Lipid oxidation reprogramming in cancer-associated fibroblasts enhances CD8+ T cell cytotoxicity and therapeutic response

Zikun Ma,Yuzhao Wang,Weikai Wang,Wanyang Sun,Rong Wang,Xue Ding,Zibin Chen,Xiangdong Li,Zhiyong Li,Yunlin Ye,Yize Mao,Jianhua Yin,Guibo Li,Rong-Rong He,Xiaoyu Liang,Zhuowei Liu

Cancer-associated fibroblasts (CAFs) are major stromal components of the tumor microenvironment, yet how their metabolic states shift during therapy and influence anti-tumor immunity remains unclear. By integrating clinical cancer samples, single-cell RNA analyses, and functional studies, we identify a chemotherapy-conditioned PTGER3 + CAF subset characterized by enhanced lipid oxidation. This metabolic reprogramming strengthens antitumor immunity by promoting CD8 + T cell activation and cytotoxicity through the suppression of PTEN-related signaling. Clinically, higher proportions of therapy-induced PTGER3 + CAFs correlate with improved treatment responses and better patient prognosis. Together, these findings reveal a previously unrecognized stromal metabolic adaptation that supports CD8 + T cell immunity and highlight CAF-driven lipid oxidation and its regulation of CD8 + T cell PTEN signaling as potential avenues to enhance chemotherapy and immunotherapy efficacy.

Astragaloside IV mitigates T-2 toxin-induced cytotoxicity by preserving mitochondrial homeostasis and suppressing apoptosis

Peixuan Li,Zhaojie Liu,Jia Yan,Haihua Huo,Yanchen Liu,Siqun Wan,Lu Guan,Feiyang Chen,Qingmei Chen,Jikai Wen,Yiqun Deng,Peiqiang Mu

T-2 toxin, a type A trichothecene mycotoxin, induces severe cytotoxicity primarily through mitochondrial dysfunction and apoptosis. In this study, the protective role of astragaloside IV (AS-IV) against T-2 toxin-induced damage was investigated using the human hepatoma cell line HepG2 as an in vitro model. Our results demonstrated that T-2 toxin significantly reduced cell viability, promoted reactive oxygen species (ROS) accumulation, and impaired mitochondrial function, as evidenced by disrupted respiratory activity, loss of mitochondrial membrane potential, and reduced ATP production. These alterations were accompanied by dysregulated mitochondrial homeostasis, characterized by enhanced mitochondrial biogenesis and excessive mitochondrial fission, along with activation of the mitochondrial apoptotic pathway. Pretreatment with AS-IV mitigated ROS accumulation, rescued impaired mitochondrial respiratory function, stabilized mitochondrial membrane potential, maintained cellular ATP level, and restored mitochondrial homeostasis. Furthermore, AS-IV suppressed T-2 toxin-triggered apoptosis by modulating the BAX/BCL-2 ratio, inhibiting cytochrome c release, and attenuating caspase activation. Together, these findings indicate that AS-IV protects HepG2 cells from T-2 toxin-induced cytotoxicity by maintaining mitochondrial homeostasis and suppressing the intrinsic apoptotic pathway, providing mechanistic evidence for its protective effect.

Non-Engineered Physiologically Modulated Nanovesicles Augment Antitumor Immune Responses via Dual-Pathway T Cell Activation

Xiaoyuan Fan,Fengxiang Liu,Qing You,Yiyang Wang,Lili Du,Xinpeng Zou,Fei Sun,Lianwen Qi,Zhonggui He,Jin Sun,Shujun Wang,Liang Sang,Kaiyuan Wang

