Methicillin-resistant Staphylococcus aureus (MRSA) has emerged as a significant pathogen of global concern, presenting substantial public health risks through foodborne transmission and contributing to elevated morbidity and mortality rates worldwide. The critical need for precise and timely identification of MRSA in food matrices has become increasingly paramount for effective public health protection. While numerous CRISPR-based detection platforms for MRSA have been recently developed, their widespread implementation has been hindered by intricate multi-step protocols and dependency on sophisticated instrumentation. In this study, we developed a direct, accurate and visual detection method for MRSA — the RPA-Cas12a-LFS method. This method comprises two main components: (1) the direct one-pot RPA-Cas12a system, which integrates bacterial lysis, RPA nucleic acid amplification, and CRISPR-Cas12a nucleic acid detection into a single step performed simultaneously at a constant temperature, and (2) the streptavidin-gold nanoparticles (SA-AuNP)-based CRISPR-specific lateral flow strip (LFS). By eliminating the need for nucleic acid extraction, this method significantly simplifies the experimental procedure and reduces the risk of cross-contamination. Through systematic optimization, this method demonstrated exceptional performance, enabling direct and specific identification of MRSA at remarkably low concentrations (10 CFU/mL) within 60 min in various milk samples. This advanced detection method, characterized by its direct sample processing, exceptional accuracy, visual interpretability, cost-efficiency, and minimal equipment requirements, is particularly suitable for on-site and real-time monitoring of pathogenic bacteria in the food industry.

A novel colorimetric platform was designed for the determination of S. aureus by utilizing a dual-recognition strategy, where wheat germ agglutinin (WGA)-functionalized magnetic beads were served as separation elements to capture and enrich S. aureus efficiently from the matrix. Horseradish peroxidase (HRP) labeled chicken anti-protein A IgY (HRP-IgY) was used to label the captured S. aureus . A chicken IgY was introduced as a signal tracer to bind with staphylococcal protein A (SPA) on the surface of S. aureus , which can circumvent the interference from protein G-producing Streptococcus . Subsequently, the colorimetric signal was achieved by an HRP-catalyzed reaction, which was amplified by HRP-IgY bound by approximately 80,000 SPA molecules on one S. aureus . The entire detection process could be accomplished within 90 min. Under optimal conditions, the linear response of different S. aureus concentrations ranged from 7.8 × 10 2 to 2.0 × 10 5 CFU/mL and the limit of detection reached down to 3.9 × 10 2 CFU/mL. Some common non-target bacteria yielded negative results, indicating the excellent specificity of the method. The developed strategy was successfully applied to the determination of S. aureus in various types of samples with satisfactory recoveries. Therefore, the novel dual-recognition strategy possessed the advantages of high sensitivity, specificity, and low cost and exhibited considerable potential as a promising tool to defend public health. Methicillin-resistant Staphylococcus aureus (MRSA) represents a serious global health challenge due to its resistance to multiple antibiotics, especially β-lactams, which limits treatment options and results in high morbidity and mortality rates. Thus, there is an urgent need for effective antibacterial agents and a deeper understanding of their mechanisms of action to combat the increasingly severe threat of bacterial resistance. In this study we have performed microbiological, molecular biology experiments and molecular dynamics simulations to investigate the antimicrobial efficacy of the sanguinarine (SAN) against MRSA and its conformational modulation of the Filamentous temperature-sensitive mutant Z (FtsZ). Our findings indicate that SAN exhibits promising anti-MRSA activity and spontaneously targets the nucleoid binding domain (NBD) of SaFtsZ to inhibit bacterial division. Additionally, SAN induces multiple single-nucleotide polymorphisms (SNPs) in the SaFtsZ coding gene, including Asp187, and site-directed mutagenesis confirms the critical role of this residue in binding. Notably, SAN causes the unfolding of SaFtsZ, significantly reducing the structural flexibility required for polymerization and triggering conformational changes. At higher dimensions, SAN disrupts protofilaments assembly, ultimately inhibiting septum formation and division. These findings provide multidimensional, high-resolution insights into the interaction between SAN and SaFtsZ, offering effective strategies for the prevention and eradication of MRSA.

