of S. sclerotiorum leads to control decline. Thus, it is critical to explore new green substitutes with different action mechanisms and high antifungal activity. Herein, the activity and the action mechanism of natamycin against S. sclerotiorum were evaluated. RESULTS Natamycin showed potent inhibition on the mycelial growth of S. sclerotiorum , and half-maximal effective concentration (EC 50 ) values against 103 S. sclerotiorum strains ranged from 0.53 to 4.04 μg/mL (mean 1.44 μg/mL). Natamycin also exhibited high efficacy against both carbendazim- and dimethachlone-resistant strains of S. sclerotiorum on detached oilseed rape leaves. No cross-resistance was detected between natamycin and carbendazim. Natamycin markedly disrupted hyphal form, sclerotia formation, integrity of the cell membrane, and reduced the content of oxalic acid and ergosterol, whereas it increased the reactive oxygen species (ROS) and malondialdehyde content. Interestingly, exogenous addition of ergosterol could reduce the inhibition of natamycin against S. sclerotiorum . Importantly, natamycin significantly inhibited expression of the Cyp51 gene, which is contrary to results for the triazole fungicide flusilazole, indicating a different action mechanism from triazole fungicides. CONCLUSION Natamycin is a promising effective candidate for the resistance management of S. sclerotiorum . © 2023 Society of Chemical Industry.

BACKGROUND How parasitoids distinguish hosts from non-hosts remains an unknown question. Chouioia cunea Yang (Eulophidae) is an important fall webworm parasitoid that attacks many forest and agricultural pests. To study the differences in the chemical clues used by C. cunea to distinguish host and non-host plants, we used gas chromatography–mass spectrometry (GC–MS) to identify volatile compounds of two C. cunea hosts ( Hyphantria cunea and Helicoverpa armigera ) and two non-hosts ( Spodoptera exigua and Spodoptera frugiperda ). Additionally, we used behavioral assays to compare the attraction of C. cunea to various compounds. RESULTS The two natural host species were more attractive than the two non-host species, in the following order: Hyphantria cunea > Helicoverpa armigera > S. exigua = S. frugiperda . The pupae of the natural hosts contained 1-dodecene, which was not produced by the two natural non-hosts. When the ‘attractants’ based on the difference between the species-specific blend emitted by pupae and the optimal blend were sprayed onto the natural non-host pupae, they significantly improved the attraction of C. cunea to the non-host pupae. CONCLUSION These results revealed that specific host-produced volatile compounds guide C. cunea to distinguish between natural hosts and non-hosts. Overall, this study provides a foundation for developing a behavior-modifying strategy to re-direct C. cunea attacks to control important non-host pests. © 2023 Society of Chemical Industry.

BACKGROUND Rhynchophorus ferrugineus , the red palm weevil (RPW), is a key pest that attacks many economically important palm species and that has evolved a sensitive and specific olfactory system to seek palm hosts. Odorant-binding proteins (OBPs) not only play crucial roles in its olfactory perception process but are also important molecular targets for the development of new approaches for pest management. RESULTS Analysis of the tissue expression profiles of RferOBP8 and RferOBP11 revealed that these two Rhynchophorus ferrugineus odorant binding proteins (RferOBPs) exhibited high expression in the antennae and showed sexual dimorphism. We analyzed the volatiles of seven host plants by gas chromatography–mass spectrometry and screened 13 potential ligands by molecular docking. The binding affinity of two recombinant OBPs to aggregation pheromones and 13 palm odorants was tested by fluorescence competitive binding assays. The results revealed that eight tested palm volatiles and ferrugineol have high binding affinities with RferOBP8 or RferOBP11 . Behavioral trials showed that these eight odor compounds could elicit an attraction response in adult RPW. RNA interference analysis indicated that the reduction in the expression levels of the two RferOBPs led to a decrease in behavioral responses to these volatiles. CONCLUSION These results suggest that RferOBP8 and RferOBP11 are involved in mediating the responses of RPW to palm volatiles and to aggregation pheromones and may play important roles in RPW host-seeking. This study also provides a theoretical foundation for the promising application of novel molecular targets in the development of new behavioral interference strategies for RPW management in the future. © 2023 The Authors. BACKGROUND Bioherbicides are becoming more attractive as safe weed control tools towards sustainable agriculture. Natural products constitute an important source chemicals and chemical leads for discovery and development of novel pesticide target sites. Citrinin is a bioactive compound produced by fungi of the genera Penicillium and Aspergillus . However, its physiological-biochemical mechanism as a phytotoxin remains unclear. RESULTS Citrinin causes visible leaf lesions on Ageratina adenophora similar to those produced by the commercial herbicide bromoxynil. Phytotoxicity bioassay tests using 24 plant species confirmed that citrinin has a broad activity spectrum and therefore has potential as a bioherbicide. Based on chlorophyll fluorescence studies, citrinin mainly blocks PSII electron flow beyond plastoquinone Q A at the acceptor side, resulting in the inactivation of PSII reaction centers. Furthermore, molecular modeling of citrinin docking to the A. adenophora D1 protein suggests that it binds to the plastoquinone Q B site by a hydrogen bond between the O1 hydroxy oxygen atom of citrinin and the histidine 215 of the D1 protein, the same way as classical phenolic PSII herbicides do. Finally, 32 new citrinin derivatives were designed and sorted according to free energies on the basis of the molecular model of an interaction between the citrinin molecule and the D1 protein. Five of the modeled compounds had much higher ligand binding affinity within the D1 protein compared with lead compound citrinin. CONCLUSION Citrinin is a novel natural PSII inhibitor that has the potential to be developed into a bioherbicide or utilized as a lead compound for discovery of new derivatives with high herbicidal potency. © 2023 Society of Chemical Industry.