The therapeutic efficacy of cancer vaccines is critically contingent upon CD8 + T cell-mediated antitumor immunity, but the effectiveness is hindered due to insufficient T cell activation. Inspired by the close interplay between cellular physiological states and immunoregulatory functions, we propose ASCENT, a nanovaccine platform integrating nanovesicles derived from activated platelets and senescent tumor cells to naturally co-present antigens and co-stimulatory molecules, thereby enabling dual-pathway T cell activation. Specifically, senescent tumor cells promote antigen presentation via upregulated major histocompatibility complex-I (MHC-I) molecules, whereas activated platelets increase the surface expression of CD40L and OX40L, thereby enhancing immune activation. Thus, without genetic or chemical engineering, ASCENT can effectively stimulate CD8 + T cells through both antigen self-presentation and dendritic cell-mediated antigen presentation, ultimately eliciting robust immune responses. This non-engineered, physiologically modulated nanovaccine exhibits broad-spectrum antitumor activity while simultaneously inducing durable systemic immune memory to effectively suppress tumor recurrence and metastasis. Overall, the ASCENT nanovaccine provides new perspectives on rational vaccine design, emphasizing the importance of engaging both direct and indirect T cell activation in reinforcing cancer immunotherapy.

Continuous and selective extraction of Cs+ using a t-BAMBP/P204/MWCNTs supported liquid membrane

Zhen Zeng,Fengfeng Gao,Yu Zhao,Xiaohan Xing,Can Wang,Xiao Du,Xiaogang Hao,Xiumin Li

A novel 4-tert-butyl-2-(α-methylbenzyl) phenol/bis(2-ethylhexyl) phosphate/ Multi-Walled Carbon Nanotubes (t-BAMBP/P204/MWCNTs) supported liquid membrane (SLM) was prepared via an in-situ impregnation method. The MWCNTs network effectively anchored the t-BAMBP/P204 synergistic extractants, enhancing membrane stability. Through proton exchange and coordination interactions, this membrane efficiently extracts Cs + in weakly alkaline environments. A systematic investigation was conducted into the effects of MWCNTs, t-BAMBP and P204 ratios, diluent ratio (kerosene: cyclohexane), pH value, and temperature on Cs + extraction performance. In this system, at equal concentrations (C Cs+, Rb+  = 200 ppm), the Cs + permeation flux reached 25.2 mmol/m 2 /h, and the Cs + /Rb + separation coefficient was 8.90. In selective permeation experiments conducted in a mixed solution containing equal concentrations of Cs + , Rb + , K + , Na + , and Li + , the separation factors for Cs + /Li + , Cs + /Na + , Cs + /K + , and Cs + /Rb + were 144.6, 45.75, 18.09, and 8.93, respectively. Furthermore, the t-BAMBP/P204/MWCNs SLM was applied to the extraction and separation of cesium from simulated lithium mica lithium-extraction tail liquor, and the system demonstrated excellent selectivity. The separation factors for Cs + /Li + , Cs + /Na + , Cs + /K + , Cs + /Rb + , Cs + /Mg 2+ , and Cs + /Ca 2+ reached 380.67, 571, 76.13, 16.79, 17.57, and 3.525, respectively. Following ten permeation experiments, both the Cs + permeation flux and the separation factors of Cs + /Rb + maintained high cycle stability. Through slope analysis and electrospray ionization mass spectrometry, it was confirmed that Cs + coordinates with t-BAMBP and P204 to form compounds with the chemical formulae [(ROCs)·(ROH)], [(ACs)·(HA)], and [CsOR·ROH(HA)]. Additionally, Cs + was found to form complexes with the two extractants individually. DFT calculations indicated that the reaction process was predominantly governed by the formation of complexes between t-BAMBP dimers and Cs + . This study presents a promising new method for Cs + recovery based on membrane separation processes.

