Constipation, a gastrointestinal function disorder, is one of the side effects of paclitaxel (PTX) chemotherapy. Shenhuang plaster (SHP) application on the Shenque acupoint promotes gut motility in clinical settings. In this study, we elucidated the ingredients in SHP and evaluated its effects on PTX-induced constipation using a tumour-bearing mouse model. SHP was prepared using the traditional Chinese plaster preparation method. The ingredients were analysed using UPLC-MS/MS and identified via screening in a standard drug database. The gastrointestinal transit was evaluated by the movement of a fluorescein-labelled dextran in the gastrointestinal tract. A histological study of the mucosa was carried out after haematoxylin and eosin staining. mRNA expression was assessed using real-time RT-PCR, and the foetal microbiota composition was elucidated through 16 s rDNA sequencing and BLAST analysis. Our results indicate that the application of SHP attenuated weight gain inhibition by PTX; however, no inhibitory effect was observed on tumour growth. PTX-induced sluggish intestine, villus, and mucosal base layer damage were significantly improved following the application of SHP. Further, SHP enhanced the stimulation efficiency of PTX on TLR4 and its downstream cytokines, as well as on IL-1β in intestinal cells. SHP combined with PTX reshaped the microbiota, which showed beneficial effects on health. Hence, these results provide evidence that SHP alleviates PTX-induced constipation and intestinal morphological damage but augments the effects of PTX on the expression of cytokines in the TLR4 pathway and IL-1β. Therefore, we propose that SHP stimulates the host immune response to eradicate cancer cells.

Pomegranate peel (PoP) is a valuable by-product rich in phenolic compounds with strong antioxidant potential. This study applied mechanochemical-assisted extraction (MCAE) to recover phenolics efficiently from PoP using eight solid reagents. The extraction parameters were optimized through response surface methodology based on a central composite design. Ultra-performance liquid chromatography coupled with triple time-of-flight mass spectrometry (UPLC-Triple-TOF/MS) tentatively identified twelve major phytochemicals. These phytochemicals include hydrolysable tannins, flavonoids, and phenolic acids. The optimal MCAE conditions were a liquid–solid ratio of 50 mL/g, an extraction temperature of 78 °C, and an extraction time of 67 min. Under these conditions, a total of 12 phytochemicals were identified and the total phenolic content reached 258.09 ± 1.35 mg GAE/g dry weight. The extract exhibited high antioxidant capacity (DPPH of 526.54 ± 13.38 μmol TE/g DW, ABTS of 1315.65 ± 15.16 μmol TE/g DW, FRAP of 798.57 ± 11.30 μmol TE/g DW, and RA of 1353.87 ± 23.43 μmol TE/g DW) and strong inhibitory activity against α-glucosidase and α-amylase. These results demonstrate that MCAE represents an eco-friendly and cost-effective technique for recovering bioactive phenolic compounds from PoP, offering promising applications as natural antioxidants and enzyme inhibitors in functional food formulations, providing new opportunities for the food industries. These findings provide a new strategy for valorizing agricultural by-products into functional ingredients for food applications.

Present studies have shown that Flos Chrysanthemi has anti-inflammatory and other effects and regulates intestinal function, while the chrysanthemum stem and leaf as non-medicinal parts of chrysanthemum have similar chemical components with chrysanthemum, but the activity and mechanisms are rarely elucidated. Therefore, this study used a DSS-induced zebrafish inflammatory bowel disease model to study the anti-inflammatory and antioxidant effects of chrysanthemum stem and leaf extracts. The results indicate that DSS induction leads to increased secretion of acidic mucin in the intestines of juvenile fish, enlargement of the intestinal lumen and the emergence of intestinal inflammation. Compared with the model group, each administration group differentially inhibited the expression of IL-1β, IL-8 and MMP9 in DSS-induced zebrafish, while upregulating the activity of superoxide dismutase. The quantitative analysis results showed that the flavonoids (including Linarin, Diosmetin-7-glucoside, Tilianin, etc.) and phenolic acids (including Isochlorogenic acid C, Isochlorogenic acid A, 1,3-Dicaffeoylquinic acid, etc.) in the alcohol extract were closely related with both anti-inflammatory and antioxidant activity, while the polysaccharides were also shown a certain anti-inflammatory and antioxidant activity. In conclusion, this study suggests that the flavonoids, phenolic acids and polysaccharides from chrysanthemum stem and leaf extracts can improve inflammatory bowel disease of zebrafish by regulating the expressions of IL-1β, IL-8 and MMP9.Keywords:Flos Chrysanthemi;chrysanthemum stem and leaf;inflammatory bowel disease (IBD);zebrafish;UPLC-TQ/MS