To enhance the antibacterial efficacy of tildipirosin against Staphylococcus aureus (S.A.) infections, optimized solid lipid nanoparticles loaded with tildipirosin (SLN-TD) were developed, using docosanoic acid (DA), octadecanoic acid (OA), hexadecanoic acid (HA), and tetradecanoic acid (TA) as lipid components. The efficacy of these nanoparticles against S.A. was evaluated using orthogonal design analysis. FTIR, DLS, HPLC, and TEM analyses confirmed that tildipirosin was successfully incorporated into the solid lipid nanoparticles, resulting in an optimal nanoparticle drug delivery system with a particle size of 322.63 ± 1.51 nm, a zeta potential of 37.83 ± 0.95 mV, an encapsulation efficiency of 82.23 ± 0.45%, and a drug loading capacity of 7.36 ± 0.18%. The SLN-TD system exhibited high stability, effective sustained release in vitro, and enhanced intracellular activity against S.A. Pharmacokinetic studies in rats administered 4 mg kg−1via intramuscular and oral routes showed that, compared to unencapsulated tildipirosin (TD), SLN-TD provided sustained release in vivo and improved gastrointestinal absorption with higher bioavailability. Additionally, in a mouse model of S.A. infection, SLN-TD demonstrated superior antibacterial activity and sustained drug delivery for effective treatment. This study offers a promising multifunctional nanoparticle drug delivery system for the effective treatment of S.A. infections and enhances the oral bioavailability of tildipirosin, with potential applications in veterinary medicine.

The emergence of antibiotic resistance represents a significant and growing threat to global public health. The development of novel and antibiotic-free antibacterial strategies is therefore highly importance. In this context, chemodynamic therapy (CDT), which is based on Fenton or Fenton-like chemistry, is a promising strategy. CDT utilizes Fe 2+ to convert H 2 O 2 into highly reactive hydroxyl radicals (•OH), which can oxidize biomolecules to achieve antibacterial effects. The antibacterial efficacy of CDT is contingent upon both the catalyst and substrate. We developed a straightforward, convenient and economical approach to fabricate Fe-caffeic acid metal–polyphenol networks (Fe-CA MPNs) by mixing CA and Fe 3+ . The Fe-CA MPNs are acid-sensitive and self-enhanced CDT agents with remarkable antibacterial efficacy against both Staphylococcus aureus ( S. aureus ) and methicillin-resistant S. aureus (MRSA). Antibacterial efficacy was evaluated via a variety of methods, including the zone of inhibition, spread plate, turbidimetric, and live/dead bacterial staining tests. Specifically, the inhibition rates of Fe-CA MPNs for S. aureus and MRSA were 96.33 ± 0.33 % and 96.33 ± 0.15 %, respectively. CaO 2 @Fe-CA nanocomposites with H 2 O 2 self-supply properties were synthesized by depositing Fe-CA MPNs on the surface of CaO 2 nanoparticles which function as a sustained-release source of H 2 O 2 . The antibacterial effect of the CaO 2 @Fe-CA nanocomposites was comparable to that of Fe-CA MPNs combined with H 2 O 2 . Both Fe-CA MPNs and CaO 2 @Fe-CA possess favorable in vitro biosafety profiles. This work will facilitate the development of innovative antibiotic-free CDT antibacterial strategies based on •OH.

In this work, a dual recognized CRISPR/Cas12a system has been proposed, in which the activation chain is cleverly divided into two parts that can serve for precise dual target recognition, and hydrazone chemistry is introduced for the formation of a whole activation chain. It has been further explored to construct a new method for the specific and sensitive detection of Staphylococcus aureus (SA) as one of the most common pathogens in infectious diseases. In virtue of proximity effect contributed by complementary base pairing, hydrazone chemistry accelerates the formation of the whole activation strand and improves the specificity of the CRISPR/Cas12a system, serving for the accurate analysis of SA. Moreover, the temporary aggregation of CRISPR/Cas12a around SA enhances its catalytical efficiency so as to further amplify signal. With high sensitivity, stability, reproducibility and specificity, the established method has been successfully applied to detect SA in complex substrates. Meanwhile, our established method can well evaluate the inhibition effect of chlorogenic acid and congo red in comparison with flow cytometry. Environmental implication Bacterial pathogens exist widely in the environment and seriously threaten the safety of human health. Staphylococcus aureus (SA) is the most common pathogen of human suppurative infection, which can cause local suppurative infection, pneumonia, and even systemic infections such as sepsis. In this work, a dual recognized CRISPR/Cas12a system mediated by hydrazone chemistry has been proposed. With high sensitivity and low detection limit, the established method can specifically detect SA and effectively evaluate the antibacterial effect of inhibitors. This method is expected to be further developed into a detection method in different scenarios such as environmental monitoring and clinical diagnosis.

