Low aqueous dispersibility and poor stability severely restrict the bioaccessibility of curcumin in health-related products. Constructing highly effective delivery carriers is essential to overcome the limitation. In this study, a novel and simple self-assembly process driven by electrostatic interaction was developed to prepare cyclodextrin succinate/chitosan (SACD/CS) nanoparticles. Spherical SACD/CS nanoparticles were obtained by adjusting the ratio of SACD:CS to obtain an average size and ζ-potential of around 500 nm and 20 mV, respectively. By analyzing the intermolecular interactions and crystalline structure of SACD/CS nanoparticles, it is indicated that SACD could act as crosslinking nodes, and contributed to transformation of molecular arrangement in the nanoparticles. The noncytotoxic SACD/CS nanoparticles were further used as curcumin carriers and showed significant improvement on curcumin stability, with the retention index of curcumin under physiological conditions increased from 19% to 62%. Moreover, the Cur-SACD/CS nanoparticles displayed controlled release behavior of curcumin during in vitro digestion, with 9%, 22%, and 44% of curcumin released at the end of the oral, stomach, and small intestine stage, respectively. This study developed a novel and effective strategy for oral delivery of curcumin, showing promising potential for improving the bioaccessibility and bioavailability of bioactive compounds like curcumin.

Oxaliplatin (OXA) has shown high effectiveness in the treatment of cancers, but its anticancer clinical effects often induce neurotoxicity leading to neuropathic pain. Oxidative damage and NLRP3 inflammasome play important roles in neuropathic pain development. Here, neuropathic pain mouse model was constructed by continuous intraperitoneal injection of OXA. OXA administration induced mechanical pain, spontaneous pain, thermal hyperalgesia and motor disability in mice. The spinal cord tissues of OXA mice exhibited the suppressed antioxidative response, the activated NLRP3 inflammasome mediated inflammatory responses, and the increased GSK-3β activity. Next, we injected curcumin (CUR) intraperitoneally in OXA mice for seven consecutive days. CUR-treated mice showed increased mechanical pain thresholds, reduced number of spontaneous flinches, increased paw withdrawal latency, and restored latency to fall. While in the spinal cord, CUR treatment inhibited the NLRP3 inflammasome mediated inflammatory response, increased Nrf2/GPX4-mediated antioxidant responses, and decreased mitochondrial oxidative generation. Additionally, CUR combined with GSK-3β through four covalent bonds and reduced GSK-3β activity. In conclusion, our findings suggest that CUR treatment inhibits GSK-3β activation, increases Nrf2 mediated antioxidant responses, inhibits oxidative damage and inflammatory reaction, and alleviates OXA-induced neuropathic pain.

To develop reversible pH-responsive emulsifiers of natural origin, alkali lignin (AL) was used to develop oil-in-water Pickering emulsions. AL was first modified to synthesize quaternized alkali lignin (QAL), which displayed pH-responsive properties and demonstrated solubility in both acidic and alkaline solutions. In contrast, QAL exhibited insolubility and formed particles in neutral solutions, thereby making it a suitable candidate for utilization as an emulsifier in doubly pH-responsive Pickering emulsions. At pH 5–9, the emulsions were stable. Above or below this pH range, the system demulsifies, resulting in a reversible Pickering emulsifier with two pH-controlled transitions. On the basis of this pH-dependent behavior, lignin-based Pickering emulsions (LPE) could be subjected to several cycles of emulsification–demulsification by alternating the pH of the aqueous phase between basic and acidic, while the droplet size and storage stability were maintained. Curcumin was used as a drug model to study the loading/release behavior of LPE, finding that 50.08% of curcumin could be encapsulated in LPE. The in vitro release of curcumin was pH-dependent. In addition, LPE exhibited an outstanding protective effect against the ultraviolet-induced degradation of curcumin.

Considering cellular uptake promotion of lecithin and high expression of phospholipase in S. aureus, we designed curcumin (Cur)-loaded soy lecithin-based mPEG-PVL copolymer micelles (MPPC). The effect of soy lecithin on the anti-S. aureus activity of the formulation was studied with cur-loaded mPEG-PVL micelles (MPC without soy lecithin) as control. It was found that MPPC enhanced the water-solubility of Cur, and showed slow and sustained release behavior of Cur. Although MPPC had the same anti-S. aureus activity as Cur, its activity was significantly higher than MPC due to the cellular uptake promotion of soybean lecithin. It was noted that MPPC had good inhibition or destruction effect on biofilm, significant cell membrane damage, strong inhibition effect on protease or lipase production, and obvious induction effect on ROS expression when compared with Cur and MPC. So, the introduction of soy lecithin could improve the antibacterial activity of Cur. The lecithin-based micelles would offer potential to deliver antibacterial drugs for improved therapeutic action.