Erythrocyte membrane–liposome coating sustains circulation stability and targeted tumor therapy of CAR-T cells

Zhiying Chen,Muya Zhou,Xiaoyu Li,Yuqi Zhao,Zhengling Wang,Jinhua Hu,Jialu Hong,Shenrui Zhou,Feiyan Pan,Zhigang Hu,Lingfeng He,Tao Gao,Xiaodong Jiang,Zhiqiang Wang,Zhigang Guo

Background CAR-T therapy is limited by off-target sequestration in liver, spleen, and lungs, which reduces tumor delivery and risks systemic toxicity. Genetic engineering approaches are complex and carry safety concerns, necessitating efficient non-genetic strategies to optimize CAR-T biodistribution. Purpose We developed red blood cell membrane–chimeric liposomes (Rlip) as a biomimetic coating system to non-genetically modify CAR-T cells, aiming to enhance immune evasion, prolong circulatory persistence, and redirect trafficking toward tumors while preserving intrinsic effector function. Methods Rlip-CAR-T cells were prepared via simple incubation. In vitro characterization assessed coating stability, CD47-mediated macrophage resistance, phenotype preservation, and antigen-specific cytotoxicity. In vivo studies evaluated biodistribution, tumor infiltration, and efficacy in Nalm-6 systemic leukemia and 1806-luc subcutaneous ovarian cancer models. Results Rlip coating stably presented CD47, functionally impairing macrophage phagocytosis and enhancing peripheral persistence without altering memory subsets, activation markers, or cytotoxicity. Rlip-CAR-T cells demonstrated markedly reduced off-target organ accumulation and increased intratumoral enrichment in both models, with superior tumor control and prolonged survival observed in the leukemia model. Conclusions Rlip surface engineering represents a universal, clinically translatable strategy to overcome CAR-T biodistribution barriers. By integrating erythrocyte-mimetic immune evasion with optimized tumor delivery, this approach improves CAR-T cell accumulation at tumor sites across hematologic and solid malignancies.

Daphnetin prevents acute graft-versus-host disease by suppressing donor T cell expansion and function

Wei Zhi-Feng,Chen Yan-Fang,Song Fei,Fu Yu,Zhang Yun-Wei,Qin Tian-Xue,Gao Su-Jun,Su Long

Background Acute graft-versus-host disease (aGVHD), which is primarily driven by donor alloreactive T cells, is a major cause of morbidity and mortality following allogeneic hematopoietic cell transplantation (allo-HCT). Daphnetin (DAPH) shows potent immunosuppressive activity by inhibiting T cell proliferation and function. However, its efficacy in preventing aGVHD remains unexplored. This study aimed to investigate the prophylactic effects of DAPH against aGVHD in murine allo-HCT models and to characterize the underlying mechanisms. Materials and methods Multiple murine allo-HCT models were established to evaluate the effects of DAPH on aGVHD progression, the graft-versus-leukemia (GVL) effect, and donor immune/hematopoietic cell recovery. Flow cytometry was employed to determine cell proliferation, immune cell alterations, apoptosis profiles, and serum cytokine levels. RNA sequencing and quantitative proteomics analyses were conducted to explore the underlying molecular mechanisms. Results DAPH significantly mitigated murine aGVHD following allo-HCT, showing efficacy comparable to that of cyclophosphamide. Notably, DAPH administration did not compromise the GVL effect while transiently delaying the recovery of donor hematopoietic cells. Mechanistically, DAPH inhibited the expansion of multiple donor immune cells, preferentially suppressing the proliferation and functioning of donor conventional T cells. The potential molecular mechanisms underlying the prophylactic efficacy of DAPH against aGVHD after allo-HCT included suppression of the T cell receptor and Janus kinase 3 (JAK3)/signal transducer and activator of transcription 3 (STAT3) signaling pathways. Conclusion These findings indicate that DAPH effectively mitigates aGVHD while maintaining the GVL effect and showing minimal influence on donor immune/hematopoietic cell reconstitution. Thus, DAPH is a promising candidate for aGVHD prophylaxis.