Aims This study aims to investigate how different wound microenvironmental factors (temperature, pH, and osmotic pressure) influence the autofluorescence of Staphylococcus aureus (S. aureus) and its underlying molecular mechanisms, specifically focusing on the porphobilinogen synthase gene (hemB) gene expression. Methods We measured the average fluorescence intensity of S. aureus colonies under varying conditions of pH (3, 5, 7, 9, 11), temperature (25°C, 31°C, 37°C, 43°C), and osmotic pressure (0.9%, 1.8%, 2.7%, 3.6%) over time. Fluorescence intensity was quantified using ImageJ software. Additionally, RT-qPCR was used to analyze the expression levels of the hemB under these conditions. Results Bacterial fluorescence intensity increased as the temperature ranged from 25°C to 43°C, with corresponding upregulation of hemB expression. At pH values between 3 and 11, fluorescence intensity decreased as pH increased, reflecting a similar trend in hemB expression. Fluorescence also diminished with higher osmotic pressures (0.9% to 3.6%), mirroring the downregulation of hemB.Conclusions Our findings indicate that temperature, pH, and osmotic pressure significantly affect the autofluorescence of S. aureus by modulating porphyrin accumulation through hemB gene expression. These environmental factors should be considered when using bacterial fluorescence for wound infection assessment.

Osteomyelitis caused by Staphylococcus aureus ( S. aureus ) is difficult to cure with antibiotics or phototherapy. Microwave (MW) has become a promising method for treating deep tissue infections due to its strong penetration ability, however, the effects of MW dynamics still need to be improved to achieve rapid and effective treatment. In this work, tin selenide/polypyrrole (SnSe/PPy) nanocomposites with MW thermoelectric catalytic performance are successfully prepared, and their ideal thermal production capability stems from increased dielectric loss, including interfacial polarization and conductive loss, as well as magnetic loss. Furthermore, the enhanced interchain electronic transport of PPy and the phonon scattering effect have improved the thermoelectric performance of SnSe/PPy. Under MW cyclic irradiation, SnSe/PPy can convert the generated temperature difference into electrical energy and further promote the ionization of sodium species, the plasma and electrons react with O 2 to produce superoxide anion (·O 2 − ), enabling SnSe/PPy to rapidly increase the temperature and effectively eliminate S. aureus infection. After irradiated circularly by MW for 20 min, the antibacterial effect of SnSe/PPy can reach 99.46 ± 0.11%. Considering the remarkable antibacterial effectiveness and excellent biosafety, it is believed that it provides new insights into the design of MW thermoelectric catalysis antimicrobial materials.

Although bacteriophage-based biosensors are promising tools for rapid, convenient, and sensitive detection of Staphylococcus aureus in food products, the effect of biosensors using temperate phages as biorecognition elements to detect viable S. aureus isolates remains unclear. In this study, three temperate S. aureus phages were isolated and their biological features (one-step growth, host range, pH stability, temperature stability, and adsorption rate) were evaluated as the biological element. The selected phage SapYZUs8 was immobilized on the nanozyme Cu-MOF via electrostatic interactions to generate SapYZUs8@Cu-MOF, and its detection performance in real food (skim milk and pork) was then evaluated. Compared with phages SapYZUm7 and SapYZUs16, phage SapYZUs8 exhibited a broader host range, greater pH stability (3–12), and a better absorption rate (92 %, 8 min) suitable for S. aureus detection, which is likely the result of the DNA replication (DNA helicase) and phage tail protein genes in the SapYZUs8 genome. Therefore, phage SapYZUs8 was fixed on Cu-MOF to generate SapYZUs8@Cu-MOF, which exhibited good sensitivity and specificity for rapid colourimetric detection of viable S. aureus. The method took <0.5 h, and the detection limit was 1.09 × 10 2 CFU/mL. In addition, SapYZUs8@Cu-MOF was successfully employed for the colourimetric detection of S. aureus in food samples without interference from different food additives, NaCl concentrations, or pH values. With these benefits, it allows rapid visual assessment of S. aureus levels.