Smart-responsive wound dressings are promising materials for rapid wound healing and infection clearance in a painless and noninvasive method. In this work, we developed a smart wound dressing, which was composed of polyaniline (PANI) and curcumin (Cur) composite nanofiber (PANI-Cur) by electrospinning. The regulation of interfacial electrons occurred through hydrogen bonds and electrostatic interaction between Cur and PANI, resulting in enhanced light absorption ability, lower bandgap, and better separation and transfer of interfacial electrons compared with that of PANI and Cur. The process of separation and transfer of interfacial electrons would be promoted under the assistance of light based on in situ Kelvin probe force microscopy (KPFM). This PANI-Cur hybrid nanofiber showed an excellent sterilization effect against 99.77 % ± 0.10 % of Staphylococcus aureus and 98.45 % ± 0.27 % of Escherichia coli under light irradiation . The PANI-Cur nanofiber exhibited an accelerated wound healing rate when used for repairing mouse skin injuries under light. This work could provide a reference for the preparation of intelligent antibacterial fibers.

In this study, the pH-responsive API-CMCS-SA (ACS) polymeric nanoparticles (NPs) based on 1-(3-amino-propyl) imidazole (API), stearic acid (SA), and carboxymethyl chitosan (CMCS) were fabricated for the effective transport of curcumin (CUR) in liver cancer. CUR-ACS-NPs with various degrees of substitution (DS) were employed to prepare through ultrasonic dispersion method. The effect of different DS on NPs formation was discussed. The obtained CUR-ACS-NPs (DSSA=12.4%) had high encapsulation rate (more than 85%) and uniform particle size (186.2 ± 1.42 nm). The CUR-ACS-NPs showed better stability than the other groups. Drug release from the CUR-ACS-NPs was pH-dependent, and more than 90% or 65% of CUR was released in 48 h in weakly acid medium (pH 5.0 or 6.0, respectively). Additionally, the CUR-ACS-NPs increased the intracellular accumulation of CUR and demonstrated high anticancer effect on HepG2 cells compared with the other groups. CUR-ACS-NPs prolonged the retention time of the drug, and the area under the curve (AUC) increased significantly in vivo. The in vivo antitumor study further revealed that the CUR-ACS-NPs exhibited the capability of inhibiting tumor growth and lower systemic toxicity. Meanwhile, CUR, CUR-CS-NPs, and CUR-ACS-NPs could be detected in the evaluated organs, including tumor, liver, spleen, lung, heart, and kidney in distribution studies. Among them, CUR-ACS-NPs reached the maximum concentration at the tumor site, indicating the tumor-targeting properties. In short, the results suggested that CUR-ACS-NPs could act a prospective drug transport system for effective delivery of CUR in cancer treatment.

Patulin (PAT), the most common mycotoxin, is widespread in foods and beverages which poses a serious food safety issue to human health. Our previous research confirmed that exposure to PAT can lead to acute kidney injury (AKI). Curcumin is the most abundant active ingredient in turmeric rhizome with various biological activities. The aim of this study is to investigate whether curcumin can prevent the renal injury caused by PAT, and to explore potential mechanisms. In vivo , supplementation with curcumin attenuated PAT-induced ferroptosis. Mechanically, curcumin inhibited autophagy, led to the accumulation of p62 and its interaction with Keap1, promoted the nuclear translocation of nuclear factor E2 related factor 2 (Nrf2), and increased the expression of antioxidant stress factors in the process of ferroptosis. These results have also been confirmed in HKC cell experiments. Furthermore, knockdown of Nrf2 in HKC cells abrogated the protective effect of curcumin on ferroptosis. In conclusion, we confirmed that curcumin mitigated PAT-induced AKI by inhibiting ferroptosis via activation of the p62/Keap1/Nrf2 pathway. This study provides new potential targets and ideas for the prevention and treatment of PAT.

Antibiotics are crucial for treating and preventing bacterial infections. However, the widespread use of antibiotics has led to serious antibiotic resistance issues. To address the growing threat of antibiotic-resistant bacterial infections, we designed and synthesized an antibacterial composite material, CCM@ZIF-8@PDA, through a one-pot method and surface modification strategy. This material combines the antibacterial properties of the natural product curcumin, the advantages of ZIF-8, and the photothermal conversion capability of PDA. It exhibits three antibacterial mechanisms against S. aureus: chemical antibacterial, ionic antibacterial, and photothermal antibacterial effects. At a low concentration of 100 ppm, CCM@ZIF-8@PDA achieves a 100% antibacterial rate. This multi-modal antibacterial strategy provides insights for developing new antibacterial materials to combat antibiotic resistance.