Biomimetic photothermal fabric from upcycled T/S waste for solar steam and thermoelectric co-generation

Bitao Zeng,Tao Xu,Yulong Ying,Lvlv Ji,Wei Li,Yuan Wei,Tao Wang,Sheng Wang

Freshwater scarcity and the high energy demand of conventional desalination technologies call for innovative and sustainable water purification strategies. Herein, we report a multifunctional, plant-transpiration-inspired photothermal fabric evaporator engineered from upcycled polyester/spandex (T/S) textile waste. Through a two-step hydrothermal–spray-coating strategy, a hierarchical asymmetric architecture is constructed: cobalt nanoneedles on the bottom layer act as root-like channels for rapid capillary pumping, the porous T/S matrix mediates intermediate water transport, and multi-walled carbon nanotubes (MWCNTs) decorated urchin-like microspheres on the top layer emulate leaf stomata for broadband photothermal harvesting and efficient interfacial evaporation. The optimized composite (CANT/S-30) delivers a high evaporation rate of 2.12 kg m −2  h −1 under one sun (82.1 % efficiency), while simultaneously generating a thermoelectric power density of 0.823 mW m −2 . In addition, the release of Co 2+ ions endowed the fabric with intrinsic antibacterial activity with >99 % bactericidal efficiency. The evaporator effectively desalinates seawater and removes organic dyes, and operates stably under outdoor solar conditions. By valorizing ubiquitous T/S waste into high-performance photothermal fabrics, this work establishes a sustainable pathway toward scalable solar desalination and distributed clean energy generation, bridging environmental remediation and circular-economy-driven textile upcycling.

Genetically engineered T cell membrane-camouflaged nanoparticles triggered cuproptosis for synergistic bladder cancer photothermal-immunotherapy

Deng Wen,Chen Yuan,Bai Yongke,Shang Haojie,Wu Jian,Zhong Zichen,Ba Xiaozhuo,Tong Yonghua,He Yu,Jiang Kehua,Tang Kun

Immunotherapy has become a promising and transformative approach for treating advanced or treatment-resistant bladder cancer (BCa). However, its efficacy remains limited due to the immunosuppressive tumor microenvironment (TME) and insufficient immune cell infiltration. Photothermal therapy (PTT), which could cause immunogenic cell death (ICD) in tumor tissue, has been explored as a synergistic approach for bladder cancer immunotherapy. Yet, thermal resistance in cancer cells often undermines the effectiveness of PTT. To address these challenges, we proposed a novel strategy that combines PTT with cuproptosis, a recently identified form of ICD, by engineering Tim-3-overexpressing T cell membrane-coated nanoparticles (Tim3@PHSM@IC) to enhance BCa immunotherapy. The overexpression of Tim-3 on the T cell membrane enabled precise targeting of tumor cells and competitively inhibited the Tim-3 receptor on T cells through recognition of Galectin-9. In vitro, Tim3@PHSM@IC nanoparticles effectively induced photothermal cytotoxicity and robust cuproptosis. In vivo, these nanoparticles significantly inhibited tumor growth in multiple BCa mouse models. Flow cytometry (FCM) and RNA sequencing (RNA-seq) analyses revealed that Tim3@PHSM@IC nanoparticles reprogrammed the TME by activating immune-related genes and enhancing ICD This study highlights the potential of Tim3@PHSM@IC nanoparticles in overcoming the immunosuppressive TME and improving the efficacy of BCa immunotherapy by integrating PTT and cuproptosis.

Anchoring metal cobalt species on defective alumina for enhanced catalytic performance in F-T synthesis

Qiuyao Jiao,Yan Liu,Ran Yu,Lei Li

For supported heterogeneous catalyst, metal-support interaction (MSI) is very important factors for catalytic performance. Commercial γ-alumina was reduced with hydrogen in advance, providing strong anchoring sites for metal (metal oxide) species. This research demonstrates the use of pre-reduced alumina as catalyst carrier to anchor metal Co species in FTS compared with unreduced one. The defective alumina supported catalyst displayed a supeior performance. The Co/RAl2O3-550 catalyst showed surprisingly high CO conversion of 56.0%, C5+ hydrocarbons selectivity of 93.2%, and a low methane selectivity of 2.7% at a remarkable high activity (CTY = 1.1 × 10-5 molCO gCo-1 s−1). It revealed the strong reduction of alumina generated more defective sites, which was strongly anchored with metal cobalt species under MSI. Besides, the reduction of alumina also boosted the number of surface L-acidity, promoting the CO absorption rate through CO dissociation. These results reveal the unique property of defective Al2O3 supported cobalt catalysts, and provide a practical strategy to explore stable and efficient FTS catalysts.