Background Honey-fried Licorice (HFL) is a dosage form of Glycyrrhizae Radix et Rhizome processed with honey, which has been recorded to exhibit better efficacy in tonifying the spleen compared to the raw product. In contrast, different processing methods of Glycyrrhizae Radix et Rhizome exhibit different efficacies and applications, but their current quality control index components remain consistent. Purpose Based on the discovery and research strategy of traditional Chinese medicine decoction piece quality marker (Q-marker), this study aimed to conduct a multidimensional integration of constituents absorbed into the body and metabolomics based on the tonifying spleen and stomach effects of HFL to effectively identify the Q-marker of HFL. Methods In this study, a spleen deficiency rat model was established using the "exhausted swimming + poor diet" method to investigate the pharmacodynamics of tonifying the spleen and stomach by HFL. The constituents absorbed into blood was conducted using UPLC-Q-TOF/MS, correlation analysis between metabolomics and constituents absorbed into blood recognized the Q-Marker of HFL. Results The pharmacodynamic data demonstrated that HFL exhibited a significant regulatory effect on the disordered levels of PP, trypsin, chymase, PL, α-Glu, MTL, GAS, VIP, IL-2, IFN-γ, and IgA in the spleen deficiency model. Furthermore, HFL was found to improve the pathological changes in the spleen and intestine in the spleen deficiency model, highlighting its significant "tonifying spleen and stomach" effect. In the serum containing HFL, a total of 17 constituents were identified as being absorbed into the blood. Among these, 11 were prototypical components, while 6 were metabolites. Metabolomics data revealed that 9 differentially expressed metabolic markers were observed. Furthermore, the analysis of endogenous metabolic markers indicated that 10 components exhibited significant correlations with these biomarkers. Conclusion The effect of "tonifying spleen and stomach" of HFL is closely related to the regulation of the material and energy metabolism pathway. The Q-Marker of HFL is glycyrrhizic acid and 18 β -glycyrrhetinic acid as the main control standards and liquiritin, isoliquiritin, liquiritin, isoliquiritin, isolicorice flavonol, licorice chalcone C and Formononetin were used as auxiliary standards.

Prunella vulgaris L. (P. vulgaris) has great application value and development prospects in improving sleep. In this study, we continued to evaluate the sleep-improvement function and mechanism of P. vulgaris from both chemical characterization and function based on sleep-improvement functional ingredients, rosmarinic acid and salviaflaside, screened out in the previous stage as the index components. The chemical constituents of P. vulgaris and its phenolic acid fraction were characterized by the UPLC-MS n technology. The quality of the sleep-improvement phenolic acid fraction of P. vulgaris was scientifically evaluated by fingerprints combined with quantitative analysis of rosmarinic acid and salviaflaside. The function of phenolic acid parts of P. vulgaris in improving sleep was verified by different insomnia models including the PCPA-induced insomnia model and surface platform sleep deprivation model. HE staining was used to observe the effect of P. vulgaris on the morphology of nerve cells in different brain regions. In vivo experiments and molecular docking explored the sedative-hypnotic effects of functional ingredients of P. vulgaris . All these results investigated the material basis and mechanism of P. vulgaris to improve sleep from multiple perspectives, which contribute to providing a basis for the development of functional food to improve sleep.

This study aimed to synthesize a novel magnetic covalent organic framework (COF)-based material and establish an efficient magnetic solid-phase extraction (MSPE) method for the determination of benzimidazoles (BZDs) in honey. Given the abundant nitrogen atoms in BZDs, the target material (Fe₃O₄@SiO₂@MPA-TAPT-COF) was designed with Fe₃O₄ as the magnetic core and a COF shell polymerized from 5-bromobenzene-1,3-dicarbaldehyde (MPA) and 4,4',4''-(1,3,5-triazine-2,4,6-triyl) trianiline (TAPT). The synthesized Fe₃O₄@SiO₂@MPA-TAPT-COF exhibited considerable specific surface area, good magnetic responsiveness, and excellent reusability. An MSPE method using Fe₃O₄@SiO₂@MPA-TAPT-COF as the adsorbent was developed for the purification of BZDs in honey, followed by quantification via ultra-performance liquid chromatography–tandem mass spectrometry (UPLC-MS/MS). This method featured simple operation, high sensitivity, and good efficiency. Under optimized MSPE conditions, It showed good linearity for 17 BZDs in the concentration range of 0.2–100 μg/L, with correlation coefficients (R²) ≥ 0.9991. The limits of detection (LODs) for BZDs in honey were as low as 0.009–0.342 μg/kg. For spiked honey samples, the recoveries of the 17 target BZDs ranged from 61.44 % to 118.90 %.These results confirm that Fe₃O₄@SiO₂@MPA-TAPT-COF is a promising adsorbent for sample pretreatment, and the developed method provides a reliable tool for the sensitive and efficient analysis of BZDs in honey.