Bacterial infections have always been a threat when it comes to public health accounting for increased morbidity and mortality rates around the world. For the first time, Polydopamine is often used as an ocular surface drug delivery medium to treat some ocular surface diseases based on its good tissue affinity. Mesoporous polydopamine nanospheres (MPDA NPs) under photothermal therapy (PTT) are demonstrated as efficient therapeutic nanoplatforms for Staphylococcus aureus (S. aureus) infection and wound healing. MPDA NPs were found to exhibit excellent photothermal performance, significantly causing an increase in temperature within a short period of NIR-I exposure (808 nm, 1 W cm−2, 6 min). The MPDA NPs under the NIR irradiation remarkably eliminated S. aureus in vitro. Moreover, these synergistic effects turnouts to be phenomenal in vivo, effectively killing and healing S. aureus-infected abscesses in mice. These revealed the combined effect of the intrinsic antibacterial activity of MPDA NPs enhanced upon NIR-I exposure. Hence, MPDA NPs under NIR-I could prove excellent therapeutic nanoplatforms for bacteria-related infections and other biomedical applications.

Owing to the fact that luteolin has antibacterial activity against Staphylococcus aureus (S. aureus) and methicillin-resistant S. aureus (MRSA), its specific mechanism in MRSA is worthy of investigation, which is the focus of this study. Initially, the collected S. aureus strains were treated with luteolin. Then, the minimum inhibitory concentration (MIC) of luteolin against the S. aureus strains was measured by the broth microdilution. The growth curves, biofilm formation, and cytotoxicity of treated S. aureus were detected using a microplate reader. The live and dead bacteria were evaluated using confocal laser scanning microscopy, the bacterial morphology was observed using scanning electron microscopy, and the S. aureus colony-forming unit (CFU) numbers were assessed. The levels of alpha hemolysin (α-hemolysin), delta hemolysin (δ-hemolysin), and hlaA were detected via western blot and RT-PCR. The mortality of mouse model with S. aureus systemic infection was analyzed, and the levels of IL-6, IL-8, IL-10, and TNF-α were quantitated using ELISA. Concretely, the MIC of luteolin against MRSA N315 was 64 μg/mL. Luteolin at 16 μg/mL did not affect the growth of MRSA N315, but inhibited the biofilm formation and CFU, and promoted the morphological changes and death of MRSA N315. Luteolin decreased the cytotoxicity and the levels of α-hemolysin, δ-hemolysin, and hlaA in MRSA N315, elevated MRSA-reduced mice survival rate, and differentially modulated the inflammatory cytokine levels in MRSA-infected mice. Collectively, luteolin inhibits biofilm formation and cytotoxicity of MRSA via blocking the bacterial toxin synthesis.

Antimicrobial resistance has become a pivotal global public health concern, significantly moderating the effectiveness of antibiotic therapies. Metallic hydrogels, as a highly representative nanozyme, have shown promise in combating drug-resistant bacterial infections. Herein, we develop an innovative AuCu@Pd hydrogel nanozyme utilizing a facile water/ethanol-phase method, achieving gelation within 40 min. The building blocks of AuCu@Pd hydrogels exhibit dendritic and core–shell structures, facilitated by the regulation of nanowire-like morphologies through galvanic replacement and ascorbic acid induction effects. The AuCu@Pd hydrogels demonstrate peroxidase-like (POD-like) and oxidase-like (OD-like) activities, facilitating the conversion of H2O2 and O2 into highly cytotoxic reactive oxygen species (ROS) to disrupt bacterial cell structures. Moreover, the AuCu@Pd hydrogels modulate the infectious microenvironment by depleting glutathione (GSH), thereby promoting the production of reactive oxygen species (ROS) and enhancing their bactericidal effects against methicillin-resistant Staphylococcus aureus (MRSA) and Escherichia coli (E. coli). Notably, AuCu@Pd hydrogels demonstrate a high photothermal conversion efficiency (η) of 61.1% and superior photothermal stability, significantly boosting the multienzyme-like activity for efficient antibacterial therapy. In addition, these hydrogels exhibit excellent biocompatibility in vivo and significantly accelerate MRSA-infected skin wound healing. This work offers novel insights into the design of highly efficient metallic hydrogels and proposes an effective strategy for the synergistic treatment of MRSA infections for biomedical applications.