A FEN1-EDCR dual-amplification strategy for ultrasensitive detection of CHCHD10 c.176C>T mutation

Han Pan,Murilege Chao,Miao He,Jinhua Chen,Feifan Yin,Yongliang Cai,Wenting Cheng,Zhongyun Wang,Yang Xiang

A single-base mutation in the CHCHD10 gene can impair thermogenic responses, and inhibit the efficient conversion of white adipose tissue to beige adipose tissue. However, achieving high-fidelity detection of such low-abundance single nucleotide polymorphisms (SNPs) within complex biological matrices remains a critical technological challenge. This study developed a novel method for detecting a single-base mutation in CHCHD1 0 by integrating the specific recognition of flap endonuclease 1 (FEN1) with the cascade amplification of an entropy-driven circuit reaction (EDCR), supplemented with magnetic separation to suppress background interference. In this process, the mutant target (MT) hybridizes with the upstream probe (UP) and a hairpin downstream probe (DP) to form a specific three-base overlapping structure. FEN1 recognizes and cleaves this structure, releasing a cleavage sequence (CS). Following magnetic purification, the CS activates the EDCR, enabling dual-signal amplification. The hairpin DP design effectively conceals the CS, significantly reducing background noise. The method achieves a detection limit of 0.23 pM, discriminates mutation abundances as low as 0.01% in mixed DNA samples, and demonstrates robust performance in complex matrices such as human serum and high-background genomic DNA. This work provides an innovative and reliable methodological tool for the detection of low-abundance point mutations in complex samples.

Ferroelastic domain switching toughening in spark plasma sintered t’-yttria stabilized zirconia/La2Zr2O7 composite ceramics

Yanfei Wang,Huan Yang,Rongjun Liu,Yingbin Cao,Siqing Wang

The poor fracture toughness of La 2 Zr 2 O 7 hinders its practical application as a thermal barrier coating topcoat material. To toughen La 2 Zr 2 O 7 , metastable tetragonal 8   wt% yttria-stabilized zirconia ( t ’-8YSZ), has been introduced into the La 2 Zr 2 O 7 matrix by plasma spray quenching of 8YSZ powders and spark plasma sintering . An evident fracture toughness improvement has been achieved due to ferroelastic domains switching. The resulting toughness increase has a quadratic relationship with t ’ phase volume fraction below a critical value (30   vol%), beyond which it is far smaller to what expected owing to the residual stress being smaller than the ferroelastic coercive stress.

Excellent Photocatalytic Efficiency of t-ZrO2/g-C3N4 Photocatalyst for Pollutants Degradation: Experiment and theory

Muyan Zhou,Yong Wu,Lingyi Shi,Shiyu Hu,Huanhuan Li,Yinyan Gong,Lengyuan Niu,Xinjuan Liu,Can Li

The large work function difference between two semiconductors will effectively adjust the interfacial electronic distribution and catalytic performances of heterojunction . In this work, a novel t -ZrO 2 /g-C 3 N 4 composite photocatalyst was prepared by integrating t -ZrO 2 nanoparticle with graphitic carbon nitride (g-C 3 N 4 ) film considering the large work function of tetragonal ZrO 2 ( t -ZrO 2 ). With the aid of experimental measurements and density functional theory calculations, the electronic distributions of g-C 3 N 4 film were explicitly adjusted by the supporting t -ZrO 2 nanoparticle on account of the large interfacial internal electric field , which leaded to the improvement of photogenerated carriers mobility and lifetime, the enhancement of reduction ability and the increase of oxidation active sites. Results showed that the photocatalytic activity of the composite sample was increased by 9.4 and 2.9 times compared with g-C 3 N 4 sample for Rhodamine B and tetracycline hydrochloride degradation under visible-light. This modification strategy shall be applied to devise other high-efficient catalysts by selecting semiconductor with suitable work function.