Ultra high performance liquid chromatography with UV detection was developed and validated for the simultaneous determination of residues of toltrazuril and its two metabolites, namely, toltrazuril sulphone and toltrazuril sulphoxide, in edible tissues (chicken and porcine muscle, liver and kidney). Acetonitrile was used to extract analytes from tissues, which were then cleaned using primary secondary amine and Oasis™ MAX solid phase extraction cartridges. Chromatographic separation was performed on a C 18 column with gradient elution. The analytes were determined using a UV detector. The regression coefficients of matrix-matched calibration curves showed linearity of >0.99. Calibration ranged from 25 to 1,000 μg kg −1 for toltrazuril and toltrazuril sulphone, and 37.5–1,500 μg kg −1 for toltrazuril sulphoxide. The accuracy was between 80 and 110 %, and the inter and intraday RSDs were lower than 15.2 and 18.3 %, respectively. The limits of detection for toltrazuril, toltrazuril sulphone and toltrazuril sulphoxide were between 10 and 37.5 μg kg −1 in all the tissues. The developed method was successfully applied to detect toltrazuril, toltrazuril sulphone and toltrazuril sulphoxide in tissues of medicated chicken.

The structural characterization of polysaccharides requires precise determination of their monosaccharide composition, including the differentiation of D- and L-configurations. Current analytical methods can identify monosaccharide types but often fail to distinguish between enantiomeric configurations, posing a significant challenge for accurate polysaccharide structural elucidation. This study introduces a robust, accurate, and reproducible ultra-performance liquid chromatography-electrospray ionization-tandem mass spectrometry (UPLC-ESI-MS/MS) method to address this limitation. By utilizing ( S )-(−)-1-phenylethylamine (S-PEA) as a chiral derivatizing reagent, monosaccharide enantiomers are converted into diastereomers through the introduction of a new chiral center. This approach enables the simultaneous determination of D/L configurations for aldoses, amino sugars, alditols, and uronic acids, achieving excellent chromatographic baseline separation. Method validation demonstrated excellent linearity (R 2 > 0.9900) within the range of 2.5–160 μg/mL. The limits of detection (LOD) and quantification (LOQ) ranged from 0.04 to 0.11 μg/mL and 0.13–0.35 μg/mL, respectively, indicating high sensitivity. Accuracy was confirmed with recovery rates between 97.23% and 100.67%, while intra- and inter-day precision RSDs were below 3.54%. The method was successfully applied to characterize the monosaccharide composition of six plant-derived polysaccharides, demonstrating its efficacy in identifying both structural types and absolute configurations. These findings establish the proposed method as a valuable tool for advancing the comprehensive structural analysis of polysaccharides.

A 25% penconazole-bupirimate microemulsion was applied three times at 7-day intervals at a rate of 300 mL/ha (equivalent to 75 g a.i. ha−1) under field conditions. Residues of penconazole, bupirimate and its metabolite ethirimol in grapes were determined using ultra-high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) following QuEChERS extraction. Mean recoveries ranged from 92.4% to 95.3% for penconazole, 91.9% to 98.5% for bupirimate, and 89.1% to 90.5% for ethirimol, with relative standard deviations (RSDs) of 1.3-2.2%, 0.95-2.0% and 0.65-3.8%, respectively. The Limits of Quantification (LOQs) for all pesticides were 0.01 mg/kg, meeting the detection requirements. The half-lives of penconazole and bupirimate in grapes were 4.6-7.5 days and 3.5-6.7 days, respectively. The dissipation of ethirimol could not be described using first-order kinetics. At 14 d post-final spray, penconazole residues ranged from < 0.01 to 0.12 mg/kg, bupirimate from < 0.01 to 0.18 mg/kg, and ethirimol from 0.015 to 0.22 mg/kg. The chronic dietary risk assessment suggested that the long-term dietary intake risk of target pesticide residues was within the acceptable threshold under the assumptions adopted in this evaluation. Notably, the RQ value of ethirimol was relatively close to the critical limit, and no national MRL for ethirimol in grape has been established in China to date.