The incidence of intrauterine adhesions (IUA) was raised accompanied by the increasing utilization of intrauterine surgery. The postoperative physical barrier methods commonly used, such as balloons and other fillers, have limited effectiveness and may even cause further damage to the remaining endometrial tissue. Herein, we developed an injectable thermosensitive hydrogel using Pluronic F127/F68 as pharmaceutical excipients and curcumin as a natural active molecule. The hydrogel effectively addresses solubility and low bioavailability issues associated with curcumin. In vitro, drug release assays revealed that the amorphous curcumin hydrogel promotes dissolution and sustained release of curcumin. In vitro experiments reveal high biocompatibility of the hydrogel and its ability to enhance vascular formation while inhibiting the expression of fibrotic factor TGF-β1. To assess the effectiveness of preventing intrauterine adhesions, in vivo experiments were conducted using IUA rats and compared with a class III medical device, a new-crosslinked hyaluronic acid gel (NCHA). According to the study, curcumin hydrogel is more effective than the NCHA group in improving the regeneration of the endometrium, increasing the blood supply to the endometrium, and reducing the abnormal deposition of fibrin, thus preventing intrauterine adhesion more effectively. This study provides a promising strategy for treating and preventing IUA.

Oral ulcers are a common oral mucosal disease that seriously affect the quality of life. Traditional drug treatments have shown unsatisfactory efficacy and potential adverse reactions. In this study, curcumin-loaded multifunctional magnesium metal–organic framework-embedded hyaluronic acid-soluble microneedles patches were developed to optimize treatment strategies for oral ulcers. This microneedles patch achieves efficient release of curcumin and Mg 2+ in the ulcer through precisely targeted delivery and controllable release mechanism, significantly regulates inflammation, promotes cell migration and angiogenesis, and accelerates the ulcer healing process. At the same time, the synergistic effect of curcumin and gallic acid effectively alleviated oxidative stress, while the backplate ε-poly- l -lysine and needle tip Mg 2+ jointly constructed an antibacterial barrier to effectively inhibit pathogens. Verification using an oral ulcer rat model showed that the microneedles patch exhibited excellent therapeutic effects. This not only opens up a new avenue for clinical oral treatment but also marks a breakthrough in nanobiomaterials science and drug delivery technology and heralds a broad prospect in the field of oral ulcer treatment in the future. Curcumin is widely recognized for its diverse antitumor properties, ranging from breast cancer to many other types of cancers. However, its role in the tumor microenvironment remains to be elucidated. In this study, we established a 3D tumor spheroids model that can simulate the growth environment of tumor cells and visualized the antitumor metabolic alteration caused by curcumin using mass spectrometry imaging technology. Our results showed that curcumin not only exerts a profound impact on the growth and proliferation of breast cancer cells but in situ multivariate statistical analysis also reveals the significant effect on the overall metabolic profile of tumor spheroids. Meanwhile, our visualization map characterized curcumin metabolic processes of reduction and glucuronidation in tumor spheroids. More importantly, abnormal metabolic pathways related to lipid metabolism and polyamine metabolism were also remodeled at the metabolite and gene levels after curcumin intervention. These insights deepen our comprehension of the regulatory mechanism of curcumin on the tumor metabolic network, furnishing powerful references for antitumor treatment.

Lyocell fiber is an important choice for functional apparel textiles because of its advantages of comfort, high strength, sustainable source and pollution-free spinning process. However, current methods for functionalization Lyocell fibers, such as blend spinning or surface modification using inorganic and synthetic materials, are generally complex, less environmentally friendly and easy to lose functionality. In this study, for the first time, we innovatively loaded the natural functional material curcumin into cationic polymer nanospheres and applied them in Lyocell fiber functionality through a simple one-step dyeing process. The fabricated curcumin@polymer nanospheres had a uniform particle distribution of approximately 148.03 nm, and exhibited a positive surface potential of +25.3 mV. Under the action of cationic polymer nanospheres, the interaction between curcumin and Lyocell fiber was obviously enhanced, and the mechanism was analyzed. The colored fibers possessed high antibacterial efficacy of 99.99 % against Staphylococcus aureus due to the synergistic antibacterial activity of curcumin and the quaternary ammonium salt groups in polymer sturcture. Furthermore, the application of the prepared curcumin@polymer nanospheres could realize the one-step process for coloration and functionalization of Lyocell fibers. Therefore, from the measurement of antibacterial activity, stability, and coloration depth, curcumin@PMBD showed great potential in the manufacture of greener antibacterial textile.