A rapid UPLC-MS/MS method for bupivacaine TDM was developed and validated (run time 2.5 min). • The assay meets FDA/Chinese Pharmacopoeia criteria with LLOQ of 1.0 μg/L using only 50 μL plasma. • Simple protein precipitation ensures high recovery (> 90%) and negligible matrix effects. • Clozapine is introduced as a cost-effective internal standard with excellent reproducibility. • Clinical application in 40 patients revealed distinct PK profiles of conventional vs. liposomal bupivacaine. Background Bupivacaine (BPC) is a cornerstone of perioperative analgesia, yet its narrow therapeutic index and high interindividual pharmacokinetic variability complicate dosing and increase toxicity risks. A robust, high-throughput analytical platform is needed to support therapeutic drug monitoring (TDM) and individualized therapy. Methods A rapid ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method for quantifying BPC in human plasma was developed and fully validated according to FDA and Chinese Pharmacopoeia guidelines. Sample preparation consisted of a simple protein precipitation step using acetonitrile containing clozapine as the internal standard. Chromatographic separation was achieved within 2.5 min on a C18 column using gradient elution, enabling high-throughput analysis suitable for routine TDM. Results The method demonstrated excellent linearity from 1.0 to 500.0 μg/L ( R 2 > 0.995) and a short run time of 2.5 min, allowing batch analysis of up to 50 samples within 3 h. Intra- and inter-day precision (coefficient of variation) were < 15%, and accuracy ranged between 85% and 115% across quality control levels. The lower limit of quantification was 1.0 μg/L. Extraction recovery was consistent (90.9%–94.5%) and matrix effects were negligible. In a pilot clinical study involving 40 patients, the method successfully differentiated the pharmacokinetic profiles of two formulations: conventional BPC (75 mg) showed rapid distribution and elimination, while liposomal BPC (133 mg) maintained sustained plasma concentrations over 24 h. Conclusion This validated UPLC-MS/MS method is a rapid, sensitive, and high-throughput tool for BPC TDM that represents a promising candidate for implementation in clinical laboratories following appropriate local validation. Its successful clinical application highlights its utility in characterizing formulation-dependent pharmacokinetics and could assist in the development of precision dosing in perioperative analgesia to optimize efficacy and safety.

Glycosidic linkage analysis is crucial for elucidating the structure-activity relationships of polysaccharides, but existing GC-MS methods have limitations in practical applications. This study developed a simple, efficient, and sensitive UPLC-QqQ-MS/MS method for glycosidic linkage analysis, validated using glucose-based oligosaccharides and polysaccharides. 16 glucose glycosidic linkage standards were synthesized using anion reconstruction, partial methylation, and PMP derivatization. Samples underwent permethylation and PMP derivatization before UPLC-QqQ-MS/MS analysis. The accuracy and feasibility of the method were validated by comparing the results with the synthesized glycosidic linkage standards and known glycosidic linkage information. Results showed that the LC-MS-MRM spectra of the 1-4-Glc glycosidic linkages in lactose, maltose, and cellulose exhibited identical retention times and characteristic fragment ions, consistent with the synthesized 1-4-Glc standard. This confirmed the accuracy of the synthesized standards and the feasibility of the LC-MS method for detecting glycosidic linkages. Similarly, the retention times and characteristic fragment ions of the T-Glc linkages in maltose and Aspergillus Niger disaccharide, the 1-3-Glc linkage in Aspergillus Niger disaccharide, and the 1-6-Glc linkages in glucan were consistent with their corresponding standards. These results demonstrate that the UPLC-QqQ-MS/MS method can be used for qualitative and quantitative analysis of both unknown and known glycosidic linkages in oligosaccharides and polysaccharides.

Bang Jian has a long-standing history of medicinal use, noted for its detoxifying effects and the treatment of various fevers, laryngitis, and thermal illnesses. Presently, Gentiana veitchiorum and G. szechenyii are the predominant varieties utilized. However, comparative data on their chemical compositions remains scarce. In this study, employing UPLC-Q-TOF/MS metabolomics, conducted a comprehensive comparison of the chemical constituents in G. veitchiorum and G. szechenyii and assessed their antioxidant activities. Results indicated both varieties exhibit significant antioxidant capabilities, with G. szechenyii displaying superior efficacy. The IC 50 values for DPPH, Hydroxyl, and ABTS in G. szechenyii were determined to be 0.16, 0.48, and 0.80 mg/mL, respectively. Through UPLC-Q-TOF/MS, 56 compounds were identified, revealing notable differences in metabolite composition between the two species. 24 differential metabolites were identified as potential chemometric markers, facilitating the accurate differentiation of G. veitchiorum and G. szechenyii . Correlation analyses identified six compounds—depressine, szechenyin A, syringic acid, isoorientin-2′'-O-glucopyranoside, isoscoparin 2″-O-glucoside, and naringenin—as significant to the variance in antioxidant activity. These findings enhance our understanding of the phytochemical and antioxidant properties of G. veitchiorum and G. szechenyii , offering a theoretical foundation for their clinical application.
